Decreased impulsiveness and MEG normalization after AI-digital therapy in ADHD children: a RCT.

Attention-deficit/hyperactivity disorder (ADHD) presents with symptoms like impulsiveness, inattention, and hyperactivity, often affecting c … driven therapy or a placebo intervention. Assessments were conducted pre- and post-intervention and magnetoencephalography (MEG) analyzed brain activity. Results showed significant reductions in impulsiveness and inattentiveness scores in the treatment group, associated with normalized MEG spectral profiles, indicating neuromaturation. Notably, improvements in inhibitor …

Synergistic association of Aβ and tau pathology with cortical neurophysiology and cognitive decline in asymptomatic older adults.

Animal and computational models of Alzheimer's disease (AD) indicate that early amyloid-β (Aβ) deposits drive neurons into a hyperactive regime, and that subsequent tau depositions manifest an opposite, suppressive effect as behavioral deficits emerge. Here we report analogous changes in macroscopic oscillatory neurophysiology in the human brain. We used positron emission tomography and task-free magnetoencephalography to test the effects of Aβ and tau deposition on cortical neurophysiology in 104 cognitively unimpaired older adults with a family history of sporadic AD. In these asymptomatic i…

Linguistic processing of task-irrelevant speech at a cocktail party.

Paying attention to one speaker in a noisy place can be extremely difficult, because to-be-attended and task-irrelevant speech compete for processing resources. We tested whether this competition is restricted to acoustic-phonetic interference or if it extends to competition for linguistic processing as well. Neural activity was recorded using Magnetoencephalography as human participants were instructed to attend to natural speech presented to one ear, and task-irrelevant stimuli were presented to the other. Task-irrelevant stimuli consisted either of random sequences of syllables, or syllable…

The Visual Word Form Area compensates for auditory working memory dysfunction in schizophrenia.

Auditory working memory impairments feature prominently in schizophrenia. However, the existence of altered and perhaps compensatory neural dynamics, sub-serving auditory working memory, remains largely unexplored. We compared the dynamics of induced high gamma power (iHGP) across cortex in humans during speech-sound working memory in individuals with schizophrenia (SZ) and healthy comparison subjects (HC) using magnetoencephalography (MEG). SZ showed similar task performance to HC while utilizing different brain regions. During encoding of speech sounds, SZ lacked the correlation of iHGP with…

A peek into premonitory urges in Tourette syndrome: Temporal evolution of neurophysiological oscillatory signatures.

BACKGROUND: Tics are the core symptom of patients with Gilles de la Tourette syndrome, yet the spatial-temporal dynamics of neural activity causing a tic remains to be determined. OBJECTIVE: Identification of cortical events preceding tic onset. METHODS: In twelve patients with Tourette syndrome we performed magnetoencephalography to trace the time course of beta oscillations (15-30 Hz) in motor cortical areas before tic onset. RESULTS: Patients showed a biphasic modulation of cortical beta activity during the second before tic onset. We observed an initial increase of beta power over the l…

The impact of Val108/158Met polymorphism of catechol-O-methyltransferase on brain oscillations during working memory.

This study investigated whether catechol-O-methyltransferase (COMT) Val/Met polymorphism was associated with variation in event-related desynchronization/synchronization (ERD/ERS) of responses during working memory (WM). 11 Val/Val and 11 Met/Met homozygous participants underwent magnetoencephalography (MEG) while performing a WM task. In contrast to small effects behaviourally, during the delay period Val/Val individuals showed lower ERS in the gamma band (Hz 30-50) in frontal regions, increased ERS in the alpha band (Hz 8-12) in the right frontal and parietal regions and increased ERD in the…

Engagement of temporal lobe regions predicts response to educational interventions in adolescent struggling readers.

Brain activation profiles obtained using magnetoencephalography were compared between middle-school students experiencing reading difficulties and non-reading-impaired students during performance of a continuous printed word recognition task. Struggling readers underwent small-group remedial instruction, and students who showed significant gains in word reading efficiency at a one-year follow-up assessment were classified as Adequate Responders whereas those not demonstrating such gains as Inadequate Responders. At baseline, compared to Inadequate Responders, the activation profiles of Adequat…

Estimates of visually evoked cortical currents.

We have measured magnetic fields evoked by the onset of checkerboard-like sectorial patterns presented at 16 locations near the center of th … ch was, 80-100 msec after the pattern onset, retinotopically organized in the occipital lobe. Since magnetoencephalography can reveal cortical locations with a precision of 2-3 mm, our procedure appears promising for further studies of cortical retinotopy and visual field defects.

Non-Invasive Mapping of the Neuronal Networks of Language.

This review consists of three main sections. In the first, the Introduction, the main theories of the neuronal mediation of linguistic operations, derived mostly from studies of the effects of focal lesions on linguistic performance, are summarized. These models furnish the conceptual framework on which the design of subsequent functional neuroimaging investigations is based. In the second section, the methods of functional neuroimaging, especially those of functional Magnetic Resonance Imaging (fMRI) and of Magnetoencephalography (MEG), are detailed along with the specific activation tasks em…

Epileptiform discharges relate to altered functional brain networks in autism spectrum disorders.

Many individuals with autism spectrum disorders have comorbid epilepsy. Even in the absence of observable seizures, interictal epileptiform discharges are common in individuals with autism spectrum disorders. However, how these interictal epileptiform discharges are related to autistic symptomatology remains unclear. This study used magnetoencephalography to investigate the relation between interictal epileptiform discharges and altered functional brain networks in children with autism spectrum disorders. Instead of particularly addressing individual brain regions, we specifically examine netw…

"When the word ends, the story begins": word offsets improve encoding of natural speech in a MEG study.

The segmentation of continuous speech into words was investigated using the temporal response function (TRF) approach for source-space magnetoencephalography (MEG) signals from 54 participants. Although the onset of words has been extensively studied, the contribution of their offset remains unclear. Acoustic and semantic predictors were used to test the relative contribution of word onsets and offsets to the processing of natural speech in the auditory temporal cortex. Circular shifting of word onsets and offsets significantly reduced predictive power (p < 0.001 for both). The corrupted word …

Abnormal hippocampo-cortical theta-gamma phase-amplitude coupling in Alzheimer's disease.

INTRODUCTION: Cognitive decline in Alzheimer's disease (AD) may arise not only from neuronal loss but also from disrupted temporal coordination across distributed networks. Prior works in healthy individuals has shown memory-related modulation of hippocampal-cortical phase-amplitude coupling (PAC). We hypothesized alteration of resting-state hippocampo-cortical PAC in AD which relates to cognitive impairment. METHODS: Resting-state magnetoencephalography (MEG) data were obtained from 78 AD patients and 70 healthy controls. Source-reconstructed hippocampal and cortical signals were analysed us…

Physical activity enhances theta-periodicity of visual attentional allocation.

The traditional notion of a sustained visual spotlight of attention has been challenged by behavioral and neural evidence showing that attention fluctuates rhythmically in the theta band (3-8 Hz). In rodents, locomotion drives hippocampal theta oscillations that support cue sampling during navigation, enhancing sensory encoding and decision-making. This raises the question of whether physical activity (PA) similarly modulates theta-based attentional sampling in humans. Using magnetoencephalography (MEG), we found that discrimination accuracy (DA) increased following PA, accompanied by stronger…

Frequency-specific cortical subnetworks support fast human swallowing.

Fast, highly constrained sensorimotor acts require rapid coordination of distributed cortical systems on subsecond timescales. Here, we used source-resolved magnetoencephalography to characterise time-locked cortical connectivity during voluntary swallowing in 74 healthy adults. Cluster-based network statistics revealed focal swallowing-related connectivity changes confined to anatomically selective subnetworks. Undirected phase-lagged connectivity identified theta- and low-gamma weighted phase lag index (wPLI) effects involving somatosensory, motor, supramarginal, and insular regions. Directe…

Prediction of mild cognitive impairment progression using time-sensitive multimodal biomarkers.

Alzheimer's disease (AD) develops silently for years before symptoms emerge, making early identification of at-risk individuals essential for prevention trials and early intervention. We tested whether combining neurophysiological, imaging, and blood biomarkers improves prediction of progression to mild cognitive impairment (MCI) in cognitively unimpaired older adults with a family history of AD (n = 102; 31 progressors; mean follow-up of 5.9 years). Magnetoencephalography, magnetic resonance imaging, plasma biomarkers, and amyloid and tau positron emission tomography each captured complementa…

Neural signatures of temporal anticipation in human cortex represent event probability density.

Temporal prediction is a fundamental function of neural systems. Recent results show that humans anticipate future events by calculating probability density functions, rather than hazard rates. However, direct neural evidence for this hypothesized mechanism is lacking. We recorded neural activity using magnetoencephalography as participants anticipated auditory and visual events distributed in time. We show that temporal anticipation, measured as reaction times, approximates the event probability density function, but not hazard rate. Temporal anticipation manifests as spatiotemporally pattern…
