Membrane bound glycogen storage in muscle is characteristic for the lysosomal storage disorder acid maltase (acid alpha-glucosidase) deficiency while in phosphofructokinase and phosphorylase deficiency, glycogen is stored free in the cytoplasm. Using immunohistochemistry, we examined whether acid maltase deficiency had an effect on early endosomes, recycling endosomes and trans-Golgi network, vesicle systems linked to lysosomes. Vacuolated glycogen containing fibres stained intensely for the lysosomal marker lysosomal-membrane-protein-1 within fibres and at the sarcolemma. There was a similar

The vast majority of well absorbed drugs are transported passively across the cell membranes. Physicochemical descriptors of drug molecules that are believed to influence transcellular transport are routinely used to predict drug absorption by means of complex mathematical models. In this paper, a new in vitro method, based on the retention data in micellar liquid chromatography (MLC), is validated for the prediction of passive drug absorption. The retention of a heterogeneous drugs set in MLC using Brij 35 as surfactant in the mobile phase is compared with the retention data reported in liter

The vesicular monoamine transporter in the brain can sequester the neurotoxin 1-methyl-4-phenylpyridinium into synaptic vesicles and protect catecholamine-containing neurons from degeneration. Mouse nigrostriatal dopaminergic neurons, and to a lesser extent locus coeruleus noradrenergic neurons, are vulnerable to toxicity produced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine. The present study sought to determine whether pharmacological inactivation of the vesicular monoamine transporter in the brain would enhance the degeneration of substantia nigra dopaminergic neurons and locus coeruleus

The movement of key transition metal ions is recognized to be of critical importance in the interaction between macrophages and intracellular pathogens. The present study investigated the role of copper in mouse macrophage responses to Salmonella enterica sv. Typhimurium. The copper chelator BCS (bathocuproinedisulfonic acid, disodium salt) increased intracellular survival of S. Typhimurium within primary mouse BMM (bone-marrow-derived macrophages) at 24 h post-infection, implying that copper contributed to effective host defence against this pathogen. Infection of BMM with S. Typhimurium or t

Calcium plays a regulatory role in several aspects of protein trafficking in the cell. Both vesicle fusion and vesicle formation can be inhibited by the addition of calcium chelators. Because the effects of calcium chelators have been studied predominantly in cell-free systems, it is not clear exactly which transport steps in the secretory pathway are sensitive to calcium levels. In this regard, we have studied the effects of calcium chelators on both anterograde and retrograde protein transport in whole cells. Using both cytochemical and biochemical analyses, we find that the anterograde-dire

The utilization of cell-free therapies derived from extracellular vesicles (EVs) has garnered mounting interest as a promising approach to address the myriad challenges associated with ischemic stroke. These Mesenchymal stem cell-derived extracellular vesicles (MSC-EVs) possess considerable therapeutic potential due to their inherent properties, including low immunogenicity, efficient cargo transportation, and the ability to cross the blood-brain barrier. This review examines the mechanisms underlying mesenchymal stem cell-derived EVs in the treatment of ischemic stroke. Future research should

By means of delta pH 14C-methylamine indicator the myometrium vesicle sarcolemma fraction was shown to be capable, while applying a "delta pH-leap", for developing in it a proton transmembrane gradient, dissipating in time. The proton gradient dissipation under Ca ions transmembrane equilibrium concentration is a driving force of these ions transposition against the concentration gradient. The blocking agents of H+ transport--Cd ions and DCCD decrease the proton-dependent 45Ca2+ accumulation in the vesicle sarcolemma fraction. The conclusion has been made about the possibility of Ca2+(H(+)-exc

Histochemical studies on tissue sections showed alterations of lectin-binding reactivities in the epididymis, seminal vesicle and ampulla of Brucella ovis-infected rams. These modifications in the carbohydrate composition of organs participating in maturation, transport, and storage of spermatozoa, could be involved in the impaired fertility observed in this disease.

Outer membrane vesicles (OMVs) have been increasingly recognized as common mediators of bacterial physiology in Gram-negative bacteria, including Vibrio species. The degree and function of OMV production can differ among strains and even within a single species. The secretion of OMVs is a prevalent trait among many Vibrio species, particularly in pathogenic organisms such as Vibrio cholerae, Vibrio vulnificus, and Vibrio parahaemolyticus. The OMVs released by these organisms are often associated with infection, transport of virulence factors into host cells, defense against stress, biofilm for

When cultured in broth to the transmissive phase, Legionella pneumophila infects macrophages by inhibiting phagosome maturation, whereas replicative-phase cells are transported to the lysosomes. Here we report that the ability of L. pneumophila to inhibit phagosome-lysosome fusion correlated with developmentally regulated modifications of the pathogen's surface, as judged by its lipopolysaccharide profile and by its binding to a sialic acid-specific lectin and to the hydrocarbon hexadecane. Likewise, the composition of membrane vesicles shed by L. pneumophila was developmentally regulated, bas

Mutations in the human gene encoding presenilin-1, PS1, account for most cases of early-onset familial Alzheimer's disease. PS1 has nine transmembrane domains and a large loop orientated towards the cytoplasm. PS1 locates to cellular compartments as endoplasmic reticulum (ER), Golgi apparatus, vesicular structures, and plasma membrane, and is an integral member of gamma-secretase, a protein protease complex with specificity for intra-membranous cleavage of substrates such as beta-amyloid precursor protein. Here, an interaction between PS1 and the Sec13 protein is described. Sec13 takes part in

Neurodegenerative diseases, including Alzheimer's disease, Parkinson's disease, amyotrophic lateral sclerosis, and Huntington's disease are among the progressive disorders of the nervous system that are characterized by the gradual destruction of neurons, the accumulation of misfolded proteins, and the limited effective therapeutic options. In recent years, numerous lines of evidence have emphasized the important role of extracellular vesicles (EVs) in the formation and progression of these diseases. These vesicles are membrane-bound nanoscale structures that are secreted by almost all cell ty

BACKGROUND: Plant extracellular vesicles (PEVs) have emerged important roles in central nervous system (CNS) disorders. PEVs are nanoscale vesicles (30-150 nm) that mediate intercellular communication and exhibit unique therapeutic potential due to their natural biocompatibility, minimal immunogenicity, and ability to cross the blood-brain barrier (BBB). With increasing interest in neurotherapeutics, PEVs offer promising applications for CNS disorders by overcoming delivery barriers and reducing adverse effects associated with synthetic nanoparticles.

The binding and transport of glycoalbumin (gA) by the endothelium of murine myocardial microvessels were studied by perfusing in situ 125I-gA or gA-gold complexes (gA-Au) and examining the specimens by radioassays and EM, respectively. After a 3-min perfusion, the uptake of radioiodinated gA is 2.2-fold higher than that of native albumin; it is partially (approximately 55%) competed by either albumin or D-glucose, and almost completely abolished by the concomitant administration of both competitors or by gA. D-mannose and D-galactose are not effective competitors. Unlike albumin-gold complexes

A rapid, simple and reproducible method has been developed for the measurement of ion tracer flux with both native membrane vesicles and reconstituted lipid vesicle systems. Following the absorption of vesicles onto glass fiber filters, tracer flux is performed directly upon the deposited samples. In contrast to the more conventional vacuum and gel filtration techniques, absorption filtration exhibits comprehensive data retrieval whereby the removal of extravesicular ions, the retention of intravesicular ions and the amount of ions fluxed can be accurately analyzed. Both influx and efflux assa

BACKGROUND: The cause of sporadic amyotrophic lateral sclerosis (ALS) is largely unknown but hypotheses about disease mechanisms include oxidative stress, defective axonal transport, mitochondrial dysfunction and disrupted RNA processing. Whereas familial ALS is well represented by transgenic mutant SOD1 mouse models, the mouse mutant wobbler (WR) develops progressive motor neuron degeneration due to a point mutation in the Vps54 gene, and provides an animal model for sporadic ALS. VPS54 protein as a component of a protein complex is involved in vesicular Golgi trafficking; impaired vesicle tr

Multidrug-resistant bacteria (MDRB) have become a global health crisis that challenges the effectiveness of conventional antibiotics. Bacterial extracellular vesicles (BEVs) are nanoscale bilayered membrane vesicles secreted by both Gram-positive and Gram-negative bacteria. They can encapsulate proteins, lipids, and nucleic acids, and transfer these molecules between bacteria and host cells without direct contact. Owing to their natural ability to transport bioactive molecules, BEVs have recently gained attention as potential anti-infective platforms. They can deliver antimicrobial agents dire

Hypertensive encephalopathy was induced in the rat by clipping one renal artery and contralateral nephrectomy. The possible changes of vascular permeability of the cerebral blood capillaries and venules were investigated by using ferritin as a tracer. The uninephrectomized rats served as controls. In controls, ferritin was never seen in the basement membranes, within plasmalemmal vesicles on the basal surface of the endothelium, or in endothelial cell junctions of cerebral capillaries and venules up to 180 min after the injection. In venules of the brain in rats with hypertensive encephalopath

17alpha-Ethinylestradiol (EE2), a component of oral contraceptives, is known to undergo considerable first-pass 3-O-sulfation in the intestine and liver. Once formed, the 3-O-sulfate conjugate (EE2-Sul) is detected in circulation at appreciable levels (versus parent EE2) and is present in bile. Therefore, hepatic uptake of EE2-Sul was assessed with suspensions of cryopreserved human primary hepatocytes. In this instance, there was evidence for active (temperature-dependent) uptake, which was described by a two-K(m) (Michaelis constant) model (K(m1) = 220 nM; K(m2) = 15.5 microM). Uptake was in

Extracellular vesicles (EVs) are now recognized as key mediators of intercellular communication, transferring cargos such as proteins, lipids, and nucleic acids between cells and thereby influencing physiological and pathological processes. While the role of EVs has been extensively investigated in physiological and pathophysiological contexts, nutrition has only recently been recognized as an important modulator of endogenous EV biology. Emerging evidence indicates that nutrition and food bioactive compounds can influence EV biology through three distinct mechanisms: (i) modulation of the bio
