Maternal life history--versus gestation-focused assessment of prenatal exposure to substances of abuse.

PURPOSE: Substance use by pregnant women is socially stigmatized and may be legally punishable. This societal condemnation raises concerns about underascertainment of prenatal substance exposure of offspring if mothers are asked specifically about their behavior during gestation, versus their life histories without reference to gestational dates. This study assessed agreement between life history-focused and pregnancy-focused assessments of prenatal exposure, and percentages of offspring classified as exposed to a range of substances by each measure, in a sample of school-aged children of meth

Relationship of prenatal cocaine exposure and maternal postpartum psychological distress to child developmental outcome.

Maternal cocaine use during pregnancy can affect the infant directly through toxic effects or indirectly through cocaine's influence on maternal psychological status. We followed 160 cocaine exposed and 56 nonexposed infants and their mothers identified at birth through interview and/or urine screen. Although cocaine exposure defined the groups, infant exposure to alcohol, marijuana, and tobacco was allowed to vary. Infants were 99% African American and poor. All mothers completed the Brief Symptom Inventory (BSI) and infants were given the Bayley Scales of Mental (MDI) and Motor (PDI) Develop

Child neurodevelopment is related to maternal manganese exposure: A Taiwan national prospective cohort study.

AIM: Prenatal exposure to air pollutants has been linked to developmental delays in early childhood. This study investigated the association between prenatal exposure to ambient manganese (Mn) and delays in achieving specific neurodevelopmental milestones. METHOD: Data were obtained from a nationwide population-based cohort study that recruited children born in 2005 and their mothers. Developmental outcomes were assessed by conducting home interviews at 6 and 18 months of age. Gestational exposure to ambient Mn was estimated using a land-use regression model enhanced with machine learning. As

Psychomotor Development in Infants Following Maternal Exposure to Biologics: Results From the DUMBO Registry.

BACKGROUND & AIMS: The impact of biologic treatment for inflammatory bowel disease (IBD) during pregnancy and lactation on infant psychomotor development is barely studied. We investigated the effect of exposure to biologics in utero or during breastfeeding on the psychomotor development of offspring during their first year. METHODS: The study included patients and infants from DUMBO, an ongoing prospective, observational registry of GETECCU enrolling pregnant women (aged ≥18 years) with IBD from 60 centers across Spain. Mothers were followed up at each trimester during pregnancy and after de

Prenatal PFAS exposure and outcomes related to maternal gut microbiome composition in later pregnancy.

The composition of the gut microbiome is dependent on factors including diet, lifestyle, and exposure to environmental chemicals, and has implications for human health. Per- and polyfluoroalkyl substances (PFAS), a class of man-made chemicals that have nonstick and flame-retardant properties may impact on gut microbiome composition. Our objective was to elucidate links between PFAS and maternal gut microbiome composition in two geographically diverse sites of the Environmental Influences on Child Health Outcomes program. The present analysis includes participants in the Atlanta African America

Maternal lead exposure impairs offspring growth and intestinal microbiota via AMPK/ULK1 pathway activation.

The pervasive presence and bioaccumulation of lead (Pb2 +) in living organisms pose a significant risk to human health. Extensive research has been conducted on the neurotoxicity and reproductive toxicity of lead, but the impact of maternal lead exposure on subsequent generations still needs to be addressed. Therefore, we aimed to explore the adverse effects and mechanisms of lead exposure in pregnant mice on the next generation. The present study indicated that lead exposure decreased growth performance and impaired the function of the colon, spleen, testes, and kidneys in the next generation

Comparison of Metformin PBPK Models Incorporating Placental Transfer to Predict Fetal and Maternal Exposure.

Physiologically based pharmacokinetic (PBPK) modeling of placental drug transfer is an evolving tool for predicting fetal drug exposure. In this study, a pregnancy-specific metformin PBPK model was developed, and the following four approaches were evaluated to predict metformin placental transfer: (1) perfusion-limited model, and permeability-limited models using (2) ex vivo cotyledon open system apparent clearance, (3) ex vivo cotyledon closed system data fit to a three-compartment model to estimate clearance, and (4) active transport kinetics and passive clearance. Simulated metformin matern

Postnatal antibiotic exposure due to maternal group B streptococcus is associated with childhood asthma.

BACKGROUND: Previous studies identified early-life antibiotic exposure as a risk factor for childhood asthma. However, this association may be confounded by an indication, as antibiotics are often prescribed for respiratory infections, which themselves promote asthma. To mitigate this bias, we aim to assess the unique contribution of postnatal antibiotic therapy, given to non-infected infants for maternal indication, on childhood asthma risk. METHODS: We screened electronic medical records to identify healthy full-term infants born during 2006-2018 to mothers with a positive group B streptoco

Physiologically based pharmacokinetic modeling to predict maternal pharmacokinetics and fetal carbamazepine exposure during pregnancy.

Carbamazepine is an antiepileptic drug commonly used in pregnant women, during which the physiological changes may affect its efficacy. The aim of this study was to establish a physiologically based pharmacokinetic (PBPK) model of carbamazepine and its active metabolite carbamazepine-10,11-epoxide, and simulate maternal and fetal pharmacokinetic changes of carbamazepine and carbamazepine-10,11-epoxide in different trimesters and propose dose adjustment. We established pregnancy PBPK models for carbamazepine and carbamazepine-10,11-epoxide in PK-Sim® and Mobi® and validated the models with obse

Maternal 129S1/SvImJ background attenuates the placental phenotypes induced by chronic paternal alcohol exposure.

Paternal alcohol use is emerging as a plausible driver of alcohol-related growth and patterning defects. Studies from our lab using an inbred C57Bl/6 J mouse model suggest that these paternally-inherited phenotypes result from paternally programmed deficits in the formation and function of the placenta. The 129S1/SvImJ genetic background is typically more susceptible to fetoplacental growth defects due to strain-specific differences in placental morphology. We hypothesized that these placental differences would sensitize 129S1/SvImJ-C57Bl/6 J hybrid offspring to paternally-inherited fetoplacen

Exacerbated lung inflammation in offspring with high maternal antibody levels following secondary RSV exposure.

Respiratory syncytial virus (RSV) is the primary cause of bronchiolitis-related hospitalizations among children under 5 years of age, with reinfection being common throughout life. Maternal vaccination has emerged as a promising strategy, delivering elevated antibody levels to newborns for immediate protection. However, limited research has explored the protective efficacy of maternal antibodies (matAbs) against secondary RSV infections in offspring. To address this gap, we employed a mouse model of maternal RSV vaccination and secondary infection of offspring to evaluate lung pathology follow

Long-term outcomes of children after prenatal exposure to maternal cancer and its treatment.

The incidence of antenatal cancer is increasing, prompting a medical-ethical evaluation. The International Network on Cancer, Infertility, and Pregnancy (INCIP) was established to study cancer treatment safety during pregnancy and its impact on maternal and child health. Pivotal research has led to a paradigm shift in clinical management, demonstrating the feasibility and safety of most antenatal oncological treatments. Short-term outcomes reveal normal growth and cardiac function in the exposed offspring, but caution is advised against first-trimester chemotherapy. Psychological impact studie

Maternal diesel particle exposure alters gut microbiota and induces lung injury in rat offspring.

Maternal air pollutant exposure inhibits fetal lung development. Diesel exhaust particles (DEP) are one of the most substantial contributors to particulate matter pollution. The effects of maternal DEP exposure on gut microbiota in mothers and offspring and fetal lung development remain unclear. In this study, time-dated pregnant Sprague Dawley rats received intranasal administration of 100 μL phosphate-buffered saline (PBS) or DEP (250 μg) in 100 μL PBS from gestational days 16-21. The dams were permitted to deliver vaginally at term. On postnatal days 0 and 7, gut microbiota was sampled from

In Utero Exposure to Maternal COVID-19 and Offspring Neurodevelopment Through Age 24 Months.

IMPORTANCE: In utero exposure to maternal infections has been associated with abnormal neurodevelopment among offspring. The emergence of a new, now endemic infection (SARS-CoV-2) warrants investigating developmental implications for exposed offspring. OBJECTIVE: To assess whether in utero exposure to maternal COVID-19 is associated with abnormal neurodevelopmental scores among children ages 12, 18, and 24 months. DESIGN, SETTING, AND PARTICIPANTS: Data were ascertained from the ASPIRE (Assessing the Safety of Pregnancy in the Coronavirus Pandemic) trial, a prospective cohort of pregnant ind

Maternal exposure to chiral triazole fungicide tebuconazole induces enantioselective thyroid disruption in zebrafish offspring.

Pesticides could induce long-term impacts on aquatic ecosystem via transgenerational toxicity. However, for many chiral pesticides, the potential enantioselectivity of transgenerational toxicity has yet to be fully understood. In this study, we used zebrafish as models to evaluate the maternal transfer risk of tebuconazole (TEB), which is a chiral triazole fungicide currently used worldwide and has been frequently detected in surface waters. After 28-day food exposure (20 and 400 ng/g) to the two enantiomers of TEB (S- and R-TEB) in adult female zebrafish (F0), increased malformation rate and

Effects of silver nanoparticles on maternal mammary glands and offspring development under lactation exposure.

The widespread applications of silver nanoparticles (AgNPs) throughout our daily lives have raised concerns regarding their environmental health and safety (EHS). Despite an increasing number of studies focused on the EHS impacts of AgNPs, there remain significant knowledge gaps with respect to their potential health impacts on susceptible populations, such as lactating mothers and infants. Herein, we aimed to investigate the deleterious effects of AgNPs with different sizes (20 and 40 nm) and surface coatings (PVP and BPEI) on maternal mice and their offspring following lactation exposure at

Prenatal Stress Exposure Amplifies Effect of Maternal Suicidal Ideation on Early Childhood Behavioral Trajectories.

The in utero environment influences fetal development and may predispose to disease later in life. This study examines whether maternal suicidal ideation during pregnancy is associated with children's behavioral trajectories across early childhood, and whether prenatal maternal traumatic stress accelerates the trajectories. The study included mother-child dyads (N = 331, 51.1% boys) from the longitudinal Stress In Pregnancy study; 31.1% (n = 103) mothers were Exposed to Superstorm Sandy. During their second trimester, 12.4% (n = 41) women reported suicidal ideation during pregnancy. Mothers co

Maternal Voice Exposure and Its Effect on Premature Infants' Feeding Milestones: A Systematic Review.

BACKGROUND: Premature infants experience alterations in maternal stimulation (including auditory sensory alteration such as talking or singing to the infant in the neonatal intensive care unit) due to admission to the neonatal intensive care unit. Because of their physiological and neurobehavioral immaturity, infants are at an increased risk of delays in reaching feeding milestones (a key developmental milestone), which often need to be achieved before discharge. PURPOSE: This systematic review evaluated the literature regarding the effect of maternal speech on achievement of feeding mileston
