Extensive use of antibiotics in clinical practice has been associated with increasing frequency of resistant microorganisms. So new strategy is needed to treat the resistant pathogens. Hence this study was conducted to determine the effect of Ethylenediaminetetraacetic acid (EDTA) in increasing the inhibition effect of some antibiotics on multi-drug resistant (MDR) gram-negative bacteria. For this purpose, 40 E. coli isolates, 40 K. pneumoniae isolates and 50 P. aeruginosa isolates were collected from different University's hospitals in Mansoura, Egypt. Antibacterial susceptibility pattern aga…

Respiratory infections in mechanically ventilated patients caused by Gram-negative bacteria are a major cause of morbidity. Rapid and unequivocal determination of the presence, localization, and abundance of bacteria is critical for positive resolution of the infections and could be used for patient stratification and for monitoring treatment efficacy. Here, we developed an in situ approach to visualize Gram-negative bacterial species and cellular infiltrates in distal human lungs in real time. We used optical endomicroscopy to visualize a water-soluble optical imaging probe based on the antim…

INTRODUCTION: Hospital-acquired and ventilator-associated bacterial pneumonia (HABP/VABP) are among the most prevalent infections in hospitalized patients, particularly those in the intensive care unit. Importantly, the frequency of multidrug resistant (MDR) Gram-negative (GN) bacteria as the bacteriologic cause of HABP/VABP is increasing. These include MDR Pseudomonas aeruginosa, Acinetobacter baumannii, and carbapenem resistant Enterobacteriaceae (CRE). Few antibiotics are currently available when such MDR Gram-negatives are encountered and older agents such as polymyxin B, colistin (polymyx

This review discusses the relationship of the efflux pump (EP) system of Gram-negative bacteria to other antibiotic resistance mechanisms of the bacterium such as quorum sensing, biofilms, two component regulons, etc. The genetic responses of a Gram-negative to an antibiotic that render it immune to an antibiotic are also discussed. Lastly, the methods that have been developed for the identification of bacteria that over-express their EP system are presented in detail. Phenothiazines are well-known antipsychotic drugs with reported activity against bacterial EPs and other ancillary antibiotic …

Antimicrobial resistance (AMR) requires environmental surveillance because aquatic systems can receive resistant bacteria, antimicrobial residues and mobile resistance genes from domestic, agricultural, hospital and industrial sources. This culture-based study investigated culturable Gram-negative bacteria recovered from 12 inland, saline, wetland and coastal aquatic sites in western Algeria between January and April 2023. Bacterial isolates were identified by conventional biochemical methods and 16 S rRNA gene sequencing. Antimicrobial susceptibility was assessed by disk diffusion and selecte…

Multidrug-resistant (MDR) Gram-negative bacteria pose significant therapeutic challenges due to their protective outer membranes and evolving resistance mechanisms. Exploiting bacterial iron uptake pathways via siderophore-antibiotic conjugates offers a promising strategy to enhance antibiotic efficacy. Here we synthesized siderophore-aztreonam, and siderophore-daptomycin conjugates and compared their activity to that of cefiderocol against select Gram-negative pathogens, including MDR Acinetobacter baumannii and Pseudomonas aeruginosa. Our in vitro studies demonstrated that siderophore conjug…

The global rise in antimicrobial resistance has increased morbidity and mortality, particularly among patients with multidrug-resistant Gram-negative infections. Colistin, a last-resort therapeutic, has faced a concerning emergence of resistance. Accurate detection is therefore critical in clinical microbiology laboratories. Although broth microdilution (BMD) is the only reference method recommended for colistin susceptibility testing, its labor-intensive process has driven the search for automated alternatives. The VITEK 2 system is widely used in routine diagnostics, but international guidel
