Compartmental Modeling of Vitamin A Stable Isotope Data from Milk or Plasma Provides Comparable Predictions of Vitamin A Stores in Theoretical Lactating Women.

BACKGROUND: Previous compartmental models describing and quantifying whole-body vitamin A (VA) metabolism have been developed from plasma retinol kinetic data after human subjects ingest stable isotope-labeled VA. For humans, models based on data obtained from other sampling sites (e.g., excreta or milk) have not been proposed. OBJECTIVES: Our objective was to determine whether comparable model predictions of VA total body stores (TBS) in theoretical lactating women were obtained using tracer data from only retinol in plasma or VA in milk. METHODS: We used Simulation, Analysis and Modeling s

Development of a Compartmental Model for Studying Vitamin A Kinetics and Status in Theoretical Lactating Women.

BACKGROUND: Low vitamin A status and suboptimal milk vitamin A concentrations are problems in many populations worldwide. However, limited research has been done on whole-body vitamin A kinetics in women of reproductive age, especially during lactation. OBJECTIVES: Goals were to develop compartmental models describing retinol kinetics in theoretical nonlactating (NL) and lactating (L) women and to determine whether the retinol isotope dilution (RID) method accurately predicted vitamin A total body stores (TBS) in the groups and individuals. METHODS: We adapted 12 previously-used theoretical

Potential role of endocrine-disrupting chemicals in preeclampsia: A hypothesis based on integration of epidemiological and experimental evidence.

Preeclampsia (PE) is a complex hypertensive disorder and a leading cause of maternal and perinatal morbidity worldwide. Emerging evidence suggests that exposure to endocrine-disrupting chemicals (EDCs) may contribute to the etiology of PE, yet this association remains underexplored. This review aimed to investigate epidemiological and experimental studies assessing the potential link between EDC exposure and PE development. A literature search was conducted across PubMed, ScienceDirect, and Google Scholar for original articles published in the last ten years. Forty studies were selected, inclu

Best practice assessment of disease modelling for infectious disease outbreaks.

During emerging disease outbreaks, public health, emergency management officials and decision-makers increasingly rely on epidemiological models to forecast outbreak progression and determine the best response to health crisis needs. Outbreak response strategies derived from such modelling may include pharmaceutical distribution, immunisation campaigns, social distancing, prophylactic pharmaceuticals, medical care, bed surge, security and other requirements. Infectious disease modelling estimates are unavoidably subject to multiple interpretations, and full understanding of a model's limitatio…

In-vivo rodent models for the experimental investigation of prenatal immune activation effects in neurodevelopmental brain disorders.

Based on the epidemiological association between maternal infection during pregnancy and enhanced risk of neurodevelopmental brain disorders in the offspring, a number of in-vivo models have been established in rats and mice in order to study this link on an experimental basis. These models provide indispensable experimental tools to test the hypothesis of causality in human epidemiological associations, and to explore the critical neuroimmunological and developmental factors involved in shaping the vulnerability to infection-induced neurodevelopmental disturbances in humans. Here, we summariz

Consequences of kriging and land use regression for PM2.5 predictions in epidemiologic analyses: insights into spatial variability using high-resolution satellite data.

Many epidemiological studies use predicted air pollution exposures as surrogates for true air pollution levels. These predicted exposures contain exposure measurement error, yet simulation studies have typically found negligible bias in resulting health effect estimates. However, previous studies typically assumed a statistical spatial model for air pollution exposure, which may be oversimplified. We address this shortcoming by assuming a realistic, complex exposure surface derived from fine-scale (1 km × 1 km) remote-sensing satellite data. Using simulation, we evaluate the accuracy of epidem

Inferring the Ecological Niche of Toxoplasma gondii and Bartonella spp. in Wild Felids.

Traditional epidemiological studies of disease in animal populations often focus on directly transmitted pathogens. One reason pathogens with complex lifecycles are understudied could be due to challenges associated with detection in vectors and the environment. Ecological niche modeling (ENM) is a methodological approach that overcomes some of the detection challenges often seen with vector or environmentally dependent pathogens. We test this approach using a unique dataset of two pathogens in wild felids across North America: Toxoplasma gondii and Bartonella spp. in bobcats (Lynx rufus) and

Cannabis- and Substance-Related Carcinogenesis in Europe: A Lagged Causal Inferential Panel Regression Study.

Recent European data facilitate an epidemiological investigation of the controversial cannabis-cancer relationship. Of particular concern were prior findings associating high-dose cannabis use with reproductive problems and potential genetic impacts. Cancer incidence data age-standardised to the world population was obtained from the European Cancer Information System 2000-2020 and many European national cancer registries. Drug use data were obtained from the European Monitoring Centre for Drugs and Drug Addiction. Alcohol and tobacco consumption was sourced from the WHO. Median household inco

Event history analysis and inference from observational epidemiology.

Systematic inclusion of time in observational epidemiological studies may help strengthen the inference to be drawn, but new epidemiological challenges arise, such as time-dependent confounders - covariates which may change from being confounders to being intermediate variables. The focus of this presentation concerns two sets of tools: event history analysis and structural nested failure time models, both applied to a particularly intricate problem in observational epidemiology, of empirically assessing the graft-versus-leukaemia effect after bone marrow transplantation.

Ethnic inequities in mathematical models for respiratory infectious disease: A systematic review.

COVID-19 was the most recent pandemic to highlight profound ethnic inequities. However, most mathematical epidemiological models do not adequately represent these inequities, limiting the potential for such models to inform equitable public health responses and interventions. This systematic review identifies mathematical models of infectious respiratory disease transmission that report results by ethnic group and surveys the different mathematical modelling approaches used to attempt to capture ethnicity-related outcomes. Only 28 studies met the inclusion criteria, which is rare relative to t…

Multiple Disease-Free Equilibria: Toward a Vector Structure of R 0.

Traditional epidemiological models rely on the basic reproductive number, R 0 , as a single threshold quantity to determine whether a disease will persist or die out. However, increased structural complexity in models can lead to the emergence of multiple disease-free equilibria (DFEs). To illustrate this dynamic diversity, we analyze a compartmental model based on the Health Belief Model, where risk perception, social imitation, and behavioral fatigue among susceptible individuals generate the coexistence of multiple DFEs. This case study exposes the limitations of a single scalar threshold …

malariasimple: An R package for fast simulations of malaria transmission.

Process-based malaria transmission models are important tools for evaluating intervention strategies, quantifying uncertainty, and informing malaria control policy. Individual-based models such as malariasimulation are computationally demanding, which limits their practicality for applications that require large numbers of simulation runs. In this paper we present malariasimple, a simplified, compartmental model implemented as an R package which approximates the epidemiological structure and parameter definitions of malariasimulation while operating at a fraction of the computational cost. Spatio-temporal modelling of dengue counts in the Central Valley of Costa Rica.

This study analyses 18 years of weekly reported dengue cases (January 2002-December 2020; 988 weeks) from Costa Rica's Central Valley to examine seasonal and multi-year patterns. To model the spatio-temporal dynamics of dengue, we employ three statistical approaches for case counts: the spatial hurdle integer-valued generalized autoregressive conditional heteroskedasticity (INGARCH) model, the spatial zero-inflated generalized Poisson (ZIGP)-INGARCH model, and the endemic-epidemic (EE) model. Covariates include rainfall and maximum temperature or alternatively seasonal Fourier terms to represe

This approach offers a novel analytical framework for fractional-order epidemiological models. The findings demonstrate that fractional order derivatives are more dependable and efficient than classical orders when it comes to explaining biological processes.

[A study of confounding effect control based on residual strategy with multivariate random forest analysis].

By using generalized estimating equation, random intercept model, random coefficients model, and generalized linear model respectively, this study obtained residuals for predictors and outcomes as new predictors and outcomes for multivariate random forest (MRF) analysis. Simulation experiments were conducted to compare the performance of these methods in confounding adjustment under scenarios with varying outcome correlations, causal effect magnitudes, numbers of confounding factors, and sample sizes. United Kingdom Biobank data were used for further validation. Simulation results showed that

Mathematical analysis and prediction of future outbreak of dengue on time-varying contact rate using machine learning approach.

This article introduces a novel mathematical model analyzing the dynamics of Dengue in the recent past, specifically focusing on the 2023 outbreak of this disease. The model explores the patterns and behaviors of dengue fever in Bangladesh. Incorporating a sinusoidal function reveals significant mid-May to Late October outbreak predictions, aligning with the government's exposed data in our simulation. For different amplitudes (A) within a sequence of values (A = 0.1 to 0.5), the highest number of infected mosquitoes occurs in July. However, simulations project that when βM = 0.5 and A = 0.1,

Gallation and B-Ring Dihydroxylation Increase Green Tea Catechin Residence Time in Plasma by Differentially Affecting Tissue-Specific Trafficking: Compartmental Model of Catechin Kinetics in Healthy Adults.

Catechins in green tea extract (GTE) (epigallocatechin gallate (EGCG), epigallocatechin (EGC), epicatechin (EC), epicatechin gallate (ECG)) vary in bioactivity. We developed a physiologically relevant mathematical model of catechin metabolism to test the hypothesis that fractional catabolic rates of catechins would be differentially affected by their structural attributes. Pharmacokinetic data of plasma and urine catechin concentrations were used from healthy adults (n = 19) who ingested confections containing 0.5 g GTE (290 mg EGCG, 87 mg EGC, 39 mg EC, 28 mg ECG). Zoonotic visceral leishmaniasis transmission: modeling, backward bifurcation, and optimal control.

Visceral leishmaniasis (VL), a vector-borne disease caused by protozoan flagellates of the genus Leishmania, is transmitted by sand flies. After malaria, VL is the second-largest parasitic killer, responsible for an estimated 500,000 infections and 51,000 deaths annually worldwide. Mathematical models proposed for VL have included the impact of dogs versus wild canids in disease dissemination and models developed to assist in control approaches. However, quantitative conditions that are required to control or eradicate VL transmission are not provided and there are no mathematical methods prop

Box-Jenkins modelling of some viral infectious diseases.

During the last few years Box-Jenkins models have become of increasing importance in such fields as economics and industry. They have been used for forecasting and for detecting relations between different time series. Similar applications are also relevant in epidemiology. Since many people who are concerned with the analysis of medical data are not familiar with this subject, a short non-technical introduction to Box-Jenkins models is given. The model-building process is demonstrated in some detail using monthly case reports of the two seasonal endemic diseases chicken-pox and mumps, and the

Improving outbreak forecasts through model augmentation.

Accurate forecasts of disease outbreaks are critical for effective public health responses, management of healthcare surge capacity, and communication of public risk. There are a growing number of powerful forecasting methods that fall into two broad categories-empirical models that extrapolate from historical data, and mechanistic models based on fixed epidemiological assumptions. However, these methods often underperform precisely when reliable predictions are most urgently needed-during periods of rapid epidemic escalation. Here, we introduce epimodulation, a hybrid approach that integrates
