Albumin competitively inhibits glycation of less abundant proteins.

Glycation, a non-enzymatic reaction between glucose and protein is the primary cause of diabetic complications. Albumin, the most abundant plasma protein undergoes glycation both in vivo and in vitro. The influence of albumin on glycation of less abundant proteins has not been addressed. For the first time, we show that albumin competitively inhibits the glycation of less abundant proteins. This study suggests that at least in the initial stages of diabetes, albumin may protect other proteins from glycation.

[Ischemia modified albumin--specific marker in cardiological diagnostics?].

Ischemia modified albumin (IMA) is a new biological marker for early identification of chest pain and ruling out myocardial infarction among patients with acute syndromes submitting to emergency department. Recently IMA has been investigated in the light of other cardiac markers (cTnT, CK-MBmas, NT-proBNP) in various states of ischemia (acute coronary syndromes, after percutaneous coronary intervention, in coronary vasospasm). Ischemia modified albumin levels were elevated in these states what suggests myocardial ischemia. However decrease in IMA concentration after exercise-induced skeletal m

[Conformational disturbances of serum albumin binding sites in schizophrenia].

The properties of serum albumin binding sites were studied using quenching of fluorescence of the molecular probe CAPIDAN (N-carboxyphenylimide of dimethylaminonaphthalic acid) with the nitrate anion. The samples of serum were obtained from 24 patients with paranoid schizophrenia and 24 healthy volunteers. In the absence of quencher the specific probe fluorescence was 1,4 times higher in patients than in volunteers. Fluorescence quenching constant for the probe bound to albumin was (M+/-m) 2,48+/-0,17 l/mol in patients versus 4,65+/-0,37 l/mol (p<0,01) in volunteers (p<0,01). The fluorescence

Effect of intravenous albumin infusion on brain salicylate concentration.

BACKGROUND: Salicylate poisoning appears to result in death, despite supportive care, once a critical brain salicylate concentration is reached. The binding of salicylate to albumin is saturable; free plasma salicylate concentrations rise disproportionately to total drug levels. Because unbound salicylate distributes into the brain, the authors questioned whether an intravenous (i.v.) infusion of albumin would cause a redistribution of salicylate from the brain back into the plasma, which might allow enough time for hemodialysis to be instituted. OBJECTIVES: To determine if i.v. albumin infus

[Microalbuminuria and urinary albumin excretion: clinical practice guidelines].

UNLABELLED: Measurement of urinary albumin excretion (UAE) may be done on a morning urinary sample or on a 24 hours-urine sample. Values defining microalbuminuria are: 24 hour-urine sample: 30-300 mg/24 hours; morning urine sample: 20-200 mg/ml or 30-300 mg/g creatinine or 2.5-25 mg/mmol creatinine (men) or 3.5-35 mg/mol (women). Timed urine sample: 20-200 microg/min. The optimal use of semi-quantitative urine test-strip is not clearly defined. It is generally believed that microalbuminuria reflects a generalized impairment of the endothelium; however, no definite proof has been shown in human

An albumin-fixed membrane for the removal of protein-bound toxins.

Established methods for kidney dialysis do not work for liver failure because kidney dialysis removes only water-soluble toxins, while the liver normally removes albumin-bound toxins. In the present study, a polysulfone dialysis membrane with a -OH reactive group was prepared by hydrolyzing the chloromethylated polysulfone membrane, and the bovine serum albumin molecules were fixed into the membrane with 1,1'-carbonyldiimidazole activation. The content of albumin of the albumin-fixed membrane was 121.3 mg (g membrane)(-1). The albumin-fixed dialysis membranes were used to remove protein-bound

A novel method for simultaneous purification of albumin and immunoglobulin G.

A novel method was developed to obtain both highly purified bovine serum albumin (BSA) and immunoglobulin G (IgG), at the same time, on a pilot scale. Heat-isopropyl alcohol was used to denature and precipitate the other plasma proteins, except for BSA and IgG; then, CM-Trisacryl was applied to further purify and isolate BSA and IgG. The new procedure produced highly purified BSA and IgG, 98% and 96.8%, respectively, and yielded ideal output, 2.18% and 0.54%, from starting plasma, respectively. The new technique is a rapid and is an available pilot process to prepare the plasma fractions devoi

Effects of albumin supplementation on microvascular permeability in septic patients.

Albumin has a stabilizing effect on endothelium and helps maintain capillary permeability to macromolecules. Critically ill patients with sepsis may have profound hypoalbuminemia, but the effect of this hypoalbuminemia on microvascular permeability is unknown. To determine the degree and potential importance of this effect, we measured the transcapillary escape rate (TER) of (125)I-labeled albumin in 12 adult patients fulfilling American College of Chest Physicians/Society of Critical Care Medicine criteria for septic shock. We measured TER over a 90-min baseline period and then repeated these

Poly(ethylene glycol): protein-repulsive or albumin-compatible?

In the literature, many papers deal with the behavior of proteins in aqueous media in the presence of poly(ethylene glycol) (PEG) molecules or poly(ethylene oxide) (PEO) segments, physically adsorbed onto, or covalently attached to, macromolecules or to solid surfaces. In particular, it is well known that PEO segments make foreign materials stealthy, i.e. they are much less detected by the immune system either through humoral reactions or, at the cell level, through opsonins. Revisiting the literature led us to challenge the largely accepted opinion that the decreased recognition of PEO segmen

Effects of physiologic albumin and hespan levels on hepatocytes in vitro.

BACKGROUND: Although albumin and hydroxyethyl starch (HES) are routinely used in critically ill, hypoalbuminemic patients, no studies have tested the effect of supplemental albumin and HES on hepatocyte function. METHODS: In this study, the effects of these agents were evaluated by using stable, rat hepatocyte cultures in a collagen sandwich configuration. Hepatocyte synthesis of albumin, urea, and intracellular triglycerides was monitored in Dulbecco's modified Eagle medium (supplemented with fetal bovine serum, hydrocortisone, L-proline, gentamycin, and insulin) without supplemental colloid

Dilutional acidosis following hetastarch or albumin in healthy volunteers.

BACKGROUND: The intent of this study was to evaluate the impact of the commonly used colloids-hetastarch and albumin-on in vivo acid-base balance. From this evaluation, a better understanding of the mechanism of dilutional acidosis was expected. METHODS: In a prospective, randomized fashion, 11 healthy volunteers were administered 15 ml/kg hetastarch solution, 6%, or 15 ml/kg albumin, 5%, intravenously over 30 min. Four weeks later, the study subjects were administered the other colloid. Arterial blood gas and electrolyte parameters were measured at baseline and at 30, 60, 90, 120, 210, and 3

Differential phototoxicity of fluorescent dye-labeled albumin conjugates.

OBJECTIVE: Fluorescent dyes are commonly used as probes for assessment of macromolecular permeability. Despite numerous examples of light-dye induced toxicity in the microvasculature, little is known regarding the relative phototoxicity of commonly used fluorescent conjugates. We, therefore, compared the phototoxicity of four fluorescent conjugates of bovine serum albumin (BSA) available commercially. METHODS: An in vitro photohemolysis assay was used, in which rat erythrocytes were incubated with fluorescently labeled BSA and exposed to epi-illumination, using an inverted microscope designed

Characterization of proteinuria in primary glomerulonephritides: urinary polymers of albumin.

In 142 patients with primary glomerulonephritis (GN), there were polymers of albumin (PAs) in the urine samples of 87% of 15 minimal-change disease (MCD) patients, 52% of 27 focal segmental glomerulosclerosis (FSGS) patients, 51% of 47 membranous glomerulonephritis (MGN) patients, 55% of 20 membranoproliferative glomerulonephritis (MPGN) patients, and 9% of 33 immunoglobulin A nephropathy (IgAN) patients (P = 0.000). In IgAN, only three patients with nephrotic syndrome were PA positive. The PAs were significantly correlated with nephrotic syndrome (NS) (P = 0.000) and with the degree of protei

[Is albumin administration useful in critical care for burnt patients?].

The most typical reaction of the organism after a major burn consists in transcapillary shift from plasma into interstitial space. Capillary hyperpermeability, but also changes in colloid osmotic gradient and decrease in interstitial hydrostatic pressure, explain the fluid shift to burned and, at minor importance, non burned areas during the first post-burn day. The extent of capillary hyperpermeability results in inefficiency of colloid infusions in reducing fluid shift to burned areas. Some groups advocate colloid supply from the eighth post-burn hour, on when hyperpermeability decreases, in

Salicylate binding by human albumin in the presence of a perfluorochemical emulsion.

The binding of 43 and 216 micrograms/ml sodium salicylate by a perfluorochemical emulsion, by human albumin, and by their mixtures, has been examined at ambient room temperature. Sodium salicylate was chosen as a model drug since it is bound by both sites I and II on human albumin. The concentration of free salicylate was determined through standard centrifugation followed by supernatant ultrafiltration. Sodium salicylate was weakly bound by the perfluorochemical emulsion droplets through an interaction with the emulsifiers. This binding was independent of salicylate concentration. Sodium sali
