Effects of exercise timing and metabolic state on post-exercise energy intake and appetite-a randomized crossover trial.

BACKGROUND: Exercise timing and metabolic state influence post-exercise energy balance, yet their combined effects remain incompletely understood. OBJECTIVES: To disentangle the effects of time of day (morning compared with evening) and nutritional state (fasted compared with fed) on post-exercise energy intake and appetite. METHODS: In a randomized crossover design, 35 healthy young adults completed 3 fasted exercise conditions: morning after a 12 h overnight fast (12 h AMEx) and evening after a 12 h (12 h PMEx) or 6-h fast. A subsample (n = 17) additionally completed fed morning and fed ev

Hypothalamic regulation of sleep.

The recent discovery linking narcolepsy, a sleep disorder characterized by very short REM sleep latency, with a neuropeptide that regulates feeding and energy metabolism, provides a way to understand how several behaviors may be disrupted as a result of a defect in this peptide. In this chapter we review the evidence linking hypocretin and sleep, including our own studies, and propose that a defect in the lateral hypothalamus that also involves the hypocretin neurons is likely to produce a disturbance in sleep, mood, appetite, and rhythms.

Anorexia in rats caused by a valine-deficient diet is not ameliorated by systemic ghrelin treatment.

Rodents exhibit aversive behavior toward a diet that lacks at least one of the essential amino acids. We sought to determine whether the particular form of anorexia caused by such diets could be ameliorated by the administration of orexigenic peptides while simultaneously analyzing the neural mechanisms underlying anorexia. Rats were fed a valine-deficient diet, which induced severe anorexia (reducing food consumption by 80%). The severe anorexia was associated with a significant decrease in the cerebrospinal fluid valine concentration and hyper-ghrelinemia. Between 6 and 12 days after initiat

Effects of day-length and food availability on food caching, mass and fat reserves in black-capped chickadees (Poecile atricapillus).

Many animals seasonally change energy stores such as food caches and fat deposits. Two factors that potentially influence energy stores are changes in day-length and food availability. We investigated food storing and fat levels in black-capped chickadees to determine how they are affected by changes in duration of light period and duration of food availability. Because previous studies have confounded the effects of photoperiod and the effects of duration of access to food we manipulated these variables independently. Birds were exposed to either long days (15 h light with 15 h of food availa

Potential role of intelectin-1 in the regulation of feeding of goldfish (Carassius auratus).

Appetite regulation in vertebrates involves central neuropeptides and peripheral metabolic and endocrine signals that act together to maintain energy homeostasis. These include orexigenic (e.g., neuropeptide Y, orexin, ghrelin) and anorexigenic (e.g., cocaine- and amphetamine-regulated transcript, corticotropin-releasing factor, cholecystokinin, leptin) factors coordinate feeding behavior. Intelectin-1 (ITLN1), a protein implicated in innate immunity and metabolic signaling in mammals, has not yet been investigated in the context of fish feeding physiology. This study examined the potential ro

Effects of atypical antipsychotics and haloperidol on PC12 cells: only aripiprazole phosphorylates AMP-activated protein kinase.

By converting changes in intracellular energy status to changes in cell membrane polarization, ATP-sensitive K(+) (K(ATP)) channels in hypothalamic appetite-regulating neurons play a critical role in linking neuronal electrochemical function, metabolic and energy status, and feeding behavior. Most atypical antipsychotics (AAPs) increase the appetite of patients with schizophrenia and thus cause obesity. This study aimed to explain the mechanism underlying AAP-induced appetite stimulation, based on the fact that the efficiency of fatty acid uptake into mitochondria generating ATP through β-oxid

Hypothalamic neuropeptide Y (NPY) and the attenuation of hyperphagia in streptozotocin diabetic rats treated with dopamine D1/D2 agonists.

1. Dopamine is an appetite suppressant, while neuropeptide Y (NPY), an appetite stimulant in the brain, is reported to be involved in anorectic action induced by a combined administration of D1/D2 agonists in normal rats. In diabetic rats, however, these factors have not been studied. 2. Rats (including normal, diabetic and insulin-treated diabetic rats) were given daily injections of saline or D1/D2 agonists for 6 days. Changes in food intake and hypothalamic NPY content of these rats were assessed and compared. 3. The D1/D2 agonist-induced anorectic responses were altered in diabetic rats co

Spontaneous peripheral neuritis in two electric eels (Electrophorus electricus).

This study represents cases with spontaneous neuritis of peripheral nerves in electric eels. Two electric eels were presented with abnormal swimming behavior and loss of appetite. Electric eels had extensive histopathologic lesions in the splenic and cardiac nerves. The lesions were characterized by swelling of neuronal cells, central chromatolysis and marked inflammatory cell infiltration consisting mainly of lymphocytes around the affected nerves. To the best of our knowledge, this is the first case report of spontaneous neuritis of peripheral nerves in electric eels.

Area postrema and adjacent solitary nucleus in water and energy balance.

Lesions of the area postrema (AP) and immediately adjacent nucleus tractus solitarii cause a syndrome of permanent weight loss with little disruption to behavioral controls of body weight regulation. There is also a permanent polydipsia and elevated salt appetite, which appear to be secondary to deficits in renal conservation of water and sodium. Neuroanatomical studies indicate that the immediately adjacent solitary nucleus and AP are sites of termination of visceral afferents from abdominal organs and the destination and origin of distant central afferent and efferent connections.

Behavioral Challenges and Adaptive Strategies in Providing Mealtime Care to Residents With Dementia: A Qualitative Study of Nursing Home Clinical and Administrative Staff.

This study examined how nursing home staff adapt to challenging mealtime behaviors in residents with dementia, such as turning away or refusing food. Ten focus groups were conducted with 52 clinical and administrative staff from three southeastern U.S. nursing homes. Transcripts were analyzed using four key codes: feeding behaviors, interventions, training, and communication. Staff described a range of mealtime challenges, including cognitive and functional decline, dysphagia, and elopement. Feeding-related behaviors like refusing to eat reflect the complexity of care required. Staff reported

Effect of tau burden and Lewy pathology on neuropsychiatric symptoms in Aβ-positive individuals.

INTRODUCTION: Neuropsychiatric symptoms (NPS) are common across the Alzheimer's disease (AD) spectrum. We aimed to evaluate the effects of amyloid beta (Aβ), tau, and Lewy body (LB) pathologies on NPS in Aβ-positive individuals. METHODS: In 336 Aβ-positive participants from the Alzheimer's Disease Neuroimaging Initiative cohort, spanning from cognitively unimpaired to those with early dementia, NPS were assessed. Aβ and tau positron emission tomography were conducted. In a subgroup of 238 participants, dichotomized LB pathology was assessed by detecting α-synuclein seeding activity. The effec

Brain histaminergic system: An emerging target for the treatment of feeding and eating-related disorders.

Feeding is an intrinsic and fundamental behavior regulated by complex neurobiological networks that integrate peripheral metabolic signals with neurotransmitter systems within key brain regions. Histamine, a biogenic amine acting as a neuromodulator in the central nervous system, plays a crucial role in the regulation of food intake and energy homeostasis. Within the hypothalamus, histaminergic neurons influence appetite-related circuits, primarily through H1R and H3R. Central administration of histamine suppresses food consumption, an effect largely mediated via H1R. The blockade of H1R has b

A longitudinal study of early postoperative symptom cluster trajectories in patients with gynecologic malignancies.

OBJECTIVE: To explore the dynamic evolution of symptom clusters in patients with gynecologic malignancies during the early postoperative period and identify key transition points and influencing factors, providing evidence for precision symptom management in clinical nursing. METHODS: A longitudinal study was conducted among 324 patients using the MDASI-PeriOp-GYN on postoperative days 1 (T1), 5 (T2), and 7 (T3). Exploratory factor analysis identified symptom clusters at each time point, and growth mixture modeling (GMM) was applied to examine trajectory patterns. Latent profile analysis (LPA

Hypothalamic mechanisms in cachexia.

The role of nutrition and balanced metabolism in normal growth, development, and health maintenance is well known. Patients affected with either acute or chronic diseases often show disorders of nutrient balance. In some cases, a devastating state of malnutrition known as cachexia arises, brought about by a synergistic combination of a dramatic decrease in appetite and an increase in metabolism of fat and lean body mass. Other common features that are not required for the diagnosis include decreases in voluntary movement, insulin resistance, and anhedonia. This combination is found in a number

Side effects of minocycline treatment in patients with fragile X syndrome and exploration of outcome measures.

Minocycline can rescue the dendritic spine and synaptic structural abnormalities in the fragile X knock-out mouse. This is a review and preliminary survey to document side effects and potential outcome measures for minocycline use in the treatment of individuals with fragile X syndrome. We surveyed 50 patients with fragile X syndrome who received minocycline for at least 2 weeks and found that the most common reported side effect is gastrointestinal difficulty, including loss of appetite. The families reported an improvement in language and behavioral areas. Outcome measures in the design of f

Evaluation of Realistic Sensation Using Biological Reaction Measurements for Food Videos Presented by KMMD.

We have prototyped a KANSEI multimedia display (KMMD) that is able to release scent through the screen in order to realize collaboration between images and scents. Two types of "sukiyaki" food videos were presented to subjects using this device, and a method for objectively evaluating the realistic sensation of the food videos was examined using biological reaction measurements. The sukiyaki scent was added to one type of video to improve appetite. Viewers' saliva flow rate, line of sight, pupil diameter, autonomic nerve activity, and cerebral blood flow were measured at the same time, and cha

The novel cannabinoid CB1 antagonist AM6545 suppresses food intake and food-reinforced behavior.

Drugs that interfere with cannabinoid CB1 transmission suppress food-motivated behaviors, and may be useful clinically as appetite suppressants. However, there may also be undesirable side effects (e.g., nausea, malaise, anxiety, and depression) that are produced by the current generation of CB1 inverse agonists such as rimonabant and taranabant. For that reason, it is important to continue research on novel cannabinoid antagonists. The present studies examined the effects of the novel compound AM6545, which is a neutral antagonist of CB1 receptors that is thought to have relatively poor penet

NTS and VTA oxytocin reduces food motivation and food seeking.

Oxytocin (OT) is a neuropeptide whose central receptor-mediated actions include reducing food intake. One mechanism of its behavioral action is the amplification of the feeding inhibitory effects of gastrointestinal (GI) satiation signals processed by hindbrain neurons. OT treatment also reduces carbohydrate intake in humans and rodents, and correspondingly, deficits in central OT receptor (OT-R) signaling increase sucrose self-administration. This suggests that additional processes contribute to central OT effects on feeding. This study investigated the hypothesis that central OT reduces food
