Symbiotic fungi underlie the regeneration potential of island rainforests.

Symbioses can be vital on islands, where low species diversity leaves few alternative partners and the failure of associations can cascade into broader community collapse. Key to the functioning of many island ecosystems is the rainforest tree, Pisonia grandis (pisonia). Pisonia attracts nesting seabirds whose guano delivers intense nutrient pulses that fuel coral reef ecosystems. Symbiotic mycorrhizal fungi have been hypothesized to be crucial for capturing and distributing these nutrients to pisonia trees. However, little is known about the factors that influence the distribution of mycorrhi

Research Progress on Arbuscular Mycorrhizal Fungi-mediated Selenium Uptake, Transport, and Metabolism in Plants.

Selenium (Se) is an essential micronutrient for human health, playing a crucial role in antioxidant defense and immune regulation. However, the uneven global distribution of bioavailable Se in soils has led to widespread dietary Se deficiency, posing a potential public health risk. Although plants are the primary dietary source of Se, their intrinsic capacity for Se enrichment is limited. This necessitates the exploration of microbial-assisted strategies for enhancing Se biofortification. Arbuscular mycorrhizal fungi (AMF) are keystone symbionts in the plant-soil continuum and are known to sig

Characterization of Bacteria Living on Arbuscular Mycorrhizal Fungal Hyphae Under Field Conditions.

The fine nature of arbuscular mycorrhizal (AM) fungal hyphae makes it challenging to directly isolate them from soil, hindering the understanding of hyphosphere bacterial communities in the real world. Here, we developed an innovative system combining hyphae in-growth tubes and a 0.45 μm microporous filter membrane, establishing a method for in situ induction of AM fungal extraradical hyphae and indoor collection. This provides an efficient platform for investigating the diversity and functionality of hyphal-associated microbiomes under in situ conditions.

Ericoid mycorrhizal symbiosis: theoretical background and methods for its comprehensive investigation.

Despite decades of intensive research (especially from 1970s to 1990s), the ericoid mycorrhizal (ErM) hair root is still largely terra incognita and this simplified guide is intended to revive and promote the study of its mycobiota. Basic theoretical knowledge on the ErM symbiosis is summarized, followed by practical advices on Ericaceae root sample collection and handling, microscopic observations and photo-documentation of root fungal colonization, mycobiont isolation, maintenance and identification and resynthesis experiments with ericoid plants. The necessity of a proper selection of the r

Convergent losses of decay mechanisms and rapid turnover of symbiosis genes in mycorrhizal mutualists.

To elucidate the genetic bases of mycorrhizal lifestyle evolution, we sequenced new fungal genomes, including 13 ectomycorrhizal (ECM), orchid (ORM) and ericoid (ERM) species, and five saprotrophs, which we analyzed along with other fungal genomes. Ectomycorrhizal fungi have a reduced complement of genes encoding plant cell wall-degrading enzymes (PCWDEs), as compared to their ancestral wood decayers. Nevertheless, they have retained a unique array of PCWDEs, thus suggesting that they possess diverse abilities to decompose lignocellulose. Similar functional categories of nonorthologous genes a

Whole-genome sequence of Rahnella aceris HPDA25 isolated from mycorrhizosphere of Gastrodia elata.

OBJECTIVES: The growth, development, and accumulation of active compounds in Gastrodia elata are influenced by its mycorrhizosphere bacteria, which indirectly enhances host nutrition by stimulating the growth of the orchid mycorrhizal fungus Armillaria gallica. However, data on the mycorrhizosphere bacterial community of G. elata, especially on key functional strains, remain limited, restricting their application in medicinal plant cultivation. Here, we report the complete genome sequence of the plant growth-promoting bacterium Rahnella aceris HPDA25, which was isolated from the mycorrhizosphe

Mycorrhizal strategy of non-native plants varies with biome and disturbance.

Predicting which non-native plant species will become established and where is critical for conserving and managing biodiversity. Theory suggests that the mycorrhizal strategy of non-native plants may predict their establishment success. Here we combine a global dataset of 440,788 vegetation plots with data on plant native status and mycorrhizal type to assess mycorrhizal strategy of non-native plants. The mycorrhizal strategy of non-native plants varies strongly across biomes. Across grassland and desert biomes, non-native species are more frequently non-mycorrhizal than native species, where

Application of Virus-Induced Gene Silencing to Arbuscular Mycorrhizal Fungi.

Downregulation of AM fungal genes using a plant viral vector is feasible. A partial sequence of a target gene is cloned into the multicloning site of the CMV2-A1 vector developed from RNA2 of Cucumber mosaic virus Y strain, and the RNA2, together with RNA1 and RNA3 of the virus, are in vitro-transcribed. Inoculation of Nicotiana benthamiana with these viral RNAs results in reconstitution of the virus in the plant, which triggers silencing of the fungal gene. Here, we describe not only the methods but also several tips for the successful application of virus-induced gene silencing to AM fungi.

Effects of ectomycorrhizal fungi on the physiological and ecological characteristics of Larix gmelinii under salt stress.

Soil salinization has become a major constraint on afforestation and forest regeneration in Northeast China, limiting the growth and development of Larix gmelinii. Enhancing the salt tolerance of L. gmelinii is therefore of considerable importance, and ectomycorrhizal fungi (ECMF) inoculation has been shown to improve plant stress resistance. However, the mechanisms by which ECMF enhance salt tolerance in L. gmelinii remain poorly understood. In this study, L. gmelinii seedlings inoculated with an ECMF consortium consisting of Boletus edulis and Chroogomphus rutilus were subjected to 120 mM Na

Intracellular Inorganic Orthophosphate Distribution in Arbuscular Mycorrhizal Root.

Phosphate is an essential macronutrient for plants and plays a key role in the establishment and regulation of arbuscular mycorrhizal (AM) symbiosis. However, conventional methods for studying phosphate levels in plant tissues often lack cellular resolution or require technically demanding procedures such as genetic transformation. Here, we present an adaptation of the recently developed inorganic orthophosphate staining assay (IOSA), for Lotus japonicus roots colonized by the AM fungus Rhizophagus irregularis. IOSA is a rapid, colorimetric method that enables high-resolution, semi-quantitativ

Methods for Single-Spore Cultures of Arbuscular Mycorrhizal Fungi Using a Superabsorbent Polymer-Based Autotrophic System.

Single-spore cultures of arbuscular mycorrhizal fungi (AMF) are essential for taxonomy, genomics, experimental ecology, and the production of well-characterized inoculum. Classical pot cultures in opaque substrates make monitoring the establishment of symbiosis and verifying culture purity difficult, whereas in vitro cultures on Ri T-DNA-transformed roots require sterile conditions and support only a limited subset of AMF species. Transparent "soils" based on superabsorbent polymers (SAPs) offer a simple alternative that combines the advantages of in vivo and in vitro systems. Here, we present

Arbuscular mycorrhizal fungi and their response to pesticides.

Arbuscular mycorrhizal fungi (AMF) form symbioses with the majority of plant species and can provide multiple benefits to the host plant. In agro-ecosystems, the abundance and community structure of AMF are affected by agricultural management practices. This review describes and discusses current knowledge on the effects of inorganic and organic chemical pesticides on AMF in the conflicting area between agricultural use and environmental concerns. Variable effects have been reported following chemical pesticide use, ranging from neutral to positive and negative. Moreover, a species-specific re

Variations in the chemical composition of cassava ( Manihot esculenta Crantz) leaves and roots as affected by genotypic and environmental variation.

The purpose of this study was to assess the quality of cassava cultivars, in terms of cyanogenic potential and composition of macro- and micronutrients, sampled from different locations in rural Mozambique. Total cyanide concentrations in fresh cassava tissues were measured using portable cyanide testing kits, and elemental nutrients were later analyzed from dried plant tissue. Variation in cyanogenic potential and nutrient composition occurred both among cultivars and across locations. The majority of cultivars contained >100 ppm total cyanide, fresh weight, and are therefore considered to be

Techniques and Strategies for the Conservation of Arbuscular Mycorrhizal Fungi.

Arbuscular mycorrhizal symbiosis between fungi of the phylum Glomeromycota and the roots of terrestrial plants is fundamental for nutrient and water absorption, thereby improving plant nutritional efficiency. Arbuscular mycorrhizal fungi (AMF) influence key ecological processes, such as nutrient cycling and carbon stabilization in soil, making them crucial to agricultural sustainability. AMF conservation is carried out using various techniques. One of the most common is the use of trap crops, where host plants that favor efficient fungal colonization are selected. This method allows for mainta

Arbuscular mycorrhizal fungi reshape carbon-nitrogen balance and metabolism in nitrogen-limited escarole (Cichorium endivia L.).

Nitrogen nutrition remains a major challenge for sustainable crop production, particularly under the current need to reduce fertilizer inputs. This study evaluated whether inoculation with arbuscular mycorrhizal fungi (AMF) improves the physiological, nutritional, and metabolic performance of escarole (Cichorium endivia L.) plants under nitrogen-limited conditions. Escarole plants were grown under two nitrogen regimes, full nitrogen supply and 25% of the optimal nitrogen concentration, either inoculated or non-inoculated with Rhizophagus irregularis. Plant growth, mineral composition, soluble

Nanoplastics shift arbuscular mycorrhizal fungal mediation of bacterial community assembly from diversity to compositional stability under arsenic stress.

Arbuscular mycorrhizal fungi (AMF) play a vital role in contaminated soil remediation, yet how NPs regulate AMF mediated arsenic (As) remediation under NP and As co-contamination remains unclear. This study aims to elucidate the effects of NP on the strategic shifts in AMF in response to As exposure. This study demonstrates that NP fundamentally redirect the strategy of AMF. The result shows that AMF inoculation significantly increased community diversity under individual NP or As stress. Instead, AMF orchestrated a profound restructuring of the microbial community, notably enriching key phyla

A Whole-Plant Culture Method to Study Structural and Functional Traits of Extraradical Mycelium.

An in vivo whole-plant bi-dimensional experimental system has been devised and tested with different host plants to obtain extraradical mycelium (ERM) produced by different isolates of arbuscular mycorrhizal fungi (AMF). In this system, a plantlet is inoculated with AMF to establish mycorrhizal symbiosis and, after colonization, newly formed extraradical hyphae and spores are removed. Then, the mycorrhizal root system is wrapped in a nylon net and placed between two membranes in a Petri dish, allowing ERM to grow on the membrane surfaces. Such extraradical hyphae may be used for in situ morpho

Collection of Arbuscular Mycorrhizal Hyphal Exudates Under in Vitro Culture Conditions.

Hyphal exudates of arbuscular mycorrhizal (AM) fungi can be collected in vitro using dual-compartment or split-plate systems. AM fungi are co-cultivated with host roots in one compartment, while hyphae grow across the central barrier into a root-free hyphal compartment. Once sufficient hyphal growth is established, sterile nutrient solution or phosphate-buffered saline solution is added to the hyphal compartment. After 24-48 h, the solution is collected. This method enables root-free access to AM fungal secretions for metabolic and functional analyses.
