Dicer-2 processes diverse viral RNA species.

RNA silencing pathways play critical roles in gene regulation, virus infection, and transposon control. RNA interference (RNAi) is mediated by small interfering RNAs (siRNAs), which are liberated from double-stranded (ds)RNA precursors by Dicer and guide the RNA-induced silencing complex (RISC) to targets. Although principles governing small RNA sorting into RISC have been uncovered, the spectrum of RNA species that can be targeted by Dicer proteins, particularly the viral RNAs present during an infection, are poorly understood. Dicer-2 potently restricts viral infection in insects by generati

Environmental concentrations of methoprene and its transformation products after the treatment of Altosid® XR Briquets in the city of Richmond, British Columbia, Canada.

Water runoff from catch basins treated with Altosid® XR Briquets for mosquito larvae control was sampled at 10 storm drainage pump stations along the outskirts of the city of Richmond, British Columbia, Canada after rainfall events in 2006 to determine the residual concentrations of methoprene and transformation products: citronellic acid, methoprene acid, and 7-methoxycitronellic acid. Runoff of prior-to-treatment, posttreatment, and 150-d-after-treatment was collected. No residues were detected in the prior-to-treatment samples. However, methoprene was detected in posttreatment, and citronel

Infectious disease in a warming world: how weather influenced West Nile virus in the United States (2001-2005).

BACKGROUND: The effects of weather on West Nile virus (WNV) mosquito populations in the United States have been widely reported, but few studies assess their overall impact on transmission to humans. OBJECTIVES: We investigated meteorologic conditions associated with reported human WNV cases in the United States. METHODS: We conducted a case-crossover study to assess 16,298 human WNV cases reported to the Centers for Disease Control and Prevention from 2001 to 2005. The primary outcome measures were the incidence rate ratio of disease occurrence associated with mean weekly maximum temperatur

Limited pathogenicity of usutu virus for the domestic goose (Anser anser f. domestica) following experimental inoculation.

Usutu virus (USUV) is a mosquito-borne flavivirus of the Japanese encephalitis virus group, which has been associated with avian mortality in Austria since 2001. The affected birds are predominantly Eurasian blackbirds (Turdus merula). In the present study, the pathogenicity of USUV for domestic geese (Anser anser f. domestica) was investigated. Eleven 2-week-old geese were inoculated intramuscularly with 5 x 10(4) 50% tissue culture infectious dose of USUV strain Vienna-2001 blackbird. No clinical signs were seen during the observation period. Four inoculated and one in-contact geese died wit

Endosymbiotic Wolbachia of parasitic filarial nematodes as drug targets.

The parasitic nematodes Wuchereria bancrofti, Brugia malayi and B. timori cause a dreadful disease in humans known as lymphatic filariasis, which afflicts more than 120 million people worldwide. As per recent epidemiologic estimates on prevalence of W. bancrofti and B. malayi, about 428 million people are at risk, with 28 million microfilaria carriers and 21 million clinical cases spread out in 13 States and 5 Union Territories of India. The Indian subcontinent that comprises Bangladesh, India, Maldives, Nepal and Sri Lanka harbours 50 per cent of the world's lymphatic filarial disease burden.

Membrane permeabilization by small hydrophobic nonstructural proteins of Japanese encephalitis virus.

Infection with Japanese encephalitis virus (JEV), a mosquito-borne flavivirus, may cause acute encephalitis in humans and induce severe cytopathic effects in various types of cultured cells. We observed that JEV replication rendered infected baby hamster kidney (BHK-21) cells sensitive to the translational inhibitor hygromycin B or alpha-sarcine, to which mock-infected cells were insensitive. However, little is known about whether any JEV nonstructural (NS) proteins contribute to virus-induced changes in membrane permeability. Using an inducible Escherichia coli system, we investigated which p

Malaria vaccine protection against intradermal or venous parasites: a randomized phase 2b human challenge trial.

Two licensed vaccines block Plasmodium falciparum malaria sporozoites through anticircumsporozoite protein antibodies. In animal models, intradermal (ID) sporozoites are more readily blocked than intravenous sporozoites. We hypothesized that this complicates human studies, where infectious mosquito bites deliver a mixture of ID and venous sporozoites. Here, to test whether vaccine efficacy varies by route of inoculation, we undertook a phase 2b open, randomized controlled trial, recruiting healthy volunteers in Kenya for randomization to the circumsporozoite protein-based R21/Matrix-M vaccine

Malaria: a review on its current epidemiological status and management strategies.

Malaria remains a global health concern, with Africa bearing the highest global burden of malaria, as Plasmodium falciparum malaria remains the leading cause of malaria-related mortality on the continent. The transmission dynamics of malaria are shaped by a combination of factors, including climate conditions, economic constraints, geographic variability, human activities, and security instability. Owing to repeated infections and widespread implementation of early interventions, there has been a notable rise in cases of clinically atypical malaria and asymptomatic Plasmodium carriers, which i

Spatial exploration of the impact of cattle on risk of malarial infection using data from a cluster randomised trial.

INTRODUCTION: In the context of stalled progress in reducing malaria burden and ongoing efforts to develop novel vector control methods, there is ongoing debate about the impact of livestock on malaria transmission. Zooprophylaxis refers to the use of livestock as an alternate source of blood that diverts mosquito vectors away from humans, potentially reducing malaria transmission. However, conflicting evidence highlights the concept of zoopotentiation, where livestock may instead increase vector populations and enhance malaria transmission. Prior studies that explore these concepts have exami

Forecasting high-risk areas for dengue outbreaks in China: A trend analysis of Aedes albopictus and Aedes aegypti distributions from 2014 to 2030.

BACKGROUND: Global warming, urbanization and the resumption of global population movements post-COVID-19 have made the prevention and control of dengue and its vector transmission more challenging. To tackle this issue, this study evaluated and predicted the suitable habitats of Aedes albopictus and Aedes aegypti in China. This study aims to identify the key influencing factors, analyze patterns of habitat expansion and contraction, and explore regions in China at risk of dengue fever outbreaks in the future. METHODOLOGY/PRINCIPAL FINDINGS: This study utilized mosquito distribution data from

Population genetic analysis of Plasmodium knowlesi reveals differential selection and exchange events between Borneo and Peninsular sub-populations.

The zoonotic Plasmodium knowlesi parasite is a growing public health concern in Southeast Asia, especially in Malaysia, where elimination of P. falciparum and P. vivax malaria has been the focus of control efforts. Understanding of the genetic diversity of P. knowlesi parasites can provide insights into its evolution, population structure, diagnostics, transmission dynamics, and the emergence of drug resistance. Previous work has revealed that P. knowlesi fall into three main sub-populations distinguished by a combination of geographical location and macaque host (Macaca fascicularis and M. ne

Exploring the key anthropological drivers of and barriers to zoonotic malaria preventative behaviour in a community exposed to Plasmodium knowlesi infection in Malaysia: protocol for a qualitative study with a participatory research design.

INTRODUCTION: Plasmodium knowlesi malaria is a zoonotic mosquito-borne disease with complex epidemiology. According to the WHO, the prevention and control of vector-borne diseases require community participation to increase coherence between malaria interventions and sustainable public health programmes. We describe a participatory research (PR) design for a study aimed at exploring the key anthropological drivers of and barriers to zoonotic malaria preventive behaviour among communities exposed to P. knowlesi infection in Malaysia. Participatory approaches can facilitate policymakers in desig

Illuminating the bacterial microbiome of Australian ticks with 16S and Rickettsia-specific next-generation sequencing.

Next-generation sequencing (NGS) studies show that mosquito and tick microbiomes influence the transmission of pathogens, opening new avenues for vector-borne pathogen control. Recent microbiological studies of Australian ticks highlight fundamental knowledge gaps of tick-borne agents. This investigation explored the composition, diversity and prevalence of bacteria in Australian ticks (n = 655) from companion animals (dogs, cats and horses). Bacterial 16S NGS was used to identify most bacterial taxa and a Rickettsia-specific NGS assay was developed to identify Rickettsia species that were ind

Genetic diversity and population structure of the primary malaria vector Anopheles sinensis (Diptera: Culicidae) in China inferred by cox1 gene.

BACKGROUND: Anopheles sinensis is a primary vector for Plasmodium vivax malaria in most regions of China. A comprehensive understanding of genetic variation and structure of the mosquito would be of benefit to the vector control and in a further attempt to contribute to malaria elimination in China. However, there is only inadequate population genetic data pertaining to An. sinensis currently. METHODS: Genetic variations and structure among populations of An. sinensis was examined and analyzed based on the nucleotide sequences of a 662 nt variable region of the mitochondrial cox1 gene among 1
