Multiple-modified Low-Density Lipoprotein as Atherogenic Factor of Patients' Blood: Development of Therapeutic Approaches to Reduce Blood Atherogenicity.

Generally, atherosclerosis first occurs by the way of accumulation of intracellular and extracellular lipids in the arterial intima. Foam cells, overloaded by lipids, are the essential harbinger of the coronary artery disease. It should be noted that lipids that are usually composed of bulk of the intracellular lipids found in human arterial cells originate from low-density lipoprotein (LDL) circulating in human blood. Nonetheless, many efforts to force cells to accumulate cholesteryl esters under the influence of native LDL have been unsuccessful. Whilst LDL modified in vitro (exposed to malo…

Nutritional control, gene regulation, and transformation of vascular smooth muscle cells in atherosclerosis.

Contractile-state smooth muscle cells (SMC), the only cell type in the arterial media, undergoes migration to the intima, proliferation, and abundant extracellular matrix production during the early stages of atherosclerosis. This involves the ingestion of low-density lipoprotein (LDL) and modified or oxidised LDL by macrophages together with SMC by several pathways including a scavenger pathway leading to accumulation of cholesterol esters and formation of foam cells. High-plasma cholesterol levels constitute a major causative risk for atherosclerosis. The membrane-bound transcription factor …

Evaluation of interface pressure and temperature management in five wheelchair seat cushions and their effects on user satisfaction.

BACKGROUND: Factors such as the manufacturing materials, shape or even the mechanical and thermal response of sitting Pressure Redistribution Support Surfaces (PRSS) can be potential contributors to pressure ulcers. However, few studies have compared a number of characteristics of the most frequently used devices. OBJECTIVE: To compare three potential contributors to pressure ulcers in five commercial PRSS: pressure redistribution, temperature and perceived comfort. METHOD: Study with a cross-over randomized design in healthy volunteer participants. Data was collected in a temperature and re

Macrophage iron retention aggravates atherosclerosis: Evidence for the role of autocrine formation of hepcidin in plaque macrophages.

Iron accumulation has been frequently found in atherosclerotic lesions, especially in macrophages/foam cells, but the exact mechanisms by which hepcidin induces iron retention in plaque macrophages and its roles in atherogenesis remain unknown. Double immunofluorescence staining showed colocalization of hepcidin-positive macrophages with ox-LDL, TLR4, p-p65 and ferritin light chain (ferritin-L) both in human and murine atherosclerotic lesions. RAW264.7 macrophages incubated with ox-LDL showed elevated expression of TLR4, p-p65, hepcidin, ferritin-L/H, CYP27A1, CD36, PPARγ, liver X receptor α (…

Femoral cementing technique for hip resurfacing arthroplasty.

The resurgence of metal-metal bearings has renewed interest in hip resurfacing, but a paucity of information exists regarding femoral cementing technique. We developed a laboratory model in which 72 open-cell foam specimens were used to simulate bone. Analyses of two cement viscosities, two foam porosities, and six cementing techniques were performed: manual cement application only, manual application and filling of one quarter of the component with cement, filling of half of the component, manual application and half component filling, full component filling, and manual application and full c

Niemann-Pick Disease Type B in a 21 Year Old Male.

Niemann-pick disease is a group of autosomal recessive disorder of lipid storage with progressive accumulation of sphingomyelin and other lipids in the lysosomes of various tissues. We are reporting a 21 year old male who had hepatosplenomegaly, cherry red macula and normal cognitive function. Bone marrow biopsy showed plenty of foam cells and sphingomyelinase level was low, thus conforming our diagnosis. Survival into adulthood and absence of gross neurological involvement suggests Niemann-Pick disease type B.

Facile synthesis of macrocellular mesoporous foamlike Ce-Sn mixed oxides with a nanocrystalline framework by using triblock copolymer as the single template.

Macrocellular mesoporous foamlike cerium-tin mixed oxide materials with well-defined porous structure and nanocrystalline frameworks are synthesized through a simple one-step self-assembly process using an amphiphilic triblock copolymer as the single template. The macrocellular pores are synthesized without the addition of any swelling agent or hazardous acids. The final mixed oxide possesses a hierarchically porous structure including macrocellular foam with ultralarge cell size, closed windows, and mesopores on the walls. This indicates that the porous structure can be notably stabilized and

Mesenteric panniculitis of the sigmoid colon. Report of two cases.

Mesenteric panniculitis is an extremely rare disease in which the normal fatty architecture of the mesentery is replaced by fibrosis, necrosis, and calcification. Grossly, the mesentery is massively thickened and rubbery, with irregular areas of reddish-brown to pale yellow foci resembling fat necrosis scattered throughout. Microscopically, inflammatory involvement of the fibroadipose tissue shows round cells, foam cells, and giant cells. Macrophages that have ingested fat, also called lipophages, are the hallmark of the diagnosis. One hundred twenty-two cases of mesenteric panniculitis have b…

Exploring the Role of ADAMTS4 Expression in Atherosclerosis.

ADAMTS4 is a secretory metalloproteinase involved in the pathogenesis of atherosclerosis. Both in vitro and in vivo model systems are used to study the regulation of ADAMTS4 in atherosclerosis. Here we describe three model systems: in vitro model system of macrophages and foam cells and in vivo model of high-fat-diet-induced atherosclerosis. We can use qPCR for studying the mRNA expression of ADAMTS4, while ELISA and Western blot can be employed to assess its protein expression.

Endogenous lipid pneumonia in opossums from Louisiana.

Endogenous lipid pneumonia was present in 19 of 27 opossums (Didelphis virginiana) trapped in the vicinity of Baton Rouge, Louisiana. The severity of lesions varied from small pleural and subpleural aggregates of foamy macrophages with minimal disruption of pulmonary architecture to large nodular accumulations of foam cells with cholesterol clefts and localized emphysema. The cause of the lipid pneumonia may be related to pulmonary nematode parasite infections, which were evident in 13 of the affected animals.

Mycosis fungoides associated with dystrophic xanthomatosis.

A 59-year-old man with no evidence or history of hyperlipidemia had a dystrophic xanthoma, an accumulation of lipid-rich foam cells in an area of damaged or abnormal skin, that was associated with untreated mycosis fungoides (MF). The patient was subsequently treated with whole-body electron beam therapy. This association of MF and dystrophic xanthoma illustrates histiocyte and xanthoma cell interaction in MF, a cutaneous T-cell lymphoma. Xanthomas may be a cutaneous sign of an underlying lymphoproliferative or myeloproliferative disease in normolipemic patients.

[Statins and antiplatelet agents: complementary mechanisms of action at the coronary artery level].

Atherothrombotic lesions associate atherosclerosis with a thrombotic reaction; they result from a complex inflammatory response to multifactorial stress. Endothelial dysfunction, lipid accumulation in the arterial intima, the recruitment of circulating monocytes and differentiation into macrophages and then foam cells, and an inflammatory reaction, all participate in the genesis of atheromatous plaque. The lipid core and fibrous parts of the plaque then contribute to its progression towards stenosis as a result of lipid infiltration, an accumulation of foam cells and the proliferation of smoot…

Macrophage polarization as a potential therapeutic target for atherosclerosis: a dynamic stochastic modelling study.

We proposed a dynamic stochastic mathematical model to evaluate the role of macrophage polarization in plaque development. The dynamic proce … of oxidized low-density lipoprotein and phagocytic effects of macrophages governed the formation of foam cells. Furthermore, we simulated targeted therapies by either directly inhibiting the polarization probability to M1 macrophages or indirectly regulating macrophage polarization due to high-density lipoprotein levels. Comparison of simulation results with …

Nephrotic syndrome caused by immune-mediated acquired LCAT deficiency.

Lecithin-cholesterol acyltransferase (LCAT) is an enzyme involved in maintaining cholesterol homeostasis. In familial LCAT deficiency (FLD), abnormal lipid deposition causes renal injury and nephrotic syndrome, frequently progressing to ESRD. Here, we describe a 63-year-old Japanese woman with no family history of renal disease who presented with nephrotic syndrome. The laboratory data revealed an extremely low level of serum HDL and undetectable serum LCAT activity. Renal biopsy showed glomerular lipid deposition with prominent accumulation of foam cells, similar to the histologic findings of…
