Synthesis and Characterization of Palmitoyl-block-poly(methacryloyloxyethyl phosphorylcholine) Polymer Micelles for Anticancer Drug Delivery.

We report the synthesis and characterization of an amphiphilic polymer comprising a hydrophobic palmitoyl (Pal) group and a zwitterionic poly(2-methacryloyloxyethyl phosphorylcholine) (pMPC) block, which is capable of forming micelles as a drug carrier system for delivering hydrophobic anticancer drugs such as doxorubicin (DOX). We hypothesize that the sharp polarity contrast between the Pal domain and the pMPC block would strengthen the micelles and improve the drug loading capacity, while the pMPC shells improve the micelle stability and cellular uptake efficiency. In this study, the Pal-pMP…

Mitigation of Cellular and Bacterial Adhesion on Laser Modified Poly (2-Methacryloyloxyethyl Phosphorylcholine)/Polydimethylsiloxane Surface.

Nowadays, using polymers with specific characteristics to coat the surface of a device to prevent undesired biological responses can represent an optimal strategy for developing new and more efficient implants for biomedical applications. Among them, zwitterionic phosphorylcholine-based polymers are of interest due to their properties to resist cell and bacterial adhesion. In this work, the Matrix-Assisted Laser Evaporation (MAPLE) technique was investigated as a new approach for functionalising Polydimethylsiloxane (PDMS) surfaces with zwitterionic poly(2-Methacryloyloxyethyl-Phosphorylcholin…

Superlubricious surface mimicking articular cartilage by grafting poly(2-methacryloyloxyethyl phosphorylcholine) on orthopaedic metal bearings.

Aseptic loosening of the artificial hip joint with osteolysis due to the wear particles from polyethylene cup has remained as a serious issue. To reduce this wear and develop a novel artificial hip joint system, we produced a superlubricious metal-bearing material: for this, we grafted a 2-methacryloyloxyethyl phosphorylcholine (MPC) polymer onto the surface of a cobalt-chromium-molybdenum (Co-Cr-Mo) alloy. For ensuring long-term benefit retention of poly(MPC) on the Co-Cr-Mo alloy for application as a novel artificial hip joint system, several issues must be considered: strong bonding between…

High lubricious surface of cobalt-chromium-molybdenum alloy prepared by grafting poly(2-methacryloyloxyethyl phosphorylcholine).

Osteolysis caused by wear particles from polyethylene in artificial hip joints is of great concern. Various bearing couple combinations, bearing material improvements, and surface modifications have been attempted to reduce such wear particles. With the aim of reducing the wear and developing a novel artificial hip-joint system, we created a highly lubricious metal-bearing material: A 2-methacryloyloxyethyl phosphorylcholine (MPC) polymer was grafted onto the surface of the cobalt-chromium-molybdenum (Co-Cr-Mo) alloy. For ensuring the long-term retention of poly(MPC) on the Co-Cr-Mo alloy, we …

Modulation of the host immune system by phosphorylcholine-containing glycoproteins secreted by parasitic filarial nematodes.

Phosphorylcholine (PC) is increasingly becoming recognised as a carbohydrate-associated component of a wide variety of procaryotic and eucaryotic pathogens. Studies employing nematode PC-containing molecules indicate that it possesses a plethora of immunomodulatory activities. ES-62 is a PC-containing glycoprotein, which is secreted by the rodent filarial nematode Acanthocheilonema viteae and which provides a model system for the dissection of the mechanisms of immune evasion induced by related PC-containing glycoproteins expressed by human filarial nematodes. At concentrations equivalent to t…

1-O-Alkyl-sn-glyceryl-3-phosphorylcholines: a novel class of neutrophil stimulants.

1-O-Alkyl-2-O-acetyl-sn-glyceryl-3-phosphorylcholine aggregates and degranulates platelets and polymorphonuclear neutrophils. Here, the bioactivities of this platelet-activating factor, its 2-O-ethyl, and its 2-lyso derivatives were examined further. Each phospholipid aggregated and degranulated rabbit platelets and neutrophils with relative potencies of about 10,000 1,000, and 1, respectively. For rabbit neutrophils, and 2-O-acetyl compound was active in nanomolar and lower concentrations; required extracellular calcium and magnesium in order to induce aggregation; and required extracellular …

Decreased nematode clearance and anti-phosphorylcholine-specific IgM responses in mannose-binding lectin-deficient mice.

Brugia malayi is a nematode that causes human lymphatic filariasis. Previously, we showed that mannose-binding lectin (MBL)-A is necessary f … endent or T-independent antigens. However, absence of MBL-A and/or MBL-C resulted in reduced IgM to phosphorylcholine, a constituent of filarial and bacterial antigens, suggesting that inability to form proficient antibody responses to this moiety leads to lack of microfilarial clearance and overall susceptibility to filariasis.

Enhanced wear resistance of modified cross-linked polyethylene by grafting with poly(2-methacryloyloxyethyl phosphorylcholine).

We developed a cross-linked polyethylene (CLPE) modified with a phospholipid polymer in order to address the serious problem of osteolysis caused by wear particles derived from the polyethylene components of artificial hip joints. Our goal of preventing aseptic loosening could be achieved by avoiding any formation of CLPE wear particles or suppressing the activation of cell systems by the wear particles. We investigated the surface and wear resistance properties of 2-methacryloyloxyethyl phosphorylcholine (MPC) polymer grafted onto the surface of CLPE (CLPE-g-MPC). The relative density of MPC …

Identification of phosphorylcholine containing antigens of Fasciola hepatica--successful tolerization against this epitope in experimental animals.

Phosphorylcholine containing antigens have been identified in the parasite Fasciola hepatica by immunoblotting and ELISA. Immunoblots probed with polyclonal and monoclonal antibodies indicate that the majority of antigens identified in both the immature and mature parasite contain both phosphorylcholine and non-phosphorylcholine epitopes. One antigen of 58 kDa appears to contain predominantly PC epitopes or at least this epitope is the major one responded to by host animals. Successful immunotolerization against the epitope PC was achieved by injecting the PC conjugate, ovalbumin PC, into neon…

Delayed Magnetic Resonance Imaging of Alzheimer's Disease by Using Poly(2-(methacryloyloxy)ethyl phosphorylcholine)-Functionalized Nanoprobes.

Alzheimer's disease (AD) is one of the most common neurodegenerative diseases, commonly affecting the aged, with pathophysiological changes presenting 15 to 20 years before clinical symptoms. Early diagnosis and intervention are crucial in effectively slowing the progression of AD. In the current study, poly(2-(methacryloyloxy)ethyl phosphorylcholine) (PMPC)-functionalized NaGdF4 nanoparticles (NaGdF4-PMPC) were developed as magnetic resonance imaging (MRI) contrast agents for targeting alpha 7 nicotinic acetylcholine receptors (α7 nAChRs) in AD mice. NaGdF4-PMPC showed excellent biocompatibil…

Specific binding sites for 1-O-alkyl-sn-glyceryl-3-phosphorylcholine on intact human blood neutrophils.

Infection, inflammation and allergy are characterized by an infiltration of neutrophils or eosinophils in the tissue and are associated with an increased level of lysophospholipids. In this study it is shown that labeled 1-O-alkyl-sn-glyceryl-3-phosphorylcholine (lyso-paf) binds to intact human blood neutrophils. Unlabeled lyso-paf(500 nM) inhibits the binding of [3H]lyso-paf to neutrophils in the presence of fatty acid-free serum albumin (0.25%). A linear Scatchard plot analysis of the specific [3H]lyso-paf binding to neutrophils revealed a KD value of 9.2 nM with 4,100 lyso-paf binding sites…

Coupled effects of molecular weight and grafting density on protein adsorption in phosphorylcholine-based zwitterionic brushes.

Zwitterionic polymer brushes are widely used to suppress nonspecific protein adsorption, yet their performance depends jointly on molecular weight and grafting density. Rational design remains difficult because these parameters are often examined separately or represented by a single conformational descriptor. Here, bovine serum albumin adsorption was systematically investigated across 22 poly(2-methacryloyloxyethyl phosphorylcholine) brush states prepared using a polydopamine-mediated grafting-to method that enabled systematic variation of molecular weight and grafting density. Quartz crystal…

Incorporation of phosphorylcholine into the lipooligosaccharide of nontypeable Haemophilus influenzae does not correlate with the level of biofilm formation in vitro.

Nontypeable Haemophilus influenzae (NTHi) is an opportunistic pathogen that causes otitis media in children and community-acquired pneumonia or exacerbations of chronic obstructive pulmonary disease in adults. A large variety of studies suggest that biofilm formation by NTHi may be an important step in the pathogenesis of this bacterium. The objective of this report was to determine the relationship between the presence of phosphorylcholine in the lipooligosaccharide of NTHi and the level of biofilm formation. The study was performed on 111 NTHi clinical isolates collected from oropharyngeal s…

Investigation of strategies with potential for producing a phosphorylcholine-free version of the filarial nematode immunomodulator, ES-62.

Phosphorylcholine (PC) is found attached to N-type glycans of proteins secreted by filarial nematodes, where it appears to act as an immunomodulator. Based on information on the structure and biosynthesis of the PC-glycan of a major secreted protein, ES-62, strategies were designed with potential for preparing PC-free material to better understand the importance of PC in filarial nematode immunomodulation. The strategies involve either enzymatic removal of PC or inhibition of its attachment during ES-62 synthesis. No method tested was found to be 100% effective although approximately 70% remov…

N-glycans of the porcine nematode parasite Ascaris suum are modified with phosphorylcholine and core fucose residues.

In recent years, the glycoconjugates of many parasitic nematodes have attracted interest due to their immunogenic and immunomodulatory nature. Previous studies with the porcine roundworm parasite Ascaris suum have focused on its glycosphingolipids, which were found, in part, to be modified by phosphorylcholine. Using mass spectrometry and western blotting, we have now analyzed the peptide N-glycosidase A-released N-glycans of adults of this species. The presence of hybrid bi- and triantennary N-glycans, some modified by core alpha1,6-fucose and peripheral phosphorylcholine, was demonstrated by…

Identification and characterization of pptA: a gene involved in the phase-variable expression of phosphorylcholine on pili of Neisseria meningitidis.

Pili of pathogenic Neisseria are major virulence factors associated with adhesion, cytotoxicity, twitching motility, autoaggregation, and DNA transformation. Pili are modified posttranslationally by the addition of phosphorylcholine. However, no genes involved in either the biosynthesis or the transfer of phosphorylcholine in Neisseria meningitidis have been identified. In this study, we identified five candidate open reading frames (ORFs) potentially involved in the biosynthesis or transfer of phosphorylcholine to pilin in N. meningitidis. Insertional mutants were constructed for each ORF in …

The effect of an aldose reductase inhibitor on lens phosphorylcholine under hyperglycemic conditions: biochemical and NMR studies.

Phosphorylcholine (P-choline) is a precursor of the phospholipids in the lens membrane. A human lens normally contains approx. 1 mM P-choline but this is significantly lowered in some cataractous lenses. A normal rat lens contains a very high concentration (11 mM). We found that rat lens P-choline was depleted drastically when the lenses were exposed to hyperglycemic conditions either in culture, with galactose or xylose, or in vivo by streptozotocin-induced diabetes. The lens P-choline level was measured by fractionating the organic phosphates in the lens homogenate using an ion exchange colu…
