Engineered probiotic-derived indole-3-propionic acid inhibits ubiquitination via AHR signaling to treat postmenopausal osteoporosis.

Postmenopausal osteoporosis (PMOP) has a high incidence in middle-aged and elderly women, leading to an increased risk of fractures and elevated rates of disability and mortality. In this work, we identified the reduction of indole-3-propionic acid (IPA) as a potential key factor contributing to the decline in bone mass observed in postmenopausal women. Mechanistically, IPA activates AhR, leading to the stabilization of key proteins in Wnt and NF-κB pathways that regulate bone formation and resorption. We evaluated efficacy in vivo using eight-week-old female C57BL/6 mice subjected to bilatera

SHBG and APOA1 as serum biomarkers for low bone mineral density in Mexican postmenopausal women: a random forest-based analysis.

BACKGROUND: Osteoporosis is a major health issue that has implications worldwide, both clinically and socioeconomically. Therefore, searching for informative bone health biomarkers that provide early information on bone changes and assess treatment efficacy is essential. Previously, our group used label-free proteomics and bioinformatic approaches to analyze the serum proteomic profile of postmenopausal women with normal bone mineral density (BMD), osteopenia, and osteoporosis. Based on their role in bone metabolism and literature review, a panel of 12 candidate serum biomarkers for low BMD wa

Abaloparatide-Zoledronate combination protects against 4-vinylcyclohexene diepoxide-induced postmenopausal osteoporosis in mice: an osteoanabolic-antiresorptive approach.

Abaloparatide, a 34-amino acid synthetic peptide analog of parathyroid hormone-related protein (PTHrP), was approved for treating postmenopausal osteoporosis in high-risk individuals or those resistant to existing drugs. This study aimed to investigate the impact of abaloparatide (20 µg/kg/d; s.c.), an osteoanabolic drug with an antiresorptive drug zoledronate (125 µg/kg i.v.) twice weekly for a month against 4-vinylcyclohexene diepoxide (VCD)-induced postmenopausal osteoporosis in mice. Female Swiss albino mice were made ovotoxic by treatment with VCD (160 mg/kg/d) for 15 days to mimic a post

Follistatin-like protein 1 (FSTL1) modulates bone remodeling and attenuates bone loss in a mouse model of postmenopausal osteoporosis.

This study systematically delineates the expression dynamics of follistatin-like protein 1 (FSTL1) in postmenopausal osteoporosis (PMOP) and elucidates its role in skeletal homeostasis. Integrated analyses of clinical serum samples and the GTEx database revealed a pronounced reduction in circulating FSTL1 levels in PMOP patients. In vivo, Fstl1+/- ovariectomized mice displayed exacerbated bone loss-evidenced by diminished BMD and trabecular volume-together with impaired osteoblastic activity and heightened osteoclast activation. Complementary in vitro experiments showed that FSTL1 over-express

Artificial intelligence models for osteoporosis risk prediction in postmenopausal women with CKD: A cross-sectional analysis of NHANES 2005 to 2018.

Artificial intelligence (AI) can transform osteoporosis (OP) screening, but its application in high-risk, complex populations like postmenopausal women with chronic kidney disease remains limited. We sought to develop and interpret an explainable AI model specifically for identifying low bone mass (LBM) or OP in this vulnerable group. Using data from the National Health and Nutrition Examination Survey (2005-2018), a nationally representative cross-sectional study, we developed and compared 8 AI models. For the purpose of algorithm development and comparison, the complex survey weights of Nati

Examining sleep disturbance as a mediator of the association of vasomotor symptoms with positive and negative emotional well-being in midlife postmenopausal women.

OBJECTIVES: Most research evaluating the effect of menopause on mental health focuses on depression. However, short-term positive and negative emotional well-being are also key aspects of mental health. The domino hypothesis posits that vasomotor symptoms (VMS) impact mental health through sleep disturbance (SD). Thus, we evaluated SD as a mediator of the association of VMS with positive and negative emotional well-being in midlife women, using both subjective and objective measures of VMS and SD. STUDY DESIGN: Midlife women (N = 242; 98% postmenopausal; mean age 59.02 ± 4.43 years) enrolled

Squamous cell carcinoma arising within a mature cystic teratoma of the ovary in a postmenopausal woman: A case report and literature review.

Malignant transformation of mature cystic teratomas (MCTs) of the ovary is rare, with squamous cell carcinoma (SCC) being the most common histologic subtype. Due to its nonspecific clinical presentation and similarity to benign dermoid cysts, preoperative diagnosis is often challenging, resulting in delayed recognition and poorer outcomes. Risk factors such as postmenopausal status, large tumour size, and elevated tumour markers should serve as red flags prompting clinical suspicion for malignant transformation within an ovarian teratoma. This report describes the case of a 52-year-old woman w
