The Molecular Architecture of Neurodegeneration: An Integrative Overview of Convergent Mechanisms.

Neurodegenerative diseases such as Alzheimer's, Parkinson's, amyotrophic lateral sclerosis, and Huntington's disease represent a major challenge in neuroscience due to their complex, multifactorial nature and the absence of curative treatments. These disorders share common molecular mechanisms, including oxidative stress, mitochondrial dysfunction, proteostasis collapse, calcium dyshomeostasis, chronic neuroinflammation, and the prion-like propagation of misfolded proteins. Together, these processes trigger a cascade of cellular damage that culminates in synaptic dysfunction and programmed neu

Distinct actin architectures across a giant syncytial cell.

The syncytial slime mold Physarum polycephalum exhibits cytosolic streaming, cell migration, and cell network remodeling within a single cell. It is unclear how cytoskeletal elements are spatiotemporally organized to mediate these processes. Here, we combine live and fixed imaging of Physarum F-actin to uncover organizational and dynamic signatures of the actin cytoskeleton across cell structures. We find circumferential actin filaments constrict and relax to drive peristaltic contractions in feeding tubes, while migratory fans are characterized by actin patches and comets. Our work presents u

Enhancer dynamics and cellular architecture in the human spinal cord.

Studies of neurodegenerative disease mechanisms require an understanding of the cellular complexity of the healthy human spinal cord. Here, we report the spatial organization of RNA transcription and associated enhancer dynamics in the adult human spinal cord at single-cell and single-molecule resolution. We expand traditional multi-omic measurements and identify epigenetically poised and bivalent active transcriptional enhancer states that define cell-type specification. Simultaneous detection of chromatin accessibility and histone modifications in spinal cord nuclei reveals previously unobse

Exploring the shared genetic architecture of migraine and autoimmune diseases.

BACKGROUND: Migraine (MIG) frequently co-occurs with autoimmune diseases (ADs), but the shared genetic basis underlying this comorbidity remains unclear. METHODS: Using publicly available GWAS summary statistics from European populations, we applied an integrated framework to investigate the shared genetic architecture between MIG and seven ADs (SLE, UC, CD, PSC, T1D, RA, MS). Genetic correlations and polygenic overlap were evaluated using LDSC, HDL, and MiXeR. Shared pleiotropic loci were identified via PLACO and annotated with FUMA. Gene-level analyses were conducted using MAGMA, followed b…

Explainable YOLO architectures for egg size measurement and classification.

The poultry industry faces growing demands for automated, high-precision egg grading systems to ensure quality control and meet market standards. Traditional methods relying on manual inspection are time-consuming, subjective, and prone to human error. Although state-of-the-art YOLO architectures have achieved high accuracy in object detection, their limited interpretability constrains adoption in agricultural contexts where transparency and biological relevance are critical. This study presents an interpretable deep learning framework based on YOLOv12 architecture for real-time egg size detec…

Molecular architecture of the ciliary base in mammalian multiciliated cells.

Multiciliated epithelial cells (MCCs) generate tens to hundreds of motile cilia to drive fluid flow in diverse physiological contexts. While the axonemal structure of motile cilia has been described extensively in recent years, the molecular architecture of the transition zone, basal body, and surrounding ciliary environment of MCCs remain more elusive. Here, we use cryo-focused ion beam (cryo-FIB) milling and cryo-electron tomography (cryo-ET) to obtain in situ 3D views of the ciliary base within intact MCCs from mammalian trachea, complemented by in situ cross-linking mass spectrometry (XL/M…

Spatially resolved chromatin architectures in mammalian brain tissues.

Unraveling 3D chromatin architectures is crucial for decoding regulatory programs of development and disease. However, current sequencing-based methods typically profile 3D genome organizations while sacrificing the spatial context of tissues. Here we present spatial high‑throughput chromosome conformation capture (Hi-C), a technology that resolves genome-wide chromatin structures directly within tissue sections. Spatial Hi-C could decipher 3D chromatin structures in mouse embryos and brains with high fidelity and reproducibility. In the cerebellum, granular layer subclusters defined by distin…

Biomimetic Architectural Cover Accelerates Osseointegration of Titanium Implants.

BACKGROUND: Although various surface modifications enhance titanium (Ti) implant osseointegration, achieving rapid bone formation comparable to native healing remains challenging. Current strategies primarily focus on micro-scale topography, often overlooking the critical role of macroscopic three-dimensional (3D) architecture in osteoconduction. METHODS: We hypothesized that mimicking native bone's porous architecture is key to accelerating osseointegration. First, we verified the impact of structural continuity by comparing standard Ti implants with decellularized bone grafts in a mouse cal…

Influence of pore architecture on bone regeneration in atelocollagen scaffolds.

We aimed to compare the bone regeneration capacity of two atelocollagen sponges with distinct porosity, pore size, and pore morphology: Atelocollagen Sponge MIGHTY® (MIGHTY) and the three-dimensional Honeycomb Boosted® scaffold (Honeycomb). Scanning electron microscopy revealed that Honeycomb exhibited a continuous linear pore structure with a uniform honeycomb-like architecture, higher porosity, and larger pores than MIGHTY. Each sponge was placed in a high-density polytetrafluoroethylene dome and fixed to the rat calvaria. X-ray micro-computed tomography showed that Honeycomb increased bone …

Epithelial architecture and signaling activity in the adult human esophagus.

Barrier epithelia function to shield the inside of our bodies from external stressors and pathogens. The esophageal epithelium is no exception, providing protection while at the same time transporting food to the stomach. Although many epithelial tissues are comparable between humans and mice, the human esophageal epithelium displays unique features in both progenitor cell organization and tissue architecture compared to the mouse. These differences have limited our understanding of the adult human esophagus, hindering the development of therapeutic strategies targeting human esophageal diseas…

The Genetic Architecture of the Human Corpus Callosum and its Subregions.

The corpus callosum (CC) is the largest set of white matter fibers connecting the two hemispheres of the brain. In humans, it is essential for coordinating sensorimotor responses and performing associative or executive functions. Identifying which genetic variants underpin CC morphometry can provide molecular insights into the CC's role in mediating cognitive processes. We developed and used an artificial intelligence based tool to extract the midsagittal CC's total and regional area and thickness in two large public datasets. We performed a genome-wide association study (GWAS) meta-analysis o

Limited Proteolysis as a Regulator of Lymphatic Vessel Function and Architecture.

Recent advances have highlighted the multifaceted roles of the lymphatic vasculature in immune cell trafficking, immunomodulation, nutrient transport, and fluid homeostasis. Beyond these physiological functions, lymphatic vessels are critically involved in pathologies such as cancer metastasis and lymphedema, rendering their structural and functional regulation of major interest. Emerging evidence suggests that limited proteolysis is a key regulatory mechanism for lymphatic vascular function. In dyslipidemic conditions, dysregulated calpain activity impairs lymphatic trafficking and destabiliz

Bacterial microcompartment architectures as biomaterials for conversion of gaseous substrates.

Bacterial microcompartments (BMCs) are protein shells encapsulating multiple enzymes of a metabolic pathway. Interpretations of early experiments on carboxysomes led to the narrative that transport of small gases (CO2, O2) across the shell membrane is restricted. Since then, this notion has been largely contradicted by studies of engineered shells, although these shell constructs lack important proteins present in the native BMCs, altering the synthetic shells' topology, surface and mechanical properties. We discuss here an updated model of gas permeability that informs the design of engineere

The Polygenic Architecture of Human Diseases Affecting the Hair Follicle.

Disorders affecting the hair follicle (HF) and pilosebaceous unit impose psychosocial and financial burdens on patients and may signal risk for other medical conditions. Human genetic studies help to identify key physiological mechanisms that govern health and are increasingly used to improve drug development. GWASs identify genetic variants that are common in the population and implicate disease mechanisms that are widely shared among patients. In this study, we synthesize knowledge about the biology of the pilosebaceous unit that has been derived from GWASs of hair-related diseases. We ident
