Mechanistic pathways of cosmetic ingredient toxicity: a critical review of in vitro and in vivo evidence.

Every day, billions of people apply cosmetic products containing dozens of bioactive compounds whose combined toxicological profile has never been formally evaluated. Human biomonitoring confirms systemic deposition of these compounds in blood, adipose tissue, placenta, and breast milk worldwide. This scoping review, following PRISMA-ScR, synthesizes in vitro and in vivo mechanistic toxicology evidence for six major cosmetic ingredient categories-preservatives, UV filters, engineered nanoparticles, fragrances, surfactants, and colorants-and maps findings onto established adverse outcome pathwa

Dexmedetomidine and Postoperative Gastrointestinal Recovery: A Critical Narrative Review of Direct Enteric and Opioid-Sparing Pathways.

Postoperative gastrointestinal dysfunction (POGD) remains an important barrier to recovery after abdominal and non-abdominal surgery. This critical narrative review evaluates whether dexmedetomidine improves postoperative gastrointestinal recovery through an independent direct prokinetic action or predominantly through opioid-sparing and other indirect recovery pathways. Positive trials and meta-analyses report earlier flatus, defecation, or oral intake in selected settings, but most also permit simultaneous reductions in opioid exposure, pain, postoperative nausea and vomiting (PONV), inflamm

The critical role of GLP-1 signaling pathways in the pathology of Parkinson's disease and diabetes.

This review assesses the roles of GLP-1 and its receptor agonists (GLP-1RAs) in the treatment of diabetes and Parkinson's disease, integrating current theories and research. GLP-1, a vital endogenous hormone, regulates insulin secretion, delays gastric emptying, and promotes satiety, showing significant potential for diabetes management. However, its brief lifespan and restricted blood-brain barrier penetration limit its clinical application. To overcome these constraints, researchers have developed GLP-1 receptor agonists that prolong its action and exhibit high efficacy in diabetes treatment

Nitric Oxide-Dependent Pathways as Critical Factors in the Consequences and Recovery after Brain Ischemic Hypoxia.

Brain ischemia is one of the leading causes of disability and mortality worldwide. Nitric oxide (NO•), a molecule that is involved in the regulation of proper blood flow, vasodilation, neuronal and glial activity constitutes the crucial factor that contributes to the development of pathological changes after stroke. One of the early consequences of a sudden interruption in the cerebral blood flow is the massive production of reactive oxygen and nitrogen species (ROS/RNS) in neurons due to NO• synthase uncoupling, which leads to neurotoxicity. Progression of apoptotic or necrotic neuronal damag

Critical signaling pathways in osteoclast differentiation and bone resorption: mechanisms and therapeutic implications for periprosthetic osteolysis.

Bone homeostasis is dynamically regulated by the balance between osteoclast-mediated bone resorption and osteoblast-driven bone formation. Periprosthetic osteolysis (PPO), a major complication following joint arthroplasty, occurs when excessive bone resorption surpasses formation, leading to implant loosening and failure. Emerging evidence highlights the pivotal roles of the RANKL/RANK/OPG axis, nuclear factor-κB (NF-κB) signaling, and mitogen-activated protein kinase/extracellular signal-regulated kinase (MAPK/ERK) cascades in osteoclast differentiation and pathological bone resorption. This

Zebrafish mutants calamity and catastrophe define critical pathways of gene-nutrient interactions in developmental copper metabolism.

Nutrient availability is an important environmental variable during development that has significant effects on the metabolism, health, and viability of an organism. To understand these interactions for the nutrient copper, we used a chemical genetic screen for zebrafish mutants sensitive to developmental copper deficiency. In this screen, we isolated two mutants that define subtleties of copper metabolism. The first contains a viable hypomorphic allele of atp7a and results in a loss of pigmentation when exposed to mild nutritional copper deficiency. This mutant displays incompletely penetrant

The GPI-anchor biosynthesis pathway is critical for syncytiotrophoblast differentiation and placental development.

The glycosylphosphatidylinositol (GPI) biosynthetic pathway in the endoplasmic reticulum (ER) is crucial for generating GPI-anchored proteins (GPI-APs), which are translocated to the cell surface and play a vital role in cell signaling and adhesion. This study focuses on two integral components of the GPI pathway, the PIGL and PIGF proteins, and their significance in trophoblast biology. We show that GPI pathway mutations impact on placental development impairing the differentiation of the syncytiotrophoblast (SynT), and especially the SynT-II layer, which is essential for the establishment of

Hedgehog Signaling, a Critical Pathway Governing the Development and Progression of Hepatocellular Carcinoma.

Hedgehog (Hh) signaling is a classic morphogen in controlling embryonic development and tissue repairing. Aberrant activation of Hh signaling has been well documented in liver cancer, including hepatoblastoma, hepatocellular carcinoma (HCC) and cholangiocarcinoma. The present review aims to update the current understanding on how abnormal Hh signaling molecules modulate initiation, progression, drug resistance and metastasis of HCC. The latest relevant literature was reviewed with our recent findings to provide an overview regarding the molecular interplay and clinical relevance of the Hh sign

Ventral aspect of the visual form pathway is not critical for the perception of biological motion.

Identifying the movements of those around us is fundamental for many daily activities, such as recognizing actions, detecting predators, and interacting with others socially. A key question concerns the neurobiological substrates underlying biological motion perception. Although the ventral "form" visual cortex is standardly activated by biologically moving stimuli, whether these activations are functionally critical for biological motion perception or are epiphenomenal remains unknown. To address this question, we examined whether focal damage to regions of the ventral visual cortex, resultin

Transient visual pathway critical for normal development of primate grasping behavior.

An evolutionary hallmark of anthropoid primates, including humans, is the use of vision to guide precise manual movements. These behaviors are reliant on a specialized visual input to the posterior parietal cortex. Here, we show that normal primate reaching-and-grasping behavior depends critically on a visual pathway through the thalamic pulvinar, which is thought to relay information to the middle temporal (MT) area during early life and then swiftly withdraws. Small MRI-guided lesions to a subdivision of the inferior pulvinar subnucleus (PIm) in the infant marmoset monkey led to permanent de

Cerebral Dopamine Neurotrophic Factor in Dorsal Root Ganglia Neurons: A Critical Appraisal of Mechanisms and Unexplored Pathways.

The recent study by de Siqueira Santos et al. (J Neurochem. 2025; 169 (8): e70194) represents a significant contribution by identifying Cerebral Dopamine Neurotrophic Factor (CDNF) as a potent inducer of neuritogenesis in dorsal root ganglion (DRG) neurons via the PI3K/Akt pathway. While we commend the authors on their robust in vitro findings, which position CDNF as a promising therapeutic agent for peripheral neuropathies, our letter provides a critical analysis to contextualize and extend these discoveries. We posit that the mechanistic narrative, while compelling, remains incomplete. The e

The role of microRNAs in the regulation of critical genes and signalling pathways that determine endometrial receptivity.

Endometrial receptivity is the ability of the endometrium to accept embryos. Thus, endometrial receptivity dysfunction is an important factor leading to embryo implantation failure. A good endometrial receptivity provides a suitable environment for embryo implantation, improving the embryo implantation rate. The "implantation window" stage, or the receptive stage of the endometrium, is regulated by various hormones, genes, proteins and cytokines, among which microRNAs (miRNAs) and their target genes have a regulatory effect on endometrial receptivity. This review outlines the relationship betw

Pathways of sleep disturbances in juvenile idiopathic arthritis and recommendations for clinical management approaches: A critical review.

Juvenile idiopathic arthritis (JIA) is the most common rheumatic disease affecting young people. It has a profound impact on their physical, mental and social lives, leading to long-term disability. With the growing awareness of the importance of sleep in all areas of functioning in young people, an emerging literature has drawn attention to the role of sleep in the pathogenesis of JIA. Sleep disturbances in children and adolescents with JIA arise from a wide range of symptoms and pathways, leading to a vicious cycle that exacerbates subclinical inflammation, symptoms and disease progression.
