The sulfhydration of cysteine residues in proteins is an important mechanism involved in diverse biological processes. We have developed a proteomics approach to quantitatively profile the changes of sulfhydrated cysteines in biological systems. Bioinformatics analysis revealed that sulfhydrated cysteines are part of a wide range of biological functions. In pancreatic β cells exposed to endoplasmic reticulum (ER) stress, elevated H2S promotes the sulfhydration of enzymes in energy metabolism and stimulates glycolytic flux. We propose that transcriptional and translational reprogramming by the

Oxidative stress plays a significant regulatory role in tumor immune responses and can influence the efficacy of immunotherapy. Accordingly, therapeutic interventions targeting oxidative stress-related mechanisms, whether alone or in combination with other modalities, represent a compelling strategy to enhance cancer therapy. In this study, we first profiled the landscape of oxidative stress responses within the tumor microenvironment by integrating pan-cancer single-cell RNA sequencing datasets, which revealed that cancer-associated fibroblasts (CAFs) possessed the highest oxidative stress re

In response to stress, defined as a real or perceived threat to homeostasis or well-being, brain systems initiate divergent physiological and behavioral processes that mobilize energy and promote adaptation. The brainstem contains multiple nuclei that engage in autonomic control and reflexive responses to systemic stressors. However, brainstem nuclei also play an important role in neuroendocrine responses to psychogenic stressors mediated by the hypothalamic-pituitary-adrenocortical axis. Further, these nuclei integrate neuroendocrine responses with stress-related behaviors, significantly impa

Dominance hierarchies in fish are established through conflicts and lead to significant differences in stress physiology, behaviour, and immune function between dominant and subordinate individuals. This review explores the relationship between social stress within these hierarchies, individual variations in stress responses and cognitive bias towards stressful situations, and the impact of conflicts on future performance and interactions between stress and the immune system. Fish express divergent stress coping styles (proactive and reactive) that differ in the reactivity of the stress axes e

Monocytes are increasingly implicated in Parkinson's disease (PD) pathogenesis, with idiopathic cases showing mitochondrial and lysosomal dysfunction. However, the impact of PD-associated mutations on monocytes remains unclear. To address this, we investigated transcriptomic and functional disturbances in peripheral monocytes from patients with GBA1- and LRRK2-associated PD and idiopathic PD. Transcriptomic data revealed shared and mutation-specific signatures, including those related to immune dysregulation, and consistent defects in lysosomal, proteasomal and mitochondrial pathways. BACKGROUND: Integrated stress response (ISR) participates in the development of tumor, however, the roles of ISR-related genes (ISR-RGs) in pituitary neuroendocrine tumor (PitNET) are unclear. The goal of this study was to identify the biomarkers related to ISR-RGs in PitNET and explore their action mechanism. METHODS: In the present study, the public datasets from Gene Expression Omnibus database (GEO) database and ISR-RGs from literature were analyzed to obtain biomarkers for PitNET using differential expression analysis, machine learning algorithm, Boruta analysis and expression validation.

Aminoacyl-tRNA synthetases, as pivotal enzymes in protein biosynthesis, have been linked to the development of certain diseases when specific isoforms are dysregulated. However, the involvement of leucyl-tRNA synthetase (LARS) in hepatic pathologies remains unexplored. In this study, a marked upregulation of LARS was observed in cholestatic liver tissues. Mice with liver-specific LARS knockdown manifested substantially attenuated hepatic injury, evidenced by reductions in serum alanine aminotransferase, aspartate aminotransferase, and alkaline phosphatase levels, as well as dysregulated bile a

This study presents the first reported case of a 3-year-old child with EIF2AK2-related dystonia treated with adjunctive luteolin supplementation. EIF2AK2-related dystonia, characterized by exacerbations during infections, is associated with disruptions in the integrated stress response (ISR). The ISR, a cellular signaling pathway activated in response to stress, culminates in the phosphorylation of eIF2α, which modulates protein synthesis and can induce cell death. Pathogenic variants in EIF2AK2 disrupt this pathway, contributing to the development of dystonia. Luteolin, a flavonoid possessing

BACKGROUND: Sepsis-induced acute respiratory distress syndrome (ARDS) is a fatal inflammatory lung injury. The integrated stress response (ISR) may play stage-dependent roles in lung injury, but its clinical relevance in sepsis-induced ARDS remains unclear. We aimed to identify ISR-related biomarkers and explore their potential mechanisms.
METHODS: Public transcriptomic datasets of sepsis-induced ARDS were analyzed. Peripheral blood mononuclear cell (PBMC) single-cell RNA sequencing (scRNA-seq) was used to define key immune cell populations and intercellular communication. Differential expres

Hepatic glucolipotoxicity, characterized by the synergistic detrimental effects of elevated glucose levels combined with excessive lipid accumulation in hepatocytes, plays a central role in the pathogenesis of various metabolic liver diseases. Despite recent advancements, the precise mechanisms underlying this process remain unclear. Using cultured AML12 and HepG2 cells exposed to excess palmitate, with and without high glucose, as an in vitro model, we aimed to elucidate the cellular and molecular mechanisms underlying hepatic glucolipotoxicity. Our data showed that palmitate exposure induced

Periodontitis serves as a model for chronic inflammatory diseases, offering critical insights into immune regulatory mechanisms at the microbe-host interface. By integrating structural immunity theory with single-cell transcriptomics and multi-omics analyses, we investigated the microbial-epithelial-immune crosstalk in periodontitis. Single-cell transcriptomic analysis revealed robust interactions between immune cells and sulcular and junctional keratinocytes (SK/JKs) in the periodontitis epithelium. SK/JKs, being directly exposed to dental plaque biofilm, exhibit significant activation of end

The integrated stress response (ISR) is implicated in age-related diseases, while the molecular chaperone heat shock protein 70 (HSP70) can facilitate proper protein folding. However, the regulatory mechanism of ISR in insufficient testosterone synthesis of aging Leydig cells (LCs) remains unclear. This study aims to elucidate the regulatory role of ISR in inadequate testosterone synthesis of aging LCs. We observed a positive correlation between testosterone and HSP70 levels, which were found to be decreased in elderly men. ISR was detected in testicular tissue from old mice. The expression of

The Sigma 2 receptor (σ2R) was described pharmacologically more than three decades ago, but its molecular identity remained obscure until recently when it was identified as transmembrane protein 97 (TMEM97). We and others have shown that σ2R/TMEM97 ligands alleviate mechanical hypersensitivity in mouse neuropathic pain models with a time course wherein maximal anti-nociceptive effect is approximately 24 hours following dosing. We sought to understand this unique anti-neuropathic pain effect by addressing two key questions: do these σ2R/TMEM97 compounds act selectively via the receptor, and wha

Inducing cell death by the sphingolipid ceramide is a potential anticancer strategy, but the underlying mechanisms remain poorly defined. In this study, triggering an accumulation of ceramide in acute myeloid leukemia (AML) cells by inhibition of sphingosine kinase induced an apoptotic integrated stress response (ISR) through protein kinase R-mediated activation of the master transcription factor ATF4. This effect led to transcription of the BH3-only protein Noxa and degradation of the prosurvival Mcl-1 protein on which AML cells are highly dependent for survival. Targeting this novel ISR path

The p53 transcription factor is a master regulator of cellular stress responses inhibited by repressors such as MDM2 and the phosphatase PPM1D. Activation of p53 with pharmacological inhibitors of its repressors is being tested in clinical trials for cancer therapy, but efficacy has been limited by poor induction of tumor cell death. We demonstrate that dual inhibition of MDM2 and PPM1D induces apoptosis in multiple cancer cell types via amplification of the p53 transcriptional program through the eIF2α-ATF4 pathway. PPM1D inhibition induces phosphorylation of eIF2α, ATF4 accumulation, and ATF

Traumatic brain injury (TBI) is a leading cause of long-term neurological disability in the world and the strongest environmental risk factor for the development of dementia. Even mild TBI (resulting from concussive injuries) is associated with a greater than twofold increase in the risk of dementia onset. Little is known about the cellular mechanisms responsible for the progression of long-lasting cognitive deficits. The integrated stress response (ISR), a phylogenetically conserved pathway involved in the cellular response to stress, is activated after TBI, and inhibition of the ISR-even wee

The ability to adapt to environmental stress, including therapeutic insult, contributes to tumor evolution and drug resistance. In suboptimal conditions, the integrated stress response (ISR) promotes survival by dampening cytosolic translation. We show that ISR-dependent survival also relies on a concomitant up-regulation of mitochondrial protein synthesis, a vulnerability that can be exploited using mitoribosome-targeting antibiotics. Accordingly, such agents sensitized to MAPK inhibition, thus preventing the development of resistance in BRAFV600E melanoma models. Additionally, this treatment

PURPOSE: The degenerative corneal disease keratoconus is a leading indicator for corneal transplant with an unknown etiology. We recently identified the activation of the integrated stress response (ISR) in ex vivo human corneas and in vitro cell culture. Utilizing small molecules to modulate the ISR we sought to investigate the effects of stimulating the ISR in healthy cells to recapitulate aspects of the in vitro keratoconic phenotype and whether relieving the ISR signaling would recover the disease phenotype.
METHODS: Corneal fibroblasts were extracted from patients undergoing corneal tran

The present study sought to determine a profile of integrated behavioral, brain and autonomic alterations in PTSD. Previous findings suggest that PTSD is associated with changes across electrophysiological (EEG and ERP), autonomic and cognitive/behavioral measures. In particular, PTSD has been associated with reduced cognitive performance, altered cortical arousal (measured by EEG), diminished late ERP component to oddball task targets (reduced P3 amplitude) and increased autonomic arousal relative to healthy controls. The present study examined measures of cognitive function, auditory oddball
