Strengthening India's pandemic preparedness with the four zonal institutes of virology: stakeholder consultations for outbreak response and collaborative research priorities.

BACKGROUND: The Pradhan Mantri Ayushman Bharat Health Infrastructure Mission (PM-ABHIM) scheme, supported by the World Bank, is establishing four zonal National Institutes of Virology (NIVs) in India. It is essential to identify the zonal disease and research priorities so as to focus research at zonal levels. This study reports a zonal viral disease prioritisation exercise to support outbreak response and collaborative research. It describes zonal-prioritised viral diseases, assesses concordance and differences in zonal priority lists and, and identifies cross‑cutting research themes and prep…

An Integrative Approach to the Study of Cognitive Abilities in a Non-Human Primate Model in a Virology Laboratory Environment.

Non-human primates, due to their similarities in immune response to humans, are the preferred model for studying infectious processes and any associated cognitive impairments. Behavioral tests are indispensable for investigating pathogenesis in neuroinfections, especially those that do not manifest with noticeable clinical symptoms, as well as in the transition to a chronic form of the disease. Modeling viral infection requires specialized experimental conditions. Our work describes techniques for investigating mnemonic functions, tiredness, attentional focus, quick-wittedness, and basic behav

Virological reactivation of the hepatitis B virus after simplification of antiretroviral treatment.

INTRODUCTION: Between 5% and 10% of people living with HIV (PLWH) have chronic hepatitis B virus (HBV) infection. Withdrawal of anti-HBV agents during antiretroviral therapy (ART) simplification may increase the risk of viral reactivation. METHODS: We report two cases of PLWH with serological evidence of past HBV infection (isolated anti-HBc positivity) who developed HBV reactivation after switching to dual ART regimens lacking anti-HBV activity. RESULTS: A 63-year-old woman developed detectable HBV DNA (659 IU/mL) one month after starting cabotegravir/rilpivirine, and a 55-year-old man show

[HPV-related oropharyngeal cancers: main characteristics and new virological tools].

Human papillomavirus (HPV)-associated oropharyngeal squamous cell carcinomas (OPSCCs), predominantly driven by HPV16, constitute a clinically and biologically distinct entity from HPV-negative OPSCCs. Although these tumors generally exhibit a more favorable prognosis and improved response to standard treatments, approximately 10-25 % of patients experience recurrence following curative therapy. Currently, no validated biomarkers are available to identify patients at risk of relapse. This review describes: (i) the molecular bases distinguishing HPV-positive from HPV-negative OPSCCs; (ii) the he

Add-on efficacy of doravirine in PLWHIV with virological failure and low-level viremia.

BACKGROUND: Doravirine is a non-nucleoside reverse transcriptase inhibitor (NNRTI) with moderate plasma protein binding and good potential for anatomical sanctuary penetration. MATERIALS AND METHODS: Adding doravirine to existing antiretroviral therapy (ART) regimens has led to virological success in two groups of people living with HIV (PLWHIV): those with recent virological rebound (n = 84) and those with prolonged low-level viremia (LLV, n = 39). Doravirine was added without any modification to the other ART drugs. RESULTS: After 6 months, 88% of PLWHIV in the recent failure group had ach

Virological characteristics of the SARS-CoV-2 Omicron EG.5.1 variant.

In middle to late 2023, a sublineage of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) Omicron XBB, EG.5.1 (a progeny of XBB.1.9.2), is spreading rapidly around the world. We performed multiscale investigations, including phylogenetic analysis, epidemic dynamics modeling, infection experiments using pseudoviruses, clinical isolates, and recombinant viruses in cell cultures and experimental animals, and the use of human sera and antiviral compounds, to reveal the virological features of the newly emerging EG.5.1 variant. Our phylogenetic analysis and epidemic dynamics modeling sug

Flow cytometry and immunohistochemistry of the mouse dural meninges for immunological and virological assessments.

The highly vascularized meninges protect the surface of the central nervous system and contain a dense network of immune cells controlling neuroinfection and neuroinflammation. Here, we present techniques for the immunological and virological assessment of mouse dural meninges. We describe steps for immunophenotyping including meninges extraction and digestion, immunostaining, and flow cytometry. We then describe viral assessment upon lymphocytic choriomeningitis virus infection including steps for fixation of the meninges in the skull, whole-mount immunohistochemistry, and confocal imaging. [Viruses present in the environment: virological considerations and examples of their impact on human health].

Animal viruses are present in most human environments. Their viability in these media is very variable and the most important element that conditions this viability is the existence or not of a phospholipid envelope surrounding the nucleocapsid. After some general considerations on the structure of viruses, their multiplication cycle and their resistance to different physico-chemical agents, some examples of the impact of animal viruses present in the environment on human health will be presented. The situations that are related concern recent epidemiological events: circulation of type 2 poli

Abnormal Magnetic Resonance Image Signature in Virologically Stable HIV Individuals.

BACKGROUND: With implementation of combination antiretroviral therapy (cART), changes to brain integrity in people with HIV (PWH) are subtle compared to those observed in the pre-cART era. T1-weighted/T2-weighted (T1w/T2w) ratio has been proposed as a measure of cortical myelin. This study examines T1w/T2w values between virologically controlled PWH and persons without HIV (PWoH). METHODS: Virologically well-controlled PWH (n = 164) and PWoH (n = 120) were compared on global and regional T1w/T2w values. T1w/T2w values were associated with HIV disease variables (nadir and current CD4 T-cell co
