Discovery of Potential Anthelmintic Agents Against Gyrodactylus kobayashii Through Computer-Aided Drug Design and In Vivo Evaluation.

Monogeneans are ectoparasitic flatworms causing significant economic losses in aquaculture. This study aimed to identify potential anthelmintic agents against these parasites by integrating computer-aided drug design (CADD) and in vivo evaluation. The β-tubulin gene, a well-established anthelmintic target, was cloned from Gyrodactylus kobayashii and its three-dimensional structure was generated using homology modelling. Virtual screening of 2319 FDA-approved drugs and nine common benzimidazoles against the modelled β-tubulin identified several promising compounds with low binding energy. Subse

Genome-Wide Protein Interaction Analysis in Parasitic Gyrodactylus Flatworms-Fish Hosts System and Drug Target Identification.

The host-parasite arms race involves complex molecular crosstalk mediated by protein-protein interactions (PPIs). Bioinformatic analyses can be used to predict both host-parasite PPIs and potential drug targets in parasite genomes, but high-quality genomic data remain scarce for parasitic monogenean flatworms. Herein, an experimental lineage of Gyrodactylus kobayashii (Monopisthocotylea: Gyrodactylidae) is set up on goldfish hosts and used to conduct phased genome assembly using long-range PacBio HiFi and Hi-C technologies. In silico analyses of genomes of three Gyrodactylus species identified

Khawia abbottinae sp. n. (Cestoda: Caryophyllidea) from the Chinese false gudgeon Abbottina rivularis (Cyprinidae: Gobioninae) in China: morphological and molecular data.

Khawia abbottinae sp. n. is described from the Chinese false gudgeon, Abbottina rivularis (Basilewsky) (Cyprinidae: Gobioninae), from the Yangtze River basin in China. The new species can be distinguished from the congeneric species mainly by the arrangements of the testes, which form two longitudinal bands (other congeneric species have the testes irregularly scattered throughout the testicular region) and their number (at maximum 85 testes versus at least 160 in the other Khawia spp.), and the morphology of the scolex, which varies from cuneiform to widely bulbate scolex, being separated fro

LPXRFamide peptide stimulates growth hormone and prolactin gene expression during the spawning period in the grass puffer, a semi-lunar synchronized spawner.

Gonadotropin-inhibitory hormone (GnIH) plays as a multifunctional neurohormone that controls reproduction in birds and mammals. LPXRFamide (LPXRFa) peptide, the fish ortholog of GnIH, has been shown to regulate the secretion of not only gonadotropin (GTH) but also growth hormone (GH) and prolactin (PRL), which are potentially important for gonadal function. To investigate the role of LPXRFa peptide on reproduction of the grass puffer, which spawns in semilunar cycles, we examined changes in the levels of gh and prl expression over the several months during the reproductive cycle, and the effec

Effect of ozonation on the naphthenic acids' speciation and toxicity of pH-dependent organic extracts of oil sands process-affected water.

The presence of naphthenic acids (NAs) and other organic constituents in oil sands process-affected water (OSPW) stored in tailings ponds, poses a serious environmental threat due to their potential toxicity to aquatic organisms and wild life. In this work, four fractions of OSPW, extracted by dichloromethane at different pHs, were ozonated to determine the ozone impact on NAs degradation. Extracts distributions showed that high carbon number NAs (14-22) were associated with higher pH fractions (pH>7) and smaller carbon number NAs (7-13) with lower pH fractions (pH≤7). Extracts showed similar

Molecular characterization of cholecystokinin in grass carp (Ctenopharyngodon idellus): cloning, localization, developmental profile, and effect of fasting and refeeding on expression in the brain and intestine.

Cholecystokinin (CCK) is a multi-functional brain-gut peptide in fish and mammals. To investigate the role of CCK in appetite regulation in fish, a 770-bp full-length cDNA sequence of CCK gene was obtained by RT-PCR and rapid amplification of cDNA ends methods in grass carp Ctenopharyngodon idellus. Homology analysis showed that the CCK cDNA sequence of grass carp had the highest similarity (90 %) to that of goldfish Carassius auratus and a higher similarity (>70 %) to those of other teleosts than to mammals. The PCR amplification using genomic DNA identified that the CCK gene of grass carp wa

Proton-mediated feedback inhibition of presynaptic calcium channels at the cone photoreceptor synapse.

Generation of center-surround antagonistic receptive fields in the outer retina occurs via inhibitory feedback modulation of presynaptic voltage-gated calcium channels in cone photoreceptor synaptic terminals. Both conventional and unconventional neurotransmitters, as well as an ephaptic effect, have been proposed, but the intercellular messaging that mediates the inhibitory feedback signal from postsynaptic horizontal cells (HCs) to cones remains unknown. We examined the possibility that proton concentration in the synaptic cleft is regulated by HCs and that it carries the feedback signal to

Psychophysics and neurophysiology of repetition noise processing in a vertebrate auditory system.

The psychophysics and neurophysiology of repetition noise (RN) processing was studied in the goldfish. RN is the sum of a noise waveform with its delayed (by T s) repetition, which may be attenuated (by A dB), and inverted relative to the undelayed signal. Such a signal has a periodic spectrum with peaks separated by 1/T Hz, and a prominence in its autocorrelation function at T s. In usual environments, RN contains information about sound-reflecting surfaces. Delays in the range of 0.5-20 ms create pitch sensations in man. Psychophysical experiments using classical respiratory conditioning inv

A novel lanthanide metal-organic framework hydrogel for "turn-on" sensing of norfloxacin antibiotics in carassius auratu and milk.

Utilizing the "turn-on" emission sensing mode, the sensor platform for norfloxacin (NOR) detection was developed by utilizing the lanthanide metal-organic framework, Eu-MOF. The Eu-MOF exhibits high specificity and anti-resistance to interference in cognizing recognition of NOR. The results show that the Eu-MOF exhibits high efficacy and precision in norfloxacin detection, with a fluorescence detection limit of 0.56 μM. Additionally, a visual detection "turn-on" emission platform, Eu-MOF@PVA/SA, was successfully established to facilitate the detection of NOR. This platform utilizes a smartphon

Decreased pathogenicity of triple-mutant of Aeromonas hydrophila flgK, flgL, flgE suggests its potential as a live attenuated vaccine for Carassius auratus.

Aeromonas hydrophila is one of the top pathogenic bacteria that causes serious economic losses in aquaculture. In this study, we constructed a live attenuated vaccine (LAV) candidate strain by mutating the flagella genes flgK, flgL, and flgE in A. hydrophila (AH-ΔKLE). This strain exhibited sufficient attenuation with a significant reduction of 35.91-fold in pathogenicity. Compared to wild type strain of A. hydrophila (AH-WT), AH-ΔKLE exhibited flagellar loss, reduced motility, diminished hemolytic activity, decreased biofilm formation, and down-regulation of pathogenicity-related genes. 144 h

Diving deep into fish bornaviruses: Uncovering hidden diversity and transcriptional strategies through comprehensive data mining.

Recently, we discovered two novel orthobornaviruses in colubrid and viperid snakes using an in silico data-mining approach. Here, we present the results of a screening of more than 100,000 nucleic acid sequence datasets of fish samples from the Sequence Read Archive (SRA) for potential bornaviral sequences. We discovered the potentially complete genomes of seven bornavirids in datasets from osteichthyans and chondrichthyans. Four of these are likely to represent novel species within the genus Cultervirus, and we propose that one genome represents a novel genus within the family of Bornaviridae

Cytoskeletal Arrest: An Anoxia Tolerance Mechanism.

Polymerization of actin filaments and microtubules constitutes a ubiquitous demand for cellular adenosine-5'-triphosphate (ATP) and guanosine-5'-triphosphate (GTP). In anoxia-tolerant animals, ATP consumption is minimized during overwintering conditions, but little is known about the role of cell structure in anoxia tolerance. Studies of overwintering mammals have revealed that microtubule stability in neurites is reduced at low temperature, resulting in withdrawal of neurites and reduced abundance of excitatory synapses. Literature for turtles is consistent with a similar downregulation of pe

Purkinje cell responses during visually and vestibularly driven smooth eye movements in mice.

INTRODUCTION: An essential complement to molecular-genetic approaches for analyzing the function of the oculomotor circuitry in mice is an understanding of sensory and motor signal processing in the circuit. Although there has been extensive analysis of the signals carried by neurons in the oculomotor circuits of species, such as monkeys, rabbits and goldfish, relatively little in vivo physiology has been done in the oculomotor circuitry of mice. We analyzed the contribution of vestibular and nonvestibular signals to the responses of individual Purkinje cells in the cerebellar flocculus of mic

Histopathological findings on Carassius auratus hepatopancreas upon exposure to acrylamide: correlation with genotoxicity and metabolic alterations.

Acrylamide is an amide used in several industrial applications making it easily discharged to aquatic ecosystems. The toxicity of acrylamide to aquatic organisms is scarcely known, although previous studies with murine models provided evidence for deleterious effects. To assess the effects of acrylamide to freshwater fish, goldfish (Carassius auratus L.) were exposed to several concentrations of waterborne acrylamide and analysed for genotoxic damage, alterations to detoxifying enzymes and histopathology. Results revealed a dose-dependent increase in total DNA strand breakage, the formation of

Restricted expression of the neuronal intermediate filament protein plasticin during zebrafish development.

In the adult goldfish visual pathway, expression of the neuronal intermediate filament (nIF) protein plasticin is restricted to differentiating retinal ganglion cells (RGCs) at the margin of the retina. Following optic nerve injury, plasticin expression is elevated transiently in all RGCs coincident with the early stages of axon regeneration. These results suggest that plasticin may be expressed throughout the nervous system during the early stages of axonogenesis. To test this hypothesis, we analyzed plasticin expression during zebrafish (Danio rerio) neuronal development. By using immunocyto

Chronic fluoride exposure induces tissue damage, oxidative stress, and immune dysfunction in juvenile crucian carp (Carassius auratus).

Fluoride pollution in aquatic ecosystems poses substantial risks to aquatic organisms. However, most previous studies have focused on acute toxicity and skeletal fluorosis, whereas the chronic toxic effects of fluoride on non-skeletal tissues in fish remain poorly understood. In particular, the mechanisms underlying fluoride-induced toxicity, especially those associated with immune dysfunction and intestinal microbiota disruption, have yet to be fully elucidated. In the present study, juvenile crucian carp (Carassius auratus) were used as an experimental model to investigate the chronic toxic

Differential distribution of potassium channels in acutely demyelinated, primary-auditory neurons in vitro.

1. Single-channel recordings of potassium channel activity were made from two populations of primary-auditory neurons maintained in tissue culture. The saccular nerve, which is the auditory component of the eighth cranial nerve in goldfish, was separated into two branches according to its peripheral innervation pattern. Neurons which innervated the rostral saccular macula corresponded to a class of cells that showed spike frequency adaptation; whereas, neurons which innervated the caudal macula were consistent with another type of cell that demonstrated bursting spontaneous firing patterns in

Development of a rapid and visual ERA-LFD assay for sensitive detection of Ichthyophthirius multifiliis with potential for application in aquaculture.

Ichthyophthirius multifiliis is a highly pathogenic ciliate responsible for "white spot disease" in freshwater fish, causing significant economic losses to the global aquaculture industry. Early and accurate detection of this parasite is critical for disease control, however, current methods often require specialized equipment and trained personnel, limiting their applicability in resource-limited settings. In this study, a rapid, sensitive, and visual detection method for I. multifiliis was developed by combining enzymatic recombinase amplification (ERA) with lateral flow dipstick (LFD) techn
