Modeling cell response to low doses of photon irradiation--Part 1: on the origin of fluctuations.

Intra- and inter-individual variability is a well-known aspect of biological responses of cells observed at low doses of radiation, whichever the phenomenon considered (adaptive response, bystander effects, genomic instability, etc.). There is growing evidence that low-dose phenomena are related to cell mechanisms other than DNA damage and misrepair, meaning that other cellular structures may play a crucial role. Therefore, in this study, a series of calculations at low doses was carried out to study the distribution of specific energies from different irradiation doses (3, 10 and 30 cGy) in t

Developmental changes in the sensitivity of Volvox to ultraviolet light.

The response of Volvox to ultraviolet irradiation was analyzed. Young individuals isolated from a synchronous culture were exposed to UV light (120 J/m2) and subjected to variable length periods of dark following irradiation. The major effect of the UV treatment was the inability of the gonidia present in the colonies at the time of irradiation to continue and complete the developmental program. Individuals show a heightened sensitivity to UV for a limited period immediately following inversion and are insensitive at other stages of development. The cytotoxic effect of UV during this interval

Andrographis paniculata Diterpenoids Protect against Radiation-Induced Transformation in BALB/3T3 Cells.

One of the most concerning side effects of exposure to radiation are the carcinogenic risks. To reduce the negative effects of radiation, both cytoprotective and radioprotective agents have been developed. However, little is known regarding their potential for suppressing carcinogenesis. Andrographis paniculata , a plant, with multiple medicinal uses that is commonly used in traditional medicine, has three major constituents known to have cellular antioxidant activity: andrographolide (AP1); 14-deoxy-11,12-didehydroandrographolide (AP3); and neoandrographolide (AP4). In our study, we tested th

Biodosimetry Based on Gamma-H2AX Quantification in Human Peripheral Blood Lymphocytes after Partial-body Irradiation.

Quantification of gamma-H2AX foci can estimate exposure to ionizing radiation. Most nuclear and radiation accidents are partial-body irradiation, and the doses estimated using the total-body irradiation dose estimation formula are often lower than the actual dose. To evaluate the dose-response relation of gamma-H2AX foci in human peripheral blood lymphocytes after partial-body irradiation and establish a simple and high throughput model to estimate partial-body irradiation dose, we collected human peripheral blood and irradiated with 0-, 0.5-, 1-, 2-, 3-, 4-, 5-, 6-, and 8-Gy gamma rays to sim

Radiation proteomics: a brief overview.

Acute biological effects caused by the exposure to high doses of radiation, either ionizing or nonionizing, are relatively well-known but the delayed effects, occurring decades after exposure, are difficult to predict. The knowledge of the acute and delayed effects of the low doses of ionizing radiation (e.g. bystander effect) or nonionizing radiation (e.g. radiation emitted by wireless communication devices) is not yet reliably established. Often the acute effects of low doses are small and difficult to discover and replicate in scientific studies. Chronic effects of prolonged exposures to lo

Quantitative investigation of efficiency of ultraviolet and visible light in eradication of Candida albicans in vitro.

OBJECTIVE: The aim of this study was to quantitatively investigate the efficiency of the ultraviolet (UV) and visible light in eradication of Candida albicans in vitro; in particular, to determine, for selected wavelengths, the specific eradication coefficients and thresholds in terms of energy density levels required to effect 3.0log10 and 4.0log10 reduction. BACKGROUND DATA: Oral candidosis is the most common infection of the oral cavity and is caused by Candida species. The widespread use of topical and systemic antifungal agents as conventional treatment for oral candidosis has resulted i

Hepatic radiation toxicity: avoidance and amelioration.

The refinement of radiation therapy and radioembolization techniques has led to a resurgent interest in radiation-induced liver disease (RILD). The awareness of technical and clinical parameters that influence the chance of RILD is important to guide patient selection and toxicity minimization strategies. "Classic" RILD is characterized by anicteric ascites and hepatomegaly and is unlikely to occur after a mean liver dose of approximately 30 Gy in conventional fractionation. By maintaining a low mean liver dose and sparing a "critical volume" of liver from radiation, stereotactic delivery tech

Radiation hormesis, or, could all that radiation be good for us?

Nuclear medicine technologists work under significant radiation protection constraints. These constraints are based on the linear no-threshold (LNT) radiation paradigm, which was developed in the 1960s and was based largely on the deleterious effects of radiation as they were understood at the time. More recently, the theory of radiation hormesis, or a beneficial effect of low-level exposure to radiation, has gained recognition. This article reviews the history of attitudes toward radiation, describes the radiation hormesis hypothesis, examines some of the evidence that supports it, and sugges

Towards a New Concept of Low Dose.

When people discuss the risks associated with low doses of ionizing radiation, central to the discussion is the definition of a low dose and the nature of harm. Standard answers such as "doses below 0.1 Gy are low" or "cancer is the most sensitive measure of harm" obscure the complexity within these seemingly simple questions. This paper will discuss some of the complex issues involved in determining risks to human and nonhuman species from low-dose exposures. Central to this discussion will be the role of communicable responses to all stressors (often referred to as bystander responses), whic

Streptococcus mutans photoinactivation using a combination of a high potency photopolymerizer and rose bengal.

BACKGROUND: Long treatment periods to reach a substantial inactivation of microorganisms are one of the critical challenges in the photodynamic therapy field. METHODS: Planktonic suspensions of Streptococcus mutans were treated in different groups: presence of rose bengal (RB at 2μM) and light exposure by a new high potency photopolymerizer (L at wavelength=440-480nm; dosimetry=96J/cm(2) - 40s of irradiation; potency density=1200mW/cm(2); dosage=48J) - RB+L+ (PDT), just with dye - RB+L-, just with light - RB-L+ and absence of light and dye RB-L- (control group). A different perception of the linear, nonthreshold hypothesis for low-dose irradiation.

Two equally useful dosimetric quantities, both of which are called dose, are used in toxicology. With radiation measurement, only one--the energy per unit mass D--is called dose. The other--the total energy in the irradiated system--is here distinguished from D by assigning it the name collective energy, epsilon. The collective energy is a more complete statement of dose because it is the product of the energy concentration D and the mass irradiated m. Especially in radioepidemiology, in which epsilon is the total energy imparted to all persons irradiated, the quantity m must be specified beca

Complications of fast neutron therapy.

The purpose of the study was to identify the tissues and organs at risk following high-energy neutron-beam therapy for selected radioresistant tumors, estimating the separate probabilities of both normal tissue injury and of tumor recurrence, each in relation to the absorbed dose. Published statistical and anecdotal reports on the incidence of serious complications observed following fast neutron treatment directed to the cranium, head and neck, chest, upper abdomen, pelvis, and extremities are reviewed and dose-response parameters derived using bivariate probit or logistic analyses. Lethal effect induced in Pseudomonas aeruginosa Exposed to Ultraviolet-A radiation.

Ultraviolet-A (365 nm, 120 kJ/m2/h) exposure caused cell death in Pseudomonas aeruginosa at doses at which Escherichia coli cell viability was not affected. We have not found that UVA induced growth delay or any other sublethal effect. Irradiated suspensions of P. aeruginosa showed a marked reduction in membrane-bound succinate dehydrogenase (SDH) and lactate dehydrogenase (LDH) activities. Succinate-driven respiration and several nutrient transport systems were also inhibited. Whereas SDH and LDH activities were independent of the irradiation conditions, cell viability, respiration and transp

The radiation-induced bystander effect: evidence and significance.

A multitude of biological effects observed over the past two decades in various in vivo and in vitro cell culture experiments have indicated that low dose/low fluence ionizing radiation has significantly different biological responses than high dose radiation. Exposure of cell populations to very low fluences of alpha-particles or incorporated radionuclides results in significant biological effects occurring in both the irradiated and nonirradiated cells in the population. Cells recipient of growth medium from irradiated cultures can also respond to the radiation exposure. This phenomenon, ter

AI-Assisted Chromosome Aberration Analysis for Rapid Biological Dose Estimation of Ionizing Radiation.

Radiation dose assessment in exposed individuals relies on the dicentric assay, the gold-standard cytogenetic biodosimeter that quantifies radiation-induced chromosomal aberrations. Although fully automated methods have been proposed, their accuracy remains suboptimal for large-scale emergency response. Here, we present a hybrid, semi-automatic framework that couples a deep-learning-based dicentric classifier with expert review. Among 2 000 metaphase images analyzed with a semi-automatic method, the specificity was 99.8%, the accuracy 98.7%, and the area under the curve (AUC) 0.977. For radiat

Evaluation of basic characteristics of 3-mm dose equivalent measuring instrument for evaluating lens exposure dose in radiotherapy.

INTRODUCTION: Despite the development of DOSIRIS™, an eye lens dosimeter, the characteristics of DOSIRIS™ in the area of radiotherapy have not been investigated. The purpose of this study was to evaluate the basic characteristics of the 3-mm dose equivalent measuring instrument DOSIRIS™ in radiotherapy. METHODS: Dose linearity and energy dependence were evaluated for the irradiation system based on the calibration method of the monitor dosimeter. The angle dependence was measured by irradiating from a total of 18 directions. Interdevice variation was repeated three times by simultaneously irr

[Cancer incidence and mortality after low-dose radiation exposure: epidemiological aspects].

Current recommendations for limiting exposure to ionizing radiation are based on the linear-no-threshold (LNT) model for radiation carcinogenesis under which every dose, no matter how low, carries with it some cancer risk. In this review, epidemiological evidences are discussed that the LNT hypothesis is incorrect at low doses. A large set of data was accumulated that showed that cancer risk after ordinarily encountered radiation exposure (natural background radiation, medical X-rays, etc.) is much lower than projections based on the LNT model. The discovery of the low-level radiation hormesis

Low-dose ionizing radiation and chromosome translocations: a review of the major considerations for human biological dosimetry.

Chromosome translocations are a molecular signature of ionizing radiation exposure. Translocations persist significantly longer after exposure than other types of chromosome exchanges such as dicentrics. This persistence makes translocations the preferred aberration type for performing radiation dosimetry under conditions of protracted exposure or when exposure assessments are temporally delayed. Low doses of radiation are inherently difficult to quantify because the frequency of induced events is low and the background level of translocations among unexposed subjects can show considerable var
