Adaptive variable-weighted support vector machine as optimized by particle swarm optimization algorithm with application of QSAR studies.

Representing a compound by a numerous structural descriptors becomes common in quantitative structure-activity relationship (QSAR) studies. As every descriptor carries molecular structure information more or less, it seems more advisable to investigate all the possible descriptor vectors rather than traditional variable selection when building a QSAR model. Based on particle swarm optimization (PSO) algorithm, a more flexible descriptor selection and model construction method variable-weighted support vector machine (VW-SVM) is proposed. The new strategy adopted in this paper is to weight all

Reagents for astatination of biomolecules. 4. Comparison of maleimido-closo-decaborate(2-) and meta-[(211)At]astatobenzoate conjugates for labeling anti-CD45 antibodies with [(211)At]astatine.

An investigation was conducted to compare the in vivo tissue distribution of a rat antimurine CD45 monoclonal antibody (30F11) and an irrelevant mAbs (CA12.10C12) labeled with (211)At using two different labeling methods. In the investigation, the mAbs were also labeled with (125)I to assess the in vivo stability of the labeling methods toward deastatination. One labeling method employed N-hydroxysuccinimidyl meta-[(211)At]astatobenzoate, [(211)At]1c, and N-hydroxysuccinimidyl meta-[(125)I]iodobenzoate, [(125)I]1b, in conjugation reactions to obtain the radiolabeled mAbs. The other labeling me

Homogeneous enzyme immunoassay for triiodothyronine in serum.

BACKGROUND: The concentration of triiodothyronine (T3) in human serum is extremely low and can be determined only by very sensitive methods. We developed a homogeneous enzyme immunoassay for T3 analysis in unextracted serum. METHODS: A T(3) derivative was conjugated to the -SH groups of glycogen phosphorylase b (GPb) from rabbit muscle. Conjugation caused inhibition of enzyme activity, and the enzyme conjugate was reactivated upon binding of anti-T3 antibody. Activation was blocked by the presence of non-antibody-bound T3; this was the basis for the development of the homogeneous enzyme immun

Tumor microenvironment responsive nano-immunoregulator for precision cancer photodynamic immunotherapy.

Spatiotemporally controlled reactive oxygen species (ROS) generation remains pivotal for advancing photodynamic immunotherapy. Herein, we present tumor microenvironment (TME)-responsive nano-immunoregulator featuring a CaCO3 shell encapsulating a mesoporous silica core modified with folic acid/chlorin e6 (FA/Ce6) and co-loaded with dBET6 and maleimide (MA). Acidic TME triggers CaCO3 shell degradation, neutralizing tumor acidity to repolarize macrophages toward M1 type, enhancing antigen presentation and T cells immune response while exposing the BM@MFC core. The resulting BM@MFC further endocy

An aggregation-induced emission-active theranostic agent for selectively detecting and intervening pathological Tau protein.

The accumulation of Tau aggregates is commonly linked with various neurodegenerative diseases, such as Alzheimer's disease, Pick's disease, and corticobasal degeneration. Notwithstanding substantial investments in the development of clinical strategies for effective intervention, traditional design paradigms are predominantly confined to molecules featuring either a solitary function or single-dimensional mode of intervention, ignoring the necessity of personalized and precise medicine. Herein, we design and synthesize a dual-functional aggregation-induced emission-active agent to serve as bot

Protein disulfide isomerase A6 (PDIA6) is essential for acrosome biogenesis and male fertility in mice.

BACKGROUND: The family of protein disulfide isomerases (PDIs) are thiol oxidoreductases located predominantly in the endoplasmic reticulum that catalyze thiol-disulfide exchange for normal protein folding. Recent studies have shown that this family enzymes such as PDI contribute to the meiosis of spermatocytes and male fertility. However, the role of PDIA6 in the PDI family in spermatogenesis has not been characterized using genetically modified animal models. METHODS: A premeiotic PDIA6 conditional knockout (Stra8-Cre/Pdia6fl/fl) mouse model was generated for showing an essential role for PD

Matrix Metalloproteinase-9-Responsive Lipid-Core Micelles for Proof-of-Concept Enzyme-Triggered Cardiomyocyte Association.

Cardiovascular diseases (CVDs) remain the leading cause of global mortality and are frequently associated with increased matrix metalloproteinase-9 (MMP-9) activity during cardiac remodeling and inflammation. Here, we developed lipid-core micelles (LCMs) functionalized with a multifunctional peptide incorporating a cardiomyocyte-targeting sequence (PCM-1) and an MMP-9 cleavable linker (GPQGIAGQ), as a proof-of-concept for enzyme-responsive targeting platform. The peptide was conjugated to a maleimide-terminated PEGylated lipid and incorporated into LCMs prepared by low-energy hot emulsificatio

uPAR-Guided Dendrimer Gel Nanoparticles Reprogram Inflammation to Stabilize Atherosclerotic Plaques.

Atherosclerosis is characterized by lipid deposition, chronic inflammation, and apoptosis within the arterial wall, leading to plaque progression and instability. Current lipid-lowering therapies fail to fully address residual cardiovascular risk driven by local inflammation and cell death. Here, we report the development of uPA-functionalized, rapamycin-encapsulated dendrimer nanoparticles (G5PM-uPA/RA) that preferentially accumulate in urokinase plasminogen activator receptor (uPAR)-enriched atherosclerotic plaque, including macrophage- and apoptosis-rich lesion microenvironments. G5PM-uPA/R

Local periodontal injection, systemic heart repair: A dual-functional hydrogel for non-invasive concurrent treatment of periodontitis and myocardial infarction.

Periodontitis and myocardial infarction (MI), the leading cause of mortality worldwide, represent globally prevalent inflammatory diseases with bidirectional pathophysiological links. Despite the urgent demand for non-invasive strategies capable of alleviating local periodontal destruction while mitigating associated systemic cardiovascular complications, no integrated treatment modality currently exists. To address this challenge, a protein-loaded antibacterial hydrogel, thiolated chitosan/AMP-PEG-maleimide (C1.5P4/BMP-2), with novel sustained protein release properties was developed. This hy

The Nonclinical Safety Assessment of a Novel Anti-CEACAM5 Antibody Exatecan Conjugate Predicts a Low Risk for Interstitial Lung Disease (ILD) in Patients-The Putative Mechanism Behind ILD.

The therapeutic window of antibody drug-conjugates (ADC) remains challenging due to safety issues such as interstitial lung disease (ILD) observed with specific deruxtecan-based ADCs. To avoid ILD, we designed M9140 by conjugating the maleimide-containing hydrophilic β-glucuronide linker to exatecan and our anti-CEACAM5 (CarcinoEmbryonic Antigen-related Cell Adhesion Molecule 5) specific antibody. Following repeated iv-infusion at 3 to 30 mg/kg of M9140 every 3 weeks, the pathological findings obtained in cynomolgus monkeys were confined to gastrointestinal and hematolymphoid tissues and resem

Catalase-encapsulated matrix metalloproteinase-9 responsive nanogels for modulation of inflammatory response and treatment of neutrophilic asthma.

Asthma is a chronic disease with typical pathological features such as airflow limitation, airway inflammation and remodeling. Of these, neutrophilic asthma is considered to be the more severe and corticosteroid-resistant subtype of asthma. Increasing evidence suggests that patients with neutrophilic asthma often accompany with dysbiosis of the internal microbiota, where the increased abundance of non-typeable Haemophilus influenzae (NTHi) is closely related to the neutrophilic asthma phenotype. Furthermore, emerging evidence suggests that reactive oxygen species (ROS) are pivotal in the patho

Image-guided therapy of colorectal cancer using a zirconium coordinated nanosensitizer.

Sonodynamic therapy (SDT) has gained attention as a promising non-invasive cancer treatment strategy due to its localized effects and superior tissue penetration. SDT induces apoptosis and immunogenic cell death (ICD) via the generation of reactive oxygen species (ROS) upon ultrasound irradiation. However, tumor heterogeneity, the immunosuppressive tumor microenvironment, and the limited accumulation of sonosensitizers at the tumor site pose significant challenges to SDT's therapeutic efficacy. In this study, a tumor-targeting nanosonosensitizer, Zr-HMME-PEG-Linaclotide (HP-LNC), was designed

ADAMTS5-specific gapmer release from an albumin biomolecular assembly and cartilage internalization triggered by ultrasound.

OBJECTIVE: Antisense oligonucleotides (ASOs) have reached the clinic; however, they lack tissue specificity. Albumin is a plasma-abundant macromolecule that has been shown to accumulate in inflamed tissues. In this work, we have designed a recombinant human albumin (rHA)-based biomolecular assembly incorporating a DNase-resistant phosphorothioate-based complementary oligonucleotide (cODN) and an anti-ADAMTS5 ASO for potential delivery to inflamed sites. Ultrasound (US) was used to trigger ASO release from the assembly and enhance internalization into articular cartilage. METHODS: A phosphorot

PEGylation of New Thrombin Inhibitor Peptide Ultravariegin for Prolonged In Vivo Circulation and Enhanced Antithrombotic Effects.

Anticoagulant therapy is commonly used to prevent and treat arterial and venous blood clots in patients with cardiovascular disease, cerebrovascular disease, and cancer. Venous blood clots are the third leading cause of cardiovascular death following acute coronary artery disease and stroke. There is a significant need for effective anticoagulant therapy with minimal risk of bleeding. Variegin and its variants are a new type of antithrombin peptide that has shown promising results in preclinical studies. Variegin and its best variant, ultravariegin (UV), can more effectively inhibit blood clot

QCM-D Investigations on Cholesterol-DNA Tethering of Liposomes to Microbubbles for Therapy.

Lipid-shelled microbubbles (MBs) offer potential as theranostic agents, capable of providing both contrast enhancement in ultrasound imaging as well as a route for triggered drug release and improved localized drug delivery. A common motif in the design of such therapeutic vehicles is the attachment of the drug carrier, often in the form of liposomes, to the microbubble. Traditionally, such attachments have been based around biotin-streptavidin and maleimide-PDP chemistries. Comparatively, the use of DNA-lipid tethers offers potential advantage. First, their specificity permits the constructio

GlmU Inhibitors as Promising Antibacterial Agents: A Review.

Bacterial infections are a major cause of mortality and morbidity in humans throughout the world. Infections due to resistant bacterial strains such as methicillin-resistant Staphyloccocusaureus vancomycin, resistant Enterococci, Klebsiella pneumoniae, Staphylococcus aureus, and Mycobacterium are alarming. Hence the development of new antibacterial agents, which act via a novel mechanism of action, became a priority in antibacterial research. One such approach to overcome bacterial resistance is to target novel protein and develop antibacterial agents that act via different mechanisms of actio

Highly Efficient and Stable Binary Cross-Linkable/ Self-Assembled Organic Nonlinear Optical Molecular Glasses.

The development of electro-optical materials with high chromophore loading levels that possess ultrahigh electro-optic coefficients and high long term alignment stability is a challenging topic. Anthracene-maleimide Diels-Alder (DA) reaction and π-π interaction of Anthracene-pentafluorobenzene and benzene-pentafluorobenzene are developed for making highly efficient binary cross-linkable/self-assembled dendritic chromophores FZL1-FZL4. A covalently or non-covalently cross-linked network is formed by DA reaction or π-π interaction after electric field poling orientation, which greatly improves t

Synergistic chemo-photo anticancer therapy by using reversible Diels-Alder dynamic covalent bond mediated polyprodrug amphiphiles and immunoactivation investigation.

Highly efficient endocytosis and multi-approach integrated therapeutic tactics are important factors in oncotherapy. With the aid of thermally reversible furan-maleimide dynamic covalent bonds and the "polyprodrug amphiphiles" concept, thermo- and reduction-responsive PEG(-COOH)Fu/MI(-SS-)CPT copolymers were fabricated by the Diels-Alder (D-A) coupling of hydrophilic Fu(-COOH)-PEG and hydrophobic MI(-SS-)-CPT building blocks. The copolymers could self-assemble to form composite nanoparticles with a photothermal conversion reagent (IR780) and maintain excellent stability. In the in vitro simula
