Molecular cloning of a full-length cDNA for dentatorubral-pallidoluysian atrophy and regional expressions of the expanded alleles in the CNS.

Dentatorubral-pallidoluysian atrophy (DRPLA) is an autosomal dominant neurodegenerative disorder characterized by genetic anticipation and variable combinations of symptoms including myoclonus, epilepsy, cerebellar ataxia, choreoathetosis, and dementia. Recently, we discovered that DRPLA is caused by unstable expansion of a CAG repeat of a gene on the short arm of chromosome 12. We determined the consensus DRPLA cDNA sequence containing the complete coding region for 1,185 amino acids. The CAG repeat, which is expanded in DRPLA, is located 1,462 bp downstream from the putative methionine initi

Periovulatory multifocal mesothelial proliferation: a possible association with malignant transformation.

This study was aimed at the identification of the site and characterization of the kinetics of tissue renewal during the repair of the ovarian surface epithelium (OSE) surrounding ovulation. Prepubescent C57BL female mice were induced hormonally either by intraperitoneal (ip) injection of pregnant mare serum gonadotropin (PMSG, 5 IU) +/- human chorionic gonadotropin (HCG, 5 IU) 46 h later, or subcutaneous (sc) diethylsilbesterol (DES) 0.1 mg or 1.0 mg in 0.5 ml corn oil, in addition to saline-injected controls. The animals were killed after various time intervals. Autoradiography was carried o

Analysis of natural and disease-associated autoantibody repertoires: anti-endothelial cell IgG autoantibody activity in the serum of healthy individuals and patients with systemic lupus erythematosus.

The present study demonstrates that natural IgG with anti-endothelial cell activity is present in the serum of healthy individuals and in pooled normal human Ig. By using a novel method that allows for the simultaneous and quantitative assessment of reactivities of antibodies with a large number of antigens in tissues, we observed that natural anti-endothelial cell antibody (AECA) recognizes a restricted set of self antigens in endothelial cells that is conserved among healthy individuals. The extent to which natural AECA activity is expressed in serum and the pattern of reactivity of AECA wit

[Breast reconstruction with a latissimus dorsi musculocutaneous flap after amputation for cancer. Apropos of 45 cases with a 9-year follow-up].

Breast reconstruction following mastectomy for cancer using a latissimus dorsi myocutaneous flap must always be completed by insertion of a prosthesis. In 76% of our cases, it was associated with surgery on the contralateral breast to produce symmetry. By following this therapeutic plan using a flap as large as possible taken in the line of the strap of the brassiere, and raised with a large expanse of muscle to completely cover the prosthesis, the aesthetic results were uniformly good. This applies to the mastectomy patients without adjuvant therapy. All the mediocre and poor results were obs

Light-stimulated cell expansion in bean (Phaseolus vulgaris L.) leaves. I. Growth can occur without photosynthesis.

Cell expansion in dicotyledonous leaves is strongly stimulated by bright white light (WL), at least in part as a result of light-induced acidification of the cell walls. It has been proposed that photosynthetic reactions are required for light-stimulated transport processes across plasma membranes of leaf cells, including proton excretion. The involvement of photosynthesis in growth and wall acidification of primary leaves of bean has been tested by inhibiting photosynthesis in two ways: by reducing chlorophyll content of intact plants with tentoxin (TX) and by treating leaf discs with 3-(3,4-

Frontal sinus obliteration using liposuction.

Frontal sinus obliteration is performed for a variety of reasons, including chronic sinus disease, traumatic injuries, mucoceles, and osteomas of the sinus. Once the decision is made to obliterate the sinus, it is paramount that all mucosal remnants be removed and that the material used to fill the irregular expanses of the frontal sinus help prevent recurrence of the disease process. The materials most commonly used for this purpose are fat, muscle, and pericranium. Fat obtained from the abdomen, gluteal area, or lateral thigh is probably the most frequently used substance. The procurement of

Anatomy and histology of the digestive system of the paddlefish (Polyodon spathula).

The paddlefish (Polyodon spathula) is one of the most primitive and unique freshwater fishes of North America. It is adapted as a plankton filter-feeder. The wide mouth and greatly expansible pharyngeal cavity allows it to process a large quantity of water. Numerous setiform gillrakers, which operate against the flattened surfaces of the gill arches and opercula, are capable of filtering out small food particles. The abundance of taste buds and mucus secreting cells in the buccal cavity and pharynx probably aids in selecting and entangling the food. The first segment of the stomach is provided

Interventions for acute non-arteritic central retinal artery occlusion.

BACKGROUND: Acute non-arteritic central retinal artery occlusion (CRAO) occurs as a sudden interruption of the blood supply to the retina and typically results in severe loss of vision in the affected eye. Although many therapeutic interventions have been proposed, there is no generally agreed upon treatment regimen. OBJECTIVES: To assess the effects of treatments for acute non-arteritic CRAO. SEARCH METHODS: We searched the Cochrane Central Register of Controlled Trials (CENTRAL) (which contains the Cochrane Eyes and Vision Trials Register) (2022, Issue 2); Ovid MEDLINE; Embase.com; PubMed;

Accumulated dose on daily iCBCT for rectal cancer: Effects of inter-fraction bowel cavity motion volume.

BACKGROUND: To assess how inter-fraction bowel cavity (BC) motion volume influences accumulated dose in rectal cancer radiotherapy and to translate these effects into a clinically interpretable risk construct for acute bowel toxicity. METHODS: This retrospective cohort included 28 consecutive rectal cancer patients treated with 50 Gy in 25 fractions on a Halcyon v3.0 linac with daily iterative cone beam CT (iCBCT). Patients with > 5% bladder-volume or > 8% external-contour variation across the course were excluded. For each fraction, per-fraction dose was recalculated on the iCBCT using a sca

Transantral, endoscopically guided balloon dilatation of the ostiomeatal complex for chronic rhinosinusitis under local anesthesia.

To date, the lack of a suitable human cardiac cell source has been the major setback in regenerating the human myocardium, either by cell-based transplantation or by cardiac tissue engineering. Cardiomyocytes become terminally-differentiated soon after birth and lose their ability to proliferate. There is no evidence that stem/progenitor cells derived from other sources, such as the bone marrow or the cord blood, are able to give rise to the contractile heart muscle cells following transplantation into the heart. The need to regenerate or repair the damaged heart muscle has not been met by adu

Next-Generation CAR-Macrophage Therapies: Design Principles and Advances in Cancer Treatment.

Macrophages possess intrinsic tumor-homing and tissue-infiltrating abilities, performing various effector functions, including phagocytosis and antigen presentation. These properties make chimeric antigen receptor macrophages (CAR-M) a promising approach to overcoming major barriers in cellular therapy for solid tumors. This review systematically compares and critically evaluates key design variables that determine the therapeutic activity and clinical feasibility of CAR-M, including CAR architecture, macrophage source, gene-delivery method, and manufacturing strategy. We also discuss emerging

Mandibular Chondrosarcoma Mimicking an Odontogenic Infection: A Case Report.

Chondrosarcoma is a malignant mesenchymal tumor exhibiting cartilaginous differentiation. It is a relatively common primary malignant bone tumor, but its occurrence in the jaws is rare, particularly in the mandible. The clinical presentation may be nonspecific and may mimic odontogenic or inflammatory conditions, potentially resulting in delayed diagnosis. We report the case of a 37-year-old woman with mandibular chondrosarcoma initially presenting with persistent pain in the region of the right mandibular first molar. The lesion was initially managed as a dental condition, and endodontic trea

The allogeneic eDN70(-) CAR-T Cells Without TCR or MHC Editing Exhibit Enhanced Persistence and Function.

Allogeneic CAR-T therapy offers an "off-the-shelf" alternative to autologous CAR-T, but faces graft-versus-host rejection (GVH) and host-versus-graft rejection (HVG). Conventional TCR/MHC elimination compromises persistence and increase NK susceptibility. Here, we developed a next-generation UCAR-T strategy that avoids TCR/MHC disruption while addressing donor-cell alloreactivity and host-mediated rejection. First, we eliminated CD4/CD8 to reduce alloreactivity while preserving TCR integrity and potentially TCR/CD3-mediated fitness, although in vivo xGvHD protection remains to be determined. I

Obesity and Atrial Fibrillation: Epidemiology, Mechanisms, and Therapeutic Implications in the Era of Metabolic Therapies.

Obesity has become a major modifiable risk factor for atrial fibrillation (AF) and contributes significantly to its rising global prevalence. Beyond epidemiological associations, obesity induces a specific form of atrial cardiomyopathy characterized by structural remodeling, fibrosis, expansion of epicardial adipose tissue, inflammation, autonomic imbalance, and metabolic dysfunction, all of which promote AF initiation and progression. This review synthesizes current evidence linking obesity to AF, focusing on epidemiology, pathophysiological mechanisms, and therapeutic implications, including

Viscoelasticity-tuned hyaluronic acid-gelatin microcarriers with thermoresponsive polymer grafts for scalable cell expansion and gentle harvesting.

Three-dimensional cell culture using microcarriers is an effective strategy for scalable cell expansion. However, conventional enzymatic detachment can compromise cell viability, surface proteins, and native signaling. We report viscoelasticity-tunable hyaluronic acid (HA)-gelatin microspheres as microcarriers, engineered with a thermoresponsive polymer coating to enhance cell attachment and enable gentle harvesting. Gelatin-only (GLA), gelatin-HA (G-HA), and gelatin-HA-L-lysine (G-HA-L) microspheres were fabricated. HA incorporation and lysine functionalization were used to tune microsphere m

Matched Analysis of Circulating and Adipose Tissue SIRT1 Protein Level in Human Obesity.

Background/Objectives: Mammalian sirtuins (SIRTs) are evolutionarily conserved proteins that are epigenetically involved in biological processes such as metabolism and longevity. SIRT1 expression is reduced in metabolic disorders and in complicated diseases such as obesity. However, whether the SIRT1 level in subcutaneous adipose tissue (SAT) matches with its circulating form in obesity is unknown. The aim of our study is to evaluate SIRT1 derived from SAT and plasma of the same subject in individuals with and without obesity to assess whether plasma measurements may provide clinically signifi

Mechanistic remodeling and immunoregulatory functions of the B cell-humoral immunity axis in inflammatory bowel disease.

Inflammatory bowel disease (IBD) is an immune-mediated disorder characterized by chronic inflammation of the intestinal mucosa, arising from dysregulation across multiple layers of immune control. In recent years, the contribution of B cells and humoral immunity to IBD pathogenesis has gained increasing attention. B cells participate in mucosal immune regulation through differentiation, clonal expansion, and antibody production, and are closely associated with the formation and structural organization of local tertiary lymphoid structures (TLSs). Beyond antigen neutralization, antibodies can m

Tau, amyloid-β and α-synuclein co-pathologies synergistically enhance neuroinflammation and hippocampal neuron loss.

Alzheimer's (AD) and Parkinson's disease (PD) pathology often co-occur. Amyloid-β and phosphorylated tau are found in 30-50% of idiopathic PD cases, while α-synuclein inclusions are present in over 50% of AD cases. These co-pathologies are linked to increased mortality and earlier onset of cognitive decline. Immune activation is a hallmark of these neurodegenerative diseases, but current model systems primarily examine each pathology in isolation. As such, how these co-pathologies drive inflammation and neuronal loss remains poorly understood. To address this gap, we developed a mouse model co

Single-cell fixed RNA profiling of the osteosarcoma clinical Spectrum identifies ADAMTS12 and RNASE1 as key drivers of the metastatic niche.

Osteosarcoma (OS) suffers from stagnant survival rates due to high metastatic potential and genomic complexity. Here, we present a high-resolution single-cell transcriptomic atlas of the OS ecosystem using single-cell fixed RNA profiling (FLEX) of 40,401 cells across four clinical stages: treatment-naïve, post-chemotherapy, recurrent, and lung metastatic. We identified a metastasis-enriched malignant subpopulation, OB_3, characterized by aggressive transcriptional signatures and governed by ADAMTS12, which we validated as a prognostic marker for reduced overall survival. Functional in vitro ex
