Retinal vasodilatory responses are inversely associated with plasminogen activator inhibitor-1: The African-PREDICT study.

AIMS: Plasminogen activator inhibitor-1 (PAI-1), traditionally associated with fibrinolysis, is increasingly implicated in impaired vascular function. However, studies on its association with microvascular function are limited to the cutaneous and coronary microvascular beds in older and diseased individuals. To better understand its potential involvement in the early stages of disease development, we investigated the associations of retinal vasodilatory responses to flicker light with PAI-1 activity (PAI-1act) in young and healthy individuals. METHODS: We included healthy Black and White wom

Identification of class III β-tubulin as a marker of angiogenic perivascular cells.

A full understanding of the functional role for pericytes in microvascular network growth requires identifying the specific cell phenotypes involved in angiogenesis. The objective of this study was to evaluate class III β-tubulin expression along remodeling adult rat mesenteric microvascular networks. Mesenteric tissues were harvested from unstimulated adult male Wistar rats and at 2, 10 and 30 days post-compound 48/80 stimulation (n=4 per experimental group). Tissues were immunohistochemically labeled with antibodies for class III β-tubulin, NG2 and PECAM. In unstimulated microvascular networ

Long-Term Oral Administration of Salidroside Alleviates Diabetic Retinopathy in db/db Mice.

Diabetic retinopathy (DR), a microvascular complication of diabetes mellitus, is the leading cause of vision loss in the working-age population worldwide. Unfortunately, current clinical treatments cannot completely prevent the occurrence and development of DR. Salidroside (Sal) is a medicinal supplement that has antioxidative and cytoprotective properties. This study aimed to investigate the therapeutic effect of Sal on DR. Briefly, Sal treatment was applied to wide-type mice and db/db mice (a widely used diabetic mice) at 25 mg/kg by oral gavage once daily from 8 weeks to 20 weeks. Mice's bo

Increased soluble fms-like tyrosine kinase 1 after ischemia reperfusion contributes to adverse clinical outcomes following kidney transplantation.

Renal ischemia reperfusion injury (IRI) adversely affects clinical outcomes following kidney transplantation. Understanding the cellular mechanisms and the changes in gene/protein expression following IRI may help to improve these outcomes. Serum soluble fms-like tyrosine kinase 1 (sFlt-1), a circulating antiangiogenic protein, is increased in the first week following kidney transplantation. We evaluated the casual relationship of elevated sFlt-1 levels with renal microvascular dysfunction following IRI in a longitudinal study of 93 kidney transplant recipients and in several animal models. Biomarkers of Preclinical Diabetic Retinopathy Detected by OCT Angiography-A Descriptive Review.

Background: Diabetic retinopathy (DR) is a leading cause of vision loss worldwide. Microvascular changes precede clinically detectable DR, creating an opportunity for early diagnosis and intervention. Optical coherence tomography angiography (OCTA) enables noninvasive, quantitative assessments of retinal and choroidal microcirculation and has emerged as a promising tool for identifying early biomarkers of DR. The goal of this study was to review the literature on OCTA-derived biomarkers associated with preclinical diabetic retinopathy in patients with type 1 and type 2 diabetes mellitus. Metho

Comparative densitometric study of iliac crest and scapula bone in relation to osseous integrated dental implants in microvascular mandibular reconstruction.

The study was designed to compare dimension and density of four frequently used bone donor regions (the iliac crest, the lateral border of the scapula, the radius and the fibula) with regard to placement of endosseous dental implants in microvascular mandibular reconstruction. The first part of the study reports on the results of bone dimension and density findings for iliac crest and scapula bone. Nearly all of the iliac crests had adequate dimensions for the positioning of four 10 mm implants. In 63% of the scapulae, it was possible to place four 10 mm implants. In the case of the scapula, h

Activation of the Wnt/β-catenin/CYP1B1 pathway alleviates oxidative stress and protects the blood-brain barrier under cerebral ischemia/reperfusion conditions.

Accumulating evidence suggests that oxidative stress and the Wnt/β-catenin pathway participate in stroke-induced disruption of the blood-bra … junction-associated proteins (zonula occludens-1 [ZO-1], occludin and claudin-5), as well as brain microvascular density in cerebral ischemia rats. Moreover, rat brain microvascular endothelial cells that underwent oxygen glucose deprivation/reoxygenation displayed intense oxidative stress, suppression of the Wnt/β-catenin pathway, aggravated cell apoptosis, downregul …

The protective role of intermedin in promoting angiogenesis during renal fibrosis.

BACKGROUND: Renal fibrosis promotes the progression of chronic renal disease to end-stage renal disease. Microvascular damage and loss play an important role in renal fibrosis. Intermedin (IMD) is expressed mainly in the heart and kidney. IMD has been shown to increase renal blood flow and reduce the loss of glomerular and surrounding renal tubules, but its role in mediating microvascular damage in renal fibrosis remains to be elucidated. Here, we investigated the effects of IMD on microvascular damage in a renal fibrosis model. METHODS: We created a rat model of unilateral ureteral obstructi

Assessment of coronary microcirculation alterations in a porcine model of no-reflow using ultrasound localization microscopy: a proof of concept study.

BACKGROUND: Coronary microvascular obstruction also known as no-reflow phenomenon is a major issue during myocardial infarction that bears important prognostic implications. Alterations of the microvascular network remains however challenging to assess as there is no imaging modality in the clinics that can image directly the coronary microvascular vessels. Ultrasound Localization Microscopy (ULM) imaging was recently introduced to map microvascular flows at high spatial resolution (∼10 μm). In this study, we developed an approach to image alterations of the microvascular coronary flow in ex v

A Validation of Spatially Compounded Volumetric Ultrasound Localization Microscopy for Glomerular Imaging With Light Sheet Microscopy.

OBJECTIVE: Glomerular structure and microvascular function are central to kidney health, yet current clinical assessments rely largely on indirect serum markers or invasive biopsy. Imaging approaches capable of resolving glomeruli in vivo across the entire kidney remain limited by spatial and temporal resolution. In this study, we perform whole-kidney ultrasound localization microscopy (ULM) with quantification of glomerular microvascular structure and function, and evaluate its performance against light-sheet microscopy (LSM), an established optical gold standard.
