The Primary Cilium: Emerging Role as a Key Player in Fibrosis.

PURPOSE OF REVIEW: The myofibroblast is the culprit in the pathogenesis of fibrosis in systemic sclerosis (SSc). Activation of morphogen signaling pathways has been shown to be critically involved in organ fibrosis. Remarkably, the cellular receptors and key molecules from these signaling pathways are localized in the primary cilium. The primary cilium is a unique cellular organelle present in virtually all cells. This article summarizes recent studies evaluating the association between primary cilia and morphogen signaling driving myofibroblast transition and subsequent fibrosis. RECENT FIND

The STING pathway in response to chlamydial infection.

The past decades have witnessed significant progress in discovery and characterize cytosolic DNA sensing and signaling, especially the understanding of the stimulator of interferon genes (STING). This pathway to foreign nucleic acids enables the initiation of robust anti-pathogenic responses to protect the host, and provides a new understanding for therapeutic intervention in a growing infectious disease, including chlamydial infection. Chlamydiae are obligate intracellular pathogenic bacterium causing widespread human diseases such as sexually transmitted infections and respiratory tract infe

Antimalarial drug mefloquine inhibits nuclear factor kappa B signaling and induces apoptosis in colorectal cancer cells.

Nuclear factor kappa B (NF-κB) signaling pathway is activated in many colorectal cancer (CRC) cells and in the tumor microenvironment, which plays a critical role in cancer initiation, development, and response to therapies. In the present study, we found that the widely used antimalarial drug mefloquine was a NF-κB inhibitor that blocked the activation of IκBα kinase, leading to reduction of IκBα degradation, decrease of p65 phosphorylation, and suppressed expression of NF-κB target genes in CRC cells. We also found that mefloquine induced growth arrest and apoptosis of CRC cells harboring ph

DNA Damage Signaling Instructs Polyploid Macrophage Fate in Granulomas.

Granulomas are immune cell aggregates formed in response to persistent inflammatory stimuli. Granuloma macrophage subsets are diverse and carry varying copy numbers of their genomic information. The molecular programs that control the differentiation of such macrophage populations in response to a chronic stimulus, though critical for disease outcome, have not been defined. Here, we delineate a macrophage differentiation pathway by which a persistent Toll-like receptor (TLR) 2 signal instructs polyploid macrophage fate by inducing replication stress and activating the DNA damage response. The Wnt inhibitor dickkopf-1: a link between breast cancer and bone metastases.

Breast cancer is the second leading cause of cancer death in women and metastasizes to bone in greater than 80 % of advanced-disease patients. Once breast cancer bone metastases are established, the disease is incurable and drives numerous complications that increase morbidity and diminish patients' quality of life. Many mechanisms have been implicated in bone metastases of breast cancer. The critical role of Wnt signalling pathway inhibition in initiating bone lesions has been demonstrated in a variety of bone diseases and tumours. Overexpression of dickkopf-1 (Dkk1) protein, a negative regul

[Effect of vitamin d receptor polymorphisms on the development and progression of malignant melanoma].

BACKGROUND: Malignant melanoma is one of the most aggressive cutaneous tumors in men and women. The risk of developing a malignant melanoma depends on several external factors along with deregulation of mutual interaction of genotype and phenotype. Nowadays, growing attention is focused on the study of the interactions of the active form of vitamin D3 with its receptor and inhibitory effect of vitamin D3 receptor polymorphisms on multiple signaling pathways involved in proliferative and metastatic processes. OBJECTIVES: This review article addresses the relationship between factors involved i

Molecular pathogenesis of spondylocheirodysplastic Ehlers-Danlos syndrome caused by mutant ZIP13 proteins.

The zinc transporter protein ZIP13 plays critical roles in bone, tooth, and connective tissue development, and its dysfunction is responsible for the spondylocheirodysplastic form of Ehlers-Danlos syndrome (SCD-EDS, OMIM 612350). Here, we report the molecular pathogenic mechanism of SCD-EDS caused by two different mutant ZIP13 proteins found in human patients: ZIP13(G64D), in which Gly at amino acid position 64 is replaced by Asp, and ZIP13(ΔFLA), which contains a deletion of Phe-Leu-Ala. We demonstrated that both the ZIP13(G64D) and ZIP13(ΔFLA) protein levels are decreased by degradation via

Pathophysiology of Trousseau's syndrome.

Clinically relevant clotting abnormalities in cancer patients are referred to as Trousseau's syndrome. While thrombotic complications such as venous thromboembolism are most frequent in every day's practice, cancer patients may also experience severe bleeding symptoms due to complex systemic coagulopathies, including disseminated intravascular coagulation, haemolytic thrombotic microangiopathy, and hyperfibrinolysis. The pathophysiology of Trousseau's syndrome involves all aspects of Virchow's triad, but previous basic research has mainly focused on the cellular and molecular mechanisms underl

Cytokine pathways and interactions in alopecia areata.

Alopecia areata (AA) is an autoimmune disease resulting in the premature arrest of the follicular growth cycle clinically resulting in patchy, non-scarring hair loss. The presence of a dense follicular T cell infiltrate and variations in cytokines have led to the hypothesis that T cell activation and alterations in inflammatory mediators are crucial participants in the etiopathogenesis of the disease. Various studies suggest that the AA pathogenesis has a dominant TH1-mediated component, with potential involvement of the TH17 pathway. However, a fully integrated view of intersecting cytokine n

Pilocytic astrocytoma: a disease with evolving molecular heterogeneity.

Pilocytic astrocytoma, the most common pediatric brain tumor, is a clinically and molecularly heterogeneous disease that occurs most often in the cerebellum and hypothalamic and chiasmatic regions. Classically, pilocytic astrocytomas are driven by the mitogen-activated protein kinase/extracellular signal-regulated kinase pathway. Recently described genetic aberrations involving this pathway are critical for tumorigenesis. Tandem duplication of 7q34 encodes BRAF and produces several KIAA1549-BRAF novel oncogenic fusions. Activating point mutations of BRAF, such as BRAF (V600E), also lead to pil

Role and therapeutic potential of PI3K-mTOR signaling in de novo resistance to BRAF inhibition.

BRAF inhibition is highly active in BRAF-mutant melanoma, but the degree and duration of responses is quite variable. Improved understanding of the mechanisms of de novo resistance may lead to rational therapeutic strategies with improved efficacy. Proteomic analysis of BRAF-mutant, PTEN-wild-type human melanoma cell lines treated with PLX4720 demonstrated that sensitive and de novo resistant lines exhibit similar RAS-RAF-MEK-ERK pathway inhibition, but the resistant cells exhibited durable activation of S6 and P70S6K. Treatment with the mTOR inhibitor rapamycin blocked activation of P70S6K an

Pulmonary hypertension: role of combination therapy.

Over the past decade different effective treatment options for use in pulmonary arterial hypertension (PAH) have been developed. Due to multiple pathophysiological pathways in PAH and unsatisfactory overall results with the use of monotherapy in a substantial number of PAH patients, there is a pharmacological rationale for combination therapy. The currently approved substances target the prostacyclin, the endothelin and the NO (nitric oxide) -pathways. Those agents have shown a varying degree of improvement in different categories of PAH associated limitations like hemodynamics, exercise capac

Multiple acquired renal carcinoma tumor capabilities abolished upon silencing of ADAM17.

Malignancy is a manifestation of acquired defects in regulatory circuits that direct normal cell proliferation and homeostasis. Most of these circuits operate through cell autonomous pathways, whereas others potentially involve the neighboring microenvironment. We report that the metalloprotease ADAM17 plays a pivotal role in several acquired tumor cell capabilities by mediating the availability of soluble transforming growth factor-alpha, an epidermal growth factor receptor (EGFR) ligand, and thus the establishment of a key autocrine signaling pathway. Silencing of ADAM17 in human renal carci

The protein secretion systems in Listeria: inside out bacterial virulence.

Listeria monocytogenes, the etiologic agent of listeriosis, remains a serious public health concern with its frequent occurrence in food coupled with a high mortality rate. The capacity of a bacterium to secrete proteins to or beyond the bacterial cell surface is of crucial importance in the understanding of biofilm formation and bacterial pathogenesis to further develop defensive strategies. Recent findings in protein secretion in Listeria together with the availability of complete genome sequences of several pathogenic L. monocytogenes strains, as well as nonpathogenic Listeria innocua Clip1

Phlorizin attenuates lupus nephritis via upregulating PI3K/Akt pathway-mediated Treg differentiation.

BACKGROUND: Lupus nephritis (LN) leads to widespread kidney damage and nephron loss, establishing it as a major contributor to acute and chronic kidney injury, which can progress to end-stage renal disease. Phlorizin (PHZ), a major pharmacologically active constituent derived from Lithocarpus polystachyus Rehd., has been shown to exhibit significant immunomodulatory and anti-inflammatory properties. Growing evidence indicates that PHZ may exert a protective influence on kidney function. However, the therapeutic effect and mechanism of PHZ in treating LN need to be elucidated. METHODS: The PHZ

[Research progress on traditional Chinese medicine in prevention and treatment of metabolic diseases via SIRT1/AMPK signaling pathway].

Metabolic diseases, characterized by systemic dysregulation of glucose, lipid, protein, and energy homeostasis, encompass a cluster of multisystem disorders and have emerged as a critical global public health challenge. The silent information regulator 1(SIRT1)/adenosine monophosphate-activated protein kinase(AMPK) signaling pathway serves as a central hub for cellular energy sensing and stress response. By synergistically promoting downstream effectors such as peroxisome proliferator-activated receptor γ coactivator-1α(PGC-1α), this signaling axis orchestrates mitochondrial biogenesis, enhanc

The diversity of STING in regulating immune cell function and its role in liver diseases: from bench to bedside.

BACKGROUND: The cyclic guanosine monophosphate-adenylate synthase (cGAS)-stimulator of interferon genes (STING) pathway is a critical innate immune signaling pathway that recognizes and transmits cytoplasmic DNA signals, triggering interferon and inflammatory responses. Immune cells enriched in the liver participate in the development of various liver diseases through the STING pathway; however, the precise regulatory mechanisms of this pathway within the immune cells remain poorly integrated. Elucidating these mechanisms holds promise for developing novel therapeutic strategies to address rel
