Brimonidine Beyond a Single Specialty: Pharmacological Profile, Dermatologic Applications, and Advances in Drug Delivery Systems.

Brimonidine, a highly selective α2-adrenergic receptor agonist originally developed for glaucoma treatment, has emerged as an important dermatological agent due to its potent vasoconstrictive and anti-inflammatory properties. This review summarizes its pharmacological characteristics, and clinical applications. By activating α2-adrenergic receptors in cutaneous vessels, brimonidine induces rapid, reversible vasoconstriction and reduces neurogenic inflammation, leading to significant improvement of facial erythema in rosacea. Beyond its approved indication, topical brimonidine demonstrates effi

Chitosan-based bioactive polymer systems for the treatment of vulvovaginitis: advances in vaginal drug delivery.

Vulvovaginitis is an inflammation of the vulva and vagina that is commonly caused by yeast infections, bacterial vaginosis, or sexually transmitted illnesses affecting approximately 70-75% of women at least once during their lifetime, with recurrence rates reaching 30-40%. Vulvovaginal candidiasis (VVC) is an infection of the vulva and vagina caused by an overabundance of Candida yeast. Available therapy is not always effective, and resistance to several antifungal drugs therefore there is a need for advanced drug delivery systems. In recent decades, chitosan, a biopolymer having intrinsic ant…

Exploring Recent Advances in Colon-targeted Drug Delivery Systems: from Traditional Methods to Advanced Technologies.

PURPOSE OF REVIEW: This review examines various colon-targeted drug delivery systems (CTDDS) and their implications for the treatment of colonic disorders. RECENT FINDINGS: Recent studies have demonstrated substantial advancements in CTDDS by the development of pH-dependent, enzyme-responsive, and time-controlled formulations designed for enhancing site-specific delivery and reducing systemic adverse effects. Advanced methodologies, such as ligand or receptor-mediated targeting and multifunctional systems that incorporate various triggering mechanisms, are demonstrating encouraging outcomes i…

Drug delivery systems loaded with plant-derived natural products for dental caries prevention and treatment.

Dental caries, driven by dysbiosis in oral flora and acid accumulation, pose a significant threat to oral health. Traditional methods of managing dental biofilms using broad-spectrum antimicrobials and fluoride face limitations such as microbial resistance. Natural products, with their antimicrobial properties, present a promising solution for managing dental caries, yet their clinical application faces significant challenges, including low bioavailability, variable efficacy, and patient resistance due to sensory properties. Advanced drug delivery systems (DDSs) are emerging to address these l…

Recent Advances in the Local Drug Delivery Systems for Diabetic Wound Healing: A Comprehensive Review.

Wound management in diabetic patients holds significant importance in both clinical and social contexts due to the delayed and compromised healing that these individuals experience. Diabetic wounds exhibit slow and incomplete healing, increasing patients' susceptibility to infections. Managing wounds in diabetic patients, particularly when complicated by diabetic foot infection or diabetic foot ulcer, becomes challenging. The ideal drug delivery systems for treating diabetic wounds should integrate diverse drugs and/or biological factors, offering advantages such as sustained and localized rel…

Label-free classification of nanoscale drug delivery systems using hyperspectral imaging and convolutional neural networks.

Label-free characterization of nanoscale drug delivery systems remains a critical challenge in pharmaceutical research. Traditional analytical methods, such as cryo-electron microscopy, are labor-intensive, low-throughput, and often require labeling, which can interfere with nanoparticle functionality. This study introduces a non-invasive hyperspectral imaging (HSI) framework combined with deep learning to classify therapeutic liposomes. A 3D convolutional neural network (3D CNN) was employed to extract spatial-spectral features, while the synthetic minority oversampling technique (SMOTE) addr…

Overview of biomembrane-derived nanoparticles as emerging drug delivery systems for treating bone aging diseases.

Bone aging diseases, including osteoporosis and osteoarthritis, present significant global health challenges, particularly in countries experiencing demographic aging. Conventional treatment strategies such as pharmacotherapy and lifestyle adjustments frequently yield suboptimal therapeutic outcomes and are carry risks of long-term adverse effects. In recent years, biomembrane-derived nanoparticles have emerged as promising candidates for advanced drug delivery systems to overcome these limitations. By encapsulating therapeutic agents within biomembranes (cell membrane, extracellular vesicles)…

Tacrolimus-loaded Drug Delivery Systems in Vascularized Composite Allotransplantation: Lessons and Opportunities for Local Immunosuppression.

Long-term systemic immunosuppression is needed for vascularized composite allotransplantation (VCA). The high rate of acute rejection episodes in the first posttransplant year, the development of chronic rejection, and the adverse effects that come along with this treatment, currently prevent a wider clinical application of VCA. Opportunistic infections and metabolic disturbances are among the most observed side effects in VCA recipients. To overcome these challenges, local immunosuppression using biomaterial-based drug delivery systems (DDS) have been developed. The aim of these systems is to…

Overview of Venous Thromboembolism and Emerging Therapeutic Technologies Based on Nanocarriers-Mediated Drug Delivery Systems.

Venous thromboembolism (VTE) is a serious health condition and represents an important cause of morbidity and, in some cases, mortality due to the lack of effective treatment options. According to the Centers for Disease Control and Prevention, 3 out of 10 people with VTE will have recurrence of a clotting event within ten years, presenting a significant unmet medical need. For some VTE patients, symptoms can last longer and have a higher than average risk of serious complications; in contrast, others may experience complications arising from insufficient therapies. People with VTE are initial

The potential use of bacteria and bacterial derivatives as drug delivery systems for viral infection.

Viral infections in humans are responsible for fatalities worldwide and contribute to the incidence of various human ailments. Controllable targeted medicine delivery against many illnesses, including viral infection, may be significantly aided by using bacteria and bacteria-derived products. They may accumulate in diseased tissues despite physical obstacles, where they can launch antiviral immunity. The ability to genetically and chemically modify them means that vaccinations against viral infections may be manufactured and delivered to affected tissues more safely and effectively. The object

Источник пометил публикацию как отозванную

CONTEXT: Eplerenone is a member of antihypertensives used individually or in combination with other medicines. Eplerenone exhibits poor solubility and is considered a class II drug. OBJECTIVE: Increasing the solubility of eplerenone by using both liquid and solid self-emulsifying drug delivery systems as an alternative to its marketed tablet product. METHODS: Solubility studies of eplerenone were done with different oils, surfactants, and co-surfactants to determine which one has the highest solubility for eplerenone and determine the preference in the formulations of liquid self-emulsifying…

Spatio-Temporal In Vivo Imaging of Ocular Drug Delivery Systems using Fiberoptic Confocal Laser Microendoscopy.

Subconjunctival injection is an attractive route to administer ocular drugs due to easy trans-scleral access that bypasses anterior ocular barriers, such as the cornea and conjunctiva. While therapeutic effects and pharmacokinetics of the drugs upon subconjunctival injection have been described in some studies, very few assess the ocular distribution of drugs or drug delivery systems (DDS). The latter is critical for the optimization of intraocular DDS design and drug bioavailability to achieve the desired ocular localization and duration of action (e.g., acute versus. prolonged). This study e…
