Because salivary glands, as well as thyroid tissue, are able to concentrate radioiodine, the treatment of thyroid diseases with iodine-131 may have secondary effects on salivary gland function which seriously impair the quality of life. Such effects include sialoadenitis and xerostomia. Salivary secretion is stimulated by prostaglandins (PGs). In this study we evaluate whether 131I therapy influences the levels of PGs in saliva. Patients who had previously received 131I for treatment of hyperthyroidism or differentiated thyroid cancer and healthy volunteers were studied. Levels of PGs [6-oxo-P

Renal function was prospectively analyzed in 26 evaluable patients, irradiated to various doses on their kidneys for neoplastic disease. Glomerular function was assessed by 99mTc-DTPA renography, creatinine clearance, and serum beta 2-microglobulin, whereas tubular function was monitored by 99mTc-DMSA scintigraphy, urine beta 2-microglobulin, urine N-acetyl glucosaminidase, and alanine aminopeptidase and a urine concentration test. In the patients given the highest irradiation dose to the entire left kidney, that is, 40 Gy in 5 1/2 weeks, glomerular and tubular functional impairment, as assess

Radiation-induced brain injury is a severe complication of cranial radiotherapy, but the redox-dependent mechanisms linking neuronal damage to microglial neuroinflammation remain incompletely understood. Here, we investigated whether radiation-induced reactive oxygen species (ROS) are associated with neuronal mitochondrial dysfunction, lipid accumulation, and subsequent microglial inflammatory activation. Using an irradiated mouse brain injury model and neuron-microglia conditioned-medium systems, we found that ionizing radiation increased neuronal ROS production and impaired mitochondrial hom

Radiotherapy for brain tumors frequently induces irreversible cognitive decline, yet the underlying mechanisms remain poorly understood. Here, we present an integrated single-cell and spatial transcriptomic atlas of radiation-induced brain injury (RIBI) in mice, spanning the acute to chronic phases. We reveal that the early-delayed phase following irradiation represents a critical window during which the blood-cerebrospinal fluid barrier (BCSFB) integrity is compromised. This disruption results from the selective depletion of a radiosensitive Tmsb4x+ epithelial subpopulation in the choroid ple

BACKGROUND: Radiation-induced intestinal injury (RIII) is a serious complication of radiotherapy and is closely associated with mitochondrial dysfunction, but the underlying mechanism remains unclear.
METHODS: Mitochondria‑related differentially expressed genes (DEGs) were identified using online databases in an RIII model. Histopathological staining analysis was performed to evaluate injury severity. The expression of Mrm2 was examined in vivo after irradiation.
RESULTS: The core functional modules of the identified DEGs included mitochondrial‑coding genes and apoptosis‑related genes. Histo

AIMS: Radiation-induced intestinal injury (RIII) severely compromises the quality of life in patients undergoing abdominal/pelvic radiotherapy and may necessitate treatment discontinuation. To date, there is no approved agent for the prevention or treatment of RIII. This study aims to clarify the protective effects of mannose on RIII and elucidate its mechanisms of action, in order to identify new safe and effective therapeutic agents and potential therapeutic targets for the prevention and treatment of RIII.
RESULTS: Here, we report that intraperitoneal administration of mannose, a natural b

BACKGROUND: Macrophages play a crucial role in the inflammation and wound repair processes of radiation-induced lung injury (RILI). The mechanosensitive ion channel Piezo1 is upregulated during these inflammatory and wound repair processes. However, the involvement of macrophage Piezo1 in the pathogenesis of RILI remains unclear. This study aims to elucidate the regulatory role of Piezo1 in the injury and repair process in RILI and to investigate the underlying mechanisms.
METHODS: We established Myeloid-specific knockout of Piezo1 (Piezo1∆ LysM) mice, and the mice were subjected to total-che

BACKGROUND: Radiation-induced lung injury (RILI) is a major dose-limiting complication of thoracic radiotherapy. Although mitochondrial damage has been implicated in RILI, the endogenous transcriptional programs that restore mitochondrial structure and bioenergetic function after irradiation remain poorly defined.
METHODS: To identify radiation-sensitive mitochondrial regulators in type II alveolar epithelial cells (AT2), we integrated single-cell RNA sequencing data from irradiated lungs with weighted gene co-expression network analysis. Transcription factor prediction, multi-omics correlati

Genitourinary injury after prostate radiation therapy encompasses a wide spectrum of disease states that present after radiation, including fistula, stricture, and bladder fibrosis, resulting in debilitating changes in quality of life and loss of function. Management often includes temporizing urinary diversion, endoscopic therapies, and ultimately, surgical intervention, including pelvic organ extirpation. Though challenging to manage, novel adjuncts and minimally invasive approaches in the treatment of the sequelae of radiation injury are evolving, offering new treatment paradigms for this c

Radiation-induced injury is a complex pathological process involving DNA damage, oxidative stress, inflammatory amplification, epithelial barrier disruption, immune dysregulation, metabolic remodeling, and impaired tissue repair. Accumulating evidence indicates that the gut microbiota is not merely altered by irradiation but may also modify host radiation responses and tissue recovery; however, direct causal support remains confined to selected microbes, metabolites, and pathways, predominantly in preclinical models. Irradiation is frequently associated with reduced microbial diversity, deplet

Radiotherapy is a critical treatment modality for abdominal and pelvic malignancies; however, it is associated with a high incidence of radiation-induced intestinal injury, which severely limits therapeutic efficacy and impairs patients' quality of life. In recent years, the bidirectional regulatory role of the gut microbiota in the development and progression of radiation-induced intestinal injury has garnered significant attention. Radiotherapy can induce a reduction in gut microbial diversity, disrupt the metabolism of short-chain fatty acids and tryptophan, and promote the expansion of opp

BACKGROUND: Lymphedema is a common but distressing sequela after cancer treatment. A cure is not yet available; thus, prevention is essential in its management.
METHODS AND RESULTS: Here, we used a mouse hindlimb model to demonstrate the effects of exercise performed before lymph node resection and irradiation on the development of lymphedema. BALB/c mice trained on a treadmill at moderate intensity, 30 minutes/day, 5 times/week for 4 weeks, were compared to a no-exercise group (n = 7/group). Weekly limb measurements and indocyanine green (ICG) lymphangiography were performed to assess lymphe

BACKGROUND: Radiation-induced skin injury (RISI) is a notable complication of cancer radiotherapy, impacting patients' quality of life. Existing interventions mainly address symptoms, with limited success in targeting the fundamental mechanisms. Berberine (BBR), a bioactive compound recognized for its anti-inflammatory, antioxidant, and anti-fibrotic characteristics, presents a compelling option for treating RISI.
METHODS: The molecular targets of BBR and RISI were identified using Swiss Target Prediction and GeneCards databases. A protein-protein interaction (PPI) network was then constructe

Radiotherapy can be limited by pneumonitis, which is impacted by innate immunity, including pathways regulated by TRAIL death receptor DR5. We investigated whether DR5 agonists could rescue mice from toxic effects of radiation and found that 2 different agonists, parenteral PEGylated trimeric TRAIL (TLY012) and oral TRAIL-inducing compound (TIC10/ONC201), could reduce pneumonitis, alveolar wall thickness, and oxygen desaturation. Lung protection extended to late effects of radiation including less fibrosis at 22 weeks in TLY012-rescued survivors versus unrescued surviving irradiated mice. Wild

Radiotherapy (RT) is a treatment method commonly used in oncology. A vast majority of patients undergoing RT suffer from radiation-induced skin injury (RISI), which results from complex biochemical reactions in the irradiated skin. Current strategies for preventing and managing RISI are insufficient for achieving full skin regeneration. Multiple studies have shown that alterations in the skin microbiome correlate with the development and severity of RISI. These studies suggest that dysbiosis is a crucial factor in promoting radiation-associated dermatitis. Targeting the skin microbiota present
