Knowledge mapping of core decompression in osteonecrosis of the femoral head: a bibliometric analysis.

BACKGROUND: Osteonecrosis of the femoral head (ONFH), caused by disrupted blood supply leading to bone cell death and joint collapse, remains a critical orthopedic challenge. While core decompression has advanced significantly in ONFH treatment, no bibliometric analysis has mapped its research landscape. METHOD: We analyzed 854 articles from the Web of Science Core Collection (1986-2023) using VOSviewer, CiteSpace, and bibliometrix. RESULTS: Publications surged from 5.4/year (1986-2001) to 52.3/year (2015-2023), reflecting intensified interest. China (253 articles, 29.6%) and the United Stat

Induced Pluripotent Stem Cell-Derived Corneal Cells: Current Status and Application.

Deficiency and dysfunction of corneal cells leads to the blindness observed in corneal diseases such as limbal stem cell deficiency (LSCD) and bullous keratopathy. Regenerative cell therapies and engineered corneal tissue are promising treatments for these diseases [1]. However, these treatments are not yet clinically feasible due to inadequate cell sources. The discovery of induced pluripotent stem cells (iPSCs) by Shinya Yamanaka has provided a multitude of opportunities in research because iPSCs can be generated from somatic cells, thus providing an autologous and unlimited source for corne

Stem cell health and tissue regeneration in microgravity.

Exposure to microgravity causes significant mechanical unloading of mammalian tissues, resulting in rapid alterations of their physiology, which poses a significant risk for long-duration manned spaceflight. The immediate degenerative effects of spaceflight we understand best are those studied during short-term low-Earth-orbit experiments, and include rapid microgravity-adaptive bone and muscle loss, loss of cardiovascular capacity, defects in wound and bone fracture healing, and impaired immune function. Over the long-term, exposure to microgravity may cause severe deficits in mammalian stem

Human pluripotent stem cell-derived retinal ganglion cells: advances in differentiation and translational applications.

Retinal ganglion cells (RGCs) are neurons that transmit visual information from the retina to the brain. Their degeneration, as seen in glaucoma and other optic neuropathies, leads to irreversible vision loss. As mature human RGCs are difficult to access, most of their studies rely on rodent models, which do not fully recapitulate human retinal biology. Human pluripotent stem cells (hPSCs) provide a promising source for generating RGCs in vitro, supporting disease modelling, drug screening, and future cell replacement therapies. This review outlines key markers that define RGC identity, matura

Emerging strategies for spinal cord injury repair: stem cells, extracellular vesicles, biomaterials, and neuromodulation.

Spinal cord injury (SCI) is a devastating condition that frequently results in permanent disability. Repair and functional recovery after SCI are hindered by the complexity of its pathophysiology and the inherent challenges of neural regeneration within the central nervous system. Despite decades of research aimed at elucidating its pathological mechanisms and developing effective strategies to promote axonal regeneration and circuit rewiring, successful therapeutic outcomes remain limited. Recent advances in bioactive materials and stem cell technology have shifted the research focus from sol

Strategies to enhance paracrine potency of transplanted mesenchymal stem cells in intractable neonatal disorders.

Mesenchymal stem cell (MSC) transplantation represents the next breakthrough in the treatment of currently intractable and devastating neonatal disorders with complex multifactorial etiologies, including bronchopulmonary dysplasia, hypoxic ischemic encephalopathy, and intraventricular hemorrhage. Absent engraftment and direct differentiation of transplanted MSCs, and the "hit-and-run" therapeutic effects of these MSCs suggest that their pleiotropic protection might be attributable to paracrine activity via the secretion of various biologic factors rather than to regenerative activity. The tran

Generation of three induced pluripotent stem cell lines from a long QT syndrome type 2 family harboring the pathogenic KCNH2 c.209A > G (p.His70Arg) variant.

Long QT Syndrome Type 2 (LQT2) is a heart rhythm disorder caused by a loss-of-function mutation in the KCNH2 gene, characterized by a prolonged QT interval on an electrocardiogram (ECG) and symptoms such as syncope and potentially life-threatening Torsades de Pointes arrhythmia. We derived three induced pluripotent stem cell (iPSC) lines from a LQT2 family: two individuals with LQT2 who carry the pathogenic variant c.209A > G (p.His70Arg), and one healthy individual. These iPSC lines exhibit normal karyotype, differentiation capability, stem cell pluripotency, and stem cell morphology, providi

Ovarian cancer stem cells and their role in drug resistance.

Ovarian cancer is typically diagnosed at advanced stages (III or IV), with metastasis ensuing at stage III. Complete remission is infrequent and is not achieved in almost half of the women diagnosed with ovarian cancer. Consequently, management and treatment of this disease is challenging as many patients are faced with tumour recurrence disseminating to surrounding organs further complicated with acquired chemo-resistance. The cancer stem cell theory proposes the idea that a drug resistant subset of tumour cells drive tumour progression, metastasis and ultimately, recurrent disease. In the ov

Current State and Issues of Regenerative Medicine for Rheumatic Diseases.

The prognosis of rheumatic diseases is generally better than that of malignant diseases. However, some cases with poor prognoses resist conventional therapies and cause irreversible functional and organ damage. In recent years, there has been much research on regenerative medicine, which uses stem cells to restore the function of missing or dysfunctional tissues and organs. The development of regenerative medicine is also being attempted in rheumatic diseases. In diseases such as systemic sclerosis (SSc), systemic lupus erythematosus (SLE), and rheumatoid arthritis, hematopoietic stem cell tra

Stem Cells for Osteochondral Regeneration.

Stem cell research plays a central role in the future of medicine, which is mainly dependent on the advances on regenerative medicine (RM), specifically in the disciplines of tissue engineering (TE) and cellular therapeutics. All RM strategies depend upon the harnessing, stimulation, or guidance of endogenous developmental or repair processes in which cells have an important role. Among the most clinically challenging disorders, cartilage degeneration, which also affects subchondral bone becoming an osteochondral (OC) defect, is one of the most demanding. Although primary cells have been clini…

Human stem cell models in cerebral cavernous malformations.

Cerebral cavernous malformation (CCM) is a rare cerebrovascular disorder characterized by abnormal endothelial architecture and clinically manifests as hemorrhage, epilepsy, and neurological deficits. Two primary factors have constrained the advancement of research in the field of CCM. Firstly, the utilization of animal models, which do not fully recapitulate human neurovascular biology, has been a major impediment. Secondly, the restricted access to patient lesion tissue has hindered progress. These constraints impede mechanistic dissection and therapeutic translation. This review discusses h

Stem Cell Transplantation Therapy for Retinal Degenerative Diseases.

In the past decade, progress in the research on human embryonic stem cells (hESCs) and induced pluripotent stem cells (iPSCs) has provided the solid basis to derive retinal pigment epithelium, photoreceptors, and ganglion cells from hESCs/iPSCs for transplantation therapy of retinal degenerative diseases (RDD). Recently, the iPSC-derived retinal pigment epithelium cells have achieved efficacy in treating patients with age-related macular degeneration (AMD). However, there is still much work to be done about the differentiation of hESCs/iPSCs into clinically required retinal cells and improveme

Oral cavity-derived stem cells and preclinical models of jaw-bone defects for bone tissue engineering.

BACKGROUND: Jaw-bone defects caused by various diseases lead to aesthetic and functional complications, which can seriously affect the life quality of patients. Current treatments cannot fully meet the needs of reconstruction of jaw-bone defects. Thus, the research and application of bone tissue engineering are a "hot topic." As seed cells for engineering of jaw-bone tissue, oral cavity-derived stem cells have been explored and used widely. Models of jaw-bone defect are excellent tools for the study of bone defect repair in vivo. Different types of bone defect repair require different stem cel

Kidney organogenesis and regeneration: a new era in the treatment of chronic renal failure?

The recent development of a strategy to establish human inducible pluripotent stem (iPS) cells has created a second surge in the field of regenerative research, which had been slowed by restrictions on the use of pluripotent embryonic stem cells. Research on regenerative nephrology offers hope for patients on dialysis. However, due to its anatomic complexity, the kidney is the most difficult organ for the application of regenerative medicine. Very recently, the establishment of a functional whole kidney has been attempted using various stem cells, which may lead to clinical applications. We re

The Role of Human Placental Mesenchymal Stem Cell Derived Exosomes in Wound Healing and Scar Therapy.

Exosomes are a subtype of extracellular vesicles, critical for intercellular communication. They are released from a cell via exocytosis, after which they travel through the extracellular fluid until they reach a target cell. Research in the fields of cellular medicine, regenerative and stem cell therapy continues to grow exponentially. Deciphering the biological properties of mesenchymal stem cells-exosomes as cell-free therapeutic tools is important for wound healing and cutaneous regeneration. The future is also bright for genetically engineering exosomes for targeted diagnostic and therape
