Loratadine dysregulates cell cycle progression and enhances the effect of radiation in human tumor cell lines.

BACKGROUND: The histamine receptor-1 (H1)-antagonist, loratadine has been shown to inhibit growth of human colon cancer xenografts in part due to cell cycle arrest in G2/M. Since this is a radiation sensitive phase of the cell cycle, we sought to determine if loratadine modifies radiosensitivity in several human tumor cell lines with emphasis on human colon carcinoma (HT29). METHODS: Cells were treated with several doses of loratadine at several time points before and after exposure to radiation. Radiation dose modifying factors (DMF) were determined using full radiation dose response surviva

Intraoperative application of different imaging techniques in sacral neuromodulation: a systematic review and meta-analysis.

BACKGROUND: Sacral neuromodulation (SNM) treats bladder dysfunction by implanting electrodes in the sacral foramen to regulate bladder reflexes. Accurate electrode placement is critical but challenging due to anatomical variations, intestinal gas interference, and radiation exposure from X-ray fluoroscopy. Alternative imaging methods are needed to improve precision and safety. METHODS: We searched PubMed, EMBASE, and Ovid Medline for studies (inception–June 2025) comparing imaging techniques for SNM. Outcomes included operative time, puncture attempts, and radiation dose. Study quality was ass

Long-Term Subjective Functional Outcomes of Prostate Cancer: Prostatectomy vs Radiation.

BACKGROUND: Long-term subjective outcomes of prostate cancer are relatively unknown. The Oregon Urology Institute (OUI) has been collecting subjective functional outcome data to help determine the long-term subjective outcomes of prostatectomy vs radiation therapy. METHODS: Patients treated at OUI completed interval post-treatment questionnaires that assessed: urinary, bowel, sexual, and hormonal function, and overall treatment satisfaction. Two cohorts were established: prostatectomy vs radiation. Results from each cohort were compared and analyzed with a linear mixed effect model. RESULTS:

Radiosensitization by BRAF inhibitors.

BACKGROUND: Increased skin toxicity during combination therapy with a BRAF inhibitor and radiation therapy has recently been reported. MATERIAL AND METHODS: We present seven melanoma patients with non-resectable stage III or IV disease and concomitant treatment with a BRAF inhibitor and radiation therapy. RESULTS: In all patients, combination therapy yielded a good local response. Only two patients, both on vemurafenib, showed severe radiation dermatitis (CTCAE grade 3/4) after one and two weeks, respectively, resulting in interruption of BRAF inhibitor treatment. The respective cumulative r

Surgery After Primary Radiation Treatment for Sporadic Vestibular Schwannomas: Case Series.

BACKGROUND: As radiation has become an increasingly popular primary treatment option for sporadic vestibular schwannomas, there is a minority of tumors that do not respond favorably to radiation. Data on repeat radiosurgery are emerging, and salvage surgery has been associated with increased technical difficulty and poor facial nerve outcomes. OBJECTIVE: To review the current literature and report our results with surgical resection of sporadic vestibular schwannomas that have failed primary radiation treatment. METHODS: This is a retrospective, single-surgeon case series of patients with sp

Radiation exposure from abdominal imaging studies in patients with intestinal Behçet disease.

BACKGROUND/AIMS: Recently, several studies have revealed that diagnostic imaging can result in exposure to harmful levels of ionizing radiation in inflammatory bowel disease patients. However, the extent of radiation exposure in intestinal Behcet disease (BD) patients has not been documented. The aim of this study was to estimate the radiation exposure from abdominal imaging studies in intestinal BD patients. METHODS: Patients with a diagnosis of intestinal BD established between January 1990 and March 2012 were investigated at a single tertiary academic medical center. The cumulative effecti

Phase II clinical trial of parenteral hydroxyurea and hyper-fractionated, accelerated external beam radiation therapy in patients with advanced squamous cell carcinoma of the head and neck: toxicity and efficacy with continuous ribonucleoside reductase inhibition.

BACKGROUND: Almost all concurrent chemoradiation regimens for head and neck are platinum based; however, cisplatin is associated with severe renal, oto-, and neurotoxicity. Hydroxyurea (HU) has been associated with fewer irreversible toxicities. We obtained HU in parenteral form to be administered continually during the radiation treatment. Intravenous HU promised better pharmacokinetics and cell cycle blockade. METHODS: Participants had biopsy-proven, untreated squamous cell carcinoma of the oral cavity, oropharynx (stage IV) and hypopharynx (stages II-IV). Radiation therapy consisted initia

Faecal calprotectin and lactoferrin as markers of acute radiation proctitis: a pilot study of eight stool markers.

BACKGROUND: Non-invasive diagnostic tools to evaluate the severity of acute, radiation-induced proctitis are not readily available. The faecal excretion of eight markers of gut inflammation was therefore examined. Five proteins and three lipid derivates were analysed in sequential stool samples taken before and during radiation therapy. METHODS: Stool samples from 15 patients with prostate cancer scheduled for radiation therapy were examined. Pretreatment and in-treatment samples (2nd and 6th weeks) were measured by enzyme-linked immunosorbent assay (ELISA) (calprotectin, lactoferrin, transfe

Seeing radiation: Applications of an imaging survey meter in radiation oncology.

BACKGROUND AND PURPOSE: Radiation shielding surveys require location and evaluation of radiation hotspots in shielding barriers. This process is normally done by using a conventional survey meter and slowly moving the survey meter manually across the face of the barrier. It would be advantageous to have a radiation image of the barrier showing hot spots. This would hasten the process, potentially reveal hidden hot spots and provide clear and unprecedented documentation for a variety of radiation sources, including HDR afterloaders, linacs, Gamma Knife and CT simulators. The purpose of this stu

Autophagy inhibition can overcome radioresistance in breast cancer cells through suppression of TAK1 activation.

BACKGROUND/AIM: Autophagy is frequently activated in radioresistant cancer cells. In the present study, we evaluated the role of autophagy and transforming growth factor-activated kinase 1 (TAK1) in radioresistance. MATERIALS AND METHODS: TAK1 phosphorylation in MDA-MB231 breast cancer cells was evaluated by western blotting. The regulatory effects of the TAK1 inhibitor and autophagy inhibitor were assessed by cell morphology, cell survival and induction of apoptosis. RESULTS: Radiation induced the phosphorylation of TAK1, whereas the inhibition of TAK1 activity enhanced the cytotoxicity of

[Efficacy of first-line radiation for non-resectable carotid paraganglioma].

BACKGROUND: Carotid paragangliomas are rare. Surgical resection is the primary treatment. However, when surgery is impossible, radiation therapy is an alternative potentially enabling local control with less morbidity. We report a case of good local control of an unresectable paraganglioma after external beam radiation. CASE REPORT: A 72-year-old-woman, had a 4-year history of right facial palsy associated with pulsatile tinnitus, episodic ear bleeding and ipsilateral hearing loss. Physical examination revealed a retro-mandibular and sub-mastoid pulsatile mass. Magnetic resonance imaging show

Intraoperative radiation therapy during pancreatectomy for borderline and locally advanced pancreatic cancer: A propensity-matched analysis.

BACKGROUND: Borderline resectable and locally advanced pancreatic ductal adenocarcinoma remains associated with high local recurrence despite modern neoadjuvant therapy and surgery. Intraoperative radiation therapy enables targeted radiation delivery to the tumor bed while minimizing exposure to surrounding structures, yet its oncologic benefit in pancreatic ductal adenocarcinoma remains unclear. This study evaluated whether intraoperative radiation therapy improves survival and recurrence outcomes in borderline resectable/locally advanced pancreatic ductal adenocarcinoma patients undergoing p

The Relationship Between Radiation History and Outcomes of Immediate Implant-Based Breast Reconstruction.

BACKGROUND: Postmastectomy Radiotherapy (PMRT) has become standard of care for patients with breast cancer. However, local recurrence following breast conserving therapy is increasing in incidence due to increased survivorship. The relationship between prior radiation therapy, PMRT and immediate breast reconstruction (IBR) outcomes has not been widely studied. We aim to assess factors that correlate with post-IBR complications in the face of radiation therapy. METHODS: An IRB approved retrospective cohort review of 262 patients was completed using our tumor registry to identify female patient
