Insights into bacterial 6-methylsalicylic acid synthase and its engineering to orsellinic acid synthase for spirotetronate generation.

The enzymes 6-methylsalicylic acid (6-MSA) synthases (6-MSASs) are involved in the building of an aryl moiety in many bioactive secondary metabolites produced by fungi and bacteria. Using the bacterial 6-MSAS ChlB1 in the biosynthesis of spirotetronate antibiotic chlorothricin (CHL) as a model, functional analysis of its dehydratase (DH) and ketoreductase (KR) domains by site-specific mutagenesis revealed that selective ketoreduction is not essential for polyketide chain extension. Promiscuity of the ketoacylsynthase domain in beta functionality recognition allows for engineering ChlB1 to an o…

Genetic engineering of a lysosomal enzyme fusion protein for targeted delivery across the human blood-brain barrier.

Mucopolysaccharidosis Type I, Hurler's Syndrome, is a lysosomal storage disorder that affects the brain. The missing enzyme, alpha-L-iduronidase (IDUA), does not cross the blood-brain barrier (BBB). To enable BBB transport of the enzyme, human IDUA was fused to the carboxyl terminus of the heavy chain of a chimeric monoclonal antibody (MAb) to the human insulin receptor (HIR). The HIRMAb crosses the BBB on the endogenous insulin receptor, and acts as a molecular Trojan horse to ferry into brain the IDUA. Transfection of COS cells resulted in high levels of IDUA enzyme activity both in the medi

Metabolic engineering of the E. coli L-phenylalanine pathway for the production of D-phenylglycine (D-Phg).

D-phenylglycine (D-Phg) is an important side chain building block for semi-synthetic penicillins and cephalosporins such as ampicillin and cephalexin. To produce d-Phg ultimately from glucose, metabolic engineering was applied. Starting from phenylpyruvate, which is the direct precursor of L-phenylalanine, an artificial D-Phg biosynthesis pathway was created. This three-step route is composed of the enzymes hydroxymandelate synthase (HmaS), hydroxymandelate oxidase (Hmo), and the stereoinverting hydroxyphenylglycine aminotransferase (HpgAT). Together they catalyse the conversion of phenylpyruv…

New perspectives on bioengineering of joint tissues: joint adaptation creates a moving target for engineering replacement tissues.

The current paradigm in tissue engineering is that "full regeneration" or "total replacement" of normal tissue is required in order to restore joint function. However, there is considerable evidence that suggests that targets other than "normality" may actually be required for tissue substitutes. Sometimes "less than normal" tissue properties of substitutes may be required following an injury, and sometimes "more than normal" may be required (following tissue degradation, damage, and failure). Diarthrodial joints function as "organs" in a physiological sense and normal individual joint tissues…

Protein engineering of a human enzyme that hydrolyzes V and G nerve agents: design, construction and characterization.

Because of deficiencies in the present treatments for organophosphorus anticholinesterase poisoning, we are attempting to develop a catalytic scavenger that can be administered as prophylactic protection. Currently known enzymes are inadequate for this purpose because they have weak binding and slow turnover, so we are trying to make an appropriate enzyme by protein engineering techniques. One butyrylcholinesterase mutant, G117H, has the desired type of activity but reacts much too slowly. This communication describes an attempt to determine the reason for the slow reaction so that a more effi…

Characterization of infectious aerosols in health care facilities: an aid to effective engineering controls and preventive strategies.

Assessment of strategies for engineering controls for the prevention of airborne infectious disease transmission to patients and to health care and related workers requires consideration of the factors relevant to aerosol characterization. These factors include aerosol generation, particle size and concentrations, organism viability, infectivity and virulence, airflow and climate, and environmental sampling and analysis. The major focus on attention to engineering controls comes from recent increases in tuberculosis, particularly the multidrug-resistant varieties in the general hospital popula…

Congenital microtia patients: the genetically engineered exosomes released from porous gelatin methacryloyl hydrogel for downstream small RNA profiling, functional modulation of microtia chondrocytes and tissue-engineered ear cartilage regeneration.

BACKGROUND: Mesenchymal stem cells (MSCs) exosomes were previously shown to be effective in articular cartilage repairing. However, whether MSCs exosomes promote mature cartilage formation of microtia chondrocytes and the underlying mechanism of action remains unknown. Additionally, some hurdles, such as the low yield and unsatisfactory therapeutic effects of natural exosomes have emerged when considering the translation of exosomes-therapeutics to clinical practices or industrial production. Herein, we investigated the roles of human adipose-derived stem cells (ADSCs) exosomes in modulating m

Highly Diastereo- and Enantioselective Imine Reductases Engineered for γ-Piperidinamines Bearing Contiguous Stereocenters.

Imine reductases (IREDs) represent a highly promising class of biocatalysts for the asymmetric synthesis of chiral amines bearing two stereogenic centers. Herein, we report an IRED-catalyzed asymmetric reductive amination strategy that converts racemic ketones into γ-piperidinamines with contiguous stereocenters and operates through a pathway consistent with a dynamic kinetic resolution-asymmetric reductive amination (DKR-ARA). The wild-type AcIRED afforded the 1-benzyl-N-cyclopropyl-3-methylpiperidin-4-amine (1a) in 60.8% conversion, but with only 78.5% enantiomeric excess (eecis) for (3S,4R)

A mechanically active microneedle platform integrated with engineered exosomes for enhanced tendon-bone regeneration.

Functional regeneration of the tendon-bone interface remains a major challenge because early healing is constrained by an adverse inflammatory microenvironment and insufficient mechanical cues. Here, we developed a mechano-regenerative strategy that combines YAP-primed engineered exosomes with localized biomaterial-based delivery and active mechanical stimulation. Exosomes derived from a YAP agonist (PY-60) preconditioned TDSCs (P-Exo) showed enhanced uptake and promoted the osteogenic and chondrogenic differentiation of bone marrow mesenchymal stem cells, accompanied by activation of YAP-asso

Mismatch-Engineered CRISPR Transistors for Ultra-Low-Frequency Single-Nucleotide Variant Detection.

Accurate detection of low-frequency single-nucleotide variants (SNVs) in a high wild-type (WT) background remains challenging. Here, we report a mismatch-engineered CRISPR transistor (MECT) for rapid, amplification-free detection of mutant RNA. MECT integrates a Cas13a-crRNA recognition interface with a graphene field-effect transistor and employs a spaced mismatch crRNA design centered on the mutation site in a nonseed region. It leverages the synergistic effect between natural and engineered mismatches at the mutation site to weaken WT binding while preserving mutant recognition, thereby amp

Ultrafast Laser-Engineered Hierarchical Interface for Synergistic Radiative Cooling and Droplet Energy Harvesting.

Passive daytime radiative cooling provides an energy-free approach for thermal management, but achieving multifunctional operation under realistic outdoor conditions remains challenging, because optical regulation, surface wetting, and energy conversion are typically realized with separate material components. Here, we develop a femtosecond laser-engineered passive radiative cooler (FLA-PRC) based on a hierarchical PTFE/Ag/SiO2 interface that simultaneously regulates photons, water droplets, and interfacial charges. Orthogonal laser scanning creates periodic flat-topped micropyramid arrays dec

Electron-Transfer Engineered Biointerface With Detonation Nanodiamonds for Next-Generation Cortisol Electrochemical Immunoassay.

Detonation nanodiamonds (DND) emerge as versatile nano building blocks for next generation biosensing, owing to their exceptional physicochemical properties and biocompatibility. In this study, a robust label-free electrochemical immunosensor for cortisol detection is based on screen printed carbon electrodes (SPE) modified with pristine DND. By utilizing EDC/NHS, efficient covalent crosslinking and stable antibody immobilization is achieved, as shown by comprehensive morphological and spectroscopic characterization (AFM, SEM, Raman, FTIR). Electrochemical analyses demonstrate that the DND int

Silicosis Among Engineered Stone Workers in Ontario, Canada: A Case Series of 20 Patients.

BACKGROUND: Silicosis is a preventable occupational lung disease caused by inhalation of respirable crystalline silica. Although traditionally associated with mining, it is increasingly recognized among workers who fabricate engineered stone, also referred to as artificial stone, for kitchen and other countertops. In Canada, updated surveillance data are limited, and no published case series have characterized engineered stone-associated silicosis. METHODS: We report a retrospective case series of 20 patients diagnosed with silicosis following occupational exposure to engineered stone in Onta

Mining Microbial Transcriptomes to Engineer Cell-Based Bacterial Biosensors in Gut-Resident Bacteroidaceae.

The gastrointestinal tract is rich in metabolic, immune, and microbiome-derived signals that can inform the design of live biotherapeutics and diagnosis of intestinal disorders. Engineered cell-based biosensors can tap into this molecular information and report on their environment, yet their development in gut-resident symbionts has been limited by a lack of validated sensor systems. Here, we present a generalizable pipeline that leverages bacterial transcriptional profiling to identify environment-responsive systems for biosensor engineering. Candidate Sensors Systems (CSSs) mined from healt…

Engineered models of skin fibrosis: from biomaterial-free to biomaterial-based in vitro systems.

Skin fibrosis is a complex and progressive disorder characterized by excessive extracellular matrix deposition, persistent fibroblast activation and pathological tissue stiffening. Its clinical manifestations such as keloids, hypertrophic scars and systemic sclerosis, can lead to functional impairment, aesthetic disfigurement and significant psychosocial burden. Unfortunately, there are currently no efficient therapies that can halt or reverse skin fibrosis, largely due to our incomplete understanding of the dynamic molecular and cellular mechanisms driving fibrosis progression. Here, we revie
