Cognitive training-induced short-term functional and long-term structural plastic change is related to gains in global cognition in healthy older adults: a pilot study.

Computerized cognitive training (CCT) is a safe and inexpensive intervention to enhance cognitive performance in the elderly. However, the neural underpinning of CCT-induced effects and the timecourse by which such neural changes occur are unknown. Here, we report on results from a pilot study of healthy older adults who underwent three 1-h weekly sessions of either multidomain CCT program (n = 7) or an active control intervention (n = 5) over 12 weeks. Multimodal magnetic resonance imaging (MRI) scans and cognitive assessments were performed at baseline and after 9 and 36 h of training. Voxel

Combined home-delivered transcranial direct current stimulation and cognitive training in older adults with mild cognitive impairment due to Alzheimer's disease: a randomized, single-blind, sham-controlled trial.

BACKGROUND: Non-pharmacological interventions such as transcranial direct current stimulation (tDCS) and cognitive training (CT) may benefit cognition in older adults with mild cognitive impairment due to Alzheimer’s disease (MCI-AD), yet evidence on combined, home-delivered protocols remains limited. This study evaluated the effectiveness of a combined home-delivered combined tDCS and CT intervention across multiple cognitive domains in individuals with MCI-AD. METHODS: We conducted a randomized, single-blind, sham-controlled trial in 41 participants (≥ 65 years) with MCI-AD, allocated to fou

Transcranial direct current stimulation plus cognitive training for cognitive symptoms in patients with post-acute sequelae of SARS-CoV-2 infection: A randomized, double-blind, sham-controlled trial.

BACKGROUND: Post-acute sequelae of SARS-CoV-2 infection (PASC) is characterized by persistent cognitive deficits alongside anxiety and depression symptoms that adversely affect quality of life. Cognitive training (CT) programs and non-invasive neuromodulation, specifically transcranial direct current stimulation (tDCS), have each shown promise for alleviating similar deficits in non-clinical populations. However, their combined efficacy has not yet been evaluated in PASC patients. Therefore, this study aimed to determine whether the combination of CT and tDCS produces benefits for cognitive an

Integrated bio-cooperative robotic platform for virtual cognitive training in Parkinson's disease: design and methodology of the OPERA project.

INTRODUCTION: Mild cognitive impairment in Parkinson's disease (PD-MCI) can affect several cognitive domains, including attention, working memory, executive functions, language, visuospatial skills, and episodic memory, resulting in a progressive reduction of autonomy and an increased risk of dementia. Cognitive training may help preserve cognitive abilities, especially when supported by innovative tools; nevertheless, standardized and engaging interventions are still lacking. The OPERA project aims to develop and evaluate the usability of PRoBio, a novel bio-cooperative platform that integrat

Prefrontal and hippocampal microstructural gray matter following cognitive training under moderate hypoxia in mood disorders: a randomized controlled trial.

BACKGROUND: Cognitive impairment persists during partial or full remission in 50-70% of individuals with mood disorders and impacts daily functioning and clinical prognosis. Preclinical evidence suggests that extended exposure to moderate hypoxia, combined with motor-cognitive learning, may elevate neuroplasticity and improve cognition. In these individuals with remitted mood disorders, we found that cognitive training under repeated moderate normobaric hypoxia improved executive function, and here investigate neurobiological mechanisms. METHODS: Participants with major depressive disorder (M

Feasibility of Internet-Based Mind-Body Training for Adults With Subjective Cognitive Decline: Protocol for a Randomized Controlled Trial.

BACKGROUND: In the United States, the prevalence of Alzheimer disease (AD) is projected to double over the next 30 years, with associated familial and societal costs estimated at US $1 trillion annually if current trends continue. Although pharmacological treatments of AD are showing promise, the adoption of healthy lifestyle behaviors, particularly during the preclinical phase of AD, may reduce dementia rates by up to 45%. Subjective cognitive decline (SCD), defined as persistent self-perceived declines in cognitive functioning compared with previously normal cognitive abilities, has been ide

Comparing skill transfer and cognitive style effects across three laparoscopic training modalities: a prospective randomized study in medical students.

BACKGROUND: Simulation-based training is an important component of modern surgical education. While virtual reality (VR) simulators, box trainers, and serious games are all used in laparoscopic training, comparative data on their effectiveness, transferability, and the role of individual cognitive learning styles remain limited. METHODS: In this prospective, randomized study, 80 medical students without prior laparoscopic experience were assigned to one of four groups: VR simulator, box trainer, serious game, or control. Each group received four one-hour training sessions. Cognitive style was

Bridging the Gap Between IND Safety Reporting and Routine Clinical Practice: Operational, Cognitive, and Training Challenges at Investigative Sites.

PURPOSE: Patient safety event documentation in routine clinical practice and safety reporting for clinical studies conducted under Investigational New Drug (IND) regulations each operate within established but fundamentally different frameworks. Each framework has distinct documentation objectives, reporting pathways, terminology, and accountability structures, and the 2 frameworks lack sufficient overlap to allow site staff to move and transition fluidly between them. This commentary distinguishes IND safety reporting obligations from routine clinical documentation, identifies common training

Brief Audiovisual Feedback Training Yields Long-Lasting Gains in Multisensory Integration and Speech Comprehension: Contributions of Different Cognitive Abilities.

The ability to integrate information from different sensory modalities, such as vision and hearing, enables us to perceive and understand complex stimuli. The relevance of this ability can be seen in speech perception, for example, which requires the integration of auditory (e.g., voice) and visual (e.g., lip movements) information. Recent studies have shown that the accuracy of this integration process, and thus speech comprehension, can be improved by brief audiovisual feedback training. However, the long-term stability as well as associated cognitive mechanisms are not yet fully understood.

Task-specific physiological signatures of perceptual and cognitive workload under cockpit noise: implications for pilot state monitoring and training.

Cockpit noise is a common operational stressor that can affect pilots' mental workload and performance. This study examined whether multimodal physiological measures could show task-demand-dependent physiological signatures under cockpit-noise interference, and whether these responses are moderated by flight experience. Eighty-five valid participants from an initial sample of 100 pilots performed a Stroop task and a logical reasoning task under no-noise and 85-90 dB(A) noise conditions. Eye-tracking measures and ECG-derived heart rate were recorded. Noise exposure was associated with increased

Improved cognitive flexibility after high-intensity interval training (HIIT) in individuals with Parkinson's disease: a pilot randomized controlled trial.

BACKGROUND: People with Parkinson's disease (PwPD) commonly experience impairments in both motor and cognitive domains, particularly executive function and cognitive flexibility. While aerobic exercise may benefit non-motor domains, it remains unclear whether high-intensity interval (HIIT) provides superior cognitive benefits compared to intensive continuous endurance training (CT). This randomized pilot study investigated whether HIIT on a stationary bike leads to greater improvements in cognitive performance in PwPD than high-intensity endurance training on a stationary bike, with the aim of

Cognitive training and promoting a healthy lifestyle in isolated REM sleep behavior disorder: The randomized controlled trial CogTrAiL-RBD.

INTRODUCTION: Isolated rapid eye movement (REM) sleep behavior disorder (iRBD) is a prodromal phase of Lewy body diseases and linked to a high risk for cognitive decline. Therefore, iRBD provides a suitable window for early intervention. METHODS: In the monocentric randomized controlled trial "Cognitive Training & a Healthy, Active Lifestyle Program for People with Isolated REM Sleep Behaviour Disorder" (CogTrAiL-RBD), the feasibility and behavioral effects of a 5-week digital, adaptive, multidomain cognitive training alongside a module promoting a healthy lifestyle (INT) were compared to a w

Online Group-Based Dual-Task Training to Improve Cognitive Function of Community-Dwelling Older Adults: Randomized Controlled Feasibility Study.

BACKGROUND: Cognitive training for older adults is crucial before cognitive impairment emerges. During periods of social distancing like the COVID-19 pandemic, cognitive stimuli are lacking. Online dual-task training is proposed as a solution to address these needs. OBJECTIVE: We aimed to explore the feasibility, acceptance, and potential effects of online group-based dual-task training as an intervention for enhancing cognitive function among community-dwelling older adults. METHODS: A randomized controlled feasibility study was conducted with 76 participants in Hong Kong, randomly assigned

Cognitive training for Alzheimer's disease and other forms of dementia: Insights from a systematic review and Bayesian meta-analysis.

BackgroundThe prevalence of individuals living with mild cognitive impairment (MCI), Alzheimer's disease (AD), and other forms of dementia is globally increasing. Four out of nine international clinical guidelines recommend non-pharmacological cognitive interventions to enhance cognition, independence, and wellbeing. However, the effectiveness of cognitive rehabilitation (CR) and cognitive training (CT) for individuals with MCI and AD and other forms of dementia is still debatable, often due to significant heterogeneity among studies.ObjectiveThis study aims to assess the effectiveness of CT a

High-intensity interval training improves cognitive dysfunction in chronically stressed mice through alleviating homocysteine-induced transcriptional repression of Cldn5.

Chronic stress-induced blood-brain barrier (BBB) dysfunction contributes to neurological disorders, with homocysteine (HCY) as a key risk factor. Considering pharmacotherapy limitations, non-invasive interventions like high-intensity interval training (HIIT) are promising. To determine whether HIIT improves stress-induced BBB dysfunction and cognitive impairment, we first established a chronic unpredictable mild stress (CUMS) model and assigned mice into four groups: Control (Ctrl), CUMS, HIIT, and HIIT + CUMS. Here, we found that HIIT significantly ameliorated cognitive impairment in male CUM
