Xanthine oxidase-induced oxidative stress causes activation of NF-kappaB and inflammation in the liver of type I diabetic rats.

We previously showed that xanthine oxidase activity increases in type I diabetic animals and that this is a significant cause of the oxidative stress which occurs in the disease. The aim of this work was to search for molecular links between xanthine oxidase-induced oxidative stress and inflammation in Type I diabetes and to assess the ability of allopurinol, a drug widely used in clinical practice, to prevent both processes. 3-month-old male Wistar rats were made diabetic by injection (i.p.) of either streptozotocin or alloxan. Allopurinol (32 mg/Kg) was administered (i.p) to diabetic rats af

Olea europaea leaf (Ph.Eur.) extract as well as several of its isolated phenolics inhibit the gout-related enzyme xanthine oxidase.

In Mediterranean folk medicine Olea europaea L. leaf (Ph.Eur.) preparations are used as a common remedy for gout. In this in vitro study kinetic measurements were performed on both an 80% ethanolic (v/v) Olea europaea leaf dry extract (OLE) as well as on nine of its typical phenolic constituents in order to investigate its possible inhibitory effects on xanthine oxidase (XO), an enzyme well known to contribute significantly to this pathological process. Dixon and Lineweaver-Burk plot analysis were used to determine K(i) values and the inhibition mode for the isolated phenolics, which were anal

Increased plasma xanthine oxidase activity is related to nuclear factor kappa beta activation and inflammatory markers in familial combined hyperlipidemia.

BACKGROUND AND AIMS: Xanthine oxidase (XO) has been described as one of the major enzymes producing free radicals in blood. Oxidative stress and inflammatory processes have been implicated in the pathogenesis of endothelial dysfunction and the progression of atherosclerosis but until now, there is little data about the influence of vascular prooxidant systems and inflammation in familial combined hyperlipidemia (FCH). Our goal was to evaluate whether XO activity was altered in FCH and if it was related to the inflammatory process represented by NFkB, IL-6 and hsCRP, and assessing the correlati

From xanthine oxidase inhibition to inflammasome regulation: exploring the potential role of allopurinol in TXNIP-NLRP3 signaling and pyroptosis in diabetes.

Diabetes mellitus (DM) is a chronic metabolic disorder characterized by persistent hyperglycemia and progressive multi-organ complications driven by oxidative stress, inflammation, and metabolic dysfunction. Accumulating experimental evidence identifies the thioredoxin-interacting protein (TXNIP)-NOD-like receptor family pyrin domain-containing 3 (NLRP3) inflammasome axis as a critical molecular link between hyperglycemia-induced oxidative stress and inflammatory cell death, although its therapeutic modulation in diabetes remains under active investigation. Activation of TXNIP promotes assembl

Discovery of a Potent and Orally Bioavailable Xanthine Oxidase/Urate Transporter 1 Dual Inhibitor as a Potential Treatment for Hyperuricemia and Gout.

The main uric acid-lowering agents in clinical use for hyperuricemia and gout are xanthine oxidase (XO) inhibitors or urate transporter 1 (URAT1) inhibitors. While these therapies can partially control the disease, they have various limitations. The development of XO/URAT1 dual inhibitors offers the potential to enhance therapeutic potency and reduce toxicity compared with single-target inhibitors. Through scaffold hopping from the XO inhibitor febuxostat (2) and the URAT1 inhibitor probenecid (3), followed by structure-activity relationship (SAR) studies, we identified compound 27 as a potent

Integration of Pharmacometrics and Pharmacoeconomics to Quantify the Value of Improved Forgiveness to Nonadherence: A Case Study of Novel Xanthine Oxidase Inhibitors for Gout.

Linked pharmacometric and pharmacoeconomic models provide a structured approach for assessing the value of candidate drugs in development. The aim of this study was to assess the utility of such an approach for identifying the properties of xanthine oxidase inhibitors (XOi) providing improved forgiveness to nonadherence and estimate the maximum reimbursement price. The pharmacometric and pharmacoeconomic models were used to simulate the time course of serum uric acid concentrations and estimate quality-adjusted life years and costs for the XOi febuxostat and a range of hypothetical analogues.

The effect of cyclic adenosine 3',5' monophosphate, methyl xanthines and nicotinic acid on plasma nonesterified fatty acids in sheep.

The intravenous injection of 3', 5' AMP, 10 mg/kg, caused an immediate rise in blood glucose in normal sheep. In sheep where liver glycogen was depleted by pretreatment with 100 mg/kg of nicotinic acid, the blood glucose response was greatly reduced. The glycogenolytic effect of 3', 5' AMP was thus easily demonstrated. The injection of 3', 5' AMP was followed by an immediate rise in NEFA, which was most pronounced in the pretreated animals. After the initial increase there was a fall in NEFA in both the normal and the pretreated animals. The role of hyperglycemia for the NEFA response to exoge

Discovery of novel xanthine oxidase inhibitors using deep learning-based composite molecular fingerprint: Screening, bioactivity assessment and binding behavior analysis.

Xanthine oxidase (XO) confirms as a critical target for the treatment of hyperuricemia (HUA). Current Xanthine oxidase inhibitors (XOIs) used in clinical practice is constrained by adverse effects or suboptimal bioavailability, necessitating the discovery of novel inhibitors. In this study, we generated composite molecular fingerprints with deep learning (DL) techniques and trained an XOI classification model for high-throughput virtual screening and combining molecular docking, ADME prediction and manual screening of predicted conformations. Three active XOIs: Hit 3, Hit 5 and Hit 6 were iden

The Assembly of Alginate-Chitosan-Xanthine Hydrogel and Halomonas Improves Soil Quality of Protected Crop Growth Under Saline-Alkali Stress.

Soil saline-alkali stress has become a great risk, leading to significant decline in crop yields, especially in protected agriculture that is frequently threatened by excessive chemical fertilization. There is an urgent need to develop friendly strategies for improving the quality of saline-alkali soil. Microbial inoculants are promising agents in attenuating soil stress, but their colonization ability in the crop rhizosphere is often limited. To improve the efficiency of microbial inoculants, this study constructed the assembly of a salt-tolerant bacterium Halomonas (Homs) and a hydrogel comp

Activation of Coronary Arteriolar PKCβ2 Impairs Endothelial NO-Mediated Vasodilation: Role of JNK/Rho Kinase Signaling and Xanthine Oxidase Activation.

Protein kinase C (PKC) activation can evoke vasoconstriction and contribute to coronary disease. However, it is unclear whether PKC activation, without activating the contractile machinery, can lead to coronary arteriolar dysfunction. The vasoconstriction induced by the PKC activator phorbol 12,13-dibutyrate (PDBu) was examined in isolated porcine coronary arterioles. The PDBu-evoked vasoconstriction was sensitive to a broad-spectrum PKC inhibitor but not affected by inhibiting PKCβ2 or Rho kinase. After exposure of the vessels to a sub-vasomotor concentration of PDBu (1 nmol/L, 60 min), the e

Long-term inhibition of xanthine oxidase by febuxostat does not decrease blood pressure in deoxycorticosterone acetate (DOCA)-salt hypertensive rats.

Xanthine oxidase and its products, uric acid and ROS, have been implicated in the pathogenesis of cardiovascular disease, such as hypertension. We have previously reported that allopurinol inhibition of XO does not alter the progression of deoxycorticosterone acetate (DOCA)-salt hypertension in rats. However other researchers have observed a reduction in blood pressure after allopurinol treatment in the same model. To resolve this controversy, in this study we used the newer and more effective XO inhibitor febuxostat, and hypothesized that a more complete XO blockade might impair hypertension
