Protective efficacy of lambdacyhalothrin-impregnated bednets against Phlebotomus orientalis, the vector of visceral leishmaniasis in Sudan.

Field investigations on the sandfly Phlebotomus orientalis (Diptera: Psychodidae), the vector of Leishmania donovani causing visceral leishmaniasis (VL) in Sudan, were undertaken in two villages (Bellow and Elgamel) and Dinder National Park, to determine the protective value of bednets (polyester, 100 denier) impregnated with lambda-cyhalothrin 10 mg a.i./m2 pyrethroid insecticide. After exposure to treated netting for 30 s, P. orientalis females all died within 1 h. When field-tested in Acacia woodland, treated bednets provided complete protection from bites of the vector. Numbers of P. orien

LLIN Evaluation in Uganda Project (LLINEUP): modelling the impact of COVID-19-related disruptions on delivery of long-lasting insecticidal nets on malaria indicators in Uganda.

BACKGROUND: Disruptions in malaria control due to COVID-19 mitigation measures were predicted to increase malaria morbidity and mortality in Africa substantially. In Uganda, long-lasting insecticidal nets (LLINs) are distributed nationwide every 3-4 years, but the 2020-2021 campaign was altered because of COVID-19 restrictions so that the timing of delivery of new nets was different from the original plans made by the National Malaria Control Programme. METHODS: A transmission dynamics modelling exercise was conducted to explore how the altered delivery of LLINs in 2020-2021 impacted malaria

Optimising the deployment of vector control tools against malaria: a data-informed modelling study.

BACKGROUND: Concern that insecticide resistant mosquitoes are threatening malaria control has driven the development of new types of insecticide treated nets (ITNs) and indoor residual spraying (IRS) of insecticide. Malaria control programmes have a choice of vector control interventions although it is unclear which controls should be used to combat the disease. The study aimed at producing a framework to easily compare the public health impact and cost-effectiveness of different malaria prevention measures currently in widespread use. METHODS: We used published data from experimental hut tri

Laboratory and semi-field evaluation of long-lasting insecticidal nets against leishmaniasis vector, Phlebotomus (Phlebotomus) duboscqi in Kenya.

BACKGROUND & OBJECTIVES: Phlebotomine sandflies are vectors of leishmaniases and other diseases. Long-lasting insecticidal nets (LLINs) as possible tools for control have not been widely tested against them. The objective of this study was to determine the efficacy of Olyset Net and PermaNet LLINs alongside a local brand, K-O Tab treated net (Supanet) against Phlebotomus duboscqi female sandflies. METHODS: Four replicates of unwashed and 20x washed Olyset Nets and PermaNets, K-O Tab-treated and untreated Supanet and 'no net' treatments were evaluated against sandflies within the laboratory by

Characterizing the escalation of pyrethroid resistance and its impact on bed nets efficacy alongside molecular basis in Anopheles funestus from Cameroon.

BACKGROUND: Malaria prevention still relies greatly on vector control interventions. However, increasing levels of resistance to pyrethroids across Africa have significantly reduced the efficacy of pyrethroid-based interventions leading to an increase of malaria burden. Consequently, it is imperative to characterize the extent and molecular basis of this resistance. METHODS: This study was conducted from 2020 to 2021 in a South-North transect across Cameroon. WHO tube assay was used to assess the susceptibility profile of Anopheles funestus to the four main classes of insecticides. The effica

Modelling the impact of different intervention packages for malaria control under varying intensities of pyrethroid resistance.

BACKGROUND: Malaria control in sub-Saharan Africa faces significant challenges from biological threats, such as insecticide resistance and adaptive vector behaviours, as well as increasing financial constraints, which necessitate strategic intervention planning to maximize impact. This study assesses the effectiveness of combining vector control methods, case management, and immunoprevention to reduce malaria in Tanzania, considering varying intensities of insecticide resistance in the main vector species. METHODS: A compartmental model was developed to simulate malaria transmission, incorpor

Identification of a rapidly-spreading triple mutant for high-level metabolic insecticide resistance in Anopheles gambiae provides a real-time molecular diagnostic for antimalarial intervention deployment.

Studies of insecticide resistance provide insights into the capacity of populations to show rapid evolutionary responses to contemporary selection. Malaria control remains heavily dependent on pyrethroid insecticides, primarily in long lasting insecticidal nets (LLINs). Resistance in the major malaria vectors has increased in concert with the expansion of LLIN distributions. Identifying genetic mechanisms underlying high-level resistance is crucial for the development and deployment of resistance-breaking tools. Using the Anopheles gambiae 1000 genomes (Ag1000g) data we identified a very recen

Chlorfenapyr bednets effectively overcome pyrethroid resistance escalation in highly resistant Anopheles malaria vectors in Uganda.

Escalating insecticide resistance threatens the efficacy of LLINs, undermining malaria control in Africa. We conducted the first experimental hut trials in Uganda using highly resistant free-flying wild Anopheles mosquitoes and F2 hybrids of FANG and Uganda An. funestus to evaluate the performance of bednets. The interceptor G2 (chlorfenapyr) bednet demonstrated superior efficacy compared to Interceptor (pyrethroid-only) net [mortality odds ratio (OR): 18.7 (8.05-48.6) P < 0.0001], achieving an overall mortality rate of 70.6% and 63.2% against An. funestus and An. gambiae respectively. The mini chamber test: a novel bioassay for bioefficacy testing of chlorfenapyr treated nets used in malaria control.

Dual-active-ingredient insecticide-treated nets (Dual-AI ITNs) combining pyrethroids with chlorfenapyr are increasingly being deployed across malaria-endemic areas. Laboratory evaluation of chlorfenapyr ITNs is challenging due to chlorfenapyr's requirement for metabolic activation. Inspired by the tunnel test, the mini chamber test is smaller, has higher throughput, and is animal-free. We optimised the primary parameters using pyrethroid-susceptible and -resistant mosquito strains exposed to new pyrethroid-chlorfenapyr ITNs versus controls. With parameters defined, we used a draft standard ope

Integrating zooprophylaxis, insecticide-treated livestock and mosquito nets for malaria control: a One Health proposal.

Malaria transmission persists despite widespread use of insecticide-treated nets (ITNs) due to multiple factors, including outdoor and early-evening biting and insecticide resistance. In this context, zooprophylaxis, which strategically uses livestock to divert mosquitoes from humans, has shown mixed epidemiological effects. Conversely, treating livestock with topical or systemic insecticides (known as insecticide-treated livestock [ITL]) has been demonstrated to significantly reduce mosquito survival. Combining ITL and zooprophylaxis with ITNs could improve vector control for species that fee

Can the performance of pyrethroid-chlorfenapyr nets be reduced when combined with pyrethroid-piperonyl butoxide (PBO) nets?

BACKGROUND: Pyrethroid-chlorfenapyr (CFP) and pyrethroid-piperonyl butoxide (PBO) nets are being scaled across endemic countries to improve control of malaria transmitted by pyrethroid-resistant mosquitoes. CFP is a pro-insecticide requiring activation by mosquito cytochrome P450 monooxygenase enzymes (P450s) while PBO improves pyrethroid potency by inhibiting the action of these enzymes in pyrethroid-resistant mosquitoes. The inhibitory action of PBO against P450s may thus reduce the efficacy of pyrethroid-CFP nets when applied inside the same household as pyrethroid-PBO nets. METHODS: Two e

Combination of different intervention strategies for malaria mosquito vector control in Uganda: A review of secondary data of two districts with moderate to high disease transmission.

BACKGROUND: Indoor residual spraying (IRS) and insecticide treated bed nets (ITNs) used either singly or in combination are the main mass mosquito vector control measures for malaria control. Despite their widespread use, malaria transmission rates remain high, the burden is unacceptably huge, and yet the disease is completely preventable. This study measured the impact of compounds used in different types and campaigns of IRS and ITNs, different permutations of IRS + ITNs on malaria test positivity rates in high disease transmission settings of Yumbe district and Gulu district. Implementation performance of routine insecticide-treated nets distribution through the health facilities in mainland Tanzania: 5 years (2018-2022) experience.

BACKGROUND: The distribution of insecticide-treated nets (ITNs) through health facilities helps increase and sustain access to ITNs, targeting biologically vulnerable populations. However, their effectiveness in terms of issuance rates has yet to be assessed over time in Tanzania. METHODS: Monthly DHIS-2 data on ITN distribution from health facilities were extracted from 6,790 facilities from January 2018 to December 2022. The Generalized Estimating Equation (GEE) was used to model the marginal performance trend over time for ANC and EPI separately. RESULTS: Across 5 years, ANC had an issuan

A mathematical model of visceral leishmaniasis transmission and control: Impact of ITNs on VL prevention and elimination in the Indian subcontinent.

Visceral Leishmaniasis (VL) is a deadly, vector-borne, parasitic, neglected tropical disease, particularly prevalent on the Indian subcontinent. Sleeping under the long-lasting insecticide-treated nets (ITNs) was considered an effective VL prevention and control measures, until KalaNet, a large trial in Nepal and India, did not show enough supporting evidence. In this paper, we adapt a biologically accurate, yet relatively simple compartmental ordinary differential equations (ODE) model of VL transmission and explicitly model the use of ITNs and their role in VL prevention and elimination. We

Implementation of a malaria prevention education intervention in Southern Ethiopia: a qualitative evaluation.

BACKGROUND: Though school-aged children (SAC) are at high risk of malaria, they are the ones that benefit the least from malaria prevention measures. A cluster randomized controlled trial was conducted to evaluate the effect of malaria prevention education (MPE) on insecticide-treated bed net (ITN) utilization and prompt diagnosis, reported incidence and treatment (PDAT) of malaria. Qualitative evaluation of the implementation of such interventions is vital to explain its effectiveness and will serve as guidance for future interventions. Therefore, this study aimed to evaluate the implementati

The influence of feeding behaviour and temperature on the capacity of mosquitoes to transmit malaria.

Insecticide-treated bed nets reduce malaria transmission by limiting contact between mosquito vectors and human hosts when mosquitoes feed during the night. However, malaria vectors can also feed in the early evening and in the morning when people are not protected. Here, we explored how the timing of blood feeding interacts with environmental temperature to influence the capacity of Anopheles mosquitoes to transmit the human malaria parasite Plasmodium falciparum. In laboratory experiments, we found no effect of biting time itself on the proportion of mosquitoes that became infectious (vector

Targeting cattle for malaria elimination: marked reduction of Anopheles arabiensis survival for over six months using a slow-release ivermectin implant formulation.

BACKGROUND: Mosquitoes that feed on animals can survive and mediate residual transmission of malaria even after most humans have been protected with insecticidal bednets or indoor residual sprays. Ivermectin is a widely-used drug for treating parasites of humans and animals that is also insecticidal, killing mosquitoes that feed on treated subjects. Mass administration of ivermectin to livestock could be particularly useful for tackling residual malaria transmission by zoophagic vectors that evade human-centred approaches. Ivermectin comes from a different chemical class to active ingredients
