[Development of gentamicin sulfate sustained release tablet remaining-floating in stomach].

Gentamicin sulfate sustained-release tablet remaining-floating in stomach (G-HBS) was developed based on the hydrodynamically balanced system. The dissolution rate of G-HBS was determined by rotary basket method (100 r/min, 37 +/- 0.5 degrees C, 0.1 mol/L HCl). The release characteristics of G-HBS showed basically first order kinetics with the dissolution rate constant (Kr) of 0.3992 h-1. The mean dissolution time (MDT) of G-HBS was 2.53 h-1. The density of G-HBS was found to have no significant influence on dissolution of G-BHS. The gamma-scintiphoto technique was used to examine the gastric

Recovery of impaired gluconeogenesis in kidney-cortex tubules of gentamicin-treated rabbits.

Rabbits were given gentamicin over a period of 10 days. At 1, 3, 5 and 10 days renal proximal tubules were isolated and glucose synthesis from several substrates was measured. A relationship between the inhibition of renal gluconeogenesis, accompanied by a decline of both pyruvate carboxylase and phosphoenopyruvate carboxykinase (PEPCK) activities, and an increased gentamicin level in kidney-cortex was noticed after 5 days of therapy. Both the rates of glucose formation from various substrates as well as pyruvate carboxylase and the cytosolic PEPCK activity recovered fully within 3 weeks after

Ciprofloxacin monotherapy for acute pelvic infections: a comparison with clindamycin plus gentamicin.

This prospective, randomized controlled study compared the efficacy and safety of ciprofloxacin alone versus a conventional two-drug regimen, clindamycin with gentamicin. The study group included 71 patients hospitalized for pelvic infections such as acute (N = 33) and chronic (N = 8) salpingitis, tubo-ovarian abscesses (N = 11), endometritis (N = 9), septic abortion (N = 3), and other categories (N = 7). Twenty-two of 35 patients on ciprofloxacin and 20 of 36 on clindamycin plus gentamicin had culturable pathogens: gonococci in 28, anaerobes in six, chlamydia in four, and associated pathogens

Photo recall-like phenomenon following the use of cefazolin and gentamicin sulfate.

A twenty-one-year-old woman being treated as an inpatient for a urinary tract infection with cefazolin and gentamicin showed a photodistributed eruption five days into her hospitalization. Photo recall phenomena are well known to occur in patients receiving chemotherapeutic agents after sustaining prior skin damage from radiation therapy or the sun. Similar reports with more commonly used agents are sparse. We report a photo recall-like phenomenon comprised of a similar entity with a different reaction pattern. After reviewing the literature on photo recall phenomena, we compare and contrast o

Inner ear effects of topical gentamicin treatment in patients with Menière's disease.

Several efforts have been made to treat patients with Menière's disease with ototoxic drugs. Lately the aminoglycoside antibiotic, gentamicin, has been favored. Several reports have proved its efficacy. In a series of 12 patients treated at the Karolinska Hospital in Stockholm, two patients failed to show improvement regarding their vertigo in spite of repeated courses of treatment. These patients both underwent labyrinthectomy where their vestibular sensory epithelia as well as portions of the cochlea were removed. The findings in these two cases were quite unanimous in that the vestibular se

High-level chromosomal gentamicin resistance in Streptococcus agalactiae (group B).

This is the first report of high-level gentamicin resistance in a group B streptococcus. Strain B128 of serotype II was isolated from an infected leg wound in 1987. B128 was resistant to high levels of gentamicin as well as of all other available aminoglycosides and was also resistant to tetracyclines. No bactericidal synergism was found between ampicillin or vancomycin and any of these aminoglycosides. Gentamicin, kanamycin, streptomycin, and tetracycline resistance determinants transferred by conjugation into a plasmid-free group B streptococcus recipient at a frequency of 10(-8) to 10(-9) t

Differential susceptibility to gentamicin ototoxicity between albino and pigmented guinea pigs.

The known chemical affinity of melanin pigment for aminoglycoside antibiotics has led to the suggestion that higher concentrations of these drugs will bind to the pigmented inner ear and produce greater ototoxicity compared to the nonpigmented albino cochlea. Although this has provided a compelling hypothesis, results from the few investigations to address this question have been equivocal. In the present study, cochlear microphonic (CM) thresholds were recorded from albino and pigmented guinea pigs both before and two weeks after exposure for 14 consecutive days to 100 mg/Kg gentamicin. Effect of glucarolactam on low-dose nephrotoxicities of dibekacin and gentamicin in Fischer rats.

Intramuscular administration of dibekacin or gentamicin to Fischer rats at doses of 10, 20 and 40 mg/kg for 11 days gave polyuria, enzymuria and cytauria. These changes were suppressed completely or partially by the simultaneous administration of glucarolactam sodium or potassium salt. The ameliorating results were supported by serum and histopathological analyses on day 12. A rise in BUN and serum creatinine levels was partially or completely suppressed and histopathological scores were improved by co-administration of glucarolactam salts.

Effect of parathyroid hormone on gentamicin plasma membrane binding and tissue accumulation.

Reportedly, the initiating event in the renal uptake of gentamicin is its binding to anionic, plasma membrane phospholipids. Because parathyroid hormone is known to affect phospholipid metabolism, the plasma membrane binding and tissue accumulation of gentamicin were examined as a function of the parathyroid hormone status of the animal. The experiments were conducted by evaluating the parameters in isolated brush border and basolateral membranes from control, hyper- and hypoparathyroid rats. Scatchard analysis revealed that [125I]gentamicin bound with equal affinity to either membrane to a si

[Synergism of ciprofloxacin with beta-lactam antibiotics, gentamicin, minocycline and pipemidic acid].

Studies on the Synergism of Ciprofloxacin with beta-Lactam Antibiotics, Gentamicin, Minocycline and Pipemidic Acid. Using Escherichia coli, Klebsiella pneumoniae/Klebsiella oxytoca, Pseudomonas aeruginosa and Staphylococcus aureus strains, we examined a possible influence of pipemidic acid, minocycline, gentamicin, cefazolin, mezlocillin and ampicillin on the antibiotic activity of ciprofloxacin. We found in all bacterial species synergistic influence by pipemidic acid. When ciprofloxacin was combined with gentamicin, synergism was observed in E. coli and in K. pneumoniae/oxytoca, additive eff

The killing effects of cefathiamidine or ampicillin alone and in combination with gentamicin against enterococci.

The in-vitro activity of cefathiamidine against Streptococcus faecalis and Str. faecium were studied in comparison with other beta-lactams. All the 56 strains of Str. faecalis tested were inhibited by 2 mg/l of cefathiamidine. MICs of ampicillin were twofold higher than those of cefathiamidine and MICs of cephaloridine and cephazolin were 8-16-fold higher. No significant inoculum effect on MICs of cefathiamidine could be seen. Two isolates of Str. faecium showed resistance to the four antibiotics. The MBCs of cefathiamidine and ampicillin for ten strains of Str. faecalis showed that the ratios

Gentamicin nephrotoxicity. II. Definition of conditions necessary to induce acquired insensitivity.

Acquired insensitivity to the nephrotoxic effects of gentamicin develops in Fischer 344 rats after 10 to 14 days' treatment after development of histologic acute tubular necrosis in a setting of extensive histologic regeneration. To determine the relative importance of aminoglycoside exposure, necrosis, and regeneration in the induction of insensitivity, we examined the effect on gentamicin toxicity of prior non-aminoglycoside-mediated tubular necrosis, antecedent nonnecrotizing aminoglycoside exposure, and unilateral Nx-induced renal tubular hyperplasia. Pretreatment with potassium dichromate

[The indications for middle ear instillation of gentamicin in Menière's disease (author's transl)].

Menière's disease treated by use of Gentamicin sulphate instillation into the middle ear is reported. This drug is injected via a small plastic tube which is inserted into the middle ear. 4 mgm (0.1 ml) of Gentamicine sulphate are instilled every 5 hours until the labyrinth begins to show a reaction. After this treatment 95 per cent of patients no longer suffer from vertigo and tinnitus and the pressure feeling are favourably influenced. The hearing mainly remains unchanged, but sometimes may even improve.

Effects of hydration on gentamicin excretion and renal accumulation in furosemide-treated rats.

The effect of furosemide on gentamicin excretion and tissue accumulation was studied with clearance techniques in anesthetized rats, at two different infusion rates of saline or Ringer solution. Gentamicin ( approximately 20 mg/kg) was administered by constant intravenous infusion over a period of 3 h. With the low fluid infusion rate, furosemide (25 mg/kg intravenously) caused severe reduction in glomerular filtration rate and diminished urinary output of gentamicin. Serum and renal tissue levels of the antibiotic were significantly elevated. High fluid infusion prevented the decline of the g

Detection of pneumococci in respiratory secretions: clinical evaluation of gentamicin blood agar.

The use of sheep blood agar containing 5 microng of gentamicin per ml has been suggested as a means of selectively isolating Streptococcus pneumoniae from respiratory secretions. We have tested this method, in parallel with standard methods, on 844 respiratory specimens in a clinical laboratory and have confirmed that the yield of pneumococci can be increased approximately 40% by using agar containing gentamicin. However, since the antibiotic suppresses the growth of staphylococci, group A streptococci, and gram-negative bacilli, gentamicin agar cannot be used as a replacement for the standard

The effect of concomitant mercuric chloride and gentamicin on kidney function and structure in the rat.

In order to define the possible effects of gentamicin on the course of experimental acute renal failure, the interaction between gentamicin and mercuric chloride was studied in rats. Acute renal failure was induced with 1 mg. of HgCl2 per kilogram intravenously. When given alone, HgCl2 produced a uniform, reproducible, nonoliguric, acute renal failure with a low mortality rate. Animals receiving gentamicin over the course of HgCl2-induced acute renal failure, in doses sufficient to produce a 1 hour postinjection serum concentration of 10 mug/ml., recovered glomerular filtration in a fashion si
