Formaldehyde (HCHO) is an important intermediate in the metabolism of one-carbon (C1) compounds such as methanol, formate, and methane. The ribulose monophosphate (RuMP) pathway is the most-studied HCHO assimilation route and the 3-hexulose-6-phosphate synthase (Hps) plays an important role for HCHO fixation. In this study, we proposed and selected a pyruvate-dependent aldolase to channel HCHO into 2-keto-4-hydroxybutyrate as an important intermediate for biosynthesis. By combining this reaction with three further enzymes we demonstrated a pyruvate-based C1 metabolic pathway for biosynthesis o

High anion gap metabolic acidosis is marked by decreased blood pH and bicarbonate concentration, with normal chloride levels and compensatory hypocapnia. It results from the accumulation of strong acids such as ketoacids, lactic acid (L- and D-isoforms), uremic acids, or toxins like ethylene glycol. Elevated inorganic phosphate may also contribute. Clinical manifestations typically reflect the underlying disorder rather than the acidosis itself. Management focuses on identifying and treating the primary disease process.

peri-Annulation of naphthalane, an important tool for realization of wide range of functional materials, is presently accomplished with limited few functional groups like imide, amide and diamine-derivative (perimidine). To increase the diversity, we have incorporated α-keto aldonitrone as a new functional group, and herein report about five peri-naphthoisatogens (PNTIs) dyes. The synthesis were accomplished using an unusual reaction of aromatic nitro group, which is nucleophilic attack of a C-nucleophile (enol) to the N-atom of nitro group. Although prostacyclin (PGI2) has been well described as a regulator of smooth muscle activity, limited data are available concerning its role in the myometrium of pigs. The present research aimed to examine profiles of PGI2 synthase (PTGIS) and PGI2 receptor (PTGIR) expression and 6-keto PGF1α (a PGI2 metabolite) concentrations in the myometrium of gilts throughout the estrous cycle and during early pregnancy using qPCR, Western blot, and/or ELISA methods. Furthermore, myometrial explants were exposed to iloprost (a stable PGI2 analog) to investigate the effect of PGI2 on the mRNA expression o

The intraerythrocytic apicomplexan Babesia microti is the primary causative agent of human babesiosis, which is an infectious disease that occurs in various regions around the world. Although the aldo-keto reductases (AKRs) of this parasite have been sequenced and annotated, their biological properties remain unknown. AKRs are a superfamily of enzymes with diverse functions in the reduction of aldehydes and ketones. In the present study, we cloned the full-length cDNA of a B. microti aldo-keto reductase-like protein (BmAKR) and analyzed the deduced amino acid sequence of the BmAKR protein. Thi

1. Gas-liquid chromatographic methods were used to measure urinary acidic and alcoholic metabolites of L-DOPA, which had been administered in high oral dosage to patients with postencephalitic and idiopathic Parkinsonism.2. The output of these compounds was normal before treatment. During drug therapy, large quantities of the dopamine metabolites, homovanillic acid and dihydroxyphenylacetic acid, were excreted but traces only of 4-hydroxy-3-methoxyphenylethanol. Noradrenaline metabolites showed little change in output other than a small increase in 4-hydroxy-3-methoxymandelic acid.3. Informati

Testosterone production by testicular Leydig cells (steroidogenesis) is vital to male fertility and overall male health. Information about how nutrition influences Leydig cell steroidogenesis is lacking. Branched chain amino acids (BCAAs - leucine, isoleucine, and valine) are essential amino acids and important regulators of protein synthesis and energy production. Circulating and tissue BCAA levels are tightly regulated by the enzyme branched chain a-keto acid dehydrogenase kinase (BCKDK), which inhibits their catabolism. This work explored how BCAAs, and especially leucine, modulate male fer

The pregnane X receptor (PXR), a key hepatic nuclear receptor, exhibits a highly plastic ligand-binding domain (LBD) that recognizes diverse endogenous and exogenous ligands, contributing to interindividual variations in xenobiotic metabolism and toxic responses. Emerging studies on the gut-liver axis reveal that microbiota metabolites regulate hepatic PXR through dual mechanisms: (1) Direct ligand-receptor interactions, where secondary bile acids (e.g., 3-keto LCA, DCA) and indole-3-propionic acid (IPA) bind PXR-LBD via hydrogen bonding to induce conformational changes, subsequently upregulat

l-Lysine oxidase (LysOX) is a FAD-dependent homodimeric enzyme that catalyzes the oxidative deamination of l-lysine to produce α-keto-ε-aminocaproate with ammonia and hydrogen peroxide. LysOX shows strict substrate specificity for l-lysine, whereas most l-amino acid oxidases (LAAOs) exhibit broad substrate specificity for l-amino acids. Previous studies of LysOX showed that overall structural similarity to the well-studied snake venom LAAOs. However, the molecular mechanism of strict specificity for l-lysine was still unclear. We here determined the structure of LysOX in complex with l-lysine

Lipopolysaccharides of eight wild-type strains of the phototrophic bacterium Rhodospirillum tenue have been analyzed. All of the lipopolysaccharides are highly lipophilic. The compositions of preparations obtained by the phenol-water or by the phenol-chloroform-petroleum ether procedure are very similar. The polysaccharide moiety, obtained by mild acid hydrolysis of lipopolysaccharide, consists mainly of aldoheptoses: L-glycero-D-mannoheptose is present in all strains, whereas D-glycero-D-mannoheptose is an additional constituent in some strains. A method for the acylation of sulfonamides and transacylation of N-acylsulfonamides has been developed. The acylation proceeds smoothly in the presence of Cu(NTf2)2 (0.1 mol%) using carboxylic anhydrides, acyl chlorides, or isopropenyl acetate as acylating agents. Alternative catalysts active in this transformation include Zn(NTf2)2, Mg(NTf2)2, La(NTf2)3, and Ce(NTf2)3. The transacylation of N-acylsulfonamides takes place smoothly at room temperature in excess acyl chlorides in the presence of Cu(NTf2)2 (20 mol%) and furnishes analogous N-acylsulfonamides through exchange of the N-acyl group.

A single-atom catalyst with generally regarded inert Zn-N4 motifs derived from ZIF-8 is unexpectedly efficient for the activation of alcohols, enabling alcohol-mediated alkylation and transfer hydrogenation. C-alkylation of nitriles, ketones, alcohols, N-heterocycles, amides, keto acids, and esters, and N-alkylation of amines and amides all go smoothly with the developed method. Taking the α-alkylation of nitriles with alcohols as an example, the α-alkylation starts from the (1) nitrogen-doped carbon support catalyzed dehydrogenation of alcohols into aldehydes, which further condensed with nit…

Maple syrup urine disease (MSUD) is an autosomal recessive inherited metabolic disorder caused by mutations in any of the genes encoding for the branched-chain keto dehydrogenase (BCKDH) components. This study screened MSUD patients throughout the whole Upper Egypt describing their symptoms, clinical and laboratory findings, genetic studies, and their treatment, with a 6-month follow-up for their responses. Screening identified three children with MSUD. Homozygous mutation in R195Q single nucleotide polymorphism (SNP) within the BCKDHA gene was found with the second MSUD patient. Follow-up for

Individuals with maple syrup urine disease (MSUD) have an inherited metabolic disorder resulting in a deficiency in the branched-chain keto-acid dehydrogenase complex. As a result, these individuals have elevated concentrations of the branched-chain amino acids valine, isoluecine, allo-isoleucine, and leucine. MSUD presents in the first few days of life and progression may lead to irreversible intellectual disability, coma, cerebral edema, and death. However, early diagnosis and intervention can mitigate or eliminate many of the potential adverse effects. Consequently, it is important to devel

BACKGROUND: Alterations in circulating amino acid profiles have been observed in ischemic stroke patients; however, whether cerebral ischemia disrupts amino acid metabolism within brain tissue and whether this disruption contributes to cellular stress and cerebral injury remain unknown. This hypothesis-testing study investigates disrupted BCAA (branched-chain amino acid) catabolism as a key mechanism of ischemic brain damage and evaluates BCKDK (branched-chain α-keto acid dehydrogenase kinase) as a novel therapeutic target.

In vivo, volume-selected 1H NMR spectroscopy employing the SPACE technique was used to monitor biochemical changes in the thiamin deficient rat brain in response to glucose loading. The concentrations of brain N-acetylaspartate, glutamate/glutamine/gamma-aminobutyric acid, lactate and glucose differed significantly from those of control animals. The results are consistent with a metabolic block at the reaction catalyzed by the thiamin dependent enzyme alpha-keto glutarate dehydrogenase soon after the onset of neurological symptoms of thiamin deficiency, and a further block at pyruvate dehydrog
