To verify the physiological role of the pore-forming protein perforin in vivo, its expression in subpopulations of human peripheral blood lymphocytes was examined by immunocytochemical staining and their cytolytic potentials compared. In addition to NK cells and gamma delta T cells, which uniformly expressed abundant perforin in their cytoplasmic granules, only a small subpopulation of CD8+ alpha beta T cells contained perforin, namely the CD11b+ subset. However, in vitro activation with an anti-CD3 antibody and IL-2 induced perforin expression in approximately 50% of the CD8+CD11b- T cells an

We have examined in vitro the effect of the proopiomelanocortin gene product, beta-endorphin (bE), on the cytotoxic activity of natural killer (NK) cells, lymphokine activated killer (LAK) cells and cytotoxic T-lymphocytes (CTL). Our studies show that bE reproducibly suppressed LAK cytotoxic activity in all donors tested. The effect of bE on the generation of CTL varied, and was negligible on CTL cytotoxic function. Our study also confirms the variable nature of the effects of bE on NK cytotoxicity. In all instances, the effects of bE were generally small, but could be blocked by opioid recept

Ectromelia virus-specific cytotoxic T (Tc) cell precursors were present in the draining popliteal lymph node of all strains of mice tested at 2 to 3 days after footpad inoculation of a high dose (10(5) p.f.u.) of the virulent Moscow strain of ectromelia virus. To detect this response it was necessary to culture lymph node cells from infected mice in the presence of T cell growth factors and to use the more sensitive neutral red assay for measuring cytotoxicity. Cells with lytic activity were virus-specific, major histocompatibility complex-restricted TC cells. C57BL/6J resistant mice, which ex

In vitro expanded T cell lines were used to determine whether antigen-specific cytolytic T lymphocytes are generated after infection with the intracellular bacterium, Listeria monocytogenes. Spleen cells from infected mice were cultured in the presence of syngeneic accessory cells, listerial antigen, and interleukin 2 containing supernatants. Cell lines were greater than 98% Thy-1+, L3T4-, Lyt-2+. Bone-marrow macrophages were used as target cells in two in vitro cytolytic assay systems. The Lyt-2+ T cells killed bone marrow macrophages only when infected with L. monocytogenes as assessed in a

Factors which enhance eosinophil-mediated killing of antibody-coated schistosomula of Schistosoma mansoni include semipurified eosinophil colony stimulating factor (CSF-alpha) and eosinophil cytotoxicity enhancing activity (E-CEA) present in supernatants from cultured mononuclear cells. We have examined the mechanism of enhancement. Both actions require oxygen in order to enhance killing and do not enhance killing under anaerobic conditions (P less than or equal to 0.005). E-CEA had no detectable effect upon oxidative metabolism. In contrast to CSF-alpha which, in our previous studies, increas

We have generated measles virus-specific T cell clones from a patient with multiple sclerosis who has been described previously as a strong responder to measles virus. T cell clones were screened on the basis of their capacity to proliferate to measles virus. The cell surface phenotype of each clone was OKT3+, OKT4+, OKT8-. The majority of these clones (11 of 14) were cytotoxic for their autologous measles virus-infected lymphoblastoid B cell target. This cytotoxicity was specific for measles virus inasmuch as these T cell clones could not lyse influenza or mumps virus-infected B cell targets.

Cell-mediated cytotoxicity is considered to be an important defence system of the body in the case of virus diseases, transplant rejection and in the early stages of cancer. Apart from T lymphocytes, two types of effector cells which lyse tissue culture cells by different mechanisms have been investigated intensively over the past decade--K (killer) and NK (natural killer) cells. K cells possess the capacity to lyse antibody-coated target cells. These effector cells have been described to occur mainly in peripheral blood and spleen. They are small lymphocyte-like cells which possess receptors

Natural killer (NK) cells have been used in several clinical trials as adaptive immunotherapy. The low numbers of these cells in peripheral blood mononuclear cells (PBMC) have resulted in various approaches to preferentially expand primary NK cells from PBMC. While some clinical trials have used the addition of interleukin 2 (IL-2) to co-stimulate the expansion of purified NK cells from allogeneic donors, recent studies have shown promising results in achieving in vitro expansion of NK cells to large numbers for adoptive immunotherapy. NK cell expansion requires multiple cell signals for survi

We studied the influence of various extracellular Ca++ and Mg++ concentrations on human natural killer (NK) cells and found that NK cell-mediated target cell lysis requires the presence of both divalent cations. The calcium channel blocker verapamil and the local anaesthetics procaine and lidocaine inhibited NK cell-mediated killing of K 562 target cells, whereas the selective K+ channel blocker tetraethylammonium and the selective Na+ channel blocker tetrodotoxin had no effect on NK activity. Our results demonstrate an important role of extracellular Ca++ and Mg++ for NK cell-mediated killing

Limited understanding of African swine fever (ASF) immunity remains a major barrier to the rational development of safe and effective vaccines. While antibody-mediated protection is still poorly defined, growing evidence highlights a central role for cellular immunity, particularly cytotoxic cells, as components associated with ASF virus (ASFV) infection control. However, the contribution of individual cytotoxic subsets across different virological and immunological contexts is not well characterized. Here, we investigated cytotoxic responses during BA71ΔCD2 live attenuated vaccine-induced pro

Thalidomide and lenalidomide are immunomodulatory drugs that show promise in mantle cell lymphoma (MCL). In this study, their potential mechanisms of action against MCL cells were investigated, both alone and in combination with rituximab. Thalidomide, lenalidomide and rituximab have no direct effect on MCL cell viability. However, both immunomodulatory drugs indirectly affect viability by enhancing peripheral blood mononuclear cell-mediated cytotoxicity, with lenalidomide inducing significantly higher levels of toxicity than thalidomide. Rituximab induces both complement-dependent and antibod

Little information is available about the generation and specificity of the cytotoxic cells that eliminate human monocytes/macrophages infected with mycobacteria. To address this we have developed a cytotoxicity assay in which 51Cr-labeled monocytes pulsed with bacillus Calmette Guerin (BCG) or Mycobacterium leprae, were used as target cells in overnight cytotoxicity assays. As effector cells, peripheral blood mononuclear cells from healthy occupational contacts or from leprosy patients stimulated with antigen for 7 days were used. Cytotoxicity against antigen-pulsed monocytes that could be in

The effect of cytomegalovirus (CMV) infection on T-lymphocyte subpopulations (employing the monoclonal antibody OKT4 to delineate helper cells and OKT8 for suppressor-cytotoxic cells) was studied in 10 normal individuals with CMV mononucleosis and four renal transplant patients with symptomatic primary CMV disease. In both clinical settings, acute CMV infection is associated with a reversal of the normal helper/suppressor-cytotoxic T cell ratio, with both a relative and absolute decrease in T helper cells and an increase in T suppressor-cytotoxic cells observed. With clinical recovery, there i

Following the recent success of genetically modified T cells in cancer immunotherapy, an increasing number of studies are focusing on natural killer (NK) cells as an alternative to T cells. The characteristics of NK cells may help overcome the disadvantages of T cell-based therapy, such as graft-versus-host disease, cytokine release syndrome, and the high cost and long preparation time of the treatment. To accelerate the development of NK cell-based therapies, the establishment of appropriate mouse models using murine NK cells would be beneficial. Although several groups have reported the esta

Cytotoxic T lymphocytes eliminate infected or malignant cells, safeguarding surrounding tissues. Although experimental and systems-immunology studies have cataloged many molecular and cellular actors involved in an immune response, the design principles governing how the speed and magnitude of T cell responses emerge from cellular decision-making remain elusive. Here, we recast the T cell response as a feedback-controlled program, wherein the rates of activation, proliferation, differentiation, and death are regulated through antigenic, pro- and anti-inflammatory cues. By exploring a broad cla

OBJECTIVE(S): This review focuses on the formation, molecular composition, and functional dynamics of T cell immune synapses within oral lichen planus (OLP) lesions. It aims to systematically elucidate how these specialized junctions direct cytotoxic T cells to recognize and eliminate basal keratinocytes, which is central to OLP pathogenesis.
DESIGN: We synthesized recent advances in the field to dissect the key cellular events at the T cell-keratinocyte interface. The review provides an in-depth analysis of the dynamic regulation of immune synapses, evaluating their role in targeted cytotoxi

Behçet's disease (BD) is a systemic large-vessel vasculitis characterized by a wide clinical spectrum including recurrent oral and genital ulcerations, uveitis, vascular, neurological, articular, renal and gastrointestinal manifestations. Therapeutic management of BD depends on the clinical presentation and organ involved. Although colchicine, nonsteroidal antiinflammatory agents and topical treatments with corticosteroids are often sufficient for mucocutaneous and joint involvements, more aggressive approach with immunosuppressive agents is warranted for severe manifestations such as posterio

In vitro and ex vivo studies assessing the impact of stress hormones on immune competence commonly replace the natural milieu of leukocytes with an artificial medium, excluding plasma factors, hormones, and cytokines. Given prevalent inconsistencies between in vitro, ex vivo, and in vivo findings, we studied whether such procedures could yield misleading outcomes regarding the impact of stress hormones on NK cell cytotoxicity (NKCC), using fresh human whole blood samples. We found that in the presence of plasma 10-30-fold higher concentrations of cortisol, epinephrine, and prostaglandin-E2 (PG

CD4(+) T cells are generally regarded as helpers and regulators of the immune response. Although cytolytic CD4(+) T cells have been described, whether those generated during the course of a viral infection play a role in virus control remains unknown. Here we show that during acute infection with ectromelia virus, the mouse homolog of the human virus of smallpox, large numbers of CD4(+) T cells in the draining lymph node and liver of resistant mice have a cytotoxic phenotype. We also show that these cells kill targets in vivo in a perforin-dependent manner and that mice with specific deficienc

The most relevant criterion for recognizing chronic granular lymphocyte (GL) proliferations, defined as lymphoproliferative disease of GL (LDGL), has been historically based on the number of the proliferating cells displaying typical GL morphology. With the extensive development of immunological and molecular techniques, two major groups of LDGL have been recognized, one belonging to the T-cell and the other to the NK cell lineage. The recent definition of a series of receptors of NK cells (NKR) and the identification of the specific targets recognized has expanded our knowledge of the propert
