A phenylketonuria mouse model exhibits EEG spike-wave discharges: Effects of sleep deprivation and low-Phe diet.

Phenylketonuria (PKU) is an inherited metabolic disorder characterized by phenylalanine hydroxylase deficiency and elevated phenylalanine (Phe) levels in blood and brain, causing neurotoxicity and brain dysfunction. Elevated Phe levels may alter brain activity, as poor dietary control is linked to abnormal EEGs. However, knowledge on these electrophysiological characteristics remains limited. The current study applied electroencephalogram (EEG) recordings of brain activity in PAHenu2 mice, which have elevated Phe levels resembling untreated PKU patients. EEG recordings revealed frequent sponta

Ontogenetic development of ponto-geniculo-occipital waves during paradoxical sleep in kittens.

STUDY OBJECTIVES: Ponto-geniculo-occipital (PGO) waves are a hallmark feature of paradoxical sleep (PS), emerging just before PS onset during the transition from slow-wave sleep (SWS) to PS (TSWS-PS) and persisting throughout the PS phase in adult cats. These waves are considered a biomarker for the maturation of PS. However, their developmental trajectory in mammals remains poorly understood. This study investigated the emergence and maturation of PGO waves during PS in developing kittens. METHODS: Electrodes were stereotaxically implanted into the lateral geniculate nucleus, accurately loca

Objectively measured sleep characteristics and prevalence of coronary artery calcification: the Multi-Ethnic Study of Atherosclerosis Sleep study.

BACKGROUND: We tested whether objectively measured indices of obstructive sleep apnoea (OSA) and sleep quality are associated with coronary artery calcification (CAC) prevalence independent of obesity, a classic confounder. METHODS: 1465 Multi-Ethnic Study of Atherosclerosis participants (mean age 68 years), who were free of clinical cardiovascular disease, had both coronary CT and in-home polysomnography and actigraphy performed. OSA categories were defined by the Apnea-Hypopnea Index (AHI). Prevalence ratios (PRs) for CAC >0 and >400 (high burden) were calculated. RESULTS: Participants wit

Objective B wave analysis in 55 patients with non-communicating and communicating hydrocephalus.

BACKGROUND: B waves, slow and rhythmic oscillations in intracranial pressure (ICP), are claimed to be one of the best predictors of outcome after surgery for normal pressure hydrocephalus (NPH). OBJECT: To determine the relation between the percentage of B waves and outcome in patients with hydrocephalus, and also the diurnal variation of B waves. METHODS: ICP and patient behaviour were recorded overnight (17 to 26 hours) in 29 patients with non-communicating hydrocephalus and 26 with NPH. The B wave activity, measured with an amplitude threshold of 0.5, 0.75, 1.0, 1.5, 2.0, 3.0, and 5.0 mm

[Effect of oral almitrine on the sleep apnea syndrome].

Almitrine stimulates breathing by activating peripheral chemoreceptors and was given orally in a dose of 200 mg/day to nine patients with sleep apnoea syndrome, after a single blind placebo sequence. No reduction in the number of respiratory events per hour of sleep was observed. On the other hand Almitrine led to a significant fall in the mean duration of respiratory events (p less than 0.02). This fall occurred principally in the obstructive apnoea group and in those with mixed apnoea only during light slow wave sleep. These results are in agreement with the hypothesis that the arousal respo

Idiopathic sleep terror in an adult patient.

We report a case of a man in his 40s presenting with idiopathic adult-onset sleep terrors manifesting as nocturnal violent behaviour, including a concerning episode of attempted strangulation of his bed partner. Polysomnography documented an event during N3 sleep (slow-wave, deep non-rapid eye movement) with autonomic hyperactivation and complex motor behaviour. A detailed psychiatric evaluation did not reveal any diagnosable anxiety, mood or other psychiatric disorder, and there was no history of substance use, recent psychosocial stressors or sleep deprivation. Notably, there were no precipi

Influence of the cholinergic agonist RS 86 on normal sleep: sex and age effects.

In 36 healthy control subjects (21 females, 15 males; age range 18-65 years; mean age 41.8 years, SD 15.6 years), a bedtime dose of 1.5 mg RS 86, an orally acting cholinergic agonist, shortened rapid eye movement (REM) latency, increased REM sleep, and decreased slow-wave sleep. Six of the subjects (greater than 40 years old) even displayed sleep-onset REM periods after the drug. Results of the present study agree well with those of studies using other cholinomimetics (i.e., physostigmine, arecholine) and confirm the importance of the cholinergic system for REM sleep regulation. Since RS 86 mi

Agrypnia excitata: a human model to explore the derailment of sleep-wake cycle integrated control.

The commemoration of the 70th anniversary of rapid eye movement sleep discovery offers a unique possibility to reassess the peculiar organic condition of agrypnia excitata. Agrypnia excitata is characterized by a severe loss of sleep leading to a complete derangement of physiological sleep-wake cycle and body homeostasis. Agrypnia excitata is a definite clinico-neurophysiological condition characterized by: (1) slow-wave sleep loss with disruption of sleepwake cycle; (2) a 24-hr motor and autonomic overactivity; and (3) peculiar episodes of oneiric stupor. Agrypnia excitata may happen within d

Rapid Eye Movement Sleep Sawtooth Waves Are Associated with Widespread Cortical Activations.

Sawtooth waves (STW) are bursts of frontocentral slow oscillations recorded in the scalp electroencephalogram (EEG) during rapid eye movement (REM) sleep. Little is known about their cortical generators and functional significance. Stereo-EEG performed for presurgical epilepsy evaluation offers the unique possibility to study neurophysiology in situ in the human brain. We investigated intracranial correlates of scalp-detected STW in 26 patients (14 women) undergoing combined stereo-EEG/polysomnography. We visually marked STW segments in scalp EEG and selected stereo-EEG channels exhibiting nor

Time-related changes in the sleep-wake cycle of rats infected with Trypanosoma brucei brucei.

Patients with human African trypanosomiasis, sleeping sickness, show a major disturbance in the circadian distribution of sleeping and waking, with sleep and wake episodes equally distributed throughout the nycthemeron. In order to develop an animal model, polysomnography was taken continuously in 8 male OFA rats in a 12:12 h light-dark cycle, during 1 baseline week and for 2 weeks after infection with Trypanosoma brucei brucei. Considerable sleep fragmentation was observed in the infected rats, with numerous sleep-wake stage changes and an increased number of wakefulness and slow-wave sleep p

[Features of cortex areas system interactions in the left and right brain hemospheres during different human sleep stages].

The article is devoted to analysis of research brain biopotential field structure during different stages of natural night sleep in adults. The special attention is given to distinctions of the spatial organization of EEG interregional relations in the left and right hemispheres which are shown most strongly in the II and III sleep stages. The special role of frontal cortical areas in reorganization of brain system activity during falling asleep is highlighted. Essential strengthening of hemispherical interaction of cortex biopotentials fluctuations during falling asleep--a stage of I(B)--alon

Impairment of respiratory rhythmogenesis and sequelae of bacterial meningitis.

A 9-year-old boy with respiratory disturbance associated with medullary lesions after pneumococcal meningitis is reported. Although he lives a normal daily life, he cannot cough or sneeze. A polysomnographic study revealed a low respiration rate and an irregular respiratory rhythm not only during REM sleep but also during slow wave sleep, and marked desaturation during sleep. Respiratory function tests including CO2 response revealed normal values. Magnetic resonance imaging demonstrated bilateral small lesions in the medulla. This patient is unusual because respiratory rhythm is impaired, wit

Distribution of REM latencies and other sleep phenomena in depression as explained by a single ultradian rhythm disturbance.

The McCarley-Hobson model, describing the alternation of NREM and REM sleep in the cat, was applied to human electroencephalographic data. The influence of initial conditions on oscillatory behavior was especially emphasized. It appears that the distribution of REM latency in depression, the abnormal accumulation of REM sleep, the variability of NREM-REM cycle duration, the frequent stage shifts, and frequent awakenings can be explained in this model by means of a decrease in the initial value of a single variable, which may be regarded as representing the strength of REM inhibition. The obser

Electrical Synapses Contribute to Sleep-Dependent Declarative Memory Retention.

Sleep supports memory formation by neurophysiological mechanisms that are yet to be fully uncovered. We investigated the contribution of the direct coupling of neurons via electrical synapses (gap junctions). The administration of mefloquine (250 mg p.o. vs. placebo), an antimalarial, which blocks electrical synapses, to healthy young men before nocturnal sleep impaired the retention of word pairs learned before drug administration and disrupted the coupling of sleep spindles to EEG slow oscillations. In control experiments, in which participants received mefloquine before a consolidation inte

Effects of modafinil and amphetamine on sleep-wake cycle after sleep deprivation in cats.

AIM: The effects of modafinil and amphetamine on sleep-wake cycle and cortical power spectrum were assessed in the cats before and after sleep deprivation. METHODS: The sleep deprivation in the cats was used with the water tank technique. Cats were administrated with modafinil (5 mg.kg-1 p.o.) or amphetamine (1 mg.kg-1) before and after sleep deprivation. RESULTS: The waking effect of 8-10 h induced by modafinil before and after sleep deprivation was similar and was not followed by an increase in sleep rebound. On the contrary, the arousal effect about 8 h evoked by amphetamine after sleep d

Electroencephalographic findings in pediatric cases of coma.

Electroencephalographic aspects of coma in 201 children are presented. The ages of the subjects ranged between 1 month and 14 years. The EEG picture was not specific. Diffuse slowing and convulsive activity were present in meningoencephalitis and postconvulsive coma. Diffuse slowing was also present in coma of metabolic origin. Mixed slow and fast activity was typical of coma due to benzodiazepines, barbiturates and alcohol. In patients with intracerebral hemorrhages, there was marked focal slowing associated with generalized delta and theta waves. In 12 patients with craniocerebral trauma, th
