COVID-19 is mainly associated with respiratory symptoms, although several reports show that SARS-CoV-2 affects the nervous system. We evaluated the effects of SARS-CoV-2 infection on human derived midbrain organoids by exposing them to the virus (multiplicity of infection 0.05, 16-h exposure) and analyzing cellular and molecular changes at 4 and 28 days post-infection using immunofluorescence microscopy and RNA sequencing. SARS-CoV-2 nucleocapsid protein preferentially colocalized with tyrosine hydroxylase-positive (TH+) dopaminergic neurons, inducing neurite fragmentation and cellular stress.…

Patient-Derived Organoids (PDO) are 3D culture systems that mimic human tissues more accurately than traditional 2D cell lines. They maintain cellular diversity, spatial layout, and patient-specific traits, making them essential for high-throughput screening (HTS). Due to tumor heterogeneity and cancer therapies resistance, combination screening has become increasingly important. Moreover, recent advancements now enable larger-scale combination studies. Standardized matrices improve uniformity and reproducibility, while smaller plate formats and automation streamline seeding, dosing, and overa…

The efficacy of standard adjuvant chemotherapy for locally advanced gastric cancer (GC) remains suboptimal, particularly in patients with signet-ring cell carcinoma (SRCC). Urgent demand exists for reliable preclinical models to predict therapeutic responses, and in vitro drug sensitivity testing using patient-derived organoids (PDOs) has emerged as a promising platform. In this study, PDOs were established from patients with locally advanced GC and analyzed via next-generation sequencing (NGS) and pharmacotyping. Seventeen GC PDOs were successfully generated, achieving a success rate of 63%. …

INTRODUCTION: Organoids are three-dimensional (3D) culture models derived from selforganizing stem cells or progenitor cells, capable of recapitulating the cellular growth microenvironment and interactions within the human body to a certain extent. These models play significant roles in biomedical research, drug screening, and related fields. Research on lung organoids has provided insights into lung development and the cellular and molecular processes underlying diseases, while studies on their transplantation in animals hold promise as novel strategies for post-lung injury therapies. However…

Early heart development involves heart tube elongation, looping, and axial patterning, yet these processes remain difficult to study experimentally. While pluripotent stem cell-derived heart organoids model cardiomyocyte differentiation, they do not recapitulate early morphogenetic events. Here, we generate elongating heart organoids (EHOs) from human induced pluripotent stem cells that undergo coordinated elongation and looping-like curvature, resembling early cardiac morphogenesis. EHOs establish a venous-to-arterial axis with spatially organized sinus venosus-like, atrial, and ventricular c…

Hematologic patient-derived organoids (PDOs) reconstruct human bone marrow and lymph node microenvironments in 3D, preserving malignant cell states, clonal diversity, and clinically relevant drug responses. Leukemia PDOs artificially mimic the complex bone marrow niche and cytokine dependencies. Lymphoma PDOs recapture immune-stromal interactions and enable testing of targeted and immunotherapies, including bispecific antibodies and chimeric antigen receptor (CAR) T-cells. Multiple myeloma PDOs model the bone marrow microenvironment and maintain plasma tumor cell survival cues that drive drug …

Cerebral organoids (COs) are valuable for studying neurodegenerative diseases and pathogens, but their limited microglia pose challenges. Here, we present a protocol to generate organoids with physiologically relevant microglia (CO-iMs). We describe steps for differentiating induced pluripotent stem cells (iPSCs) into mesoderm, seeding embryoid bodies (EBs), and harvesting hematopoietic progenitor cells (HPCs). We detail procedures for co-culturing HPCs and iPSCs to generate CO-iMs, followed by their infection and treatment. The model provides a platform to study viral infections and neuroinfl…

BACKGROUND: Traditionally, transformed cell line monolayers have been the standard model for studying epithelial barrier and transport function. Recently, intestinal organoids were proposed as superior in recapitulating the intestine. Typically, 3D organoids are digested and seeded as monolayers on gelatinous matrix pre-coated surfaces for anchorage. As this coat could potentially act as a diffusion barrier, we aimed to generate robust human duodenum-derived organoid monolayers that do not need a gelatinous matrix for anchorage to improve upon existing models to study epithelial transport and …

Chronic pancreatitis (CP) affects ~3 million people worldwide, yet altering the course of disease is challenging. We developed a patient-derived organoid (PDO) platform to investigate the molecular pathogenesis of this disease and identify therapeutic strategies. We generated 36 PDOs from patients with idiopathic, hereditary, and alcohol-related CP with high genetic concordance. PDOs retained inflammation-associated transcriptional and proteomic features. Transcriptomic profiling revealed three molecular subtypes of CP independent of etiology. We discovered widespread dysfunction of the cystic

Recent advancements in organoid technology have revolutionized our understanding of organ development and disease modelling. However, despite significant progress, retinal organoids continue to face several limitations, including imperfect architecture, incomplete maturation, limited functional integration, and lack of vascularization. These challenges hinder their potential as accurate models of human retinal biology and disease. In this review, we examine the currently available models of age-related macular degeneration, evaluate the potential of retinal organoids, and discuss their respect…
