The PI3K/Akt and MAPK-ERK1/2 pathways are altered in STZ induced diabetic rat placentas.

Diabetic pregnancy is associated with complications such as early and late embryonic death, fetal growth disorders, placental abnormalities, and embryonal-placental metabolic disorders. Excessive apoptosis and/or changes of proliferation mechanisms are seen as a major event in the pathogenesis of diabetes-induced embryonic death, placental weight and structural anomalies. Akt and ERK1/2 proteins are important for placental and fetal development associated with cellular proliferation and differentiation mechanisms. The mechanism underlying the placental growth regulatory effects of hyperglycemi

Integrated RNA-seq and ATAC-seq analyses reveal muscle development differences between european meat pigeon and shiqi pigeon during embryonic to post-hatching stages.

Poultry muscle growth and development critically depend on skeletal muscle formation during embryonic and hatching stages. However, early embryonic muscle development differences between pigeon breeds remain unclear. We used European meat pigeons and Shiqi pigeons, which differ in body size, as models. Pectoral muscles from both breeds were subjected to ATAC-seq and RNA-seq at embryonic day 14 (E14) and post-hatching day 1 (P1). At P1, Shiqi pigeons had a significantly greater proportion of myofibers with 1-50 μm diameters, higher myofiber density, and a predominant 50-100 μm distribution acro

Persistence of the omphalomesenteric duct: another cause of acute abdomen.

The omphalomesenteric or vitelline duct is an embryonic structure that communicates the yolk sac with the midgut, obliterating between 5-9 weeks of intrauterine life. Persistence of the omphalomesenteric duct, which can exist in up to 2% of population, is a congenital anomaly of the gastrointestinal tract that can present as abdominal pain, intestinal obstruction, or gastrointestinal bleeding.

[Ganglioneuroma in a newborn with early inborn syphilis].

A case is reported of a retroperitoneal sympathic nodes ganglioneuroma in a newborn with classic early inborn syphilis involving practically all systems of the viscera, skin, placenta. Some peculiar morphological features of the tumour are described. Possible teratogenic and oncogenic effects of treponema pallidus on embryonal fetal structures during intrauterine development are suggested.

Ultraviolet-B radiation induces transcriptional modulation of components associated with the extracellular matrix in embryos of decapod Macrobrachium olfersii.

The extracellular matrix (ECM) is a non-cellular and three-dimensional structure, constituted by a macromolecular dynamic network that involves the cells in all animal tissues, including embryonic ones. Several studies with vertebrates and cell cultures have reported deleterious effects of ultraviolet-B (UVB) radiation on the components associated with the ECM. However, studies focusing on the UVB radiation effects on ECM components of crustaceans during embryonic development are very scarce. Thus, the aim of this study was to identify the coding sequences of components associated with the ECM

Beta-keratin localization in developing alligator scales and feathers in relation to the development and evolution of feathers.

Beta-keratins form large part of the corneous material of scales and feathers. The present immunocytochemical study describes the fine distribution of scale- and feather-keratins (beta-keratins) in embryonic scales of the alligator and in avian embryonic feathers. In embryonic scales of the alligator both scale-keratin and feather-keratin can be immunolocalized, especially in the subperiderm layer. No immunolabeling for feather keratin is instead present in the adult scale after the embryonic epidermis is lost. The embryonic epidermis of feather folds into barb ridges while subperiderm or subs

Laminar organization of the anterior olfactory nucleus-the interplay between neurogenesis timing and neuroblast migration.

INTRODUCTION: The anterior olfactory nucleus (AON) is a laminar structure embedded within the olfactory peduncle which serves as the conduit for connectivity between the olfactory bulb (OB) and the central processing centers of the brain. The largest portion of the AON is a ring of neurons and fibers that surround the core of the peduncle, the pars principalis (AONpP). The AONpP is further subdivided into an outer plexiform layer, or layer 1 (L1), that contains axons and dendrites, and an inner cell zone, or layer 2 (L2), formed by densely packed pyramidal cells. Relative to other regions of t

An engineered multicellular stem cell niche for the 3D derivation of human myogenic progenitors from iPSCs.

Fate decisions in the embryo are controlled by a plethora of microenvironmental interactions in a three-dimensional niche. To investigate whether aspects of this microenvironmental complexity can be engineered to direct myogenic human-induced pluripotent stem cell (hiPSC) differentiation, we here screened murine cell types present in the developmental or adult stem cell niche in heterotypic suspension embryoids. We identified embryonic endothelial cells and fibroblasts as highly permissive for myogenic specification of hiPSCs. After two weeks of sequential Wnt and FGF pathway induction, these

Engineering the Developmental Continuum: From Embryo-Like Structures to Functional Tissue Interfaces.

Advanced in vitro developmental models, including embryo-like structures, organoids, and organ-on-chip platforms, have emerged as powerful systems for studying human development under controlled conditions. These models are enabled by advances in stem cell biology and bioengineering, allowing multicellular self-organization that recapitulates selected aspects of embryonic patterning and tissue formation. Key engineering parameters-such as stem cell source, signaling control, geometric confinement, matrix composition, and microfluidic regulation-define the boundary conditions that guide develop

Morphogenesis of the Vertebrate Intestine: From Form to Function.

Vertebrate gut morphogenesis involves complex tissue transformations that convert a simple gut tube into a functionally sophisticated organ with a multilayer architecture optimized for nutrient absorption. The formation of the functional gut requires coordinated development at multiple scales, from cellular behaviors to tissue-level deformations. In this review, we examine recent advances in our understanding of three fundamental aspects of gut development: the formation of the primitive gut tube from embryonic germ layers, establishment of left-right asymmetry driving gut rotation and looping

Established, New and Emerging Concepts in Brain Vascular Development.

In this review, we discuss the state of our knowledge as it relates to embryonic brain vascular patterning in model systems zebrafish and mouse. We focus on the origins of endothelial cell and the distinguishing features of brain endothelial cells compared to non-brain endothelial cells, which is revealed by single cell RNA-sequencing methodologies. We also discuss the cross talk between brain endothelial cells and neural stem cells, and their effect on each other. In terms of mechanisms, we focus exclusively on Wnt signaling and the recent developments associated with this signaling network i

Adipose derived stem cells to construct parathyroid organoid for hypoparathyroidism.

Hypoparathyroidism is one of the most common postoperative complications of thyroid surgery, and organoid transplantation is a frontier field expected to treat hypoparathyroidism. Organoids are three-dimensional cell aggregates derived from embryonic stem cells, pluripotent stem cells, or tissue precursor cells, possessing similar structures and functions to organs. Thus they can replace diseased organs to play a role. Adipose-derived stem cells (ASCs) are a population of postnatal stem cells residing in the fat tissue, capable of differentiating into parathyroid-like cells with parathyroid ho

Ifit1 inhibits Japanese encephalitis virus replication through binding to 5' capped 2'-O unmethylated RNA.

The interferon-inducible protein with tetratricopeptide (IFIT) family proteins inhibit replication of some viruses by recognizing several types of RNAs, including 5'-triphosphate RNA and 5' capped 2'-O unmethylated mRNA. However, it remains unclear how IFITs inhibit replication of some viruses through recognition of RNA. Here, we analyzed the mechanisms by which Ifit1 exerts antiviral responses. Replication of a Japanese encephalitis virus (JEV) 2'-O methyltransferase (MTase) mutant was markedly enhanced in mouse embryonic fibroblasts and macrophages lacking Ifit1. Ifit1 bound 5'-triphosphate

Enforced expression of the homeobox gene Mixl1 impairs hematopoietic differentiation and results in acute myeloid leukemia.

Mixl1, the sole murine homologue of the Xenopus Mix/Bix family of homeobox transcription factors, is essential for the patterning of axial mesendodermal structures during early embryogenesis. Gene targeting and overexpression studies have implicated Mixl1 as a regulator of hematopoiesis arising in differentiating embryonic stem cells. To assess the role of Mixl1 in the regulation of adult hematopoiesis, we overexpressed Mixl1 in murine bone marrow using a retroviral transduction/transplantation model. Enforced expression of Mixl1 profoundly perturbed hematopoietic lineage commitment and differ

The C. elegans evl-20 gene is a homolog of the small GTPase ARL2 and regulates cytoskeleton dynamics during cytokinesis and morphogenesis.

The in vivo functions of ARF-like members of the Ras superfamily of GTPases are relatively unexplored. Here we describe the analysis of C. elegans evl-20 gene that encodes a functional homolog of human ARL2. Elimination of evl-20 function results in abnormal vulval, gonad, and male tail development and disrupts embryonic proliferation, hypodermal enclosure, and elongation. Loss of evl-20 function causes specific defects in the microtubule cytoskeleton, which is the likely molecular basis for the observed defects. EVL-20 is closely associated with both the cell cortex and astral microtubules, s

Intracranial extracerebral neuroglial heterotopia: A case report and review of the literature.

Heterotopic masses of neuroglial tissue are uncommon and most frequently involve extracranial midline structures. We report an unusual case of an intracranial, extracerebral neuroglial heterotopia involving the middle and anterior cranial fossae of a 5-year-old girl who presented with facial asymmetry. The lesion was composed of mature but disorganized gray and white matter admixed with surrounding soft tissues and exhibited histologic features reminiscent of cortical dysplasia. These rare lesions have been postulated to arise from a protrusion of tissue from the neuraxis through a pial defect

Female splenic-gonadal fusion of the discontinuous type.

Splenic-gonadal fusion is an extremely rare congenital anomaly that results from fusion between the splenic and gonadal anlage during embryonic development. Approximately 90 cases have been reported in the literature. The condition has been divided into two major subgroups: continuous splenic-gonadal fusion in which a continuous cord-like structure connects the spleen and the gonadal-mesonephric structures, and discontinuous splenic-gonadal fusion in which no such connection exists. We report a case of discontinuous splenic-gonadal fusion in an adult women with no evidence of other anomalies o

A betaPix Pak2a signaling pathway regulates cerebral vascular stability in zebrafish.

The vasculature tailors to the needs of different tissues and organs. Molecular, structural, and functional specializations are observed in different vascular beds, but few genetic models give insight into how these differences arise. We identify a unique cerebrovascular mutation in the zebrafish affecting the integrity of blood vessels supplying the brain. The zebrafish bubblehead (bbh) mutant exhibits hydrocephalus and severe cranial hemorrhage during early embryogenesis, whereas blood vessels in other regions of the embryo appear intact. Here we show that hemorrhages are associated with poo

Placental defects in alpha7 integrin null mice.

The alpha7beta1 integrin is a heterodimeric transmembrane receptor that links laminin in the extracellular matrix to the cell cytoskeleton. Loss of the alpha7 integrin chain results in partial embryonic lethality. We have previously shown that alpha7 integrin null embryos exhibit vascular smooth muscle cell defects that result in cerebral vascular hemorrhaging. Since the placenta is highly vascularized, we hypothesized that placental vascular defects in alpha7 integrin null embryos may contribute to the partial embryonic lethality. Placentae from embryonic day (ED) 9.5 and 13.5 alpha7 integrin

Intraplacental Mature Cystic Teratoma.

INTRODUCTION: Placental teratomas are benign germ cell tumors composed of mature tissues from multiple germ layers. They may mimic a cardiac fetus but lack organized fetal structures and an umbilical cord. CASE PRESENTATION: A 25-year-old G5P1224 woman delivered at 34 weeks of gestation following PPROM. Examination of the placenta revealed a 3.5 cm intraplacental, mixed solid and cystic lesion without a pedicle. Histology showed mature tissues including cartilage, ganglion cells, choroid plexus, and ciliated epithelium, consistent with a mature cystic teratoma. DISCUSSION: The absence of an
