Trifluoromethylation of alkenes by visible light photoredox catalysis.

A method for trifluoromethylation of alkenes has been developed employing visible light photoredox catalysis with CF(3)I, Ru(Phen)(3)Cl(2), and DBU. This process works especially well for terminal alkenes to give alkenyl-CF(3) products with only E-stereochemistry. The mild reaction conditions enable the trifluoromethylation of a range of alkenes that bear various functional groups.

Type-2 alkenes mediate synaptotoxicity in neurodegenerative diseases.

Synaptic dysfunction appears to be an early pathogenic event in Alzheimer's disease, amyotrophic lateral sclerosis and Parkinson's disease. Although the molecular mechanism of this synaptotoxicity is not known, evidence suggests that these diseases are characterized by a common pathophysiological cascade involving oxidative stress, lipid peroxidation and the subsequent liberation of alpha,beta-unsaturated carbonyl derivatives such as acrolein and 4-hydroxy-2-nonenal (HNE). A diverse body of in vivo and in vitro data have shown that these soft electrophilic chemicals can cause nerve terminal da

Mechanoredox Enables Sulfonylation of Alkenes in a Heterogeneous System.

The sulfur dioxide insertion strategy serves as a significant approach for the construction of C-S bonds. Nevertheless, current liquid-phase reaction systems utilizing inorganic SO2 sources frequently suffer from poor solubility, necessitating an excess of reagents and resulting in low atom economy. Furthermore, these systems frequently rely on additional bases or transition metal additives, which may inadvertently trigger side reactions. To address these limitations, we have reported a novel method for sulfonylation of alkenes that employs ball milling in conjunction with a piezoelectric mate

Cobaltaphotocatalysed intramolecular hydroarylation of alkenes for the construction of tetrahydroquinolines.

1,2,3,4-Tetrahydroquinolines are a class of heterocycles that commonly feature in both biologically active natural products and pharmaceuticals. However, their synthesis is often dominated by either high-pressure hydrogenation reactions or acid-mediated Friedel-Crafts-like hydroarylations. Recently, cobalt-salen catalysed cycloisomerisation reactions have been reported as a promising method to construct heterocycles, but there are limited examples of tetrahydroquinolines. Moreover, these protocols often rely on stoichiometric additives and proceed through radical pathways in which catalyst con

Intramolecular Radical Amidooxygenation of Alkenes for the Construction of Pyrrolidinones.

An intramolecular 1,2-amidooxygenation of unactivated alkenes for the construction of the pyrrolidinone scaffold containing a masked 5-hydroxymethyl functionality is reported. γ,δ-Unsaturated N-aryloxyamides react with sodium 2,2,6,6-tetramethylpiperidin-1-olate (TEMPONa) to afford alkoxyaminated pyrrolidinones. The cascade proceeds through reductive amidyl radical generation with TEMPONa, 5-exo cyclization, and TEMPO trapping. No transition metal is required to perform these transformations, and complex (fused, bridged) pyrrolidinones are accessible in moderate to good yields. The product alk

Controlled silicon surface functionalization by alkene hydrosilylation.

Immobilization of indene ligands onto two types of hydrogen-terminated surfaces, oxide-free Si [H/Si(111)] and oxidized Si [H/SiO2/Si], has been investigated by infrared absorption spectroscopy. The activity of a common catalyst (H2PtCl6) is shown to depend critically on the nature of the solvent. For instance, 2-propanol preferentially reacts with the surface, preventing any ligand attachment. Chlorobenzene is more stable, allowing some ligand attachment, but the H2PtCl6 catalyst also fosters silicon oxidation. In contrast, UV irradiation on oxide-free surfaces promotes a cleaner and more eff

Homologation of Alkenes Using Acetylene as a C2 Feedstock.

The preparation of diverse homologs from lead compounds has been a common and important practice in medicinal, mechano, polymeric, and many other branches of chemistry. The homologation of alkenes, though important, remains challenging due to the difficulty of (CH2)n insertion into the main carbon chain of alkenes compared to chains containing other polar functional groups. Here, we report the homologation of both terminal and internal alkenes by using acetylene as a cheap and abundant C2 feedstock. The process involves an in-situ transformation of alkenes into boranes and then into elongated

Applications of sulfonyl hydrazides in radical cyclization of alkenes.

Radical cyclization is regarded as a powerful and promising strategy for the assembly of diverse important cyclic structures because of its high atom- and step-economy. As excellent radical acceptors, alkenes offer two potential directions, pushing the research domain of radical cyclization. In this context, as a radical precursor, sulfonyl hydrazide plays an important role in accomplishing radical cyclization of alkenes in a facile and efficient way. This review focuses on the applications of sulfonyl hydrazides in radical cyclization of alkenes, which generally has two radical conversion mod

Activation parameters for additions of ambiphilic methoxychlorocarbene to alkenes.

Activation parameters are determined for the additions of methoxychlorocarbene (MeOCCl) to four alkenes, including tetramethylethylene (TME). MeOCCl is the first carbene to exhibit appreciable activation energy in addition to TME (5.8 kcal/mol), although this reaction is still "dominated" by entropy (-TDeltaS(double dagger) approximately 7 kcal/mol).

Metabolism of chlorinated alkenes and alkanes as related to toxicity.

The chlorine substitution in aliphatic compounds results, by its electron attracting effect, in a destabilization in alkanes and a stabilization in alkenes. Thus, with alkanes the main pathways of metabolic transformation to reactive intermediates are radical formation by C-C break or dechlorination, or dehydrochlorination. In alkenes, the stability of the molecule increases with the number of chlorine substitutions. In the series of chlorinated ethylenes, the first step of metabolic transformation is the oxidation to electrophilic oxiranes which may be hydrolized enzymatically or non-enzymati

Photocatalytic Phosphine-mediated Hydroxymethylation of Alkenes With Formaldehyde Hydrate.

The mild and convenient generation of unprotected hydroxymethyl radical from simple C1 feedstocks remains a fundamental challenge. Here we report a visible-light-induced, phosphine-mediated hydroxymethylation of electron-deficient alkenes using aqueous formaldehyde as the hydroxymethyl radical precursor. The reaction is enabled by single-electron oxidation of phosphine to phosphine radical cation (PRC), which engages formaldehyde hydrate to release hydroxymethyl radical via β-scission. Subsequent regioselective radical addition to C─C double bonds, single-electron reduction, and protonation de

NiH-Catalyzed Enantio- and Regioselective Hydroselenylation of Unactivated Alkenes.

Chiral alkyl selenides are valuable motifs in medicinal chemistry and redox-active molecular design, yet enantioselective hydroselenylation of unactivated alkenes remains challenging due to limited control over regio- and enantioselectivity. Here, we report nickel-hydride-catalyzed enantio- and regioselective hydroselenylation of unactivated alkenes directed by native amide groups using readily available diselenides. The method accommodates both terminal and internal alkenes as well as both aryl and alkyl diselenides, exhibiting broad functional-group tolerance and delivering structurally dive

Kharasch-Type Haloalkylation of Alkenes by Photoinduced Copper Catalysis.

The simultaneous construction of C-C and C-X bonds in a single step facilitates multistep synthesis through rapid carbon chain growth and subsequent transformations of halide functionalities. Traditional Kharasch addition requires simple polyhalogenated compounds or those with electron-withdrawing groups at the α-carbon. Herein, we present a Kharasch-type reaction utilizing a broad range of carboxylic acid-derived redox-active esters as the alkyl source, which enables the efficient introduction of highly functionalized alkyl groups. Our method produces α-halo carbonyls that enable versatile nu

Synthesis of aminoalcohols from substituted alkenes via tungstenooxaziridine catalysis.

Herein we report the WO2Dipic(H2O) promoted oxyamination of alkenes using sulfonamides as the quantitative source of N. The reaction works for activated and unactivated alkenes in high yields, diastereoselectivities, and stereospecificity. A catalytic cycle involving the formation of tungstenooxaziridine complex 1 as the active catalyst and hydrolysis of tungstenooxazolidine intermediate A as the rate-determining-step has been proposed. Initial kinetic and competition experiments provide evidence for the proposed mechanism.

Cu-Electrocatalytic Diazidation of Alkenes at ppm Catalyst Loading.

The 1,2-diamine motif is prevalent in natural products, small-molecule pharmaceuticals, and catalysts for asymmetric synthesis. Transition metal catalyzed alkene diazidation has evolved to be an attractive strategy to access vicinal primary diamines but remains challenging, especially for practical applications, due to the restriction to a certain type of olefins, the frequent use of chemical oxidants, and the requirement for high loadings of metal catalysts (1 mol % or above). Herein we report a scalable Cu-electrocatalytic alkene diazidation reaction with 0.02 mol % (200 ppm) of copper(II) a
