Survey of Parvovirus B19 Infection in a Cohort of Pregnant Women in Belém, Brazil.

In our study, 300 pregnant women were screened for the presence of human parvovirus B19 IgG and IgM antibodies by an enzyme-linked immunosorbent assay (ELISA). Overall, 253 (84.3%) were found to be IgG-positive and IgM-negative (IgG+gM ), 42 (14%) had neither IgG nor IgM antibodies (IgG /IgM ) and 5 (1.7%) were both IgM- and IgG-positive (IgG+/IgM+). Maternal serology was performed routinely for cytomegalovirus, rubella, toxoplasmosis and syphilis. All IgG /IgM and IgG+/IgM+ women were followed up till the time of delivery, venous blood sample being taken monthly from each one; one IgG /IgM mo…

Use of parvovirus-like particles for vaccination and induction of multiple immune responses.

Expression of the VP2 gene of autonomous parvoviruses in insect cells with the use of the baculovirus system has led to the production of virus-like particles (VLPs) formed by the self-assembly of VP2. These VLPs are expressed at high levels and can easily be purified by salt fractionation. They are highly immunogenic in the corresponding host, being fully protective at doses as low as 1-2 microg of purified material per animal. No special adjuvants are required. An interesting property of these particles is their usefulness as a diagnostic reagent for ELISA kits, which have successfully repla…

Transient blood plasmacytosis in parvovirus B19 infection: a report of two cases.

Two patients with sickle cell disease were diagnosed with aplastic crisis caused by acute parvovirus B19 infection. One patient also developed thrombocytopenia associated with hemophagocytic histiocytosis in the bone marrow biopsy. Both patients developed transient blood plasmacytosis and hypocomplementemia soon after admission to the hospital. Transient blood plasmacytosis may be an immunological response to acute parvovirus B19 infection.

Rheumatic manifestations due to human parvovirus B19. A report of four cases.

Human parvovirus B19 has been incriminated in the genesis of hematologic, dermatologic, neurologic, and rheumatic disorders. We report four cases in which inflammatory rheumatic manifestations developed during the course of human parvovirus B19 infection documented by the presence of IgM and IgG antibodies. There was one case each of monoarthritis, oligoarthritis, polyarthritis, and enthesitis. Three patients had a favorable outcome under nonsteroidal antiinflammatory drug therapy, and one developed reflex sympathetic dystrophy syndrome. In patients with inflammatory rheumatic manifestations t…

Experimental infection of pigs with a parvovirus isolated from the diarrheic feces of a pig.

A parvovirus (the H-45 strain) isolated from an outbreak of epizootic diarrhea in swine was examined to observe the infectivity and pathogenecity in swine. The virus infection by intranasal route was demonstrated in each group of 2- and 5-day-old colostrum-deprived pigs, 30- and 100-day-old pigs by virus recovery from the nasal and rectal swabs, and detecting seroconversion. The virus was recovered from rectal swabs up to 14 days after inoculation and from nasal swabs up to 9 days. Uninoculated pigs were infected with the virus by contacting with the inoculated pigs. Between 1 and 5 days after…

Diagnosis and incidence of fetal parvovirus infection in an autopsy series: I. Histology.

This study was undertaken to obtain additional information about the incidence and characteristics of fetal parvovirus B19 infection in an unselected autopsy series, and to assess the distribution and quantitation of inclusions in various organs. Autopsy records from 673 fetal and neonatal autopsies performed at Women and Infants' Hospital during 1985 through 1990 were reviewed. Thirty-two cases of hydrops fetalis were identified, and, of these, 5 had parvovirus infection. This gives an incidence of fetal parvovirus infection resulting in hydrops fetalis of 0.7% among all autopsies, and a 16% …

Prevalence of human parvovirus B19 in sickle cell disease and healthy controls.

The serological HPV status of 35 patients with SCA (Hb SS), randomly chosen from a sickle cell clinic in rural south-west Togo and of 13 household members with normal haemoglobin type (Hb AA) was assessed. No difference in HPV-IgG seropositivity rate was found. In 30 urban hospital patients from Abidjan, Ivory Coast, who were in acute painful sickle cell crisis HPV-IgG seropositivity rate increased with age. In only one (HIV positive) patient HPV-IgM antibodies were detected. It is concluded that HPV-infection exists in West Africa (1) and that there is no evidence for increased seroprevalence

[A family with hereditary spherocytosis discovered after an infection with human parvovirus B19].

Hereditary spherocytosis was diagnosed in five of six children in one family after a human parvovirus B19 (B19-virus) infection. The diagnosis was made on the basis of severe anaemia, demonstrable haemolysis, decreased osmotic fragility and an increased number of spherocytes. Since in the serum of the patients an increased level of IgG and IgM antibodies against B19 virus was detectable two weeks after the crisis it was concluded that the B19-virus infection caused the severe anaemia.

Detection of human parvovirus B19 DNA by using the polymerase chain reaction.

The polymerase chain reaction (PCR) was investigated for detecting human parvovirus B19 (B19) DNA in sera. Three pairs of oligonucleotides were evaluated as primers. The best oligonucleotide pair spanned 699 nucleotides, including the region common to VP1 and VP2. After PCR amplification of B19 DNA in serum, a 699-nucleotide DNA fragment was detected on agarose gels. This DNA fragment was B19 DNA, because after Southern transfer it hybridized to a 19-nucleotide internal probe and contained a single PstI cleavage site. Dot blot hybridization with a radiolabeled cloned portion of the B19 genome …

Infection with parvovirus-like virus and aplastic crisis in chronic hemolytic anemia.

From 1980 to 1982 seven adults with chronic hemolytic anemia were admitted to Cook County Hospital, Chicago, with aplastic crisis. Six of these patients had sickle cell anemia, the seventh patient had beta thalassemia intermedia. Virologic studies showed that six patients had acute infection with the human parvovirus-like virus; in the remaining patient the lack of appropriate specimens precluded viral diagnosis. We describe the features of the virus infection and accompanying erythroid aplasia, and discuss the role of parvovirus-like virus as the etiologic agent in the arrest of erythrocyte p…

Parvovirus B19 and inflammatory arthritis: from viral mimicry to infection-associated autoimmunity.

INTRODUCTION: Parvovirus B19 is a clinically important viral mimic of systemic inflammatory rheumatic diseases, frequently presenting as an abrupt-onset inflammatory polyarthritis in adults. While typically self-limiting, B19V infection can induce transient autoantibodies, cytopenias, and phenotypes resembling rheumatoid arthritis or systemic lupus erythematosus. In specific contexts, it may also lead to persistent or atypical musculoskeletal manifestations. AREAS COVERED: This narrative review summarizes the epidemiology, immunopathogenesis, clinical spectrum, diagnostic approach, and manage…

Design of Multi-Epitope DIVA-Compatible Vaccine Candidate Against Canine Parvovirus 2.

Canine parvovirus type 2 (CPV-2) is a highly contagious and often fatal canine pathogen, with antigenic variants (CPV-2a, 2b, 2c), maternally derived antibody (MDA) interference, and lack of Differentiating Infected from Vaccinated Animals (DIVA) capability limiting current vaccines. Using an immunoinformatics-based reverse vaccinology approach, we identified conserved B-cell, cytotoxic T lymphocyte (CTL), and helper T lymphocyte (HTL) epitopes from CPV-2 structural (VP1, VP2) and nonstructural (NS1, NS2) proteins. These were screened for antigenicity, immunogenicity, safety, and stability. Th…

A recombinant Fc-fused nanobody provides complete protection against feline parvovirus infection.

Feline panleukopenia (FPL), caused by feline parvovirus (FPV), is a fatal infectious disease in cats characterized by leukopenia, fever, and severe enteritis, for which no specific antiviral treatment is available. Here, we report the development of a potent neutralizing nanobody-based therapeutic against FPV. A high-diversity camelid VHH library (2.95 × 109 CFU) was constructed and screened to identify Nb35, a nanobody with high affinity and specificity for FPV. To enhance its stability and in vivo persistence, Nb35 was fused to the human IgG1 Fc domain, yielding the recombinant fusion antibo…

Fulminant parvovirus B19 myocarditis in infants -report of three post-pandemic cases.

Lack of immune stimulation ("immune debt") in the wake of the COVID-19 pandemic has led to a resurgence of many paediatric infectious diseases, recently including parvovirus B19 (PVB19). We observed three cases of fulminant PVB19 myocarditis in infants. All had experienced a short prodromal illness and succumbed to cardiogenic shock within 2-72 h after admission. The risk of fulminant myocarditis appears to be increased in infants. Further studies are needed to assess the incidence of fulminant myocarditis caused by PVB19, potentially linked to immune debt.

Intravenous immunoglobulin treatment of congenital parvovirus B19 induced anemia - a case report.

BACKGROUND: Parvovirus is a common childhood infection that could be very dangerous to the fetus, if pregnant women become infected. The spectrum of effects range from pure red blood cell aplasia with hydrops fetalis to meningoencephalitis, with many symptoms in between. Severe anemia in the setting of pure red blood cell aplasia is one of the more common effects that neonatal experience (if infected intrapartum), with the current gold standard treatment being intrauterine or postnatal packed red blood cell (PRBC) transfusions, yet intravenous immunoglobulin (IVIG) may be a superior treatment …

Molecular Mechanisms behind Persistent Presence of Parvovirus B19 in Human Dilated Myocardium.

The role of parvovirus B19 (PVB19) in the pathogenesis of idiopathic dilated cardiomyopathy (DCM) remains poorly understood. Therefore, we have measured the levels of inflammation, fibrosis, apoptosis, and necrosis in endomyocardial biopsies (EMBs) and sera of nonischemic PVB19-positive (n = 14) and PVB19-negative (n = 18) DCM patients. Chronic persistence of PVB19 in myocardium did not induce significant infiltration of T cells (CD3 and CD45Ro) and macrophages (CD68), and did not secrete TNFα, IL-6, and CRB. The fibrosis in PVB19-positive EMBs was also lower compared to the virus-negative one…
