Intra-ChIP: studying gene regulation in an intracellular pathogen.

Intracellular bacteria that reside within a host cell use a variety of strategies to exploit this unique niche. While these organisms are technically challenging to study in the context of an infected host cell, recent advances have led to an improved understanding of how the intracellular environment impacts bacterial gene expression. We recently demonstrated that chromatin immunoprecipitation (ChIP) can be used to quantify transcription factor binding in the obligate intracellular pathogen Chlamydia trachomatis within infected cells. Furthermore, we showed it was possible to experimentally m

Insights into the pathogenesis of disease in human lymphatic filariasis.

Although two thirds of the 120 million people infected with lymph-dwelling filarial parasites have subclinical infections, ∼40 million have lymphedema and/or other pathologic manifestations including hydroceles (and other forms of urogenital disease), episodic adenolymphangitis, lymphedema, and (in its most severe form) elephantiasis. Adult filarial worms reside in the lymphatics and lymph nodes and induce lymphatic dilatation. Progressive lymphatic damage and pathology results primarily from the host inflammatory response to the parasites but also perhaps from the host inflammatory response t

Intestinal colonization resistance in the context of environmental, host, and microbial determinants.

Microbial communities that colonize the human gastrointestinal (GI) tract defend against pathogens through a mechanism known as colonization resistance (CR). Advances in technologies such as next-generation sequencing, gnotobiotic mouse models, and bacterial cultivation have enhanced our understanding of the underlying mechanisms and the intricate microbial interactions involved in CR. Rather than being attributed to specific microbial clades, CR is now understood to arise from a dynamic interplay between microbes and the host and is shaped by metabolic, immune, and environmental factors. This

Are novel and co-xenic associations common in alien fungal and fungus-like plant pathogens?

Impacts of alien fungal and fungus-like plant pathogens depend on host interactions, including novel associations with native plants and co-xenic associations with alien plants that do not share the same area of origin, both largely unquantified globally. Combining global distribution and association data, we characterised associations across pathogens' introduced regions, calculated host species richness (SR) and phylogenetic divergence, and examined predictors of novel/co-xenic associations. Novel/co-xenic alien pathogen-plant association records were common (53%) and dominated by widespread

Species-specific barriers restrict virus spillover potential across the Caenorhabditis genus.

Host range expansion and virus emergence depend on whether viral life-cycle processes align with the ecological, developmental and physiological traits of new hosts. Spillover therefore succeeds only when pathogens clear a hierarchy of mechanistic barriers, from entry and replication to transmission and evolutionary persistence, but how these barriers map onto whole-organism host competence remains poorly resolved. Here we dissect spillover barriers experimentally using the Caenorhabditis-Orsay virus system, integrating within-host viral kinetics, cellular progression, transmission, virulence

Listeria monocytogenes: at the coalface of host-pathogen research.

Listeria monocytogenes is a highly adaptable food-borne pathogen that causes the life threatening illness listeriosis in infected individuals. Within the host this bacterium invades cells, escapes into the host cell cytosol and replicates intracellularly. To achieve this L. monocytogenes has evolved a sophisticated set of molecular weaponry that allows it to interact with and manipulate the cell biology of the host to its own advantage. Many of these interactions are well understood, putting this pathogen at the forefront of host-pathogen research, but fascinating new interactions are still em

Cyclic β-glucans at the bacteria-host cells interphase: One sugar ring to rule them all.

Cyclic β-1,2-D-glucans (CβG) are natural bionanopolymers present in the periplasmic space of many Proteobacteria. These molecules are sugar rings made of 17 to 25 D-glucose units linked exclusively by β-1,2-glycosidic bonds. CβG are important for environmental sensing and osmoadaptation in bacteria, but most importantly, they play key roles in complex host-cell interactions such as symbiosis, pathogenesis, and immunomodulation. In the last years, the identification and characterisation of the enzymes involved in the synthesis of CβG allowed to know in detail the steps necessary for the formati

New drug-strategies to tackle viral-host interactions for the treatment of influenza virus infections.

The influenza virus (IV) is a highly contagious virus causing seasonal global outbreaks affecting annually up to 20% of the world's population and leading to 250,000-500,000 deaths worldwide. Current vaccines have variable effectiveness, and, in particular during a pandemic outbreak, they are probably not available in the amounts needed to protect the world population. Therefore we need effective small molecule drugs to combat an IV infection and that can be produced, in case of pandemic, rapidly and in large quantities. Unfortunately, natural occurring IV becomes more and more resistant to cu

Multi-comparative systems biology analysis reveals time-course biosignatures of in vivo bovine pathway responses to B.melitensis, S.enterica Typhimurium and M.avium paratuberculosis.

BACKGROUND: To decipher the complexity and improve the understanding of host-pathogen interactions, biologists must adopt new system level approaches in which the hierarchy of biological interactions and dynamics can be studied. This paper presents the application of systems biology for the cross-comparative analysis and interactome modeling of three different infectious agents, leading to the identification of novel, unique and common molecular host responses (biosignatures). METHODS: A computational systems biology method was utilized to create interactome models of the host responses to Br…

Structure-resolved virus-host interactomics by cross-linking mass spectrometry.

Viruses depend on host protein networks to replicate, assemble progeny, and spread between cells and organisms. Defining these virus-host protein interactions is challenging because they are highly dependent on infection stage, cell type, species, and because mechanistic interpretation requires information about structural interfaces and conformational states. Cross-linking mass spectrometry (XL-MS) addresses these challenges by adding a spatial and structural dimension to virus-host interactomics in native systems. In this review, we discuss how XL-MS has advanced from targeted analysis of vi

When a protozoan pathogen imitates cancer.

A few specialized tick-transmitted protozoa cause fatal infectious diseases in cattle through a unique cell biological mechanism, the transformation of host leukocytes into cancer-like cells. These 'transformative' parasites, such as Theileria annulata, hijack host signaling pathways and, during the schizont stage of their complex life cycle, cause uncontrolled proliferation and immortalization of infected cells, likely through a broad repertoire of fine-tuned effector proteins. This review summarizes recent advancements on the parasite-host interactions proposed to underlie this remarkable tr

Impact of climate change on the tick-host-pathogen complex: distribution patterns, disease incidence, and host infestation.

Ticks, being ectothermic, are highly sensitive to climate variables, such as temperature, humidity, and precipitation. Over the past century, fossil fuel use has altered the climate and significantly affected the tick-host-pathogen system. These changes influence tick lifecycles, behavior, vector competency, host dynamics, and pathogen transmission. Consequently, tick-borne diseases (TBDs) have experienced shifts in their geographical range, incidence, and host preferences, particularly in the Northern Hemisphere. While climate change drives the emergence of vector-borne diseases, key aspects,

Antibody profiles induced by Trypanosoma cruzi in chagasic patients with previous or current exposure to mycobacteria.

Since the immune response mounted by the host to a particular microorganism might be influenced by the acquired immunological experience due to previous contact with other microorganisms, we performed a cross-sectional study to explore the pattern of Trypanosoma cruzi infection-related antibodies in T. cruzi-seropositive individuals presenting concomitant tuberculosis, or the antecedent of BCG vaccination. Sampled individuals were grouped as follows: patients with Chagas disease, not vaccinated with BCG, who further developed pulmonary tuberculosis; individuals with Chagas disease, BCG-vaccina

Host-microbe interactions in the appendix of children with inflammatory bowel diseases.

The human appendix is traditionally considered a vestigial organ; however, clinical observations link it to inflammatory bowel diseases (IBDs), including Crohn disease (CD) and ulcerative colitis (UC), as suggested by periappendicular inflammation and the reported protective effect of appendectomy in UC. Despite these associations, its functional contribution remains poorly defined. Here, we performed a multiomics analysis of appendix tissue from pediatric patients with IBD and non-IBD surgical controls (n = 15) to characterize microbial composition and host molecular landscape. Manipulation of Host Cell Organelles by Intracellular Pathogens.

Pathogenic intracellular bacteria, parasites and viruses have evolved sophisticated mechanisms to manipulate mammalian host cells to serve as niches for persistence and proliferation. The intracellular lifestyles of pathogens involve the manipulation of membrane-bound organellar compartments of host cells. In this review, we described how normal structural organization and cellular functions of endosomes, endoplasmic reticulum, Golgi apparatus, mitochondria, or lipid droplets are targeted by microbial virulence mechanisms. We focus on the specific interactions of Salmonella, Legionella pneumop

Papain-like cysteine proteases: key players at molecular battlefields employed by both plants and their invaders.

Papain-like cysteine proteases (PLCPs) play crucial roles in plant-pathogen/pest interactions. During these parasitic interactions, PLCPs act on non-self substrates, provoking the selection of counteracting inhibitors and other means to evade proteolysis. We review examples of PLCPs acting on molecular battlefields in the extracellular space, plant cytoplasm and herbivore gut. Examples are maize Mir1 (Maize inbred resistance 1), tomato Rcr3 (Required for Cladosporium resistance-3), Pseudomonas AvrRpt2 and AurPphB, insect DvCAL1 (Diabrotica virgifera cathepsin L-like protease-1) and nematode Mi
