Enteropathogenic Escherichia coli infection triggers host phospholipid metabolism perturbations.

Enteropathogenic Escherichia coli (EPEC) specifically recognizes phosphatidylethanolamine (PE) on the outer leaflet of host epithelial cells. EPEC also induces apoptosis in epithelial cells, which results in increased levels of outer leaflet PE and increased bacterial binding. Consequently, it is of interest to investigate whether EPEC infection perturbs host cell phospholipid metabolism and whether the changes play a role in the apoptotic signaling. Our findings indicate that EPEC infection results in a significant increase in the epithelial cell PE level and a corresponding decrease in the p

Effect of hyperpnea on enzymes of the CDPcholine path for phosphatidylcholine synthesis in rat lung.

We have examined the activity of three enzymes in pulmonary surfactant phosphatidylcholine synthesis following the hyperpnea induced by having rats either inspire 5%CO2/13%O2/82%N2 for 24 hr or swim in thermoneutral water for 30 min. Both stimuli markedly increase frequency and tidal volume of breathing and promote the release of surfactant. Lungs were perfused to remove blood, lavaged, and then homogenized in 1 mM Hepes, 0.15M KCl at pH 7.0. The homogenate was centrifuged at 9,000 g (av) for 10 min to sediment the mitochondria and lamellar bodies and at 100,000 g (av) for 60 min to obtain the

Role of Citicoline in an in vitro AMD model.

Citicoline is the exogenous form of the nootropic, Cytidine 5'-diphosphate-choline that exerts its neuroprotective effects in the brain as well as in the eye. The current study characterized the cytoprotective effects of purified Citicoline in transmitochondrial AMD (Age-related Macular Degeneration) RPE cybrid cells which carry diseased mitochondria from clinically characterized AMD patients. The effects of Citicoline were examined via flow cytometry analysis of AnnexinV/ PI-stained cells, IncuCyte live-cell imaging analysis to quantify cells undergoing caspase-3/7-mediated apoptosis, analyse

Brain Targeting of Citicoline Sodium via Hyaluronic Acid-Decorated Novel Nano-Transbilosomes for Mitigation of Alzheimer's Disease in a Rat Model: Formulation, Optimization, in vitro and in vivo Assessment.

BACKGROUND: Alzheimer's disease (AD) is one of the furthermost advanced neurodegenerative disorders resulting in cognitive and behavioral impairment. Citicoline sodium (CIT) boosts the brain's secretion of acetylcholine, which aids in membrane regeneration and repair. However, it suffers from poor blood-brain barrier (BBB) permeation, which results in lower levels of CIT in the brain. PURPOSE: This study targeted to encapsulate CIT into novel nano-platform transbilosomes decorated with hyaluronic acid CIT-HA*TBLs to achieve enhanced drug delivery from the nose to the brain. METHODS: A method

[Position of cyticholine in pneumologic therapy].

The position of cyticholine in lung lipid metabolism and hence in pneumological therapy (the subject of a personal hypothesis) is discussed. A summary is given of personal experimental work and papers read at the 1974 Padua meeting (11th December) held by the Societas Europae Physiologiae Clinicae Respiratoriae. It is shown that cyticholine may: 1) prevent the onset of acute neonatal respiratory distress; 2) improve chronic pneumogenous respiratory insufficiency; 3) prevent postoperative hypoventilation; 4) improve ventilation mechanics in hyperelastic lung syndromes; 5) increase the gas perme

Neurosurgical trauma in Japan.

According to the statistics compiled by the Institute for Traffic Accident Research and Data Analysis, the total number of head trauma patients has stayed virtually the same for the last 10 years in Japan, although a fraction of people suffering minor head trauma has shown a trend to increase. The Japan Society of Neurotraumatology is in the process of establishing a guideline for head trauma management. No major difference is noted in surgical procedures selected by neurosurgeons in Japan as compared to those in other countries. It appears, however, that the ventriculostomy may be less freque

[Efficacy of Recognan in the therapy of neurological disorders after traumatic brain injury].

Traumatic brain injury (TBI) is a brain injury caused by external mechanical stress, which leads to a deterioration in the patient's health (temporary or irreversible), and is the main cause of death and disability. Along with neurodegenerative and cerebrovascular diseases, TBI is one of the main causes of cognitive impairment, as a result of injury. Among the neuroprotectors prescribed for the correction of neurological disorders after a TBI, the medicine drug (MD) Recognan (citicoline), which has neuroprotective and reparative effects, is of particular interest. The effectiveness of citicoli

Citicoline is not protective in experimental models of Huntington's disease.

We have evaluated the neuroprotective effects of citicoline in relevant phenotypic models of Huntington's disease induced by either the mitochondrial inhibitor 3-nitropropionic acid or the N-methyl-D-aspartate agonist quinolinic acid, which, respectively, reproduce the metabolic defect or the excitotoxicity seen in the disease. We found that citicoline failed to reverse behavioural and histological alterations induced by both neurotoxins. In addition, citicoline did not reduce PC12 cell death induced by the expression of an N-terminal fragment of mutated Huntingtin. Altogether, our results sug

CDP-choline: pharmacological and clinical review.

Cytidine 5'-diphosphocholine, CDP-choline or citicoline, is an essential intermediate in the biosynthetic pathway of the structural phospholipids of cell membranes, especially in that of phosphatidylcholine. Upon oral or parenteral administration, CDP-choline releases its two principle components, cytidine and choline. When administered orally, it is absorbed almost completely, and its bioavailability is approximately the same as when administered intravenously. Once absorbed, the cytidine and choline disperse widely throughout the organism, cross the blood-brain barrier and reach the central

Effect of hypoxia on choline metabolism and phosphatidylcholine biosynthesis in isolated adult rat ventricular myocytes: effects of trifluoperazine.

The purpose of this study was to test the hypothesis that hypoxia alters choline metabolism in adult rat ventricular myocytes and that this is subject to modification by trifluoperazine. Ventricular myocytes were isolated from adult Wistar rats. Choline uptake and metabolism were examined, using [methyl-3H]choline. Choline metabolism to phosphatidylcholine (PC), an important phospholipid for cellular structure and function, was reduced by mild hypoxia while choline uptake was not altered. Trifluoperazine at 10(-5) or 10(-6) M reduced PC biosynthesis and did not reduce or prevent the effects of
