ANAPHYLAXIS IN THE ISOLATED HEART.

The isolated hearts of guinea pigs sensitized to horse serum have been shown to react characteristically upon exposure to small amounts of antigen. The cardiac rate is temporarily accelerated and transient alterations in amplitude of contraction are to be observed. Electrocardiographic abnormalities, previously recorded by remote leads during anaphylactic shock in the intact animal, have been recorded by direct leads from the isolated perfused hearts of sensitized animals during this reaction. An additional effect of anaphylaxis in the isolated heart of the guinea pig is reported: a striking r

Mitochondrial Cx43 hemichannels contribute to mitochondrial calcium entry and cell death in the heart.

Mitochondrial connexin 43 (Cx43) plays a key role in cardiac cytoprotection caused by repeated exposure to short periods of non-lethal ischemia/reperfusion, a condition known as ischemic preconditioning. Cx43 also forms calcium (Ca2+)-permeable hemichannels that may potentially lead to mitochondrial Ca2+ overload and cell death. Here, we studied the role of Cx43 in facilitating mitochondrial Ca2+ entry and investigated its downstream consequences. To that purpose, we used various connexin-targeting peptides interacting with extracellular (Gap26) and intracellular (Gap19, RRNYRRNY) Cx43 domains

The AMP-activated protein kinase modulates hypothermia-induced J wave.

BACKGROUND: The mechanism underlying the occurrence of the J wave in low temperature remains unclear. However, low temperature is associated with metabolic disorder and 5' AMP-activated protein kinase (AMPK), which modulates ionic currents and cardiac metabolism. This study investigated whether AMPK regulation can modulate the occurrence of the J wave at low temperature. METHODS: Unipolar and bipolar leads were used to record monophasic action potential (the endocardium and epicardium) and pseudo-electrocardiograms (inferior leads) to study the cardiac electrical activity. Measurements were t

Isolated perfused working hearts provide valuable additional information during phenotypic assessment of the diabetic mouse heart.

Although murine models for studying the development of cardiac dysfunction in diabetes mellitus are well established, their reported cardiac phenotypes vary. These reported divergences may, in addition to the severity of different models, also be linked to the methods used for cardiac functional assessment. In the present study, we examined the functional changes using conventional transthoracic echocardiography (in vivo) and isolated heart perfusion techniques (ex vivo), in hearts from two mouse models; one with an overt type 2 diabetes (the db/db mouse) and one with a prediabetic state, wher

Large-Animal Biventricular Working Heart Perfusion System with Low Priming Volume-Comparison between in vivo and ex vivo Cardiac Function.

BACKGROUND: Existing large-animal, ex vivo, cardiac perfusion models are restricted in their ability to establish an ischemia/reperfusion condition as seen in cardiac surgery or transplantation. Other working heart systems only challenge one ventricle or require a substantially larger priming volume. We describe a novel biventricular cardiac perfusion system with reduced priming volume. METHODS: Juvenile pig hearts were cardiopleged, explanted, and reperfused ex vivo after 150 minutes of cold ischemia. Autologous whole blood was used as perfusate (minimal priming volume 350 mL). After 15 minu

Factors determining the utilization of glucose in isolated rat hearts.

Glucose utilization by the isolated rat heart is restricted by flux through the membrane transport, phosphofructokinase and glyceraldehyde-3-P dehydrogenase reactions. These reactions can be accelerated by hormones, such as insulin, by mechanical factors, such as the "garden-hose effect", heart rate, and perhaps ventricular pressure development, and by oxygen deprivation. Glucose utilization is restricted by provision of exogenous non-carbohydrate substrates and by utilization of endogenous substrate stores. Isolated hearts perfused as Langendorff preparations with or without a ventricular dra

Cardiac inotropic and hypertrophic effects of Nigella sativa supplementation in rats.

The intrinsic cardiac responses to 2-months Nigella sativa (black seed) oral supplementation (800 mg/kg) in rats were investigated. The isolated hearts were perfused in a Langendorff preparation to demonstrate the effects of N. sativa on the baseline inotropic and chronotropic functions and the myocardial flow rate. N. sativa supplementation induced moderate global (homogenous) cardiac hypertrophy, evident by significant increases in left ventricular and whole heart weights as well as the relative heart weight/body weight ratio. The isolated perfused hearts of Nigella-treated rats showed enhan

Bradycardic and Antiarrhythmic Effects of the D-Limonene in Rats.

BACKGROUND: D-limonene (DL) is a monoterpene and is the major component in the essential oil of citrus fruit. It presents antihyperglycemic and vasodilatation activities. OBJECTIVES: This study evaluated the cardiovascular effects and potential antiarrhythmic of DL in rats. METHODS: Hemodynamic and electrocardiographic (ECG) parameters were measured in male Wistar rats, which under anesthesia had been cannulated in the abdominal aorta and lower vena cava and had electrodes subcutaneously implanted. In the in vitro approach, the heart was removed and perfused using the Langendorff technique.

Respiratory and cardiovascular effects of 3-methylxanthine, a metabolite of theophylline.

The effects of 3-methylxanthine (3-MX), a major metabolite of theophylline in man, were studied and compared with those of theophylline in isolated preparations of human and guinea-pig airways, isolated guinea-pig hearts, and in anaesthetized cats. 3-MX was pharmacologically active, producing effects similar to those of theophylline. The guinea-pig and human airways preparations were relaxed; the rate and force of contraction of guinea-pig hearts were increased. In anaesthetized cats, 3-MX decreased the respiratory overflow and arterial blood pressure, and increased the heart rate. Theophyllin

Rapid mitochondrial repolarization upon reperfusion after cardiac ischemia.

The mitochondrial membrane potential (ΔΨm) drives oxidative phosphorylation and alterations contribute to cardiac pathologies, but real-time assessment of ΔΨm has not been possible. Here we describe noninvasive measurements using mitochondrial heme bL and bH absorbances, which rapidly respond to ΔΨm. Multi-wavelength absorbance spectroscopy enabled their continuous monitoring in isolated mitochondria and the perfused heart. Calibration of heme b absorbance in isolated mitochondria revealed that reduced heme bL relative to total reduced heme b (fbL = bL/(bL + bH)) exhibits a sigmoidal relations

Metabolic dependency of malonate protection against cardiac ischemia-reperfusion injury.

Malonate, an inhibitor of mitochondrial succinate dehydrogenase (SDH), protects against ischemia-reperfusion injury (IRI) when given at early reperfusion, making it a promising therapeutic agent for acute myocardial infarct (AMI) and stroke patients. Prior to clinical studies, it is essential to identify potential translational roadblocks. One such barrier is the metabolic signature of the heart. Here, we explored whether and how the metabolic signature of the heart drive cardiac IRI and modulate malonate protection. Langendorff-perfused mouse hearts were subjected to 30 min ischemia (I) follo

Some pharmacological properties of brain ganglioside.

The effects of a preparation of purified brain ganglioside and of neuraminic acid have been studied on various pharmacological test preparations. In concentrations of 10 mug/ml. or more, ganglioside stimulates the isolated guinea-pig ileum; graded responses are usually obtained. Various substances, known to antagonize the actions of other stimulant substances, fail to affect the response to ganglioside. Ganglioside and neuraminic acid neither stimulate the superior cervical ganglion of the cat nor do they affect ganglionic transmission. Ganglioside and neuraminic acid did not change the respon

Attenuating loss of cardiac conduction during no-flow ischemia through changes in perfusate sodium and calcium.

Myocardial ischemia leads to conduction slowing, cell-to-cell uncoupling, and arrhythmias. We previously demonstrated that varying perfusate sodium (Na+) and calcium (Ca2+) attenuates conduction slowing and arrhythmias during simulated ischemia with continuous perfusion. Cardioprotection was selectively associated with widening of the perinexus, a gap junction adjacent nanodomain important to ephaptic coupling. It is unknown whether perfusate composition affects the perinexus or ischemic conduction during nonsimulated ischemia, when coronary flow is reduced or halted. We hypothesized that alte

Perfusion of rabbit hearts with pig blood results in complement mediated hyperacute xenograft rejection.

Investigation of hyperacute rejection of discordant xenografts has been hampered by the lack of a model for the study of rapid time course events. In vivo models are unsuitable for observation of early rejection processes and, along with most ex-vivo perfusion preparations, are insensitive since no functional demand is placed on the organ which may have undergone extensive damage whilst still appearing viable. For this reason a blood perfused isolated working heart preparation has been developed. With left atrial and left ventricular loading the heart performs measurable work as it ejects into

Existence of the Frank-Starling mechanism in the failing human heart. Investigations on the organ, tissue, and sarcomere levels.

BACKGROUND: The Frank-Starling mechanism is one of the most important physiological principles for regulation of contractile performance. We therefore studied the question of whether this mechanism may be absent or attenuated in end-stage failing human left ventricular myocardium. METHODS AND RESULTS: Different methodological approaches were used to analyze the effects of this mechanism on the organ, tissue, and sarcomere levels: (1) In excised human whole left ventricles (2 donor hearts, 5 failing hearts), diastolic and systolic pressure-volume relationships were obtained. (2) In isolated mu

[State of the glycolytic system in the hypertrophied heart].

A turnover of glycogen was distinctly increased in hypertrophic rat heart within 2 months after coarctation of aorta; this was corroborated by increased 4.5-fold incorporation of 14C-glucose into glycogen and by removing of label from glycogen in heart perfusion using Langendorf preparation. Due to stationary activation of glycolysis secretion of lactate from isolated hypertrophic heart was twice more than that from intact one in aerobic contitions. Activities of glycolytic enzymes, extracted from intact and hypertrophic heart muscles in optimal conditions of incubation, were approximately of

Methods for the Isolation, Culture, and Functional Characterization of Sinoatrial Node Myocytes from Adult Mice.

Sinoatrial node myocytes (SAMs) act as the natural pacemakers of the heart, initiating each heart beat by generating spontaneous action potentials (APs). These pacemaker APs reflect the coordinated activity of numerous membrane currents and intracellular calcium cycling. However the precise mechanisms that drive spontaneous pacemaker activity in SAMs remain elusive. Acutely isolated SAMs are an essential preparation for experiments to dissect the molecular basis of cardiac pacemaking. However, the indistinct anatomy, complex microdissection, and finicky enzymatic digestion conditions have prev

[Sarcocystis cruzi (Protozoa: Apicomplexa: Sarcocystiidae) infection in european bison (Bison bonasus) from Białowieza Forest, Poland].

Samples of oesophagus, diaphragm and heart muscles were taken from one European bison from Białowieza Forest during seasonal European bison elimination in 2008. Five gram of each muscle was examined by staining small samples in 0.2% aqueous solution of methylene blue. After that they were placed between compressor glasses and examined under a dissecting microscope in order to detect Sarcocystis sp. infection. All sarcocysts observed were counted. The 5-g sample of the heart muscle yielded a total of 756 sarcocysts while that of the diaphragm contained 107 of them and that of the oesophagus--89
