Exosomes, rich in diverse bioactive molecules, have emerged as a promising "liquid biopsy" approach for early cancer screening. However, relying solely on a single biomarker type can easily lead to missed diagnoses or misjudgments. Therefore, there is an urgent need to develop multiple biomarkers detection strategies to facilitate precise diagnosis. Herein, a microfluidic chip integrating two signals (ALP-driven chemiluminescence and nanobeacons-based fluorescence) into a single device was developed using breast cancer exosomes as a model. A simple sample injection enabled capture and quantita

Live cell imaging is a powerful tool to elucidate dynamics of protein(s). Our group has concentrated on dynamics of two major cell-matrix adhesion devices, hemidesmosome and focal contact in the keratinocytes. Firstly, we observed the fate of hemidesmosome protein or focal contact protein by single-color live cell imaging in the physiological setting of keratinocytes. Both hemidesmosome proteins and focal contact proteins were highly dynamic. Next, in order to observe the interaction between hemidesmosome protein and focal contact protein, we observed the fate of these proteins at the same tim

Nanosized drug delivery systems have offered promising approaches for cancer theranostics. However, few are effective to simultaneously maximize tumor-specific uptake, imaging, and therapy in a single nanoplatform. Here, we report a simple yet stimuli-responsive anethole dithiolethione (ADT)-loaded magnetic nanoliposome (AML) delivery system, which consists of ADT, hydrogen sulfide (H2S) pro-drug, doped in the lipid bilayer, and superparamagnetic nanoparticles encapsulated inside. HepG2 cells could be effectively bombed after 6 h co-incubation with AMLs. For in vivo applications, after prefere

A variety of biopolymers is imaged using noncontact atomic force microscopy. Samples are prepared by aerosol spray deposition of aqueous solutions on freshly cleaved mica followed by air drying. The distributions of contour lengths and chain or fibril thicknesses normal to the mica substrate can be measured for individual polymer molecules or molecular assemblies. In many cases it is possible to conclude that the structures imaged and quantitatively analyzed are representative of those present in solution and not artifacts of the deposition/dessication process. Imaging of linear and cyclic tri

Plasma membrane transporters play an essential role in plant development and all physiological processes. Their activity is tightly controlled by a variety of mechanisms, including the rate of transporter endocytosis. There is evidence that the local membrane environment, in particular the sterol content, influences transporter endocytosis. Here, we analyzed the sucrose transporters StSUT1 (potato) and AtSUC2 (Arabidopsis) transiently expressed in Nicotiana benthamiana epidermal cells. Using live-cell imaging of fluorescent protein fusions combined with extracellular quenching by Trypan Blue,

Chromatin remodeling governs how eukaryotic genomes are accessed and interpreted by dynamically modulating nucleosome architecture. Adenosine triphosphate (ATP)-dependent remodeling complexes are highly conserved molecular machines built around Snf2-family ATPases that use the energy of ATP hydrolysis to slide, evict, or restructure nucleosomes, thereby shaping transcription, replication, and DNA repair. Their importance is such that their malfunctioning is linked to different diseases, including cancer. Recent discoveries reveal that chromatin remodelers are far more dynamic than previously a

Amorphous red phosphorus (ARP) has recently been identified as a zig-zag ladder polymer, yet its function beyond a low-cost phosphorus source remains unexplored. Here we report the first host-guest chemistry of ARP. In water, hydrophobic ARP threads through α- or β-cyclodextrin (CD) to produce water-soluble pseudo-polyrotaxanes, as evidenced by nuclear magnetic resonance (NMR) spectroscopy, atomic force microscopy (AFM) imaging and AFM-based single-molecule force spectroscopy (SMFS). β-CD, whose cavity diameter (0.60 nm) matches the 0.582 nm van-der-Waals width of ARP, assembles readily with A

Diabetic cataract (DC) is a major cause of vision impairment, partly driven by the pathological roles of aldose reductase (AR) and inducible nitric oxide synthase (iNOS). Gigantol, a bibenzyl compound derived from Dendrobium, has shown potential in preventing DC, yet its molecular mechanism remains incompletely understood. In this study, we investigated the modulation of conformational integrity and aggregation propensity of AR/iNOS by gigantol using fluorescent labeling, fluorescence resonance energy transfer (FRET), single-molecule imaging in living cells, fluorescence spectroscopy, confocal

RNA localization adds a fundamental layer to gene expression by determining when and where translation-ready mRNAs become available, yet how this timing is coordinated with nuclear architecture and cell-cycle progression remains unclear. Here we identify a subnuclear RNA niche at the nuclear speckle periphery that couples intron retention to cell-cycle-timed RNA release. Using compartment-resolved transcriptional inhibition, sequence-based deep learning and single-molecule and super-resolution RNA imaging in human pluripotent stem cells, we define a class of nuclear RNAs with long-lived retain

Epithelial-to-mesenchymal transition (EMT) is a fundamental process in development and disease; however, its study is often complicated by the intrinsic heterogeneity of commonly used epithelial cell models. Here, we show that pre-existing heterogeneity contributes to divergent outcomes following transforming growth factor-beta 3 (TGFβ3) treatment. Flow cytometry, gene expression profiling and imaging revealed persistent variability in E-cadherin (E-cad) and epithelial cell adhesion molecule (EpCAM) expression, with differentially sorted subpopulations displaying distinct transcriptional respo

Atherosclerosis remains the leading cause of ischemic syndromes such as myocardial infarction or brain stroke, mainly promoted by plaque rupture and subsequent arterial blockade. Identification of vulnerable or high-risk plaques constitutes a major challenge, being necessary to identify patients at risk of occlusive events in order to provide them with appropriate therapies. Clinical imaging tools have allowed the identification of certain structural indicators of prone-rupture plaques, including a necrotic lipidic core, intimal and adventitial inflammation, extracellular matrix dysregulation,

Carbon nanotubes (CNT) are intensively being developed for biomedical applications including drug and gene delivery. Although all possible clinical applications will require compatibility of CNT with the biological milieu, their in vivo capabilities and limitations have not yet been explored. In this work, water-soluble, single-walled CNT (SWNT) have been functionalized with the chelating molecule diethylentriaminepentaacetic (DTPA) and labeled with indium ((111)In) for imaging purposes. Intravenous (i.v.) administration of these functionalized SWNT (f-SWNT) followed by radioactivity tracing u

RNA translation to protein is paramount to creating life, yet RNA and protein correlations vary widely across tissues, cells, and species. To investigate these perplexing results, we utilize a time-series fixation method that combines static stimulation and a programmable formaldehyde perfusion to map pseudo-Signaling with Omics signatures (pSigOmics) of single-cell data from hundreds of thousands of cells. Using the widely studied nuclear factor kappa B (NFκB) mammalian signaling pathway in mouse fibroblasts, we discovered a novel asynchronous pseudotime regulation (APR) between RNA and prote

Fluorescence resonance energy transfer (FRET) is the highly distance dependent (3-10 nm) transfer of energy from a donor to an acceptor fluorophore, with transfer efficiency inversely proportional to the distance between the fluorophores. Consequently FRET serves as a powerful spectroscopic ruler for probing molecular interactions. Whilst cell based FRET assays report bulk relative changes in FRET efficiency in a population, single molecule FRET (smFRET) is capable of deconvoluting these population averages into distinct structural states. However, the lack of universal benchmarks prevents the

Tissue stiffness assessment plays a critical role in the clinical diagnosis of liver fibrosis and the differentiation of benign from malignant tumors, among other conditions. Conventional magnetic resonance elastography (MRE) enables noninvasive evaluation of tissue stiffness but relies on dedicated external vibration devices, which limits its widespread clinical adoption. Virtual magnetic resonance elastography (vMRE) has emerged as a promising branch of MRE and has attracted increasing attention in recent years. vMRE is primarily based on microstructural analysis using diffusion-weighted ima

BACKGROUND: The tumour-suppressor protein p53 can form amyloid aggregates resulting in loss of tumour-suppressing functions and leading to tumour formation. The detection of p53 aggregates in cancer cells has been demonstrated but these aggregates have not been detected in liquid biopsies to date, due to the lack of sufficiently sensitive methods.
METHODS: We developed an ultrasensitive immunoassay based on the single-molecule array (SiMoA) technology to detect p53 aggregates in plasma, based on antibody capture of the aggregates. We confirmed that the assay detects p53 aggregates using super

Regulator of telomere length 1 (RTEL1) helicase facilitates telomere replication by disassembling DNA secondary structures, such as G-quadruplexes and telomeric loops (t-loops), at the ends of the chromosomes. The recruitment of RTEL1 to telomeres occurs during the S-phase of the cell cycle, but the dynamics of the process has not been studied. Here, we utilized CRISPR genome editing and single-molecule imaging to monitor RTEL1 movement within human cell nuclei. RTEL1 utilizes rapid three-dimensional diffusion to search for telomeres and other nuclear targets. Only 5% of the chromatin-bound RT

Enteroviruses dramatically remodel the cellular infrastructure for efficient replication and curtailing host antiviral responses. The roles of viral proteins in these processes have been studied mostly in vitro, by ectopic overexpression, or by surrogate infection systems, all of which have shortcomings. Here, we replace the essential 2A cleavage site at the P1-P2 junction with an internal ribosome entry site (IRES), 3CD cleavage site, or T2A sequence, allowing us to catalytically inactivate 2Apro in the virus context. Viruses with an inactive 2Apro are hampered in replication in cell lines an

The TRPA1 ion channel is a sensitive detector of reactive chemicals, found primarily on sensory neurons. The phenotype exhibited by mice lacking TRPA1 suggests its potential as a target for pharmacological intervention. Antibody-based detection for distribution analysis is a standard technique. In the case of TRPA1, however, there is no antibody with a plausible validation in knockout animals or functional studies, but many that have failed in this regard. To this end we employed the single molecule in situ hybridization technique RNAscope on sensory neurons immediately after detection of calc

Expansion microscopy (ExM) is a powerful technique to overcome the diffraction limit of light microscopy that can be applied in both tissues and cells. In ExM, samples are embedded in a swellable polymer gel to physically expand the sample and isotropically increase resolution in x, y, and z. The maximum resolution increase is limited by the expansion factor of the gel, which is four-fold for the original ExM protocol. Variations on the original ExM method have been reported that allow for greater expansion factors but at the cost of ease of adoption or versatility. Here, we systematically exp
