Neurological complications of cardiovascular drugs.

Cardiovascular drugs are used to treat arterial hypertension, hyperlipidemia, arrhythmias, heart failure, and coronary artery disease. They also include antiplatelet and anticoagulant drugs that are essential for prevention of cardiogenic embolism. Most neurologic complications of the cardiovascular drugs are minor or transient and are far outweighed by the anticipated benefits of treatment. Other neurologic complications are more serious and require early recognition and management. Overtreatment of arterial hypertension may cause lightheadedness or fatigue but often responds readily to dose

High COVID-19 morbidity and mortality risk among smoked drug users in Brazil.

In much of the West, including Brazil, drug use has increased since social distancing began in response to the pandemic. Use of smoked and modified drugs, and their impacts on health, may contribute to aggravate the effects of the pandemic. However, studies on the relationship between use of smoked drugs and the new coronavirus are still scarce and have not received enough attention in global health recommendations. This paper aims to briefly review the relationship between use of smoked drugs and acute respiratory syndrome coronavirus 2 [SARS-CoV-2]. Recent studies also suggest that drug cons

Multidrug and toxin extrusion proteins as transporters of antimicrobial drugs.

INTRODUCTION: Antimicrobial drugs are essential in the treatment of infectious diseases. A better understanding of transport processes involved in drug disposition will improve the predictability of drug-drug interactions with consequences for drug response. Multidrug And Toxin Extrusion (MATE; SLC47A) proteins are efflux transporters mediating the excretion of several antimicrobial drugs as well as other organic compounds into bile and urine, thereby contributing to drug disposition. AREAS COVERED: This review summarizes current knowledge of the structural and molecular features of human MAT

A research agenda for malaria eradication: drugs.

Antimalarial drugs will be essential tools at all stages of malaria elimination along the path towards eradication, including the early control or "attack" phase to drive down transmission and the later stages of maintaining interruption of transmission, preventing reintroduction of malaria, and eliminating the last residual foci of infection. Drugs will continue to be used to treat acute malaria illness and prevent complications in vulnerable groups, but better drugs are needed for elimination-specific indications such as mass treatment, curing asymptomatic infections, curing relapsing liver

GC-MS-FID characterization and antibacterial activity of the Mikania cordifolia essential oil and limonene against MDR strains.

The present study evaluated the effect of the essential oil of Mikania cordifolia (EOMc) and its major constituent limonene alone or associated with antibacterial drugs against Multidrug Resistant Bacteria (MDR). To evaluate the antibacterial activity, the minimum inhibitory concentrations (MIC) of the oil and limonene against Pseudomonas aeruginosa, Escherichia coli and Staphylococcus aureus were determined. The antibiotic-modulating activity was assessed using subinhibitory concentrations (MIC/8) of these substances in combination with conventional antibacterial drugs. Although no relevant a

Potent and Selective Inhibitors of Trypanosoma cruzi Triosephosphate Isomerase with Concomitant Inhibition of Cruzipain: Inhibition of Parasite Growth through Multitarget Activity.

Triosephosphate isomerase (TIM) is an essential Trypanosoma cruzi enzyme and one of the few validated drug targets for Chagas disease. The known inhibitors of this enzyme behave poorly or have low activity in the parasite. In this work, we used symmetrical diarylideneketones derived from structures with trypanosomicidal activity. We obtained an enzymatic inhibitor with an IC50 value of 86 nm without inhibition effects on the mammalian enzyme. These molecules also affected cruzipain, another essential proteolytic enzyme of the parasite. This dual activity is important to avoid resistance proble

Pesticide poisoning in the developing world--a minimum pesticides list.

In parts of the developing world, pesticide poisoning causes more deaths than infectious diseases. Use of pesticides is poorly regulated and often dangerous; their easy availability also makes them a popular method of self-harm. In 1985, the UN Food and Agriculture Organisation (FAO) produced a voluntary code of conduct for the pesticide industry in an attempt to limit the harmful effects of pesticides. Unfortunately, a lack of adequate government resources in the developing world makes this code ineffective, and thousands of deaths continue today. WHO has recommended that access to highly tox

Metoprolol and propranolol in essential tremor: a double-blind, controlled study.

Single oral doses of propranolol (120 mg), metoprolol (150 mg) and placebo were given in a randomised, double-blind fashion to 23 patients with essential tremor. Both beta blockers were significantly more effective than placebo in reducing the magnitude of tremor. The decrease in tremor produced by metoprolol (47, sem 9%, n = 23) was not significantly different from that observed propranolol (55, sem 5%, n = 23). Tachycardia on standing was antagonised by both drugs to a similar extent. These findings suggest that metoprolol may represent a valuable alternative to propranolol in the treatment

Carotid and vagal afferents and drug action on transcallosally evoked cortical potentials.

The use of transcallosally evoked cortical potentials to study the action of intracarotidly injected drugs on cerebral synapses has necessitated the demonstration that vagal, baroreceptor, and chemoreceptor influences do not play essential roles in the drug effects observed-for example, the cerebral synaptic inhibitory action of serotonin.

Antifungal activities and chemical composition of some medicinal plants.

The use of and search for drugs and dietary supplements derived from plants have accelerated in recent years. Ethnopharmacologists, botanists, microbiologists and natural-products scientists are combing the earth for phytochemicals and leads, which could be developed for treatment of infectious diseases. The aim of this study was to investigate the antifungal activities of the essential oils of some medicinal plants such as Stachys pubescens, Thymus kotschyanus, Thymus daenensis and Bupleurum falcatum against Fusarium oxysporum, Aspergillus flavus and Alternaria alternata. The essential oils w

Diagnostic accuracy for drug detection using liquid chromatography/mass spectroscopy in overdose patients.

AIM: Information about the causative drugs is essential for appropriate treatment for drug overdose, but patients sometimes cannot provide information about overdosed drugs owing to disturbed consciousness or an unwillingness to cooperate with treatment. The purpose of this study was to decide whether liquid chromatography/mass spectroscopy (LC/MS) is useful as a detection method for overdosed drugs. METHODS: Overdose patients (n = 279) treated in our facility were retrospectively studied. Specimens from gastric lavage, blood serum, and urine were tested using LC/MS. The matching rates betwee

Aprepitant, a Neurokinin-1 Receptor Antagonist, as a Disruptive Drug for the Treatment of Pediatric Cancer.

Although advances in pediatric oncology have resulted in cure rates exceeding 80% among children with cancer, progress in survival outcomes has begun to plateau in recent years. Mortality in this population remains largely associated with aggressive disease features, particularly tumor resistance to chemotherapy and metastatic spread. At the same time, the growing population of childhood cancer survivors has drawn increasing attention to the persistent and potentially serious late effects associated with conventional chemotherapy. Thus, it is crucial to discover drugs with specific antitumor a

Advancement in peptide-based therapeutics for the treatment of type 2 diabetes mellitus: current progress and future prospects.

Diabetes is a chronic medical disorder caused by insufficient production of the hormone insulin by the pancreas. Although there are various treatment options available for controlling diabetes, including non-peptide-based medications, the majority of these have adverse effects and are limited in comparison to peptide-based drugs. Protein drugs offer numerous benefits, including weight loss, significant reductions in blood glucose levels, and an extremely low risk of hypoglycemia. This article discusses treatment modalities, presents existing therapies, provides an in-depth comparison of peptid

Profiling of the effects of antifungal agents on yeast cells based on morphometric analysis.

The incidence of fungal infection and evolution of multidrug resistance have increased the need for new antifungal agents. To gain further insight into the development of antifungal drugs, the phenotypic profiles of currently available antifungal agents of three classes-ergosterol, cell wall and nucleic acid biosynthesis inhibitors-were investigated using yeast morphology as a chemogenomic signature. The comparison of drug-induced morphological changes with the deletion of 4718 non-essential genes not only confirmed the mode of action of the drugs but also revealed an unexpected connection amo

Blood viscosity and platelet function in thrombolytic therapy of acute myocardial infarction.

The mechanisms by which thrombolytic drugs accomplish their impressive clinical effects in the treatment of acute myocardial infarction are as yet incompletely appreciated. Factors other than lysis of thrombi and restoration of blood flow most likely play a role, and blood viscosity and platelet function probably are important factors. Blood viscosity is inversely related to oxygen supply to myocardial tissue and therefore a decrease in viscosity might contribute to preservation of myocardial function. Since fibrinogen is a major determinant of blood viscosity and this protein is largely degra

Heat illness: tips for recognition and treatment.

Heat stroke, an acute, life-threatening emergency, results from an overload or impairment of heat-dissipating mechanisms. At risk are the elderly, infants, the obese, people with hyperthyroidism, and those taking certain drugs. Early recognition and rapid cooling are essential--the more rapid the cooling, the lower the mortality.

A Perspective on Evaluating Life Stage Differences in Drug Dosages for Drug Labeling and Instructions.

Drug labeling and instructions provide essential information for patients regarding the usage of drugs. Instructions for the dosage of drug usage are critical for the effectiveness of the drug and the safety of patients. The dosage of many drugs varies depending on the patient's age. However, as our understanding of human biology deepens, we believe that these instructions need to be modified to incorporate different life stages. This is because human biology and metabolism differ significantly among different life stages, and their responses to drugs also vary. Additionally, the same age of d

[The role of CYP2C19 polymorphism in the development of adverse effects to drugs and the risk for diseases].

There are a great number of polymorphic genes in the human genome. Many of them codify enzymes that metabolizes drugs and xenobiotic agents, including carcinogens. Among the better known of them, there are a number of isozymes of the microsomal oxidative system (CYP3A4, CYP2C9, CYP2C19 y CYP2D6). This article reviews the following issues: a) frequency of presentation of the "poor metabolizer" genotype and/or phenotype for substrates of CYP2C19; b) role of CYP2C19 polymorphism on the metabolism of some drugs (mephenytoine and other antiepileptic drugs, proton pump inhibitors, several antidepres
