Decreased serum angiotensin converting enzyme in adult respiratory distress syndrome associated with sepsis: a preliminary report.

Serum angiotensin converting enzyme (ACE) levels were obtained in 24 control patients who were critically ill, in 11 patients with cardiogenic pulmonary edema, in 8 patients with status postcardiopulmonary bypass, and in 12 patients with adult respiratory distress syndrome (ARDS). Mean values in cardiogenic pulmonary edema (24.3 +/- 3.9 SD) in cardiopulmonary bypass (19.5 +/- 3.1) and in patients with ARDS and no sepsis (n = 7, 19.0 +/- 5.5) were not significantly different from controls (20.7 +/- 2.8). In contrast, patients with ARDS and sepsis had markedly decreased serum ACE levels which fe

Value of measuring serum angiotensin I converting enzyme and serum lysozyme in the management of sarcoidosis.

Serum angiotensin I converting enzyme (ACE) and lysozyme have been measured in 23 controls, 115 patients with sarcoidosis, and 64 with other chest diseases. Both enzymes were significantly raised in sarcoidosis. ACE was raised above the normal range in 21 of 72 (29%) patients with definite sarcoidosis and in 17 of 38 (45%) of those who were untreated and seen within one year of presentation. The rise discriminated usefully between those with stable and progressive disease (5% and 62% respectively). Lysozyme was raised in 50 of 72 (69%) patients with sarcoidosis but also in 11 of 54 (20%) patie

Elevation of angiotensin-converting enzyme in granulomatous lymph nodes and serum in sarcoidosis: clinical and possible pathogenic significance.

A statistically highly significant elevation of serum ACE was found in a group of 58 patients with sarcoidosis (serum ACE was elevated in 34% of patients), as compared with normal controls and patients with tuberculosis and various other common diseases. The results suggest that serum ACE is a useful aid for the diagnosis of sarcoidosis when elevated, but that a normal value does not rule out the condition and may occur in more than one-half of monitored patients. There is a trend to diminution of serum ACE with increasing duration of disease with or without steroid therapy, perhaps correlatin

Diagnostic value of angiotensin-converting enzyme levels for assessing organ involvement in sarcoidosis: A retrospective single-centre study.

BACKGROUND: Angiotensin-converting enzyme (ACE) is the most widely studied biomarker in sarcoidosis; however, its association with epidemiology and multi-organ involvement remains unclear. The aim of our study was to analyse whether elevated ACE levels are associated with epidemiological factors, as well as with pulmonary, extrathoracic, and multi-organ involvement in patients with sarcoidosis. METHODS: A retrospective single-centre study was conducted on patients diagnosed with sarcoidosis between 2000 and 2024. Multi-organ disease was defined as involvement of three or more organs, and elev

Recurrent Hemorrhagic Stroke in a Patient with Neurosarcoidosis.

A 45-year-old Japanese woman had sudden-onset subarachnoid hemorrhage and subsequently developed recurrent intracerebral hemorrhages and multiple infarctions in the left and right cerebral hemispheres, respectively. Angiotensin-converting enzyme (ACE) levels in the cerebrospinal fluid (CSF) were elevated, and a pathological examination of brain biopsy samples demonstrated multinucleated giant cell infiltration and epithelial granuloma formation in thickened pial vessel walls. After treatment with intravascular methylprednisolone 1,000 mg for 3 days, followed by oral prednisolone, she had no re

Chemical analysis and angiotensin I-converting enzyme inhibitory activity of enzymatic hydrolysates derived from meat of goat-kids with supplemental selenium.

BACKGROUND: The effects of selenium (Se) on animal health due to its antioxidant and immune system regulatory properties are very well-documented. However, there is still a lack of scientific evidence about the effect of Se on muscle tissue. Se supplementation in ruminants will enhance the antioxidant activity of myocytes and increase angiotensin-converting enzyme (ACE) inhibitory activity. Generating bioactive peptides derived from meat could prevent the production of angiotensin II, a key player in the development of cardiovascular diseases. METHODS: Forty-five suckling goat kids were rando

Identification of novel ACE inhibitory peptides from Nannochloropsis oculata through peptidomics, in silico screening and molecular docking.

The protein of Nannochloropsis oculata, a single-cell marine microalga, was hydrolyzed by different enzymes. The low-molecular-weight fraction of the trypsin hydrolysate exhibited the best angiotensin-converting enzyme inhibitory (ACEI) activity and possessed diverse peptide sequences. After in silico screening and in vitro verification, three novel high-activity peptides were identified. Among them, GPGPFTVF exhibits the lowest IC50 value (39.77 ± 1.42 μM), followed by PGPAIF (52.03 ± 2.19 μM) and WDPLGF (107.96 ± 4.84 μM). Molecular docking shows that these peptides are bound to ACE via the

Mechanistic insights into angiotensin-converting enzyme inhibition by Apium graveolens through an integrative in silico-in vitro study.

Angiotensin-converting enzyme (ACE) plays a central role in the renin-angiotensin system (RAS) and contributes to hypertension, a multifactorial disorder characterized by chronically elevated arterial pressure. Hypertension affects over one billion people worldwide and is a leading cause of cardiovascular morbidity and mortality. Although synthetic ACE inhibitors are effective, their long-term use is often limited by side effects such as cough and dizziness. Apium graveolens (AG) has been traditionally used to manage cardiovascular disorders, hyperlipidemia, and asthma, and is known for its an

Olive-derived bioactive combinations: synergistic angiotensin-I-converting enzyme (ACE) inhibition and distinct mechanisms of triterpenic acids and phenolics.

BACKGROUND: Cardiovascular diseases, largely driven by hypertension, remain the leading cause of mortality worldwide. Angiotensin-I-converting enzyme (ACE) inhibition is a well-established therapeutic approach; however, synthetic inhibitors are frequently associated with adverse effects. Natural bioactive compounds, particularly those derived from olives, have been proposed as potential alternatives or complementary agents. RESULTS: This study evaluated the ACE-inhibitory potential of olive-derived triterpenic acids (oleanolic acid (OA) and maslinic acid (MA)) and phenolic compounds (hydroxyt

Impact of Lactiplantibacillus plantarum and casein fortification on angiotensin converting enzyme inhibitory peptides in yogurt: identification and in silico analysis.

In this study, the effects of Lactiplantibacillus plantarum M11 (Lb. plantarum M11) in conjunction with sodium caseinate on the characteristics and angiotensin converting enzyme (ACE) inhibitory activity of yogurt were investigated. ACE inhibitory peptides (ACEIPs) in yogurt were identified by nano-LC-MS/MS and potential ACEIPs were predicted by in silico and molecular docking methods. The results showed that the ACE-inhibitory activity of yogurt was significantly enhanced (p < 0.05), while maintaining the quality characteristics of the yogurt. Thirteen ACEIPs in the improved yogurt (883 + M11

Proteomic Analysis of Human Macrophages Overexpressing Angiotensin-Converting Enzyme.

Angiotensin converting enzyme (ACE) exerts strong modulation of myeloid cell function independently of its cardiovascular arm. The success of the ACE-overexpressing murine macrophage model, ACE 10/10, in treating microbial infections and cancer opens a new avenue into whether ACE overexpression in human macrophages shares these benefits. Additionally, as ACE inhibitors are a widely used antihypertensive medication, their impact on ACE expressing immune cells is of interest and currently understudied. In the present study, we utilized mass spectrometry to characterize and assess global proteomi

Effect of Various Fruit Extracts on Angiotensin I-Converting Enzyme (ACE) and Kallikrein (KLK) Activities.

Hypertension is one of the main risk factors for cardiovascular disease and causes widespread morbidity and mortality worldwide. The aim of this work was to screen the fruit with high angiotensin I-converting enzyme (ACE) inhibitory activity and kallikrein (KLK) promotion activity by three different extraction methods from 22 kinds of fruits. Results showed that the aqueous extracts of fresh kiwifruit significantly inhibited ACE activity (47.71%), whereas the KLK activity was also inhibited (4.56%). This indicated that the substances inhibiting ACE activity existed in kiwifruit might be small

The Impact of the Angiotensin-Converting Enzyme Inhibitor Lisinopril on Metabolic Rate in Drosophila melanogaster.

Evidence suggests that angiotensin-converting enzyme inhibitors (ACEIs) may increase metabolic rate by promoting thermogenesis, potentially through enhanced fat oxidation and improved insulin. More research is, however, needed to understand this intricate process. In this study, we used 22 lines from the Drosophila Genetic Reference Panel to assess the metabolic rate of virgin female and male flies that were either fed a standard medium or received lisinopril for one week or five weeks. We demonstrated that lisinopril affects the whole-body metabolic rate in Drosophila melanogaster in a genoty

Deepstack-ACE: A deep stacking-based ensemble learning framework for the accelerated discovery of ACE inhibitory peptides.

Identifying angiotensin-I-converting enzyme (ACE) inhibitory peptides accurately is crucial for understanding the primary factor that regulates the renin-angiotensin system and for providing guidance in developing new potential drugs. Given the inherent experimental complexities, using computational methods for in silico peptide identification could be indispensable for facilitating the high-throughput characterization of ACE inhibitory peptides. In this paper, we propose a novel deep stacking-based ensemble learning framework, termed Deepstack-ACE, to precisely identify ACE inhibitory peptide

Experimental and clinical data analysis for identification of COVID-19 resistant ACE2 mutations.

The high magnitude zoonotic event has caused by Severe Acute Respitarory Syndrome CoronaVirus-2 (SARS-CoV-2) is Coronavirus Disease-2019 (COVID-19) epidemics. This disease has high rate of spreading than mortality in humans. The human receptor, Angiotensin-Converting Enzyme 2 (ACE2), is the leading target site for viral Spike-protein (S-protein) that function as binding ligands and are responsible for their entry in humans. The patients infected with COVID-19 with comorbidities, particularly cancer patients, have a severe effect or high mortality rate because of the suppressed immune system. N

Simple, Fast and Convenient Magnetic Bead-Based Sample Preparation for Detecting Viruses via Raman-Spectroscopy.

We introduce a magnetic bead-based sample preparation scheme for enabling the Raman spectroscopic differentiation of severe acute respiratory syndrome coronavirus type 2 (SARS-CoV-2)-positive and -negative samples. The beads were functionalized with the angiotensin-converting enzyme 2 (ACE2) receptor protein, which is used as a recognition element to selectively enrich SARS-CoV-2 on the surface of the magnetic beads. The subsequent Raman measurements directly enable discriminating SARS-CoV-2-positive and -negative samples. The proposed approach is also applicable for other virus species when t

Blood pH and COVID-19.

The coronavirus disease 2019 (COVID-19) pandemic is a worldwide war. Raising the blood pH might be a crucial strategy to chase COVID-19. The human blood is slightly alkaline, which is essential for cell metabolism, normal physiology, and balanced immunity since all of these biological processes are pH-dependent. Varieties of physiologic derangements occur when the blood pH is disrupted. Severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) proliferates in acidic blood that magnifies the severity of COVID-19. On the other side, blood acidemia is linked to increased morbidity and mortalit

Targeted protein S-nitrosylation of ACE2 inhibits SARS-CoV-2 infection.

Prevention of infection and propagation of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is a high priority in the Coronavirus Disease 2019 (COVID-19) pandemic. Here we describe S-nitrosylation of multiple proteins involved in SARS-CoV-2 infection, including angiotensin-converting enzyme 2 (ACE2), the receptor for viral entry. This reaction prevents binding of ACE2 to the SARS-CoV-2 spike protein, thereby inhibiting viral entry, infectivity and cytotoxicity. Aminoadamantane compounds also inhibit coronavirus ion channels formed by envelope (E) protein. Accordingly, we developed
