Perfluoroalkyl acid (PFAA) levels and profiles in breast milk, maternal and cord serum of French women and their newborns.

One major concern regarding perfluoroalkyl acids (PFAAs) is their potential role in onset of health troubles consecutive to early exposure during the perinatal period. In the present work, the internal exposure levels of 18 targeted PFAAs were determined in ca. 100 mother-newborn pairs recruited in France between 2010 and 2013. In serum, the cumulated concentrations of the 7 most frequently detected compounds were 5.70ng/mL and 2.83ng/mL (median values) in maternal and cord serum, respectively. Perfluorooctanesulfonic acid (PFOS), perfluorooctanoic acid (PFOA), perfluorohexylesulfonic acid (PF

Estrogenic compounds determination in water samples by dispersive liquid-liquid microextraction and micellar electrokinetic chromatography coupled to mass spectrometry.

In this work, a group of 12 estrogenic compounds, i.e., four natural sexual hormones (estrone, 17β-estradiol, 17α-estradiol and estriol), an exoestrogen (17α-ethynylestradiol), a synthetic stilbene (dienestrol), a mycotoxin (zearalenone) and some of their major metabolites (2-methoxyestradiol, α-zearalanol, β-zearalanol, α-zearalenol and β-zearalenol) have been separated and determined by micellar electrokinetic chromatography (MEKC) coupled to electrospray ion trap mass spectrometry. For this purpose, a background electrolyte containing an aqueous solution of 45 mM of perfluorooctanoic acid (

Alterations of hippocampal glucose metabolism by even versus uneven medium chain triglycerides.

Medium chain triglycerides (MCTs) are used to treat neurologic disorders with metabolic impairments, including childhood epilepsy and early Alzheimer's disease. However, the metabolic effects of MCTs in the brain are still unclear. Here, we studied the effects of feeding even and uneven MCTs on brain glucose metabolism in the mouse. Adult mice were fed 35% (calories) of trioctanoin or triheptanoin (the triglycerides of octanoate or heptanoate, respectively) or a matching control diet for 3 weeks. Enzymatic assays and targeted metabolomics by liquid chromatography tandem mass spectrometry were

A two-center study of the development of acute irritation responses to fatty acids.

BACKGROUND: A human 4-hour patch test has been developed for evaluating the acute irritation potential of chemicals. This method was developed for comparative irritation assessments. Although skin irritation responses in human subjects can be quite variable, this test method has proven robust in both intra- and interlaboratory tests. However, the previous interlaboratory studies were not optimal in that slightly differing protocols were used and the studies were not controlled for time of year or source of test chemicals. As a result, some variation in acute irritation responses were seen that

Inhibition of citrulline synthesis by octanoate and its modulation by adenine nucleotides.

Liver mitochondria from octanoate-treated rabbits showed an impaired ability to synthesize citrulline. Two methods were used to evaluate citrulline synthesis in rat liver mitochondria. Under these conditions octanoate inhibited citrulline synthesis by over 50%. When ATP was included in the assay medium the inhibitory effect of octanoate was prevented. In the absence of ATP in the suspending medium, octanoate did not significantly lower total adenine nucleotides in rat liver mitochondria. However, under these conditions octanoate caused a change in the adenine nucleotide profile such that ATP c

CRIP1 knockdown enhances glycolytic dependence and increases sensitivity to 2-Deoxy-D-Glucose in acute myeloid leukemia.

BACKGROUND: Acute Myeloid Leukemia (AML) is an aggressive hematologic malignancy with suboptimal treatment outcomes, necessitating the development of novel therapeutic strategies. Metabolic reprogramming, particularly a dependency on glycolysis, is a hallmark of cancer cells. The cysteine-rich intestinal protein 1 (CRIP1) gene exhibits dual roles in cancer, but its function in AML metabolism remains unexplored. This study investigated the metabolic consequences of CRIP1 knockdown and the subsequent efficacy of the glycolytic inhibitor 2-deoxy-D-glucose (2-DG) compared to the oxidative phosphor

Construction of MGO@ZIF-8@MIPs as peroxidase-like nanozyme for the targeted colorimetric detection of perfluorooctanoic acid in water and milk.

Perfluorooctanoic acid (PFOA) poses a threat to the environment and human health due to its persistence, bioaccumulation, and reproductive toxicity. Developing and fabricating straightforward and selective sensors for the immediate identification of PFOA is substantial and demanding. In this paper, the advancement in the creation of molecularly imprinted sensors (MGO@ZIF-8@MIPs) utilizing magnetic graphene oxide (MGO) modified with metal-organic frameworks as substrates and catalytic agents is described. They were fabricated through surface imprinting technology for the quick identification an

Investigation of the toxicological effects of PFAS on cholangiocarcinoma based on network toxicology, bioinformatics, and molecular docking.

Previous studies have identified an association between per- and polyfluoroalkyl substances (PFAS) and the occurrence of intrahepatic cholangiocarcinoma (ICC), but the specific mechanisms remain unclear. This study aims to analyze the toxicological mechanisms of PFAS, particularly perfluoroctanoic acid (PFOA) and perfluorooctane sulfonate (), on ICC through network toxicology and bioinformatics. Relevant targets for PFOS and PFOA were retrieved from the CTD, ChEMBL, and Super-PRED databases. Targets related to ICC were gathered from the GeneCards, NCBI, and OMIM databases. Common targets betwe

Impact of Redox Imbalance and Mitochondrial Dysfunction on Neurotoxicity Induced by Perfluorooctanoic Acid in Male Wistar Albino Rats (Rattus norvegicus).

Perfluorooctanoic acid is a perfluoroalkyl substance that finds many applications across numerous industrial and household products. Environmental persistence and capacity to accumulate in biological systems are well documented. Increasing evidence links PFOA exposure to neurotoxicity, though its underlying mechanisms remain unclear. The purpose of this research was to assess the neurotoxic implications of PFOA in male Wistar rats by investigating oxidative stress, mitochondrial function, histopathological changes, and glial activation in the hippocampus of the forebrain. Animals received oral

Two novel in vitro assays to screen chemicals for their capacity to inhibit thyroid hormone transmembrane transporter proteins OATP1C1 and OAT4.

Early brain development depends on adequate transport of thyroid hormones (THs) from the maternal circulation to the fetus. To reach the fetal brain, THs have to cross several physiological barriers, including the placenta, blood-brain-barrier and blood-cerebrospinal fluid-barrier. Transport across these barriers is facilitated by thyroid hormone transmembrane transporters (THTMTs). Some endocrine disrupting chemicals (EDCs) can interfere with the transport of THs by THTMTs. To screen chemicals for their capacity to disrupt THTMT facilitated TH transport, in vitro screening assays are required

Identification of a non-host semiochemical from tick-resistant donkeys (Equus asinus) against Amblyomma sculptum ticks.

Amblyomma sculptum is a tick affecting animal and human health across Argentina, Bolivia, Paraguay and Brazil. Donkeys, Equus asinus, are known to be resistant to A. sculptum, suggesting that they can produce non-host tick semiochemicals (allomones), as already demonstrated for some other vertebrate host/pest interactions, whereas horses, Equus caballus, are considered as susceptible hosts. In this study, we tested the hypothesis that donkeys produce natural repellents against A. sculptum, by collecting sebum from donkeys and horses, collecting the odour from sebum extracts, and identifying do

Associations between serum perfluoroalkyl acids and LINE-1 DNA methylation.

Perfluoroalkyl acids (PFAAs) are persistent, synthetic compounds that are used in a number of consumer products. Perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS) have been associated with cardiovascular risk factors, and changes in gene expression and DNA methylation in animals and cellular systems. However, whether PFAA exposure is associated with LINE-1 DNA methylation, a potential marker of cardiovascular risk, in humans remains unknown. We sought to evaluate the cross-sectional associations between serum PFAAs and LINE-1 DNA methylation in a population highly exposed to P

Prenatal exposure to perfluorinated chemicals and behavioral or coordination problems at age 7 years.

OBJECTIVE: Potential neurotoxic effects of perfluorinated compounds (PFCs) have been reported in highly exposed animals, but whether these chemicals are neurotoxic in humans is not known. We therefore investigated whether prenatal exposure to perfluorooctanoic acid (PFOA) or perfluorooctane sulfate (PFOS), two of the most prevalent PFCs, are associated with behavioral or coordination problems in early childhood. METHODS: We used data from the Danish National Birth Cohort, which enrolled mothers in early pregnancy, and we measured maternal blood levels of PFOA and PFOS using specimens drawn ar
