A phase 1/2 study of an adjuvanted varicella-zoster virus subunit vaccine in autologous hematopoietic cell transplant recipients.

Recombinant herpes zoster (HZ) vaccines may be an alternative to the live-attenuated HZ vaccine for immunocompromised individuals. This was a phase 1/2, randomized, observer-blind, placebo-controlled study in adults with multiple myeloma, non-Hodgkin lymphoma (B- or T-cell), Hodgkin lymphoma, or acute myeloid leukemia who had undergone autologous hematopoietic stem-cell transplant 50 to 70 days earlier. Subjects (N = 121) were randomized 1:1:1:1 to receive (at months 0, 1, 3) three doses of 50 μg varicella-zoster virus glycoprotein E (gE) adjuvanted with AS01B, 3 doses of gE adjuvanted with AS

Subunit vaccine candidate AMM down-regulated the regulatory T cells and enhanced the protective immunity of BCG on a suitable schedule.

Mycobacterium bovis bacillus Calmette-Guérin (BCG) priming and subunit vaccine boosting strategies are urgently needed to improve the protective efficacy of BCG in adult population. However, the schedule of subunit vaccine boosting is not well investigated, especially the optimal immune responses and vaccine immunization schedules are still not clear. We have constructed a novel subunit vaccine candidate consisting of fusion protein Ag85B-Mpt64 (190-198)-Mtb8.4 (AMM) in a complex adjuvant composed of dimo-thylidioctyl ammonium bromide (DDA) and BCG polysaccharide nucleic acid (BCG-PSN). Mucosal targeting of a BoNT/A subunit vaccine adjuvanted with a mast cell activator enhances induction of BoNT/A neutralizing antibodies in rabbits.

BACKGROUND: We previously reported that the immunogenicity of Hcβtre, a botulinum neurotoxin A (BoNT/A) immunogen, was enhanced by fusion to an epithelial cell binding domain, Ad2F, when nasally delivered to mice with cholera toxin (CT). This study was performed to determine if Ad2F would enhance the nasal immunogenicity of Hcβtre in rabbits, an animal model with a nasal cavity anatomy similar to humans. Since CT is not safe for human use, we also tested the adjuvant activity of compound 48/80 (C48/80), a mast cell activating compound previously determined to safely exhibit nasal adjuvant acti

Protection against lethal subcutaneous challenge of virulent Y. pestis strain 141 using an F1-V subunit vaccine.

In this study, we designed and engineered a two-component recombinant fusion protein antigen as a vaccine candidate against the possible biological threat of Yersinia pestis. The recombinant F1-V protein was formulated with Alhydrogel. A four-time injection with a dosage of 10, 20 and 50 microg/mouse in about two months was adopted for vaccination. Serum antibodies and subclass of T helper cells were measured and analyzed. After the final vaccination, the mice were challenged by 141 strain with 25-600 LD(50). In conclusion, the recombinant vaccine was capable of inducing protective immunity ag

Mucosal antitoxic and antibacterial immunity after cholera disease and after immunization with a combined B subunit-whole cell vaccine.

Mucosal and systemic immune responses to a new oral cholera vaccine, consisting of the B subunit plus killed vibrios, were studied in Bangladeshi volunteers and compared with those to clinical cholera. A single peroral dose of vaccine induced a local IgA antitoxin response in intestinal-lavage fluid of seven of eight vaccinees; the response closely mimicked that of patients convalescing from cholera, and evidence of the induction of local immunologic memory was found as well. Two peroral doses were needed for stimulation of an intestinal IgA immune response to the lipopolysaccharide of Vibrio

Distinct intracellular and extracellular maturation of goose astrovirus 2 capsid protein and the potential of virus-like particles as a novel subunit vaccine.

Goose astrovirus 2 (GAstV-2) is the primary causative agent of gosling gout in China, causing severe economic losses, yet no commercial vaccines are available. Virus-like particles (VLPs) represent a promising subunit vaccine platform, but the capsid maturation mechanism and VLP immunogenicity of GAstV-2 remain unknown. Herein, we expressed the GAstV-2 ORF2 capsid protein utilizing the baculovirus/insect cell system and systematically characterized its intracellular and extracellular maturation dynamics. Western blot analysis with domain-specific antibodies revealed distinct maturation pathway

Evaluate of the protective effect of an RSV Pre-F/Alum +CpG subunit vaccine in Balb/C mice with and without RSV prexposure.

Respiratory syncytial virus (RSV) poses a substantial global health threat. Most individuals have been infected with RSV at least once prior to vaccination, resulting in the presence of pre-existing antibodies. In this study, Balb/C mice were either pre‑exposed or non-pre-exposed to RSV on day 0, followed by a two-dose regimen of an RSV subunit vaccine on days 28 and 56. Two prefusion RSV F proteins (Pre-F-1 and Pre-F-2) were designed as vaccine antigens, formulated with Alum + CpG adjuvant to promote a Th1-biased response. Results showed that once pre-exposed to RSV (PC group), a robust immun

A Recombinant Subunit Vaccine Against Chicken Infectious Anemia Virus Elicits Protective Immunity via VP2-Assisted VP1 Refolding.

BACKGROUND: Chicken infectious anemia virus (CIAV) is a globally significant immunosuppressive pathogen that causes substantial economic losses to the poultry industry, with particularly severe outbreaks in China in recent years. Given the limitations of existing vaccines, especially the residual virulence associated with live attenuated vaccines, there is an urgent need to develop novel, safer, and more effective vaccine strategies. METHODS: In this study, the VP1 and VP2 genes of CIAV were cloned and expressed in Escherichia coli to develop a cost-effective subunit vaccine. Since VP1 primar

Immunoinformatic-based design of a multi-epitope subunit vaccine against Ruminococcus torques using subtractive proteomics and molecular dynamics simulations.

Ruminococcus torques is an anaerobic, Gram-positive gut bacterium that has been associated with gastrointestinal diseases including irritable bowel syndrome (IBS) and inflammatory bowel disease (IBD). Its mucolytic effect disrupts the integrity of the intestinal barriers, which are involved in dysbiosis and pathogenicity. Recent clinical evidence has also reported its involvement in extraintestinal infections like pneumonia, keratitis, bacteremia and ocular infection in immunocompromised individuals. Although, R. torques has emerged as a clinically more significant pathogen, no vaccine is yet

Protective efficacy of the optimized GluB-MTAP fusion subunit vaccine against Nocardia seriolae infection in largemouth bass (Micropterus salmoides).

Nocardia seriolae is the predominant causative agent of piscine nocardiosis, exhibiting broad piscine host specificity along with prolonged latency periods and high mortality rates, thereby posing a severe threat to global fish health. However, reliably effective control measures remain limited. Subunit vaccines, derived from virulence factors or immunodominant proteins of the pathogen, are widely acknowledged as one of the most secure and potent strategies for mitigating disease outbreaks. Here we focused on five T7SS gene cluster proteins (MTAP, HP05, HP23, HP24, and VST) and the dominant N.

Augmenting Subunit-Vaccine-Induced Immunity through a Dual Strategy of Gold Nanoparticle Conjugation and Chitosan Microneedle-Mediated Sustained Delivery.

Subunit vaccines offer high safety but often exhibit low immunogenicity and rapid clearance and require adjuvants. In this study, we developed a dual strategy for augmenting subunit-vaccine-induced immune responses by integrating self-adjuvanting gold nanoparticle (GNP)-antigen conjugates with implantable chitosan (CS) microneedles (MNs) to achieve sustained intradermal antigen exposure. Conjugation of a model antigen, namely, ovalbumin (OVA), onto the GNP surface (GNP-OVA) resulted in virus-mimicking multivalent antigen display, which substantially enhanced dendritic cell maturation, as evide

Antigen Format Determines Immunogenicity of AAV-Based SARS-CoV-2 Vaccines: Full-Length Spike Versus Truncated Subunits.

BACKGROUND: Antigen format strongly influences the immunogenicity of gene-based vaccines. Full-length Spike is widely used in licensed COVID-19 vaccines, while truncated subunits such as S1 or the receptor-binding domain (RBD) may simplify vector design but risk reduced potency. We aimed to compare these antigen formats in an AAV9 delivery platform. METHODS: BALB/c mice were immunized intramuscularly with recombinant AAV9 encoding full-length Spike, S1, or RBD at doses of 1 × 1010 or 1 × 1011 viral genomes. Immune responses were assessed by serology, virus neutralization, T-cell profiling, an

Chemical cross-linking facilitates antigen uptake and presentation and provides improved protection from Mpox with a dual-antigen subunit vaccine.

Antigen uptake, processing, and presentation are crucial for the immune responses of protein-based vaccines. Herein, we introduced a reversible chemical cross-linking strategy to engineer protein antigens, which can be tracelessly removed upon antigen-presenting cell (APC) uptake and cellular reduction. The chemically cross-linked antigen proteins presented significantly enhanced uptake and epitope presentation by APC. We applied this strategy to monkeypox virus antigens A29L and A35R to construct dual-antigen subunit vaccines. Our results revealed that chemical cross-linking was robust enough

PRRSV GP4 subunit vaccine combined with adenovirus heterologous prime-boost immunization strategy induced a significant immune response in mice.

BACKGROUND: The porcine reproductive and respiratory syndrome virus (PRRSV) continues to cause widespread infections in the pig industry worldwide. Currently, multiple PRRSV vaccine candidates are in preclinical or clinical trials, and each has different advantages and limitations. Glycoprotein 4 (GP4) is rich in epitopes, which can induce the body to produce neutralizing antibodies, plays a vital role in causing the host immune response, and is a key target for PRRSV vaccine development. In this study, we developed a novel candidate vaccine immunization strategy combining a subunit vaccine wi

Intranasal vaccine combining adenovirus and trimeric subunit protein provides superior immunity against SARS-CoV-2 Omicron variant.

Mucosal immunity provides efficient protection against upper-airway infections, limiting viral shedding and transmission. However, currently, no nasal spray COVID-19 vaccines are approved by WHO for global use. Here we develop a two-component intranasal vaccine that combines an adenovirus vector expressing the spike protein of the XBB.1.5 variant (Ad5XBB.1.5) with a self-assembled trimeric recombinant protein derived from the receptor binding domain (RBDXBB.1.5-HR). This two-component vaccine elicits superior humoral and cellular immunity against XBB.1.5 variants compared with the individual c

A composite subunit vaccine confers full protection against Buruli ulcer disease in the mouse footpad model of Mycobacterium ulcerans infection.

Buruli ulcer (BU) disease, a neglected necrotizing tropical skin infection caused by Mycobacterium ulcerans, is the third most common mycobacterial disease after tuberculosis and leprosy. Infections mostly occur in remote, rural areas of Central and West Africa, but also in Australia, Japan and Papua New Guinea. There is currently no vaccine against Buruli ulcer disease and all previous attempts using closely related bacteria and subunit proteins have been partially successful only. Here, we tested in mice a composite subunit formulation incorporating the Mycobacterium ulcerans toxin mycolacto
