Cholangiocarcinoma (CCA) is a fatal disease with high resistance to anticancer drugs. This is probably in part due to enhanced resistance to apoptosis. We have previously shown that galectin-3 (Gal-3), a beta-galactoside-binding lectin, is highly expressed in CCA tissues. In this study, we demonstrated further that Gal-3 plays a direct role in anti-apoptosis regardless of the apoptotic insults. The anti-apoptotic activity and chemoresistance of CCA cells were related to Gal-3 expression level. Suppression of Gal-3 expression with siRNA stimulated apoptosis. siGal-3-K626 transiently depleted Ga

Galectin-3, a beta-galactoside-binding lectin, is a multifunctional protein implicated in a variety of biological functions, including tumour cell adhesion, proliferation, differentiation, cancer progression and metastasis. The present study was performed to clarify the impact of galectin-3 expression on patients with liver fluke-associated cholangiocarcinoma. Galectin-3 expression was examined immunohistochemically in 53 patients with intrahepatic cholangiocarcinoma, who had undergone surgery without pre-operative therapy. All bile duct epithelium expressed galectin-3 with different intensiti

One of the major causes of neuronal death in neurodegenerative disease is excitotoxicity from the neurotransmitter glutamate. This form of cell death could arise from either excess levels of glutamate due to decreased astrocyte clearance or due to increased susceptibility. We have identified galectin-1, a galactose-binding lectin, as a potential neuroprotective factor secreted by astrocytes. Our results show that both native and recombinant galectin-1 protects mouse and rat cerebellar neurons from the toxic effects of glutamate. Galectin-1 applied to neurons increased their expression of the N

Galectin-3 has been found recently to be neo-expressed in thyroid cancers compared to benign lesions and it could therefore be considered a marker of malignancy. We report the case of a 36-year-old woman with a history of occasional dysphagia and dysphonia first observed in November 2000. Thyroid ultrasound scan revealed two subcentimetric nodules in the right lobe. The larger one underwent ultrasound-guided fine-needle aspiration cytology (FNAC). Due to the nature of the lesion, only a few cells could be collected. Their morphological aspect was suspicious but not conclusive for malignancy, t

Sezary cells, the malignant T cells in mycosis fungoides/Sezary syndrome, resist a variety of apoptosis-inducing agents, a feature that contributes to the poor response to therapy in mycosis fungoides. Galectin-1 is a mammalian lectin that triggers T cell apoptosis. For T cells to be susceptible to galectin-1-induced apoptosis, the T cells must express specific glycoprotein receptors, such as CD7, that bear the specific oligosaccharides recognized by galectin-1. Because Sezary cells are characteristically CD7(-), lack of CD7 expression has been proposed to render Sezary cells resistant to gale

Galectin 3 is a beta-galactoside binding protein which localizes to the cytoplasm of proliferative, mature, and hypertrophic chondrocytes in the growth plate cartilage of developing long bones. To elucidate the function of galectin 3 during bone development, we examined the epiphyseal femurs and tibias of fetal mice carrying a null mutation for the galectin 3 gene. Detailed histological and ultrastructural studies identified abnormalities in the cells of the proliferative, mature, and hypertrophic zones and in the extracellular matrix of the hypertrophic zone, as well as a reduction in the tot

Galectins, beta-galactoside-binding lectins, are extensively distributed in the animal kingdom and share some basic molecular properties. Galectin-3, a member of this family, is generally associated with differentiation, morphogenesis, and metastasis. In this study, galectin-3 was isolated from ovine placental cotyledons round the middle of the gestation period by lactose extraction followed by affinity chromatography on lactosyl-agarose, and separated from galectin-1 by size exclusion chromatography on a Superose 12 column. Under native conditions this lectin behaved as a monomer with an appa

BACKGROUND: Delayed graft function (DGF) is a major early complication after kidney transplantation. Galectin-3 (Gal3), a mediator of inflammation and fibrosis, is associated with renal injury, but its role in predicting DGF remains unclear. This study evaluated whether early postoperative Gal3 levels predict DGF and clinical outcomes.
METHODS: This single-center retrospective cohort study included adult kidney transplant recipients from deceased donors (January 2021-December 2022). Gal3 was measured preoperatively, at intensive care unit (ICU) admission, and at ICU discharge. The primary out

BACKGROUND/AIM: Acute lymphoblastic leukemia (ALL) is the most common pediatric malignancy. Among its genetic subtypes, KMT2A (formerly MLL)-rearranged ALL (KMT2Ar-ALL) is associated with poor outcomes. Immunotherapy approaches are being studied and used in KMT2Ar-ALL; however, there is evidence that leukemic cells can escape immune control. Our previous study identified Galectin-1, an immune checkpoint protein, as highly expressed in KMT2Ar-ALL, suggesting it as a potential therapeutic target. To date, the exact mechanism of Galectin-1 regulation in KMT2Ar-ALL is unknown. Therefore, the prese

Galectin-3 (Gal-3) is a carbohydrate-binding protein which plays crucial roles in inflammation, immune response, and cell signaling. At the ocular surface, Gal-3 is also known to crosslink transmembrane mucins to support barrier function. However, the biological role of Gal-3 in circulating tear fluid remains largely unexplored. Similarly, whether Gal-3 engages extracellular glycoprotein ligands in tears to affect biological processes is unknown. Herein, we combine lectin ELISA, affinity enrichment, mass spectrometry (MS)-based glycoproteomics, and lectin blotting to uncover Gal-3 interactors

To generate drug-targeting agents with a modular and efficient design for disease treatments, including cancer, we used protein coil-coiled dimerization motifs to eliminate the need for chemical conjugation techniques, generating a versatile theranostic system: a targeting moiety genetically fused to an E3 coil and a fusion protein of maltose-binding protein and a monomeric streptavidin that includes a K3 coil. The system can be directed to other targets by changing the targeting moiety. The second component enables simultaneous binding to two different ligands (maltosylated or biotinylated).

BACKGROUND: Refractory epilepsy in children remains a major clinical challenge, often resistant to standard antiseizure therapies. Identifying novel biomarkers may open new avenues for understanding disease mechanisms and predicting treatment response. This study investigated serum Galectin-3 (Gal-3) and cadherins as potential biomarkers in pediatric epilepsy.
METHODS: We analyzed serum samples from 30 children with refractory epilepsy, 33 patients on antiseizure drug monotherapy (valproic acid, levetiracetam, carbamazepine), and 66 age- and sex-matched healthy controls. Commercial ELISA assa

Developing high-affinity drug lead critically depends on the efficient structural optimization of a lead compound. However, the dynamic nature of the process by which drugs bind to target molecules often complicates the rational design of effective substituents, even if their three-dimensional structure is available. Herein, we report a streamlined strategy for the optimization of a galectin-3 binder through the integration of in situ chemistry and native mass spectrometry. This method involves the simultaneous synthesis of multiple analogues in a single reaction vessel, followed by the direct

Burn injuries often lead to hypertrophic scars (HTS), which negatively affect quality of life and skin function. Hypertrophic scars result from abnormal wound healing characterized by excess, disorganized collagen confined to the original wound. Fractional ablative CO₂ laser revision (FLSR) has shown promise in improving burn scar symptoms by inducing micro-wound healing, but its specific mechanism remains unclear. Progress is limited by a lack of standardized, objective metrics to evaluate treatment response. We hypothesized that FLSR would shift HTS collagen structure toward that of normal s

BACKGROUND/OBJECTIVES: Acute intravascular hemolysis is associated with the release of labile heme, which contributes to systemic inflammation and organ dysfunction. Galectin-3 (Gal-3) is a known modulator of inflammatory responses. However, its specific role in heme-induced organ injury remains to be fully elucidated.
METHODS: We used a phenylhydrazine (PHZ)-induced model of acute hemolysis in wild-type (WT) and Gal-3 knockout (KO) mice to investigate the influence of Gal-3 on tissue alterations and the inflammatory response.
RESULTS: Despite equivalent levels of hemolysis and anemia in bot

BACKGROUND: Surfactant Protein D (SP-D) is a critical immunomodulatory collectin maintaining alveolar homeostasis. Obstructive sleep apnea (OSA)-related intermittent hypoxia (IH) disrupts pulmonary surfactant integrity; however, severity-dependent SP-D dynamics remain incompletely characterized. This study explores SP-D as a potential indicator of IH-induced alveolar stress and evaluates whether Galectin-3 (Gal-3) inhibition modulates surfactant homeostasis.
METHODS: Forty adult male Sprague-Dawley rats (8 per group) were randomized to Control (normoxia), Moderate IH (MIH; 15-30 events/hour),
