Organelle stress in NLRP3 inflammasome: a central mediator of neurodegenerative diseases.

Organelle stress and NLRP3 inflammasome activation have been established as pivotal contributors to inflammatory responses and play a significant role in the pathogenesis of neurodegenerative diseases (NDDs), including Alzheimer’s disease (AD) and Parkinson’s disease (PD). However, the involvement of multiple organelles and their interorganellar signaling in NLRP3 inflammasome activation has not yet been systematically elucidated. The causal mechanisms by which these organelles promote NLRP3 activation and subsequently drive the progression of NDDs remain unclear. Furthermore, therapeutic inte

UBE2O-mediated monoubiquitination licenses NLRP6 inflammasome activation in the intestine.

The NLRP6 inflammasome in intestinal epithelial cells (IECs) is a key sentinel of enteric pathogens, yet its activation mechanism remains enigmatic. Here, we identify monoubiquitination as a critical post-translational switch for NLRP6 inflammasome activation. We demonstrate that the E3 ligase UBE2O catalyzes dual-site monoubiquitination of NLRP6: at K680-687 to drive oligomerization via a conformational change, and at K115/130 within the nuclear localization signal to enforce cytoplasmic sequestration through steric hindrance. Mice harboring UBE2O deficiency or monoubiquitination-resistant NL

Colchicine attenuates kidney injury by modulating the NLRP3 inflammasome pathway.

BACKGROUND: Colchicine, a well-known anti-inflammatory drug, has emerged as a therapeutic option in various inflammatory diseases. However, its role in chronic kidney disease (CKD) remains unclear. This study aims to investigate the reno-protective effects of colchicine in an experimental CKD model and its potential role in modulating the NOD-like receptor, pyrin domain containing protein 3 (NLRP3) inflammasome pathway. METHODS: A CKD animal model was established in C57BL/6 mice by feeding a 0.2% adenine-containing diet. Colchicine (0.3 mg/kg/day) or saline was administered intraperitoneally

Piperine inhibits NLRP3 inflammasome activation and alleviates inflammatory disease.

BACKGROUND: Excessive or aberrant activation of the NLRP3 inflammasome is implicated in the initiation and progression of various inflammatory diseases. PURPOSE: This study aimed to investigate whether piperine (PIP) inhibited NLRP3 inflammasome activation and disrupting the NEK7-NLRP3 interaction, and to evaluate its anti-inflammatory effects in vivo. METHODS: We identified NLRP3 as a potential target of PIP through COMET database and RNA sequencing. A classic NLRP3 inflammasome activation model was established in immortalized bone marrow-derived macrophages (iBMDMs) primed with lipopolysac

Structural mechanisms for ATP-mediated inhibition of human NLRP6 inflammasome.

Inflammasomes ignite innate immune defense in response to infectious pathogens and noninfectious dangers, primarily through sensors composed of nucleotide-binding domain (NBD), leucine-rich repeat (LRR)-containing (NLR) family proteins. NLRP6 is an inflammasome sensor that plays critical roles in regulating intestinal inflammation, and its overactivation is linked to autoinflammatory diseases such as inflammatory bowel disease. However, how NLRP6 is maintained in an inhibited structure is unknown. Here we report two cryogenic-electron microscopy structures of human NLRP6 monomer in the adenosi

4-Methylcatechol attenuates NLRP3 inflammasome activation to limit inflammation.

NACHT-, LRR- and pyrin domain-containing protein 3 (NLRP3) is an intracellular sensor that detects exogenous pathogenic invasions and endogenous cellular damage, leading to the NLRP3 inflammasome activation, which plays vital roles in host defense and contributes to the pathogenesis of inflammatory diseases, such as acute peritonitis and gout. Therefore, targeting NLRP3 or other signaling molecules downstream has the potential therapeutic benefit. However, effective, low-toxic and clinically promising targeted inhibitors of NLRP3 inflammasome need to be further studied. Through compound librar

Respiratory toxicity of perfluorooctanesulfonic acid (PFOS) via Inflammasome involvement.

Per- and polyfluoroalkyl substances (PFAS), widely used as durable additives in various consumer products for their water- and oil-repellent properties, persist environmentally and bioaccumulate, raising substantial health concerns. The primary route of human exposure towards PFAS is via contaminated food and drinking water, however, mounting evidence highlights inhalation as additional, critical route of exposure. In particular, inhalation of perfluorooctane sulfonate (PFOS) adversely affects respiratory health through immune disruption, oxidative stress, and impaired barrier function, partic

DPP3 restrains the non-canonical inflammasome through PEBP1 cleavage.

The non-canonical inflammasome is a protein complex involved in bacterial infections, and its activation leads to excessive inflammatory responses during sepsis. The precise regulation of the non-canonical inflammasome in the body remains unclear. Here, we found that some chemicals that chelate zinc ions positively regulated the activation of the non-canonical inflammasome. These chemicals acted by inhibiting the activity of dipeptidyl peptidase 3 (DPP3). DPP3 cleaved phosphatidylethanolamine binding protein 1 (PEBP1) to generate an N-terminal fragment, which could bind to caspase-4/11 and inh

Antioxidant defenses of Francisella tularensis perturb Aim2 Inflammasome Activation.

Francisella tularensis is a Gram-negative bacterium that causes tularemia, a fatal zoonotic disease. F. tularensis has been used in the bioweapon programs of several countries. Its potential use as a bioterrorism agent led the CDC to classify F. tularensis as a Tier 1 Select Agent. The cytosolic sensor absent in melanoma 2 (Aim2) detects double-stranded DNA in the cytosol of infected cells and subsequently assembles a multiprotein complex known as the inflammasome. Inflammasome activation drives the secretion of IL-1β and IL-18, key proinflammatory cytokines required for controlling F. tularen

Salmonella Enteritidis SpaO protein regulating inflammasome activation in macrophage.

Activation of the inflammasome plays a critical role in host defense against Salmonella infection, yet the bacterial factors governing this process remain poorly defined. Thus, we constructed a transposon mutant library of Salmonella enterica subspecies enterica serovar Enteritidis (S. Enteritidis C50336ΔfliC) to identify the genes involved in inflammasome activation in the macrophage J774A.1. Out of 2639 mutants, seven strains exhibited influence the inflammasome activation, and spaO was selected for further investigation. Compared with the ΔfliC infection group, infection with the ΔfliCΔspaO

The Mechanism of NLRP3 Inflammasome Activation and Its Roles in Chronic Rhinosinusitis.

NLRP3 (NLR family pyrin domain containing 3) is one of the crucial receptors in pathogen recognition receptor (PRR) families which can recognize the pathogen-associated molecular patterns (PAMPs) and the damage-associated molecular patterns (DAMPs), thus triggering innate immune response. After NLRP3 activation, it recruits the adaptor protein ASC (apoptosis-associated speck-like protein containing a CARD) and the effector protein caspase-1 (CASP-1) to form a multiprotein complex, which is named NLRP3 inflammasome, enables the cleavage of caspase-1. Furthermore, the active c-caspase-1 proteoly

Role of NLRP3 Inflammasome in Chronic Pain and Alzheimer's Disease-A Review.

The coexistence of Alzheimer's disease (AD) and chronic pain (CP) in the elderly population has been extensively documented, and a growing body of evidence supports the potential interconnections between these two conditions. This comprehensive review explores the mechanisms by which CP may contribute to the development and progression of AD, with a particular focus on neuroinflammatory pathways and the role of microglia, as well as the activation of the NLR family pyrin domain containing 3 (NLRP3) inflammasome. The review proposes that prolonged pain processing in critical brain regions can d

Targeting the NLRP3 inflammasome abrogates cardiotoxicity of immune checkpoint blockers.

BACKGROUND: Immune checkpoint inhibitors (ICIs) have revolutionized the treatment of many malignant tumors. However, ICI-induced hyper-immune activation causes cardiotoxicity. Traditional treatments such as glucocorticoids and immunosuppressants have limited effectiveness and may even accelerate tumor growth. This study aimed to identify approaches that effectively reduce cardiotoxicity and simultaneously preserve or enhance the antitumor immunity of ICI therapy. METHODS: ICI injection in melanoma-bearing C57BL/6J female mice was used to simulate cardiotoxicity in patients with tumor undergoi

M464 inhibits activation of NLRP3 inflammasome and inflammatory response in mice.

Non-steroidal anti-inflammatory drugs (NSAIDs) function as antipyretic, analgesic and anti-inflammatory agents. However, its long application might result in gastrointestinal side effects. Therefore, it is imperative to hunt for novel and safer NSAIDs. Here, we report a new class of NSAIDs, M464, which was equipped with a hydrogen sulfide-releasing radical. We delved into its anti-inflammatory properties and underlying molecular mechanisms. In vitro, the relevance of the anti-inflammatory effects of M464 through the NLRP3 signaling pathway was explored by lactate dehydrogenase (LDH) release, W

[The role of NLRP3 inflammasome in the pathogenesis of lupus nephritis and research progress].

Lupus nephritis (LN), one of the most severe complications of systemic lupus erythematosus (SLE), has a complex pathogenesis involving various endogenous factors including autoimmune complex deposition, inflammatory cell infiltration, and cellular damage. Recent research has increasingly highlighted the prominent role of inflammasomes, particularly the nucleotide-binding oligomerization domain-like receptor protein 3 (NLRP3) inflammasome, in LN pathogenesis. Substantial evidence has confirmed its significant role in both the onset and progression of LN. Given that the NLRP3 inflammasome is a c

Nlrc4 Inflammasome Expression After Acute Myocardial Infarction in Rats.

Acute myocardial necrosis activates the immune response and inflammatory processes. Although the initial response is helpful in restoring tissue injury, dysregulated and exacerbated inflammation contributes to the progression of cardiac remodeling. Inflammasomes play important roles in post-infarction inflammation. NALP1/NLRP1, NLRP 3, and NLRC4 are the best-known inflammasomes. NLRP3, which has received the most study in cardiovascular disease, has been linked to increased IL-1β (IL1B) production and caspase-1 activity, as well as impaired cardiac function. The role of NLRP1 and NLRC4 inflamm

Discovery of DFV890, a Potent Sulfonimidamide-Containing NLRP3 Inflammasome Inhibitor.

The discovery of DFV890 ((R)-1), a potent and selective NLRP3 antagonist, is described. Replacement of the sulfonyl urea core from the first-generation NLRP3 antagonist CRID3 with a sulfonimidamide core afforded a novel and potent series of NLRP3 antagonists. The (R)-enantiomers of the sulfonimidamide series were found to be consistently more potent than structurally related sulfonyl ureas. Replacement of the furan unit of CRID3 with a 5-substituted thiazole unit led to DFV890 ((R)-1), which potently inhibited IL-1β production in THP-1 cells and in primary human cells, blocked multiple downstr

NLRP3 inflammasome activity and periodontal disease pathogenesis-A bidirectional relationship.

OBJECTIVE: Periodontitis is an inflammatory oral disease that occurs as a result of the damaging effects of the immune response against the subgingival microflora. Among the mechanisms involved, the nucleotide-binding oligomerization domain, leucine-rich repeat-containing proteins family member NLRP3 (NLR family pyrin domain-containing 3), proposed as the key regulator of macrophage-induced inflammation, is strongly associated with periodontal disease due to the bacterial activators. This paper aimed to present key general concepts of NLRP3 inflammasome activation and regulation in periodontal
