Adoptive Transfer of B Cells In Vivo for Assessment of Their Immune Function.

B lymphocytes are a critical part of the adaptive immune response elicited by the immune system to fight various pathogens. The main effector function of the B lymphocytes is the ability to secrete antibodies, whether nonclass-switched immunoglobulin M (IgM) or class-switched immunoglobulin isotypes. To understand the function of B cells in vivo, mice are subjected to bone marrow depletion (using radiation or chemical radiation) before being adoptively transferred with donor bone marrow. Alternatively, B cells can be isolated from spleens and adoptively transferred to B-cell-deficient recipien

Advances in adoptive cellular immunotherapy and therapeutic breakthroughs in multiple myeloma.

The basic idea of modulating the immune system to better recognize and fight tumor cells has led to the successful introduction of adoptive cellular immunotherapy (ACT). ACT-based treatment regimens, in which the patient's own immune cells are isolated and subsequently expanded (ex vivo) and reinfused, have also contributed significantly to the development of a personalized treatment strategy. Complementing this, the unprecedented advances in ACTs as chimeric antigen receptor (CAR)-T cell therapies and their derivatives such as CAR-NK, CAR-macrophages, CAR-γδT and CAR-NKT have further maximize

Therapeutic potentials of adoptive cell therapy in immune-mediated neuropathy.

Immune-mediated neuropathy (IMN) is a group of heterogenous neuropathies caused by intricate autoimmune responses. For now, known mechanisms of different IMN subtypes involve the production of autoantibodies, complement activation, enhanced inflammation and subsequent axonal/demyelinating nerve damages. Recent therapeutic studies mainly focus on specific antibodies and small molecule inhibitors previously approved in rheumatoid diseases. Initial strategies based on the pathophysiologic features of IMN should be explored. Adoptive cell therapy (ACT) refers to the emerging immunotherapies in whi

Solitary plasmacytoma: should new approaches in diagnosis and treatment be adopted?

BACKGROUND: Radiotherapy (RT) is the gold standard for solitary plasmacytomas (SP) with great local control. The influence of radiotherapy as well as factors on multiple myeloma (MM) progression is unknown. MATERIALS AND METHODS: We present a retrospective study of 27 patients with SP (bone-SBP- and extramedullary-SEP-), treated since 1995 to 2021. We aim to analyze prognostic factors affecting local control and progression to MM in patients treated with radiotherapy (RT). RESULTS: Mean age was 57.3 years. 22 were SBP and 5 SEP. 13 patients were treated with definitive RT, and 14 with a comb

Health of Post-Institutionalized Haitian Children Adopted to the United States.

OBJECTIVES: Haiti remains a principal placement country for intercountry adoptees to the United States. This project reports the health status of children adopted from Haiti arriving to the U.S. and compares them to intercountry adoptees from other regions. METHODS: A retrospective chart review was conducted of adoptees placed in the U.S. from Haiti (n=87), age and sex matched with intercountry adoptees placed in the U.S. from Asia (n=87) and Latin America (n=87) between January 2010 and November 2019. Data on immunization status, contagious diseases, and nutrition and growth were analyzed vi

Hymenoptera venom skin testing: Adopting an accelerated test protocol.

BACKGROUND: The standard method of Hymenoptera venom intradermal skin test is performed at a starting concentration of 0.001 to 0.01 μg/mL and increased by 10-fold concentrations until positive or a maximum concentration of 1 μg/mL. Accelerated methods that start at higher concentrations have been reported as safe; however, many institutions have not adopted this approach. OBJECTIVE: To compare the outcome and safety of standard and accelerated venom skin test protocols. METHODS: This was a retrospective chart review of patients with suspected venom allergy who underwent skin testing at 4 al

Is it time for Africa to adopt primaquine in the era of malaria control and elimination?

Primaquine is a gametocytocidal drug known to significantly reduce malaria transmission. However, primaquine induces a dose-dependent acute hemolytic anemia (AHA) in individuals with glucose-6-phosphate dehydrogenase (G6PD) deficiency that has led to a limited use of the drug especially in Africa where the condition is common. The World Health Organization (WHO) now recommends a single low dose (SLD) of primaquine (0.25 mg/kg) as P. falciparum gametocytocidal without the need for prior screening of G6PD status. Adoption and implementation of SLD primaquine in Africa may probably reduce malaria

Practical Considerations in Adopting New Technology for Facial Cosmetic Procedures.

The field of facial plastic and reconstructive surgery encompasses both surgical and nonsurgical facets, creating a supplemental level of care and an additional layer of complexity. Determining the "best course of care" can be very difficult in experienced situations, but even more so when considering adopting an emerging technology. A basic and practical method of analyzing a new technology requires investigating the risk-to-benefit assessment, the utility and clinical outcomes compared with other treatment options, and an introspective ethical appraisal of whether the technology is foremost

Variation in complications and mortality following ALPPS at early-adopting centers.

BACKGROUND: Various, often conflicting, estimates for post-operative morbidity and mortality following ALPPS have been reported in the literature, suggesting that considerable center-level variation exists. Some of this variation may be related to center volume and experience. METHODS: Using data from seventeen centers who were early adopters of the ALPPS technique, we estimated the variation, by center, in standardized 90-day mortality and comprehensive complication index (CCI) for patients treated between 2012 and 2018. RESULTS: We estimated that center-specific 90-day mortality following

Barriers to the Adoption of Technological Innovations in Corrections: A Review and Case Study.

Over 600,000 criminal justice involved individuals are released from state and federal prisons each year, and close to 5,000,000 former offenders are placed under some form of community-based supervision. Access to services that may facilitate the reentry process is complicated and more often than not, returning citizens have significant and wide-ranging needs left unaddressed that require a comprehensive approach. In the current paper, we discuss the adoption and implementation processes of technological innovations noted in the criminal justice and correctional literature, as well as other d

Beyond the borders: Lessons from various industries adopted in anesthesiology.

Since the first public demonstration of anaesthesia in Boston, USA which happened around 172 years back, the field of anesthesiology has rapidly progressed, with many developments that have improved the quality and safety of anesthesia care. This has enabled tremendous advances in the surgical disciplines and increasing the life expectancy and quality of life of humans. This is a result of learning and constantly evolving. There are several similarities between healthcare and other industries, though there are several distinguishing characteristics that set it apart from other industries. Ther

Adoptive Treg Cell Therapy in a Patient With Systemic Lupus Erythematosus.

OBJECTIVE: Adoptive Treg cell therapy has great potential to treat autoimmune disease. Currently, very little is known about how these cells impact inflamed tissues. This study was undertaken to elucidate how autologous Treg cell therapy influences tissue inflammation in human autoimmune disease. METHODS: We describe a systemic lupus erythematosus (SLE) patient with active skin disease who received adoptive Treg therapy. We comprehensively quantified Treg cells and immune activation in peripheral blood and skin, with data obtained at multiple time points posttreatment. RESULTS: Deuterium tra

Glucose-6-phosphate dehydrogenase deficiency in internationally adopted children.

There are conflicting guidelines about screening of internationally adopted children for glucose-6-phosphate dehydrogenase (G6PD) deficiency, a common genetic disorder. In a multi-ethnic population of 2,169 internationally adopted children, we found that the prevalence of G6PD deficiency was 1.6% overall and 2.2% in males. Prevalence differed by country or region of origin, ranging from 0 to 13% overall and 0 to 22% in males. The prevalence in females was 1%. A diagnosis of G6PD deficiency informs the treatment of malaria and enables education and counseling to prevent morbidity and mortality
