Integrated Virtual Screening Approach Identifies New CYP19A1 Inhibitors.

The human cytochrome P450 19A1 (CYP19A1, aromatase) is a heme-containing protein catalyzing the final steps of the biosynthesis of the steroid hormone 17β-estradiol. It is a key target for the treatment of sex-hormone-related disorders due to its role in mediating the conversion of androgens to estrogens. Here, we report the development of a virtual screening workflow incorporating machine learning and structure-based modeling that has led to the discovery of new CYP19A1 inhibitors. The machine learning models were built on comprehensive CYP19A1 data sets extracted from the ChEMBL and PubChem

True horizontal or gravity plane registration for transferring the maxillary scan into the virtual articulator by using a facial scanner without the need for an additional device.

Different reference planes can be used to transfer the maxillary cast into the analog articulator, including the true horizontal or gravity reference plane. Different techniques have been described to record the gravity reference plane for transferring the maxillary scan into the virtual articulator by using facial scanning techniques. However, these digital facebow procedures require the use of an extraoral scan body system, printed reference device, or orientation reference board. This manuscript describes a technique for recording the gravity reference plane by using a facial scanner withou

Towards the Development of a Learning-Based Intention Classification Framework for Pushrim-Activated Power-Assisted Wheelchairs.

There has been a growth in the design and use of power assist devices for manual wheelchairs (MWCs) to alleviate the physical load of MWC use. A pushrim-activated power-assisted wheel (PAPAW) is an example of a power assist device that replaces the conventional wheel of a MWC. Although the use of PAPAWs provides some benefits to MWC users, it can also cause difficulties in maneuvering the wheelchair. In this research, we examined the characteristics of wheelchair propulsion when using manual and powered wheels. We used the left and right wheels' angular velocity to calculate the linear and ang

MolTarPred: A web tool for comprehensive target prediction with reliability estimation.

Molecular target prediction can provide a starting point to understand the efficacy and side effects of phenotypic screening hits. Unfortunately, the vast majority of in silico target prediction methods are not available as web tools. Furthermore, these are limited in the number of targets that can be predicted, do not estimate which target predictions are more reliable and/or lack comprehensive retrospective validations. We present MolTarPred ( http://moltarpred.marseille.inserm.fr/), a user-friendly web tool for predicting protein targets of small organic compounds. It is powered by a large

Effects of repeated snowboard exercise in virtual reality with time lags of visual scene behind body rotation on head stability and subjective slalom run performance in healthy young subjects.

CONCLUSION: After repeated snowboard exercises in the virtual reality (VR) world with increasing time lags in trials 3-8, it is suggested that the adaptation to repeated visual-vestibulosomatosensory conflict in the VR world improved dynamic posture control and motor performance in the real world without the development of motion sickness. OBJECTIVES: The VR technology was used and the effects of repeated snowboard exercise examined in the VR world with time lags between visual scene and body rotation on the head stability and slalom run performance during exercise in healthy subjects. E-virtual reality exposure therapy in acrophobia: A pilot study.

Virtual reality therapy is already used for anxiety disorders as an alternative to in vivo and in imagino exposure. To our knowledge, however, no one has yet proposed using remote virtual reality (e-virtual reality). The aim of the present study was to assess e-virtual reality in an acrophobic population. Six individuals with acrophobia each underwent six sessions (two sessions per week) of virtual reality exposure therapy. The first three were remote sessions, while the last three were traditional sessions in the physical presence of the therapist. Anxiety (STAI form Y-A, visual analog scale,

Delays in Admittance-Controlled Haptic Devices Make Simulated Masses Feel Heavier.

In an admittance-controlled haptic device, input forces are used to calculate the movement of the device. Although developers try to minimize delays, there will always be delays between the applied force and the corresponding movement in such systems, which might affect what the user of the device perceives. In this experiment we tested whether these delays in a haptic human-robot interaction influence the perception of mass. In the experiment an admittance-controlled manipulator was used to simulate various masses. In a staircase design subjects had to decide which of two virtual masses was h

Surface electromyographic control of speech synthesis.

Individuals with very high spinal cord injuries (e.g. C1-C3) may be ventilator-dependent and therefore unable to support speech breathing. However, their facial musculature is intact, given that these muscles are innervated by cranial nerves. We developed a system using surface electromyography (sEMG) recorded from facial muscles to control a phonemic interface and voice synthesizer and tested the system in healthy individuals. Users were able to use five facial gestures to control an onscreen cursor and the phonemic interface. Users had mean information transfer rates (ITRs) of 59.5 bits/min

Interactive electronic storybooks for kindergartners to promote vocabulary growth.

The goals of this study were to examine (a) whether extratextual vocabulary instructions embedded in electronic storybooks facilitated word learning over reading alone and (b) whether instructional formats that required children to invest more effort were more effective than formats that required less effort. A computer-based "assistant" was added to electronic storybooks. The assistant posed extratextual vocabulary questions. Questions were presented in a multiple-choice format so that children could respond by clicking on the picture that best represented the target word. In Experiment 1 (N=

Internet delivered images for exposure in specific phobia, design considerations for self-directed therapy.

Exposure to phobic stimuli in subjects with specific phobia typically results in increased anxiety, ranging from mild to severe, followed by gradual habituation. The Internet is a candidate medium for the delivery of phobic stimuli to phobic subjects, such as pictures, video clips or computer animations. Delivery of such images in home settings warrants careful attention to the range and time course of anxiety responses elicited, and to tailoring of progression through hierarchies of images. The agency of the user is paramount, they need to have the final say at all stages of exposure as to wh

Virtual reality and acrophobia: one-year follow-up and case study.

We present a study with 10 subjects being exposed to three sessions of simulated heights in a virtual reality (VR) system. Among the participants we highlight a 66-year-old man blind in his left eye. The participants show significant progress in anxiety, avoidance, and behavior measurements when confronted with real height circumstances. The results obtained 1 year later at follow-up are statistically significant in the Behavioral Avoidance Test (BAT) and the Attitudes Toward Heights Questionnaire (ATHQ), but not the Acrophobia Questionnaire (AQ).

Cardiac responses induced during thought-based control of a virtual environment.

Cardiac responses induced by motor imagery were investigated in 3 subjects in a series of experiments with a synchronous (cue-based) Brain-Computer Interface (BCI). The cue specified right hand vs. leg/foot motor imagery. After a number of BCI training sessions reaching a classification accuracy of at least 80%, the BCI experiments were carried out in an immersive virtual environment (VE), commonly referred as a "CAVE". In this VE, the subjects were able to move along a virtual street by motor imagery alone. The thought-based control of VE resulted in an acceleration of the heart rate in 2 sub

Comparison of virtual treatment planning outcomes designed by two commercially available software in treatment of anterior teeth crowding using in-house aligners.

Virtual treatment planning (VTP) has revolutionized clear aligner therapy (CAT) by facilitating patient communication and enhancing treatment predictability. The accuracy and reproducibility of VTP are highly dependent on the 3D design software used, particularly in in-house aligner workflows.Aim This study aimed to evaluate and compare the surface-based geometric accuracy of VTPs generated by two 3D designing software packages commonly used in clear aligners treatment planning; Maestro 3D ortho studio® and Dentone® in mild to moderate anterior teeth crowding versus ClinCheck®Pro.Methods This

SmoothRide: A Versatile Solution to Combat Cybersickness in Elevation-Altering Environments.

Cybersickness continues to bar many individuals from taking full advantage of virtual reality (VR) technology. Previous work has established that navigating virtual terrain with elevation changes poses a significant risk in this regard. In this paper, we investigate the effectiveness of three cybersickness reduction strategies on users performing a navigation task across virtual elevation-altering terrain. These strategies include static field of view (FOV) reduction, a flat surface approach that disables terrain collision and maintains constant elevation for users, and SmoothRide, a novel tec
